Polyamidoamine (PAMAM) dendrimers of different generations (G2 and G4) conjugated with β-cyclodextrin (β-CD), PAMAM (G2, G4)-CD, were synthesized using substitution reaction from mono-6-iodine-β-cyclodextrin and PAMAM dendrimers. The resulting molecular structures were characterized by NMR, IR. The molecular interaction between various dendrimers and levofloxacin lactate (LFL) were investigated by monitoring the fluorescence of LFL in the presence of dendrimers in buffer solution (pH 7.4) at 25?°C. It was found that the PAMAM (G2, G4)-CD possesses higher sensitizing ability than that of the corresponding parent dendrimers and natural β-CD, and increases concomitantly with the increases of generation and content of β-CD, suggesting that the PAMAM (G2, G4)-CD possesses stronger inclusion ability with LFL. The possible interaction mechanism between PAMAM-CD and LFL was proposed by 1H NMR analysis and theoretical calculation. The results show that the LFL molecule is located at the amine end of dendrimer molecule and along the side of cyclodextrin cavities to form supramolecular complexes. Furthermore, results indicate that the main driving force of the complex could be attributed to the electrostatic interactions and hydrogen bonding between LFL and PAMAM-CD, as well as the synergistic effect of intermolecular forces.  相似文献

Novel polyamidoamine (PAMAM) dendrimers (G=0.5–2.5) with a naphthalene core unit have been prepared. They were found to display acid as well as metal ion sensitive fluorescence signal amplification, making them of potential use as chemosensing materials. PAMAM dendritic wedges as well as naphthalene-centered PAMAM dendrimers were characterized by FT-IR, 1H and 13C NMR spectroscopic methods and elemental analysis.  相似文献

Abstract

A simple, accurate and rapid colorimetric method using ninhydrin reagent was developed for the determination of polyamidoamine (PAMAM) dendrimers (G4, G5, and G6) and their derivates in aqueous medium. This method was based on the interaction of the primary amino group of PAMAM dendrimers with ninhydrin reagent to form a blue‐colored product with λmax at 570 nm. Beer's law was obeyed in the concentration range of 25–200 µg/ml of all the three investigated PAMAM dendrimers. The effects of experimental parameters such as reagent concentration and reaction time were studied to optimize the colorimetric method. Accuracy and precision of the colorimetric method were assessed by statistical analysis. Acetylated G5 PAMAM dendrimers with various acetylated rates were simultaneously measured by the described ninhydrin assay and NMR studies and the data obtained by the two methods approximately accorded with each other. Results showed that the suggested procedures were suitable for the determination of PAMAM dendrimers and their derivates in aqueous solutions with satisfactory accuracy and precision.  相似文献

G5.0‐OH PAMAM dendrimers were used to prepare fluorescent silver clusters with weaker ultraviolet irradiation reduction method, in which the molar ratio of Ag+ to PAMAM dendrimers was the key factor to determine the geometry and properties of silver nanoparticles. The results showed that because of G5.0‐OH PAMAM dendrimers as strong encapsulatores, when the molar ratios of Ag+ to PAMAM dendrimers was smaller than 5, the obtained Agn clusters (n<5) had line structures and "molecular‐like" properties, which were highly fluorescent and quite stable in aqueous solution. Whereas when the molar ratios were between 5 and 8, the obtained Agn clusters were 2D structures and their fluorescence was weaker. When the molar ratio was larger than 8, the structure of silver nanoparticles was 3D and no fluorescence was observed from the obtained silver nanoparticles.  相似文献

A novel type of rhenium-containing dendrimers has been prepared and characterized. A series of Schiff base-terminated PAMAM dendrimers were prepared by condensing the terminal amine groups of PAMAM dendrimers with pyridine-2-carboxaldehyde. Complete condensation of the terminal amines was confirmed by 1H NMR spectroscopy. Bromotricarbonylrhenium(I) moieties were introduced onto the surface of these modified PAMAM derivatives by refluxing with bromopentacarbonylrhenium(I). These complexes had been characterized by a variety of analytical and spectroscopic techniques and their IR, NMR, and mass spectra discussed. The crystal structure of the model compound [(CH3CONHCH2CH2NCHPy)ReBr(CO)3] confirms a facial configuration of the three carbonyl ligands. UV-Vis absorption spectroscopy suggests that the {Re(CO)3} moieties are quite far apart even in the second generation PAMAM dendrimers and do not interact with one another. In fact, the intensity of the metal-to-ligand (dπ-pπ) charge-transfer is a linear function of the number of {Re(CO)3} chromophores.  相似文献

