Pestalofones I – K ( 1 – 3 ), three new dimeric meroterpenoids with 2‐(7‐benzoyl‐2,3‐dihydrobenzofuran‐2‐yl)‐1‐phenylethan‐1‐one (in 1 ) and 2‐(7‐benzoyl‐2,3‐dihydrobenzofuran‐2‐yl)‐1‐(3,8‐dioxatricyclo[5.1.0.02,4]oct‐4‐yl)ethan‐1‐one (in 2 and 3 ) skeletons, were isolated from the solid cultures of the plant endophytic fungus Pestalotiopsis fici. The structures of 1 – 3 were elucidated by NMR experiments. Compound 1 was found to be present as a racemic mixture. The absolute configurations of 2 and 3 were deduced by analogy to the previously isolated metabolites pestalofones G and H ( 4 and 5 ) from the same fungus. Biogenetically, compounds 1 – 3 are derived from the same precursors (co‐isolated compounds 9 and 10 ) as the previously isolated compounds 4 – 8 . Compounds 2 and 3 showed weak cytotoxic activities against four human tumor cell lines T24, HeLa, A549, and MCF‐7.  相似文献

Six new ( 2 , 4 – 8 ) and two known polyketides with a basic structure of an anthraquinone‐xanthone were isolated from mycelia and culture broth of the fungus Engyodontium album strain LF069. The structures and relative configurations of these compounds were established by spectroscopic means, and their absolute configurations were defined mainly by comparison of quantum chemical TDDFT calculated and experimental ECD spectra. Compounds 2 and 4 – 8 were given the trivial names engyodontochone A ( 2 ) and B–F ( 4 – 8 ). Compounds 5 – 8 represent the first example of a 23,28 seco‐beticolin carbon skeleton. The relative and absolute configurations of two known substances JBIR‐97/98 ( 1 ) and JBIR‐99 ( 3 ) were determined for the first time. All isolated compounds were subjected to bioactivity assays. Compounds 1 – 4 exhibited inhibitory activity against methicillin‐resistant Staphylococcus aureus (MRSA) that was 10‐fold stronger than chloramphenicol.  相似文献

Complexity‐increasing Domino reactions comprising C?H allenylation, a Diels–Alder reaction, and a retro‐Diels–Alder reaction were realized by a versatile catalyst derived from earth‐abundant, non‐toxic manganese. The C?H activation/Diels–Alder/retro‐Diels–Alder alkyne annulation sequence provided step‐economical access to valuable indolone alkaloid derivatives through a facile organometallic C?H activation manifold with transformable pyridines.  相似文献

The reactivity of alkynyl and enynyl Fischer carbene complexes towards 8‐azaheptafulvenes is examined. Alkynyl carbenes 1 a – f undergo regioselective [8+2] heterocyclization with 8‐aryl‐8‐azaheptafulvenes 2 a , b providing cycloheptapyrroles 3 and 4 with metal carbene or ester functionality at C3. Moreover, consecutive cyclization reactions are involved when enynyl carbenes are used. Thus, the cyclopenta[b]pyrrole framework 7 is formed by the consecutive [8+2] cyclization and cyclopentannulation reactions. The initially formed cyclopentannulation adduct can be intercepted through a Diels–Alder reaction with classic dienophiles to afford increasing structural complexity (compounds 8 and 9 ). More importantly, the construction of the indole skeleton is accomplished with a high degree of substitution and functionalization (compounds 11 – 15 ) by a one‐pot sequence that involves [8+2] cyclization, R? NC or CO insertion, and ring closure.  相似文献

An enantioselective synthesis of (+)‐8‐epi ‐xanthatin hinging on a chiral phosphoric acid catalyzed tandem allylboration/lactonization reaction is reported. With (+)‐8‐epi ‐xanthatin as the precursor, the collective synthesis of a series of synthetically challenging xanthanolides was also accomplished. Among them, xanthipungolide, one of the most complex xanthanolide monomers, was accessed through a bioinspired tandem double‐bond isomerization/6π electronic cyclization/intramolecular Diels–Alder reaction, and pungiolides A, B, D, E, and L–N, a group of xanthanolide dimers, were assembled through a bioinspired Diels–Alder dimerization followed by late‐stage diversification.  相似文献

