The aim of this study was to evaluate the effects of photobiomodulation on the repair of induced root resorption (RR) after orthodontic tooth movement. Twenty male rats were used in this study. Forty right and left upper first molars were evaluated and divided into four groups (n = 10): negative control group (NC), no tooth movement or irradiation; positive control group (PC), induced tooth movement and root resorption; conventional treatment group (CT), force was removed after 7 days; and photobiomodulation group (PBM) after force removal molars were irradiated every 48 h for 7 days using GaAlAs diode laser (810 nm). Energy per point was 1.5 J (100 mW, 15 s, 75 J cm?2). NC and PC were euthanized on day 7; CT and PBM on day 14. Histomorphometric and immunohistochemsitry analyses showed increase in area of root resorption in all groups (P < 0.05) compared to NC. RR lacunae were larger in CT compared to PC and PBM at the compression side of the distal root. OPG was higher in PBM group (P < 0.05). PBM group showed low expression of RANKL compared to PC and CT on the tension side. PBM can potentially affect RR progression by increasing OPG expression in the compression area and decreasing number of clastic cells in the root surface.  相似文献

The water extract obtained from the aerial parts of Sideritis euboea (Lamiaceae), which is known in Greece as 'mountain tea', was investigated by determining the in vitro effect of this extract on the IL-6, OPG and RANKL secretion by osteoblasts, three important molecules in osteoblast-osteoclast interplay. The results showed that this extract reduced significantly the secretion of IL-6 by KS-483 osteoblasts, while it also suppressed RANKL secretion, with both effects being dose-dependent and more potent at the higher concentrations tested (50, 100 microg/mL). We further determined the chemical profile of the extract by applying an analytical U-HPLC-DAD-ESI-MS/MS method using the high resolution hybrid LTQ-Orbitrap Discovery spectrometer. An ESI source in negative mode was employed. The analysis indicated that the water extract of S. euboea is rich in flavonoid glycosides, and phenylpropanoid glycosides.  相似文献

We report the results of an investigation of the binding of a series of bisphosphonate drugs to human bone using 2H, 13C, 15N, and 31P nuclear magnetic resonance spectroscopy. The 31P NMR results show that the bisphosphonate groups bind irrotationally to bone, displacing orthophosphate from the bone mineral matrix. Binding of pamidronate is well described by a Langmuir-like isotherm, from which we deduce an approximately 30-38 A2 surface area per pamidronate molecule and a deltaG = -4.3 kcal mol(-1). TEDOR of [13C3, 15N] pamidronate on bone shows that the bisphosphonate binds in a gauche [N-C(1)] conformation. The results of 31P as well as 15N shift and cross-polarization measurements indicate that risedronate binds weakly, since it has a primarily neutral pyridine side chain, whereas zoledronate (with an imidazole ring) binds more strongly, since the ring is partially protonated. The results of 2H NMR measurements of side-chain 2H-labeled pamidronate, alendronate, zoledronate, and risedronate on bone show that all side chains undergo fast but restricted motions. In pamidronate, the motion is well simulated by a gauche+/gauche- hopping motion of the terminal -CH2-NH3(+) group, due to jumps from one anionic surface group to another. The results of double-cross polarization experiments indicate that the NH3(+)-terminus of pamidronate is close to the bone mineral surface, and a detailed model is proposed in which the gauche side-chain hops between two bone PO4(3-) sites.  相似文献

Following extracorporeal photopheresis (ECP), lymphocytes become apoptotic and upregulate class I MHC antigenic peptides. Conversely, ECP treated monocytes demonstrate activation markers and have an increased avidity for the phagocytosis of apoptotic T cells. Processing of apoptotic T cells by monocytes, following ECP, is thought to induce an immunomodulatory response, which targets untreated, but clonal T cells. Recently we detected apoptotic lymphocytes immediately post ECP. Although enhanced CD95 (Fas) expression has been observed 24 h post ECP, CD95 and Fas-ligand (Fas-L) expression have not been determined at this very early apoptotic stage. Exposure of monocytes to UV has previously suppressed expression of the co-stimulatory molecules required for the presentation of processed antigens to T cells. Our data demonstrate no increase in CD95 or Fas-L expression on T cells tested immediately following ECP. However, the number of T cells expressing Fas-L significantly increased 24 h post ECP (P<0.005). The expression of the co-stimulatory molecules, CD54, CD80 and CD86, remained unaltered on monocytes treated by ECP. Although the mechanism responsible for early induction of lymphocyte apoptosis remains unclear, the later apoptosis involves Fas-L expression. The maintenance of co-stimulatory molecules, on treated monocytes, indicates that they retain the ability to induce an immunomodulatory response.  相似文献