Poly(amidoamine)(PAMAM) dendrimers with a cinnamoyl shell were prepared by reacting full generation PAMAM dendrimers (G=3.0) with 2‐chloroethanol and cinnamoyl chloride, which resulted in densely packed polymerizable unsaturated groups on the periphery. The cinnamoyl shell of the dendrimers dimerized when irradiated under a UV light by using 5‐nitroacenaphthylene as an initiator in dilute dimethylformamide (DMF). FTIR, 1H NMR, UV‐Vis, SEC, and a viscosity test certified that the photocycloaddition of the cinnamoyl shell of the dendrimers took place within the molecules with the disappearance of double bond signals in the FTIR. 1H NMR spectra as well as the intrinsic viscosity and polydispersity value of the products both before and after irradiation showed no difference. It was further found that the cinnamoyl shell‐modified dendrimers possessed fluorescence property, and the fluorescence intensity became stronger when the shell was photocyclized under UV‐ irradiation. © 2000 John Wiley & Sons, Inc. Dendrimers or biofunctionalized dendrimers can be assembled onto magnetic iron oxide nanoparticles to stabilize or functionalize inorganic nanoparticles. Carboxylated poly(amidoamine) PAMAM dendrimers (generation 4.5) have been used for the synthesis of iron oxide nanoparticles, resulting nanocomposites with potential biomedical applications. The present paper aims to systematically investigate the thermal behaviour of nanostructured hybrids based on ferric oxide and PAMAM dendrimers, by differential scanning calorimetry (DSC) technique. The novelty consists both in synthesis procedure of hybrid nanostructures as well as in DSC approach of these nanocomposites. For the first time, we propose a new method to prepare Fe2O3??dendrimer nanocomposite, using soft chemical process at high pressure. Commercial PAMAM dendrimers with carboxylic groups on its surface were used. When high pressure is applied, polymeric structures suffer morphological changes leading to hybrid nanostructures' formation. In the same time, crystallinity of inorganic nanoparticles is provided. DSC results showed an increase in thermal stability of composites as compared to commercial dendrimers. This could be due to the formation of strong interactions between ferric oxide and carboxyl groups, as confirmed by Fourier transform infrared spectroscopy. Electron microscopy analysis (SEM/EDX) and size measurements were performed to demonstrate the existence of nanosized particles.  相似文献

采用发散法合成了0.5~4.0代(G)聚酰胺-胺(PAMAM)树枝状高分子,通过薄层色谱分析产物组成,并进一步选用柱色谱法对半代产物进行了提纯与精制.用红外,核磁,高效液相色谱(HPLC)和热重分析等方法对提纯后产物进行了表征.结果表明,提纯得到的各PAMAM产物具有较完整的分子结构,小分子含量很少;且0.5GPAMAM产物HPLC测试纯度达到97.81%.  相似文献

Terminal amine groups of poly(amidoamine) (PAMAM) dendrimers can be substituted with different functional groups for various applications. In this study, PAMAM derivatives with acetamide, hydroxyl, and carboxyl termini were synthesized from ethylenediamine (EDA) core generation 4 and 5 primary amine-terminated PAMAM dendrimers. The reaction products were purified with dialysis and subsequently characterized by polyacrylamide gel electrophoresis (PAGE), capillary electrophoresis (CE), size exclusion chromatography (SEC), matrix-assisted laser desorption ionization time-of-flight (MALDI-TOF) mass spectrometry, potentiometric titration, 1H NMR, and 13C NMR. PAGE and CE electropherograms provide data regarding the purity, charge distribution, and electrophoretic mobility of the dendrimers and their derivatives. SEC and MALDI-TOF mass spectrometry detect the average absolute molar mass and the individual mass fractions, respectively. The combination of SEC with potentiometric titration provides quantitative evidence of the degree of the functional group substitution, while NMR techniques (both 1H NMR and 13C NMR) confirmed the changes in dendrimer surface functionalization. This study provides a general example for the comprehensive characterization of surface-functionalized PAMAM dendrimer nanoparticles. The synthesized dendrimer derivatives hold promise for environmental and medical applications.  相似文献

