We described here a new method for the determination of total calcium in plasma. The method is based on the precipitation of calcium with excess oxalate and the measurement of residual oxalate by flow injection analysis with Ru(bpy)32+ electrochemiluminescent detection. It has the advantages of extremely stable reagent, user‐friendly instrument, high selectivity, good analytical recovery, wide dynamic range, and nice correlation with atomic absorption spectroscopy. The calibration plot for calcium is linear over a concentration range from 0.5 mmol L?1 to 4.8 mmol L?1, which is wider than those obtained by most other methods. The analytical recoveries for plasma calcium are 98.4–101.2% with coefficients of variation (CVs) of 1.96–2.52%. The within‐day CVs range from 0.76% to 0.95%, and between‐day CVs were from 1.12% to 1.46%. The time for each injection is one minute. Because the proposed method can be readily carried out on increasingly popular instruments for Ru(bpy)32+ ECL immunoassays and DNA probe assays, Ru(bpy)32+ ECL method is suitable for routine clinical analysis of calcium.  相似文献

A flow injection spectrophotometric procedure with symmetric merging zones for dipyrone determination in pharmaceutical formulations is proposed. The determination is based on the formation of a blue complex (monitored at a wavelength of 642 nm) yield in the complexation reaction of dipyrone with Fe(III) in acid medium. Under optimum conditions, a calibration curve was obtained from 3.5 to 281 mg L?1 with a detection limit of 2.8 mg L?1 and the samples throughput was 80 h?1. The analytical results obtained for commercial formulation samples by applying the proposed method were in good agreement with labeled values and those obtained by a comparative procedure at a 95% confidence level.  相似文献

A post- column chemiluminescent technique for thedetection of compounds that are poor chromoshores using electorogenerated chemiluminescence following separation by capillartgy electrophoresis is described. The luminrescent signal is generated followintg the reaction of anlyres (e.g. amines) with Ru(bpy)33+, which isx electrochemically generated post-columan from Ru(bpy)32+. Tripropylamine and proline are used as two model compounds to demostrate the feasibility of the method. Detection limits for the prototype system were in the micromolar rage, suggesting that this technnique offers an alternative to indirect detection of compounds that are poor chromophores with an added selectivity advangage. The system includes the use of a conductive joint to isolate the separation field from the potential necessary to drive the elctrogenerated chemiluminescent reactiion. Addition of the chemiluminescent reagent Ru(bpy)32+ post-column did not decrease the efficiency of the separation. The design and favrication of the novel cell is discussed.  相似文献

A novel electrogenerated chemiluminescence (ECL) sensor based on natural clay and ionic liquid was fabricated. Tris(2,2′‐bipyridine)ruthenium(II) (Ru(bpy)32+) was immobilized on natural clay surface through simple adsorption. An ECL sensor was prepared by mixing Ru(bpy)32+‐incorporated clay, graphite powder and an ionic liquid (1‐butyl‐3‐methylimidazolium hexafluorophosphate) as the binder. The electrochemical behavior and ECL of the immobilized Ru(bpy)32+ was investigated. It was observed that the ECL of immobilized Ru(bpy)32+ was activated by the ionic liquid. The proposed ECL sensor showed high sensitivity to tri‐n‐propylamine (TPrA) and the detection limit was found to be 20 pM. In addition, the ECL sensor displayed good stability for TPrA detection and long‐term storage stability.  相似文献

儿茶酚胺是一类非常重要的神经递质,在人体的心血管系统、神经系统、内分泌腺、肾脏、平滑肌等组织系统的生理活动中起着广泛的调节作用。肾上腺素为儿茶酚胺的一种,建立灵敏、高效的肾上腺素检测技术具有重要的临床意义。本文将银(Ⅲ)配合物与鲁米诺组成新的流动注射化学发光体系,利用碱性介质中肾上腺素对三价银配合物-鲁米诺化学发光体系有明显的增强效应来测定肾上腺素的含量,并据此建立了高效测定肾上腺素的流动注射化学发光新方法。在优化的条件下,该方法测定肾上腺素的线型范围为1.0×10-9~1.0×10-7 mol L-1,检出限为8.0×10-10 mol L-1,对1.5×10-8 mol L-1肾上腺素11次平行测定,其相对标偏差为2.9%。利用建立的分析方法测定了药物肾上腺素,并对三价银-鲁米诺化学发光新体系测定肾上腺素的反应机理进行了讨论。  相似文献