Seven new prenylated indole alkaloids, taichunamides A–G, were isolated from the fungus Aspergillus taichungensis (IBT 19404). Taichunamides A and B contained an azetidine and 4‐pyridone units, respectively, and are likely biosynthesized from notoamide S via (+)‐6‐epi‐stephacidin A. Taichunamides C and D contain endoperoxide and methylsulfonyl units, respectively. This fungus produced indole alkaloids containing an anti‐bicyclo[2.2.2]diazaoctane core, whereas A. protuberus and A. amoenus produced congeners with a syn‐bicyclo[2.2.2]diazaoctane core. Plausible biosynthetic pathways to access these cores within the three species likely arise from an intramolecular hetero Diels–Alder reaction.  相似文献

An efficient and short entry to polyfunctionalized linear triquinanes from 2‐methoxyphenols is described by utilizing the following chemistry. The Diels–Alder reactions of masked o‐benzoquinones, derived from 2‐methoxyphenols, with cyclopentadiene afford tricyclo[5.2.2.02,6]undeca‐4,10‐dien‐8‐ones. Photochemical oxa‐di‐π‐methane (ODPM) rearrangements and 1,3‐acyl shifts of the Diels–Alder adducts are investigated. The ODPM‐rearranged products are further converted to linear triquinanes by using an O‐stannyl ketyl fragmentation. Application of this efficient strategy to the total synthesis of (±)‐Δ9(12)‐capnellene was accomplished from 2‐methoxy‐4‐methylphenol in nine steps with 20 % overall yield.  相似文献

Three new indole‐containing diketopiperazine alkaloids, named variecolorins M–O ( 1 – 3 ), together with the eight known analogues 4 – 11 , were isolated from a deep‐ocean sediment‐derived fungus, Penicillium griseofulvum. Their structures were determined by analysis of the spectroscopic data. The 2,2‐diphenyl‐1‐picrylhydrazinyl (DPPH) radical‐scavenging activities and the cell‐proliferation inhibitory activities of the three new compounds 1 – 3 were investigated.  相似文献

7‐Silyl‐ and 7‐silylmethylcycloheptatrienes were shown to react with acylnitroso reagents at room temperature, through their norcaradiene forms, to generate the corresponding cycloadducts 5 a – b and 6 a – b as single diastereomers. The course of the reaction was dramatically modified by changing the reaction conditions. Using a polar medium, functionalized cyclohexa‐1,3‐dienes 7 a – b and bicyclic compounds 13 a – b were instead generated, incorporating one or two amino groups. Similar behavior was observed by using other dienophiles, including triazolinedione, but also activated aldehydes and ketones. A tentative mechanism has been proposed to rationalize the formation of both classes of products that relies on a domino process involving four consecutive elementary steps, in this order: 1) electrocyclic process, 2) hetero‐Diels–Alder reaction, 3) cyclopropane ring opening, and 4) hetero‐Diels–Alder reaction. Trapping of the cationic intermediate and isolation of the primary cycloadduct provide support for this hypothesis. An enantioselective version of the cascade using cycloheptatriene 4 b and aldehydes and ketones, under copper(II) catalysis was also carried out, leading to cyclohexa‐1,3‐dienes 21 , 28 , and 30 with enantioselectivities up to 93 % ee. Finally, elaboration of the intermediates above has been carried out, opening a straightforward access to sugar mimics 42 – 43 and complex polycyclic systems 36 and 39 .  相似文献

