Bicalutamide is an oral nonsteroidal, anti-androgen drug used for prostate cancer. It binds to the androgen receptor. During the bulk synthesis of bicalutamide, various impurities are formed. The present work details the development of simple processes for the preparation of impurities of bicalutamide, viz bical-sulfoxides (6), bical-deshydroxy (10), bical-desfluoro (10a), bical-2-fluoro (10b), and bical-3-fluoro (10c).  相似文献

Secondary and tertiary alkylamines are privileged substance classes that are often found in pharmaceuticals and other biologically active small molecules. Herein, we report their direct synthesis from alkenes through an aminative difunctionalization reaction enabled by iron catalysis. A family of ten novel hydroxylamine-derived aminating reagents were designed for the installation of several medicinally relevant amine groups, such as methylamine, morpholine and piperazine, through the aminochlorination of alkenes. The method has excellent functional group tolerance and a broad scope of alkenes was converted to the corresponding products, including several drug-like molecules. Besides aminochlorination, the installation of other functionalities through aminoazidation, aminohydroxylation and even intramolecular carboamination reactions, was demonstrated, further highlighting the broad potential of these new reagents for the discovery of novel amination reactions.  相似文献

The androgen receptor (AR) is a pivotal target for the treatment of prostate cancer (PC) even when the disease progresses toward androgen-independent or castration-resistant forms. In this study, a series of 15 bicalutamide analogues (sulfide, deshydroxy, sulfone, and O-acetylated) were prepared and their antiproliferative activity evaluated against four different human prostate cancer cell lines (22Rv1, DU-145, LNCaP, and VCap). Bicalutamide and enzalutamide were used as positive controls. Seven of these compounds displayed remarkable enhancement in anticancer activity across the four PC cell lines. The deshydroxy analogue (16) was the most active compound with IC50 = 6.59–10.86 µM. Molecular modeling offers a plausible explanation of the higher activity of the sulfide analogues compared to their sulfone counterparts.  相似文献

A trichloromethylative olefin lactonization reaction using an iridium photoredox catalyst was developed. The reactions proceeded at room temperature for olefins with various substituents and substitution patterns, and a variety of lactones with a tetrasubstituted carbon and trichloromethyl group were obtained regio- and stereoselectively. The reaction mechanism was elucidated through isotope labeling experiments. The chemical properties of the lactones containing the trichloromethyl groups were investigated, and synthetic transformations of the product were realized.  相似文献

A three‐component carboetherification of unactivated alkenes has been developed allowing the rapid building of complexity from simple starting materials. A wide range of α‐substituted styrenes underwent smooth reactions with unactivated alkyl nitriles and alcohols to afford γ‐alkoxy alkyl nitriles with concomitant generation of a quaternary carbon center. A radical clock experiment provided clear‐cut evidence that the reaction proceeds through a tertiary alkyl radical intermediate.  相似文献

A protocol has been established for oxygen‐mediated amidoselenation and amidotelluration of alkenes under mild conditions. This method provides a simple route to a series of structurally diverse β‐amido selenides and β‐amido tellurides in moderate to high yields. The wide substrate scope, good functional group tolerance, ease of large‐scale preparation and potential for product derivatization make this reaction attractive for the synthesis of nitrogen‐, selenium‐ and tellurium‐containing molecules.  相似文献

The trifluoromethyl group is found in many synthetic bioactive compounds, and the difunctionalization of a C?C bond, as a powerful strategy for the construction of compounds with various functional groups, has been intensively investigated. Therefore, the difunctionalizing trifluoromethylation of alkenes has attracted growing interest because of the potential of the products as building blocks for bioactive molecules. Absorption and fluorescence spectra of bicalutamide (BCA) in different solvents and aqueous β-cyclodextrin (β-CD) solutions are reported. The solid inclusion complex of BCA with β-CD is prepared and investigated by FT-IR, 1H NMR, differential scanning colorimetry (DSC) and scanning electron microscopy (SEM). The Stokes shift of BCA is correlated with various solvent polarity scales like E T(30), BK and f(D,n). β-CD studies show that (i) the absorption and emission maxima of BCA are shifted to red, (ii) the absorbance is slightly decreased whereas emission intensity is increased largely with an increasing β-CD concentration, (iii) BCA forms 1:1 inclusion complex with β-CD and (iv) intramolecular charge transfer (ICT) emission is present in the excited state.  相似文献