报道了一种以钌(Ⅱ)-联吡啶[Ru(bpy)2+3]为催化剂的B-Z化学发光振荡新现象. 研究了B-Z化学发光振荡反应的影响因素, 并对体系的UV光吸收振荡进行了对比研究, 探讨了化学发光振荡反应的可能机理. 结果表明, 该体系的化学发光振荡是由于氧化态的钌(Ⅱ)-联吡啶被振荡反应过程中的强还原性中间体还原所引起的, 化学发光振荡随时间增加呈现周期性变化.  相似文献

Flow injection methodology is described for the determination of proline in red and white wines using tris(2,2′-bipyridyl)ruthenium(II) chemiluminescence detection. Selective conditions were achieved for proline at pH 10, while other amino acids and wine components did not interfere. The precision of the method was less than 1.00% (R.S.D.) for five replicates of a standard (4 × 10−6 M) and the detection limit was 1 × 10−8 M. The level of proline in white and sparkling wines using the developed methodology was equivalent to those achieved using HPLC-FMOC amino acid analysis. SPE removal of phenolic material was required for red wines to minimize Ru(bipy)33+ consumption and its associated effect on accuracy.  相似文献

A novel chemiluminescence (CL) method is developed for determination of ofloxacin and levofloxacin with Ag(III) complex in H2SO4 solution medium. The CL intensity is proportional to drug concentration in a wider range with a correlation coefficient of 0.999. The limit of detection (s/n = 3) for ofloxacin and levofloxacin was 5.3 × 10?9 g ml?1 and 8.3 × 10?9 g ml?1, respectively, and their recoveries from urine and serum samples were in the range of 90.1–112% with the RSDs of 1.0–2.8%. The proposed method was applied for analysis of real samples with satisfactory result. The possible CL mechanism was discussed.  相似文献

A novel tris(2,2'-bipyridyl)ruthenium(III) [Ru(bipy)(3) (3+)]-based chemiluminescence (CL) detection in CE using an on-line chemical generation scheme has been demonstrated. Two continuous streams respectively containing solutions of Ru(bipy)(3) (2+) and acidic cerium(IV) used as a homogeneous chemical oxidant are employed to generate Ru(bipy)(3) (3+), which is delivered into the reaction capillary of a coaxial flow interface and then reacted with analytes at the end of the separation capillary to yield light. The important operational parameters for separation and detection are identified and optimized. Four alpha-ketocarboxylic acids used as models, outside of the amine-containing compounds, are successfully separated and detected to evaluate the feasibility of the approach. The excellent resolution and detection sensitivity was achieved by using 50 mmol/L phosphate running buffer (pH 9.5) with 0.7 mmol/L CTAB, and CL reagent solution streams containing 0.15 mmol/L Ru(bipy)(3) (2+) and 0.8 mmol/L cerium(IV) (0.25 mol/L H(2)SO(4)), respectively. The concentration detection limits for alpha-ketocarboxylic acids were below 3.7x10(-8) mol/L (S/N = 3). The proposed method was applied to the determination of alpha-ketocarboxylic acids in five different honey samples with satisfactory results.  相似文献

A flow injection (FI) system combined with two photochemical processes is developed for the sensitive and rapid determination of carbaryl. It is based on the on-line photo-conversion of carbaryl into methylamine which subsequently reacts with Ru(bpy)33+ generated through the on-line photo-oxidation of Ru(bpy)32+ with peroxydisulphate. The linear concentration range of application was 0.04-4.0 μg ml−1 of carbaryl, with an R.S.D. of 1.2% (for a level of 0.50 μg ml−1) and a detection limit of 0.012 μg ml−1. The sample throughput was 200 injections per hour. The applicability of the method was demonstrated by determining carbaryl in commercial formulations, water, soil, grain and blood serum.  相似文献