Halotolerant fungus Cladosporium cladosporioides OUCMDZ‐187 was isolated from the mangrove plant Rhizophora stylosa collected in Shankou, Guangxi Province of China. Three new fatty acid esters cladosporesters A–C ( 1 – 3 ) and 5 new fatty acids cladosporacids A–E ( 4 – 8 ) were isolated from the ethyl acetate extract of the fermentation broth of OUCMDZ‐187 in a hypersaline (10% salt) medium. Their structures were elucidated by UV, IR, MS, specific rotation, and 1D and 2D NMR data.  相似文献

We report the late-stage chemical modification of ribosomally synthesized and post-translationally modified peptides (RIPPs) by Diels–Alder cycloadditions to naturally occurring dehydroalanines. The tail region of the thiopeptide thiostrepton could be modified selectively and efficiently under microwave heating and transition-metal-free conditions. The Diels–Alder adducts were isolated and the different site- and endo/exo isomers were identified by 1D/2D 1H NMR. Via efficient modification of the thiopeptide nosiheptide and the lanthipeptide nisin Z the generality of the method was established. Minimum inhibitory concentration (MIC) assays of the purified thiostrepton Diels–Alder products against thiostrepton-susceptible strains displayed high activities comparable to that of native thiostrepton. These Diels–Alder products were also subjected successfully to inverse-electron-demand Diels–Alder reactions with a variety of functionalized tetrazines, demonstrating the utility of this method for labeling of RiPPs.  相似文献

The total synthesis of the smallest cytochalasin isolated so far, periconiasin G, which bears a seven‐membered ring in lieu of the usual macrocycle, has been performed from both enantiomers of citronellal, relying on an intramolecular Diels–Alder reaction in favor of the natural endo stereochemistry. We show that, among the four synthesized stereoisomers, including the exo isomers, the one matching the NMR data of the natural product was not that assigned in the original report, imposing structure revision. The natural product, previously isolated from a plant‐mutualistic fungus, was biologically investigated taking into account its natural history, showing significant effects against the phytopathogenic fungus Botrytis cinerea and thus opening new opportunities in combating this pest.  相似文献

The asymmetric Friedel–Crafts reaction between a series of substituted indoles 2 a – l and methyl (E)‐2‐oxo‐4‐aryl‐3‐butenoates 3 a – c has been efficiently catalyzed by the scandium(III) triflate complex of (4′S,5′S)‐2,6‐bis[4′‐(triisopropylsilyl)oxymethyl‐5′‐phenyl‐1′,3′‐oxazolin‐2′‐yl]pyridine (pybox; 1 ). Substituted 4‐(indol‐3‐yl)‐2‐oxo‐4‐arylbutyric acid methyl esters 4 a – n were usually formed in excellent yields and the enantioselectivity was up to 99 % ee, irrespective of the electronic character of the substituent and its location on the indole ring, albeit with the exclusion of position 2. The adducts could be obtained as stable enol tautomers and the equilibrium with the keto structure is discussed. The X‐ray crystal structure determination of 4 m indicated the 4R absolute configuration, thus confirming the proposal of Jørgensen for 4 i . The sense of the stereoinduction can be rationalized by the same octahedral complex 5 between 3 , pybox 1 , and scandium triflate already proposed for the Diels–Alder/hetero‐Diels–Alder and the Mukaiyama–aldol reactions of pyruvates.  相似文献

A bioinsipred gold‐catalyzed tandem Diels–Alder/Diels–Alder reaction of an enynal and a 1,3‐diene, forming the highly‐strained benzotricyclo[3.2.1.02,7]octane skeleton, was reported. In contrast, a Diels–Alder/Friedel–Crafts tandem reaction occurred instead when silver salts were used as the catalyst. Although both reactions experienced the similar Diels–Alder reaction of a pyrylium intermediate with a 1,3‐diene, they have different reaction mechanisms. The former proceeded with a stepwise Diels–Alder reaction, while the latter one with a concerted one.  相似文献