In many clinical situations which cause thymic involution and thereby result in immune deficiency, T cells are the most often affected, leading to a prolonged deficiency of T cells. Since only the thymic-dependent T cell production pathway secures stable regeneration of fully mature T cells, seeking strategies to enhance thymic regeneration should be a key step in developing therapeutic methods for the treatment of these significant clinical problems. This study clearly shows that receptor activator of NF-kappaB ligand (RANKL) stimulates mouse thymic epithelial cell activities including cell proliferation, thymocyte adhesion to thymic epithelial cells, and the expression of cell death regulatory genes favoring cell survival, cell adhesion molecules such as ICAM-1 and VCAM-1, and thymopoietic factors including IL-7. Importantly, RANKL exhibited a significant capability to facilitate thymic regeneration in mice. In addition, this study demonstrates that RANKL acts directly on the thymus to activate thymus regeneration regardless of its potential influences on thymic regeneration through an indirect or systemic effect. In light of this, the present study provides a greater insight into the development of novel therapeutic strategies for effective thymus repopulation using RANKL in the design of therapies for many clinical conditions in which immune reconstitution is required.  相似文献

A lipid present in meconium and capable of inhibiting the agglutination of human M and N erythrocytes by rabbit anti-M and anti-N antibodies has been isolated and identified as cholesteryl sulfate. The reaction of human anti-A and anti-B antisera with the respective human red blood cells is not impaired by cholesteryl sulfate. Cholesteryl hemisuccinate has been found to be an hemagglutination inhibitor qualitatively similar to but less active than cholesteryl sulfate.

Zusammenfassung Eine lipophile Substanz, welche die Agglutination humaner Erythrocyten durch Kaninchen anti-M und anti-N Antiseren inhibiert, konnte aus Mekonium isoliert und als Cholesterylsulfat identifiziert werden. Die Reaktion humaner anti-A und anti-B Antiseren mit den jeweiligen A und B Erythrozyten wurde durch Cholesterylsulfat nicht beeinflußt. In bezug auf die Agglutinationshemmung verhält sich der Monocholesterylester der Bernsteinsäure ähnlich wie der der Schwefelsäure, ist jedoch erst in höherer Konzentration wirksam.

Electrophoresis is widely used to determine the electrostatic potential of colloidal particles. Oil droplets in pure water show negative or positive electrophoretic mobilities depending on the pH. This is commonly attributed to the adsorption of hydroxyl or hydronium ions, resulting in a negative or positive surface charge, respectively. This explanation, however, is not in agreement with the difference in isoelectric point and point of zero charge observed in experiment. Here we present molecular dynamics simulations of oil droplets in water in the presence of an external electric field but in the absence of any ions. The simulations reproduce the negative sign and the order of magnitude of the oil droplet mobilities at the point of zero charge in experiment. The electrostatic potential in the oil with respect to the water phase, induced by anisotropic dipole orientation in the interface, is positive. Our results suggest that electrophoretic mobility does not always reflect the net charge or electrostatic potential of a suspended liquid droplet and, thus, the interpretation of electrophoresis in terms of purely continuum effects may need to be reevaluated.  相似文献

A series of novel zoledronic acid (ZL) derivatives 1-hydroxy-3-(2-methyl-1H-imidazol-1-yl)propane-1,1-diyldiphosphonic acid (MIPrDP), 1-hydroxy-4-(2-methyl-1H-imidazol-1-yl)butane-1,1-diyldiphosphonic acid (MIBDP), and 1-hydroxy-5-(2-methyl-1H-imidazol-1-yl)pentane-1,1-diyldiphosphonic acid (MIPeDP) were prepared and successfully labeled with (99m)Tc in high labeling yields. The in vitro stability and in vivo biodistribution of (99m)Tc-MIPrDP, (99m)Tc-MIBDP and (99m)Tc-MIPeDP were investigated and compared. The biodistribution studies indicate that the radiotracer (99m)Tc-MIPrDP has highly selective uptake in the skeletal system and rapid clearance from soft tissues. The present findings indicate that (99m)Tc-MIPrDP holds great potential for use in bone imaging.  相似文献

The inhibitive action of the aqueous extract of Ruellia tuberosa L (ART) on the corrosion of copper in 0.5 ​M HCl was investigated. The inhibition efficiency increased with the extract concentration, acid concentration, as well as increasing the temperature. The Polarization studies revealed that the ART act as a mixed-type inhibitor. Based on the analysis of electrochemical impedance spectroscopy, an equivalent circuit is suggested. The adsorption of the inhibitor ART on the copper surface obeyed the Langmuir adsorption isotherm. From the results of scanning electron microscopy, Fourier transform infrared spectroscopy, and energy-dispersive X-ray spectroscopy the adsorption of ART on the copper surface is confirmed.  相似文献

