The first enantioselective total syntheses of (−)-secologanin ( 1 ), (−)-5-carboxystrictosidine ( 2 ), and (−)-rubenine ( 3 ) were accomplished in 10, 9, and 14 steps, respectively. The key transformation in the synthesis of 1 was a sequential anti-selective organocatalytic Michael reaction/Fukuyama reduction/spontaneous cyclization to form an optically active dihydropyran ring. In addition, the secologanin tetraacetate ( 16 ), which is a potential key intermediate for the bioinspired divergent syntheses of monoterpenoid indole alkaloids, was prepared in gram-scale in seven steps. The total syntheses of 2 and 3 , which are classified as glycosylated monoterpenoid indole alkaloids, were achieved through bioinspired transformations such as a diastereoselective Pictet–Spengler reaction, a site- and stereoselective epoxidation, and a site-selective epoxide ring-opening followed by a lactonization reaction.  相似文献

The tetrahydro and hexahydro derivatives with respect to the nitrogen-containing ring were obtained by reduction of 3-azafluorene and 3-azafluorenone methiodides, respectively, with sodium borohydride. Indenoindolizines with linear and angular structures were synthesized from the N-phenacyl quaternary salts of 3-azafluorenones via 1,3-dipolar cycloaddition and by the Chichibabin method. 7-Nitro-3-azafluorenone and 3-azafluorenone N-oxide were obtained.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 2, pp. 252–255, February, 1983.  相似文献

The substituted phenanthrene-9-carboxyaldehydes are very important intermediates for the syntheses of phenanthroindolizidine and phenanthroquinolizidine alkaloids. The novel title compound was prepared from the reaction of 5 steps starting from the condensation of 3-methoxyl-4-methyl-phenylacetic acid and 4-(benzyloxy)-2-iodobenzaldehyde, followed by esterification, cyclization, reduction, and oxidation. A new method for the preparation of phenanthrene ring via palladium-catalyzed intramolecular Heck reaction was described. The title compound was characterized by IR, ^1H NMR, ^13C NMR, elemental analysis, and MS.  相似文献

For the purpose of organic syntheses of some corynanthe-type indole alkaloids, sesquiterpenes, and steroids, optically active intermediates cyclopentanone 3-allylalcohol, α-methylene-γ-butylo lactones andtrans-hydrindanone-propionic acid, were synthesized from (R)- and (S)-isopropylideneglyceraldehyde derived fromD-mannitol andLascorbic acid, respectively, utilizing Claisen rearrangement as a key reaction. Concise asymmetric total syntheses of the fungal metabolites (-)-stephacidin A, (+)-stephacidin B, and (+)-notoamide B are described. Key features of these total syntheses include (1) a facile synthesis of (R)-allyl proline methyl ester, (2) a revised route toward the pyranoindole ring system, (3) a novel cross-metathesis strategy for the introduction of important functional groups, and (4) an SN2' cyclization to form the [2.2.2] bridged bicyclic ring system. Furthermore, our synthesis has taken advantage of microwave heating to shorten reaction times as well as increase yields for the preparation of vital intermediates.  相似文献

A new method is reported for the preparation of indolo[2,3-a]quinolizidines based on radical cyclization of a 2-acyl-1-phenylthiotetrahydro-β-carboline bearing a pendent α,β-unsaturated ester. The required radical cyclization precursor is efficiently assembled from E-5-ethoxycarbonyl-4-pentenoic acid and 3,4-dihydro-β-carboline through a DCC/HOBt-activation/N-acylation and BF3·Et2O/PhSH iminium-ion trapping sequence. Tin-mediated radical cyclization of the radical cyclization precursor affords stereoselectively a cis-lactam (dr = 7:1) in good yield (81%), bearing the correct D/E ring fusion stereochemistry for the Tacaman alkaloids. The methodology has been applied to formal syntheses of the indoloquinolizidine alkaloids, (±)-eburnaminol and (±)-larutensine.  相似文献

An improved method for obtaining optically pure (S)-(l-p-menthen-8-yl)amine ( 12 ) has led to expedient syntheses of two hypothetical biogenetic intermediates on the way to aistoteline ( 7 ), namely (S)-(N)-(l-p-menthen-8-yl)-2-(3-indolyl)ethylamine ( 3 ) and (S)-(N)-(l-p-menthen-8-yl)-2-(3-indolyl)ethylideneamine ( 4 ). The latter has been transformed into (?)-hobartine ( 6 ) in 64% yield via a stereoselective biomimetic cyclization by treatment with HCOOH. This unambiguous synthesis establishes the hitherto unknown absolute configuration of (?)-hobartin ( 6 ). Several model cyclization reactions of N-substituted α-(terpen-8-yl)imine derivatives yielding unsaturated 3azabicyclo [3.3.1]nonane compounds are described.  相似文献