为了研究非配位型溶剂对聚酰胺-胺(PAMAM)树形分子的模板法制备CdS量子点的影响, 分别以水、甲醇及二者的混合物( ∶ =2∶1)为溶剂, 以4.5代PAMAM树形分子为模板制备了CdS量子点. 结果表明, 相同条件下, 以甲醇为溶剂时制备的CdS量子点为单晶, 平均直径2.7 nm, 尺寸分布窄, 发光强度高; 以水为溶剂制备的CdS量子点为多晶, 平均直径为5.7 nm, 尺寸分布宽, 发光强度低; 在甲醇与水的混合溶剂中制备的CdS量子点为单晶和多晶共存, 平均直径为4.1 nm, 尺寸分布及发光强度都居中. 这主要是由于树形分子的模板作用不同造成的. 树形分子在甲醇中能充分伸展, 起到内模板作用; 树形分子与水之间由双氢键作用而产生交联, 不利于Cd2+与树形分子内部基团的配位, 主要起到外模板作用; 在甲醇与水的混合溶剂中, 树形分子则同时起到了内模板和外模板作用.  相似文献

New high-molecular-weight contrast agents based on polyamidoamine (PAMAM) dendrimers for targeted imaging of malignant tumors characterized by overexpression of human epidermal growth factor receptor (EGFR) and human alpha-fetoprotein receptor (RECAF) were designed. Conjugates of second (G2) and third (G3) generation polyamidoamine dendrimers with 1,4,7,10-tetraazocyclodecane-1,4,7,10-tetraacetic acid (DOTA) were obtained. The quantitative composition of the conjugates was determined by 1HNMR spectroscopy. It was shown that four out of the 16 terminal NH2 groups in G2-DOTA and nine out of the 32 groups in G3-DOTA were modified with DOTA. The morphology, size, and charge of the synthesized macromolecules were characterized by dynamic light scattering and electrophoresis. Gadolinium(III) was loaded into the conjugates and the Gd content was determined by atomic emission spectroscopy. For increasing the selectivity of accumulation in the tumor cells, two recombinant proteins able to bind selectively to EGFR and RECAF, namely, human recombinant epidermal growth factor (rEGF) and human recombinant 3rd domain of alpha-fetoprotein (3dAFPpG), were conjugated with G2 and G3 dendrimers. The conjugates containing vector molecules were mainly accumulated via clathrin-dependent endocytosis, whereas G2-DOTA and G3-DOTA were absorbed via caveolin-dependent endocytosis and macropinocytosis. The dendrimer conjugates with vector molecules were intensely accumulated in A549 cells characterized by high expression of EGFR (Herl) and RECAF, whereas the accumulation of conjugates in the control K562 cells (with low expression of Her1) and in the CD14? population of human unstimulated mononuclear white blood cells was insignificant. The 3dAFPpG-conjugated dendrimers were partly recycled. All synthesized conjugates had a rather low toxicity in the range of 350–450 µmol L?1 (IC50).

This paper describes an investigation of the uptake of Cu(II) by poly(amidoamine) (PAMAM) dendrimers with an ethylenediamine (EDA) core in aqueous solutions. We use bench scale measurements of proton and metal ion binding to assess the effects of (i) metal ion-dendrimer loading, (ii) dendrimer generation/terminal group chemistry, and (iii) solution pH on the extent of binding of Cu(II) in aqueous solutions of EDA core PAMAM dendrimers with primary amine, succinamic acid, glycidol, and acetamide terminal groups. We employ extended X-ray absorption fine structure (EXAFS) spectroscopy to probe the structures of Cu(II) complexes with Gx-NH2 EDA core PAMAM dendrimers in aqueous solutions at pH 7.0. The overall results of the proton and metal ion binding measurements suggest that the uptake of Cu(II) by EDA core PAMAM dendrimers involves both the dendrimer tertiary amine and terminal groups. However, the extents of protonation of these groups control the ability of the dendrimers to bind Cu(II). Analysis of the EXAFS spectra suggests that Cu(II) forms octahedral complexes involving the tertiary amine groups of Gx-NH2 EDA core PAMAM dendrimers at pH 7.0. The central Cu(II) metal ion of each of these complexes appears to be coordinated to 2-4 dendrimer tertiary amine groups located in the equatorial plane and 2 axial water molecules. Finally, we combine the results of our experiments with literature data to formulate and evaluate a phenomenological model of Cu(II) uptake by Gx-NH2 PAMAM dendrimers in aqueous solutions. At low metal ion-dendrimer loadings, the model provides a good fit of the measured extent of binding of Cu(II) in aqueous solutions of G4-NH2 and G5-NH2 PAMAM dendrimers at pH 7.0.  相似文献