Chitooligosaccharides (CHOS) added in diet have been found as potent calcium fortifiers in conditions of Ca2+ deficiency such as osteoporosis. In this study, we found that pharmaceutical intervention using CHOS prevented ovariectomy (OVX)-induced bone mineral density loss and the deterioration of trabecular microarchitecture in a dose-dependent manner (p < 0.05 or 0.01). CHOS (125, 250 mg/kg) suppressed the serum levels of bone resorption biomarkers CTx and TRACP5b induced by OVX (p < 0.05), but increased the levels of osteogenic markers ALP and OC by 11.3–11.6% and 10.7–15.2% of OVX group (p < 0.05), suggesting the exact pharmacological action of CHOS in the control of osteoporosis which may be the result of both promoting bone formation and suppressing bone resorption. Bone turnover-modulating effects of CHOS appear related to their anti-inflammatory capacity to down-regulate mRNA and protein expression of COX-2 (17.2–32.2% and 16.4–21.9% of OVX group, p < 0.05 or 0.01), a key mediator linking between inflammation and osteoporosis.  相似文献

Background and aimsBone turnover is strongly affected by pH of surrounding fluid, and in turn plays a role in maintaining systemic pH, however the quantitative contribution of bone processes to pH regulation is not known. Our goal was to develop a mathematical model describing pH regulation in the interstitial fluid and to examine the contribution of hydroxyapatite dissolution and precipitation to pH regulation.Materials and methodsWe modeled twelve reversible equilibrium reactions of sixteen calcium, phosphate, hydrogen and carbonate species in the interstitial fluid and examined the buffering capacity and range. The effect of hydroxyapatite dissolution and precipitation was modeled by assuming that the calcium, phosphate and hydroxide contained in the bone volume adjacent to the interstitial fluid is instantaneously added to or removed from the interstitial fluid.ResultsThe carbonate buffer was found to dominate electrochemical buffering system of the bone interstitial fluid. Nevertheless, the phosphate added during dissolution of bone hydroxyapatite significantly improved the interstitial fluid buffering capacity. In contrast, hydroxyapatite precipitation had limited effect on the interstitial fluid pH regulation.ConclusionThis study provides mechanistic insights into the physicochemical processes underlying the known role of bone turnover processes in regulation of body pH homeostasis.  相似文献

Exact equations and several computer programs were developed for use in studies on copolymerizations carried to high conversion when the concentration of one of the monomers (A) remains constant. Simple ACSL® and DESIRE® programs are described for simulating such copolymerizations, and their output was used to test programs and procedures that were developed to evaluate monomer reactivity ratios for such copolymerization systems. Based on an integrated form of the copolymer equation, Excel® and Fortran programs were developed for evaluating monomer reactivity ratios from information about initial monomer compositions, copolymer compositions, and the fractions of the second monomer (B) that reacted. A graphical procedure for evaluating monomer reactivity ratios from such data was also developed. A previous program developed for calculating information about monomer sequence distributions in copolymers was modified so that it would apply to copolymerization at high conversion when the concentration of one monomer remains constant. © 2000 John Wiley & Sons, Inc. Decreasing enantioselectivity (E-value) by increasing conversion has been observed in transesterification reactions of secondary alcohols catalyzed by a pure protein formulation of lipase B from Candida antarctica (Novozym 525 F). Addition of a range of enantiopure alcohols caused a temporary increase in selectivity of the transesterification reaction of 3-chloro-1-phenoxy-2-propanol with vinyl butanoate. The corresponding immobilized lipase B, (Novozym 435) showed a similar relationship between the E-value and degree of conversion.  相似文献

Bisphosphonates have been confined in siliceous ordered mesoporous materials, and the drug release rate of these systems has been investigated. The bisphosphonate adsorption rate has been increased from 1% in oral administration to around 40% locally delivered. Drug dosage can be modulated through amine modification of the material surface, leading to a bisphosphonate adsorption in the ordered mesoporous matrices 3 times larger than that for unmodified materials. The use of these drug delivery systems opens new fields with new possibilities for tissue engineering.  相似文献

Treatment of substituted benzyl alcohols with tosyl chloride resulted in the formation of the corresponding chlorides, not the usual tosylates. A series of experiments demonstrated that it was possible to predict whether chlorination or tosylation would occur for substituted benzyl alcohols and pyridine methanols. Treatment of electron withdrawing group-substituted benzyl alcohols with tosyl chloride gave the corresponding chlorides in moderate yields under mild conditions, which provided a simple way to directly prepare chlorides from alcohols.  相似文献