Cytostatics are toxic pharmaceuticals, whose presence in surfaces puts healthcare workers at risk. These drugs might also end up in hospital effluents (HWW), potentially damaging aquatic ecosystems. Bicalutamide is a cytostatic extensively consumed worldwide, but few analytical methods exist for its quantification and most of them require advanced techniques, such as liquid chromatography mass spectrometry (LC-MS), which are very complex and expensive for large monitoring studies. Therefore, a simple but reliable multi-matrix high performance liquid chromatographic method, with fluorescence detection, was developed and validated to rapidly screen abnormal concentrations of bicalutamide in HWW and relevant contamination levels of bicalutamide in indoor surfaces (>100 pg/cm2), prior to confirmation by LC-MS. The method presents good linearity and relatively low method detection limits (HWW: 0.14 ng/mL; surfaces: 0.28 pg/cm2). Global uncertainty was below 20% for concentrations higher than 25 ng/mL (HWW) and 50 pg/cm2 (surfaces); global uncertainty was little affected by the matrix. Therefore, a multi-matrix assessment could be achieved with this method, thus contributing to a holistic quantification of bicalutamide along the cytostatic circuit. Bicalutamide was not detected in any of the grab samples from a Portuguese hospital, but an enlarged sampling is required to conclude about its occurrence and exposure risks.  相似文献

The first regiodivergent oxyboration of unactivated terminal alkenes is reported, using copper alkoxide as a catalyst, bis(pinacolato)diboron [(Bpin)2] as a boron source, and (2,2,6,6‐tetramethylpiperidin‐1‐yl)oxyl (TEMPO) as an oxygen source. The reaction is compatible with various functional groups. Two regioisomers are selectively produced by selecting the appropriate ligands on copper. The products may be used as a linchpin precursor for various other functionalizations, and net processes such as carbooxygenation, aminooxygenation, and dioxygenation of alkenes can be achieved after C?B bond transformations. Mechanistic studies indicate that the reaction involves the following steps: 1) Transmetalation between CuOtBu and (Bpin)2 to generate a borylcopper species; 2) regiodivergent borylcupration of alkenes; 3) oxidation of the thus‐generated C?Cu bond to give an alkyl radical; 4) trapping of the resulting alkyl radical by TEMPO.  相似文献

Summary: Within this work we aimed to study different RAFT agents for the potential use in coupled block copolymerization. Starting from S-(thiobenzoyl)thioglycolic acid as a commercial available CTA it should easily be possible to modify the substituent due to the thiocarboxylic group. Different chain transfer agents CTA's were synthesized and RAFT polymerizations of methyl methacrylate, butyl acrylate, N,N-dimethyl acrylamide, N,N-diethyl acrylamide, and N-isopropyl acrylamide were carried out and compared with each other.  相似文献

Two radical‐mediated processes of activated alkenes, namely arylphosphonylation and arylazidation, are described. The difunctionalization of alkenes by a tandem process that involves radical addition, 1,4‐aryl migration, and desulfonylation generates α‐aryl‐β‐heterofunctionalized amides bearing a quaternary stereocenter when the substituent on the nitrogen atom is an aryl group. Alternatively, heterooxindoles or spirobicycles can be obtained with excellent regioselectivity in the presence of an alkyl substituent on the nitrogen atom.  相似文献

Herein, we report an unprecedented regioselective and entirely atom‐economic cobalt(III)‐catalyzed method for the non‐annulative, intermolecular carboamination of alkenes. The methodology enables the direct synthesis of unnatural amino acid derivatives and proceeds under redox‐neutral conditions with a completely regioselective C?C bond and C?N bond formation. Furthermore, this reaction exemplifies the inherently different mechanistic behavior of the Cp*CoIII catalyst and its Cp*RhIII counterpart, especially towards β‐H‐elimination.  相似文献