Biomimetic acryloyloxyethyl phosphorylcholine (APC) was used to react with generation 5 poly(amido amine) (PAMAM) dendrimers (G5) via the Michael addition reaction between primary amino group of PAMAM dendrimers and acrylic functional group of APC. FTIR and (1)H NMR confirmed the success of surface modification of G5. The primary amino and phosphorylcholine (PC) group numbers of the surface engineered PAMAM dendrimers (G5-PC) were calculated to be 56 and 50 via (1)H NMR and potentiometric titration. Cell viability and cell morphology studies indicated that biomimetic phosphorylcholine surface engineering successfully lowered the cytotoxicity of G5 PAMAM dendrimers. The hydrophobic interior of G5-PC was used to incorporate anti-cancer drug Adriamycin (ADR) and the G5-PC showed sustained releasing behavior for ADR. Cell morphology and viability tests indicated that the drug-loaded G5-PC conjugate could effectively enter the cancer cells and inhibit the growth of cancer cells. Biomimetic phosphorylcholine surface engineered PAMAM dendrimers with lowered cytotoxicity and high cellular penetrating ability showed great potential for the biomedical applications as nanocarrier system.  相似文献

Novel Fréchet–PAMAM hybrid dendrimers linked by triazole units as unimolecular micelles with a hydrophobic core surrounded by a hydrophilic shell were prepared. The dendritic cores with 3 and 6 alkyne terminal groups were synthesized from 1,3,5-tribromomethyl-benzene (tBrMeB), in one case by direct coupling with 17α-ethynylestradiol (EE); in the second one the tBrMeB was reacted with bis(hydroxymethyl) phenol followed by chlorination of the hydroxyl groups and subsequent coupling to EE. With this strategy, the core can be grown by further substitutions of bis(hydroxymethyl) phenol over the halogenated terminals as Fréchet dendrimer. The hydrophilic shells used were PAMAM type dendrons of 0.5 and 1.5 generations with azide as focal point and tert-butyl ester as end groups. The unimolecular micelles were obtained by cycloaddition between an azide in the selected dendron and the alkyne terminal in the hydrophobic core to obtain a 1,4-disubstituted 1,2,3-triazole. Once the coupling was achieved, the tert-butyl ester groups were hydrolyzed in trifluoroacetic acid and the corresponding dendrimers with carboxylic acid as end groups were completely soluble in phosphate buffer solutions of pH 7.0, 7.4, and 8.0. All hybrid dendrimers were characterized by High Resolution Mass Spectrometry, 1H and 13C NMR, and FTIR.  相似文献

Poly(N,N′-methylenebisacrylamide-co-acrylic acid) microsphere-supported polyamidoamine (PAMAM) dendrimers up to third generation (G) were grown onto the surface as well as the gel-layer of P(MBA-co-MAA) microspheres by a divergent method. The P(MBA-co-MAA) supported PAMAM dendrimers were used as heterogeneous stabilizers for the gold nanoparticles by an in situ reduction of HAuCl4 via the efficient coordination interaction between the amino groups of the supported PAMAM dendrimers and the gold atoms. The effects of the generations of the P(MBA-co-MAA) supported PAMAM dendrimer on the loadings and the catalytic activity of the heterogeneous Au nanoparticles were systematically investigated with the reduction of 4-nitrophenol to 4-aminophenol as a model reaction.  相似文献

The electron capture dissociation (ECD) and collision-induced dissociation (CID) of complexes of polyamidoamine (PAMAM) dendrimers with metal ions Ag+, Cu2+, Zn2+, Fe2+, and Fe3+ were determined by Fourier transform ion cyclotron resonance mass spectrometry. Complexes were of the form [PD + M + mH]5+ where PD = generation two PAMAM dendrimer with amidoethanol surface groups, M = metal ion, m = 2−4. Complementary information regarding the site and coordination chemistry of the metal ions can be obtained from the two techniques. The results suggest that complexes of Fe3+ and Cu2+ are coordinated via both core tertiary amines, whereas coordination of Ag+ involves a single core tertiary amine. The Zn2+ and Fe2+ complexes do not appear to involve coordination by the dendrimer core.  相似文献

在低代端酯基PAMAM树形分子(G1.5-COOCH3)存在时,用氢气还原AgNO3制备出银纳米颗粒。用透射电子显微镜(TEM),电子衍射(ED),紫外-可见吸收光谱(UV-Vis)和红外光谱(FT-IR)对所制备的银纳米颗粒进行了表征。实验结果表明,当用氢气作为还原剂时,以低代树形分子为保护剂,通过优化还原条件,可成功制备尺寸稳定、均一的银纳米颗粒,其粒径为2.9±0.5 nm,且所制备的银纳米颗粒的粒径分布较窄。根据树形分子的理论尺寸与制备的银纳米颗粒的粒径关系,可推断出大多数的银纳米颗粒是由多个树形分子所包围而稳定存在。  相似文献