A mass-spectrometric study of 2- and 4-azafluorenones and their mono- and polymethyl derivatives showed that the presence of a methyl group in the benzene ring leads to a sharp increase in the relative intensity of the [M — H]+ ion peak. In contrast to the fragmentation of 2- and 4-azafluorenes, the mass spectra of monomethyl-substituted compounds do not contain an [M — CH3]+ fragment; this is probably associated with expansion of the pyridine or benzene ring to a seven-membered ring in the step involving the formation of the molecular ion due to inclusion of the methyl group. The intensity of the [M — CO]+ ion peak in the mass spectra of the 4-azafluorenones is higher by a factor of two with respect to the 2-azafluorenone isomers, and the [M — HCN]+ and [M — H, -HCN]+ ion peaks observed in the mass spectra of 2-azafluorenones are absent in them.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 1, pp. 89–95, January, 1979.  相似文献

An efficient short total synthesis of benzo[c]phenanthridine alkaloids including oxyavicine, oxynitidine, and oxysanguinarine is described. Thus, N‐methyl‐o‐bromobenzaldimines 1 b – d undergo regioselective cyclization with 4‐(benzo[d][1,3]dioxol‐5‐yl)but‐3‐yn‐1‐ol ( 2 b ) in the presence of [Ni(cod)2] (cod=1,5‐cyclooctadiene). In situ oxidation of the resultant isoquinolinium salts gives isoquinolinone derivatives 5 b – d with benzo[d][1,3]dioxol‐5‐yl substitution at the C3 atom and a (CH2)2OH group at the C4 atom. Later, oxidation of the alcohol group in 5 b – d to the aldehyde moiety followed by acid‐catalyzed cyclization and dehydration completes the total syntheses to give oxyavicine, oxynitidine, and oxysanguinarine in 67, 65, and 60 % yields, respectively. The synthesis requires four steps from o‐bromobenzaldehyde derivatives. Transformations of these alkaloids to the other alkaloids in this family are also discussed herein.  相似文献

A facile one‐pot approach based on a thermally induced metal‐ and solvent‐free 5‐endodig cyclization reaction of the amino propargylic alcohols in combination with Dess–Martin periodinane‐promoted oxidative dearomatization of 4,5,6,7‐tetrahydroindole intermediates provides an efficient and robust access to 5,6‐dihydro‐1H‐indol‐2(4H)ones. Green, relatively mild and operationally simple characteristics of the synthetic sequence are the major advantages, which greatly amplify the developed methodology. The utility of obtained indolones as unified key precursors is demonstrated by the application of these products to the formal total syntheses of a whole pleiad of Erythrina‐ and Lycorine‐type alkaloids, namely (±)‐erysotramidine, (±)‐erysotrine, (±)‐erythravine, (±)‐γ‐lycorane, and abnormal erythrinanes (±)‐coccoline and (±)‐coccuvinine.  相似文献

The ring deprotonation of 2-(2- and 4-pyridyl)benzoic acids using lithium dialkylamides in THF at rt, and the in situ cyclization afforded 4- and 2-azafluorenones, respectively. 1-Azafluorenone was obtained from ethyl 2-(3-pyridyl)benzoate using a similar protocol.  相似文献

We report herein a nonbiomimetic strategy for the total synthesis of the plicamine-type alkaloids zephycarinatines C and D. The key feature of the synthesis is a stereoselective reductive radical ipso-cyclization using visible-light-mediated photoredox catalysis. This cyclization enabled the construction of a 6,6-spirocyclic core structure through the addition of a carbon-centered radical onto the aromatic ring. Biological evaluation of zephycarinatines and their derivatives revealed that the synthetic derivative with a keto group displays moderate inhibitory activity against LPS-induced NO production. This approach could offer future opportunities to expand the chemical diversity of plicamine-type alkaloids as well as providing useful intermediates for their syntheses.  相似文献

[structures: see text] The highly convergent stereocontrolled total synthesis of (-)-vincamajinine (7), (-)-11-methoxy-17-epivincamajine (9), and the oxygen-bridged (+)-dehydrovoachalotine (22) are described. Key steps in the synthesis of 7 and 9 involved the stereospecific enolate-driven palladium-catalyzed cross-coupling reaction, a Tollens reaction, an acid-assisted intramolecular cyclization to form the C(7)-C(17) quaternary center, and two stereospecific reductions. The efficiency of this strategy is illustrated by the completion of the synthesis of 7 and 9 in 16 [from d-(+)-tryptophan methyl ester 17] and 17 (from the Sch?llkopf chiral auxiliary 27) reaction vessels, respectively. This constitutes the first total synthesis of these indole alkaloids and provides the first regiospecific route to 11-methoxy-substituted ajmaline/vincamajine-related alkaloids. The synthesis of 22 required a novel DDQ-mediated cyclization to furnish the C(6)-O(17) bond, executed in stereospecific fashion. Completion of these syntheses illustrates a concise and versatile strategy for the synthesis of vincamajine-related alkaloids, which has also been employed to prepare the related compounds quebrachidine diol (53), vincamajine diol (56), and vincarinol (59).  相似文献

The annelation of α-pyridones to five membered ring systems is demonstrated by the synthesis of a series of 2-azafluorenones. These tricyclic heterocycles could be optionally substituted with methyl or phenyl groups by varying the orthoester reagent in the cyclization reaction. The conversion of the resultant cyanopyridones to pyridonecarboxamides and their N-methyl homologs is also described.  相似文献