Three new metabolites, microsphaeropsones A–C ( 1 – 3 ) with a unique oxepino[2,3‐b]chromen‐6‐one (ring‐enlarged xanthone) skeleton, were isolated from the endophytic fungus Microsphaeropsis species, co‐occurring with their putative biogenetic anthraquinoide precursors citreorosein ( 4 ) and emodin ( 5 ). From another Microsphaeropsis species, large amounts of fusidienol A ( 8 a ), smaller amounts of emodin ( 5 ), the known aromatic xanthones 9 a and 9 b , the new 3,4‐dihydrofusidienol A ( 8 b ), and the new aromatic xanthone 9 c were isolated. The endophyte Seimatosporium species produced a new aromatic xanthone, seimatoxanthone A ( 10 ), and 3,4‐dihydroglobosuxanthone A ( 12 ), closely related to α‐diversolonic ester ( 13 ) from Microdiplodia sp.. The structures were determined mainly by extensive 1D and 2D NMR experiments and supported by X‐ray single‐crystal analysis of 1 and the oxidation product 7 . The absolute configurations of the microsphaeropsones A–C ( 1 – 3 ) were established by comparison of the electronic and vibrational circular dichroism (ECD and VCD) spectra of 1 with time‐dependent DFT (TDDFT) and DFT calculations by using either the solid‐state structures or DFT‐optimized geometries as inputs. Preliminary studies indicated that 1 , 2 , and enone 7 showed antibacterial, fungicidal, and algicidal properties.  相似文献

Eight new benzoylated gentisyl alcohol (=2‐(hydroxymethyl)benzene‐1,4‐diol) glucosides, itosides A–H ( 1 – 8 ), together with the new pyrocatechol (=benzene‐1,2‐diol) glycoside itoside I ( 9 ) were isolated from the bark and twigs of Itoa orientalis (Flacourtiaceae). In itosides B–D ( 2 – 4 ), the gentisyl alcohol moiety was esterified by 1‐hydroxy‐6‐oxocyclohex‐2‐ene‐1‐carboxylic acid, while itosides E–H ( 5 – 8 ) contained instead an additional 2‐hydroxybenzoic acid moiety. The compounds were accompanied by the known derivatives 4‐hydroxytremulacin ( 10 ), poliothyrsoside ( 11 ), poliothyrsin ( 12 ), homaloside D ( 13 ), tremulacin, and pyrocatechol β‐D ‐glucopyranoside. The structures of the new compounds were elucidated by spectral and chemical methods.  相似文献

Five new mixed terpenoids, tricycloalternarenes (TCAs) A–E ( 1 – 5 ), together with two known compounds, TCA 1b ( 6 ) and TCA 2b ( 7 ), were isolated from the solid‐state‐cultured endophytic fungus Alternaria alternata of Maytenus hookeri. Their structures were identified by extensive spectroscopic (especially 2D‐NMR) experiments. Compound 7 showed weak activity against human tumor cell lines by MTT assay.  相似文献

A catalytically active aluminum‐based system for Diels–Alder transformations is reported. The system was generated by mixing a β‐diketiminate‐stabilized aluminum bistriflate compound with Na[BArCl4] (ArCl=3,5‐Cl2C6H3). Solid‐state analysis of the catalytic system reveals a unique structure incorporating a two‐dimensional coordination polymer. According to the experimental results obtained from several Diels–Alder transformations, the aluminum‐based system appears to be a more practical and more robust alternative to the recently reported compounds based on carbon and silicon cations.  相似文献

Rate constants and derived activation parameters of organic reactions in aqueous media, in particular Diels–Alder reactions, are sensitive to the presence of cosolvents in water. To enhance the solubility window of water, we introduced ionic liquids as cosolvents in the aqueous Diels–Alder reaction between anthracene‐9‐carbinol and N‐ethylmaleimide. The reactive potentials of the organic compounds are parameterized by using semi‐empirical quantum chemical methods. The principle of Savage–Wood additivity of group interactions is used to quantify the pairwise group interactions among chemically inert ionic liquids and organic reactants, both at initial and transition states of the reaction. The present approach shows promise, as the use of simple calculations from easily available kinetic data can help researchers to understand the versatility of green ionic‐liquid alternatives to volatile organic solvents.  相似文献