An operationally simple and efficient microwave‐assisted protocol for the oxidative cyclization of acrylamide derivatives with non‐activated ketones to generate 3,3‐disubstituted oxindoles is described. Alkynyl-B(aam) (aam=anthranilamidato) derivatives are readily available bench-stable compounds that undergo remarkably selective reactions with Bu3SnH in the presence of [Cp*RuCl]4 as the catalyst. The addition follows a stereochemically unorthodox trans-selective course; in terms of regioselectivity, the Bu3Sn- unit is delivered with high fidelity to the C-atom of the triple bond adjacent to the boracyclic head group (“alpha,trans-addition”). This outcome is deemed to reflect a hydrogen bonding interaction between the protic −NH groups of the benzo-1,3,2-diazaborininone ring system and the polarized [Ru−Cl] bond in the loaded catalyst, which locks the substrate in place in a favorable orientation relative to the incoming reagent. The resulting isomerically (almost) pure gem-dimetalated building blocks are amenable to numerous downstream functionalizations; most remarkable is the ability to subject the −B(aam) moiety to Suzuki-Miyaura cross coupling without need for prior hydrolysis while keeping the adjacent Bu3Sn- group intact. Alternatively, the tin residue can be engaged in selective tin/halogen exchange without touching the boron substituent; the fact that the two -NH entities of −B(aam) do not protonate organozinc reagents and hence do not interfere with Negishi reactions of the alkenyl halides thus formed is another virtue of this so far underutilized boracycle. Overall, the ruthenium catalyzed trans-hydrostannation of alkynyl-B(aam) derivatives opens a practical gateway to isomerically pure trisubstituted alkenes of many different substitution patterns by sequential functionalization of the 1-alkenyl-1,1-heterobimetallic adducts primarily formed.  相似文献

自由基氟烷基化是向有机分子中引入氟烷基的一类非常重要的方法,也是目前有机化学研究的热点之一.近几年来,由于广泛的官能团兼容性和温和的反应条件等优点,可见光促进的氧化还原催化反应得到了长足的发展,已经成为化学键的构建和活化的有力工具.因此,光氧化还原催化的自由基氟烷基化反应,作为向有机化合物中引入氟烷基的有效途径,受到了广泛关注.本文报道了我们发展的氟烷基砜作为一类方便易得的新型氟烷基自由基前体,在可见光氧化还原催化下实现对烯烃的自由基氟烷基化反应.该反应可以高效地向芳基烯烃中引入三氟甲基、二氟甲基、1,1-二氟乙基、苯基二氟甲基等各种含氟烷基基团,并实现对芳基烯烃的双官能团化转化.  相似文献

1,3‐Dehydro‐o‐carborane is a useful synthon for selective cage boron functionalization of o‐carboranes. It reacts readily with alkenes or alkynes to give a variety of cage B(3)‐alkenyl/allenyl o‐carboranes by ene reactions in very high yields and excellent regioselectivity. In this study, a row of four analogous dopamine acryl- and methacrylamide derivatives, namely N-(3,4-dihydroxyphenyethyl) acrylamide, N-(3,4-dihydroxyphenyethyl) meth acrylamide, N-phenethyl methacrylamide, N-(4-hydroxyphenethyl) methacrylamide were synthesized and characterized by 1H-NMR and 13C-NMR, followed by further solvent-based radical polymerization with N-hydroxyethyl acrylamide. All copolymers were characterized by 1H-NMR, dynamic differential calorimetry, and gel permeation chromatography. The dependency of the used comonomer ratios to the molecular mass of the corresponding copolymers has been described. The synthesis of the various polymers serves as a feasibility study and provides important data for a future biometric application in the medical field. We synthesized N-(3,4-dihydroxyphenyethyl) acrylamide copolymer up to 80 mol% by free radical polymerization without using any protecting groups. All polymers show identical perfect adhesive properties by a simple scratch test. Further, the monomers were used as a photo reactive glue formulation to test its adherence to a medical titanium surface sample by tensile shear test.  相似文献