A flow injection (FI) method is reported for the determination of thyroxine based on its enhancement of chemiluminescence (CL) from the Ru(bpy)33+-NADH system. The calibration graph was linear over the range 2.0-10 × 10−8 mol L−1 (r2 = 0.9989) with relative standard deviations (R.S.D.) in the range 2.0-4.5% (n = 4). The limit of detection (3σ blank) was 1.0 × 10−9 mol L−1 with sample throughput of 120 h−1. The effect of some organic compounds, anions and cations were studied for l-thyroxine determination. The method was applied to pharmaceutical preparations and the results obtained were in reasonable agreement with the amount labeled. The method was statistically compared with the results obtained by RIA; no significant disagreement at 95% confidence limit was observed. A calibration graph of NADH over the range 1.3 × 10−8-1.3 × 10−6 mol L−1 was also established (r2 = 0.9992) with R.S.D. in the range1.0-3.5% (n = 4). The limit of detection (3σ) was 1.0 × 10−10 mol L−1 NADH.  相似文献

A novel flow injection chemiluminescence (CL) system for the determination of isoniazid has been proposed. It is based on the direct CL reaction of isoniazid and Mn(III) in sulfuric acid medium. The unstable Mn(III) was on-line electrogenerated by constant current electrolysis. The CL emission intensity was linear with isoniazid concentration in the range 0.1–10 μg/mL; the detection limit was 3.2 × 10−2 μg/mL. The whole process could be completed in 1 min with a relative standard deviation of less than 5%. The proposed method is suitable for automatic and continuous analysis and has been applied successfully to the analysis of isoniazid in pharmaceutical preparation.  相似文献

In this paper, we report the first attempt to use humic acid (HA) as modifiers to prepare the organic‐inorganic hybrid modified glassy carbon electrodes based on HA‐silica‐PVA (poly(vinyl alcohol)) sol‐gel composite. Electroactive species of tris(2,2′‐bipyridyl)ruthenium(II) (Ru(bpy) ) can easily incorporate into the HA‐silica‐PVA films to form Ru(bpy) modified electrodes. The amount of Ru(bpy) incorporated in the composite films strongly depends on the amount of HA in the hybrid sol. Electrochemical and electrogenerated chemiluminescence (ECL) of Ru(bpy) immobilized in HA‐silica composite films coated on a glassy carbon electrode have been studied with tripropylamine (TPA) as the coreactant. The analytical performance of this modified electrode was evaluated in a flow injection analysis (FIA) system with a homemade flow cell. The as‐prepared electrode showed good stability and high sensitivity. The detection limits (S/N=3) were 0.050 μmol L?1 for TPA and 0.20 μmol L?1 for oxalate, and the linear ranges were from 0.10 μmol L?1 to 1.0 mmol L?1 for TPA and from 1.0 μmol L?1 to 1.0 mmol L?1 for oxalate, respectively. A sensitive flow injection chemiluminescence (CL) method is proposed for the determination of bovine serum albumin (BSA) using Copper(II)-Alizarin Red S (ARS) complex as an efficient chemiluminescent probe. The detection is based on the binding of the copper(II)-ARS complex to proteins and the catalytic activity of copper(II)-ARS in the luminol-H2O2 CL system. Under the selected conditions, the CL intensity is linear with the concentration of BSA in the range of 5.0 × 10?11 to 1.0 × 10?9 mol · L?1. The detection limit was 2.0 × 10?11 mol · L?1. Automated sequential injection (SIA) method for chemiluminescence (CL) determination of nonsteroidal anti-inflammatory drug indomethacin (I) was devised. The CL radiation was emitted in the reaction of I (dissolved in aqueous 50% v/v ethanol) with intermediate reagent tris(2,2′-bipyridyl)ruthenium(III) (Ru(bipy)33+) in the presence of acetate. The Ru(bipy)33+ was generated on-line in the SIA system by the oxidation of 0.5 mM tris(2,2′-bipyridyl)ruthenium(II) (Ru(bipy)32+) with Ce(IV) ammonium sulphate in diluted sulphuric acid. The optimum sequence, concentrations, and aspirated volumes of reactant zones were: 15 mM Ce(IV) in 50 mM sulphuric acid 41 μL, 0.5 mM Ru(bipy)32+ 30 μL, 0.4 M Na acetate 16 μL and I sample 15 μL; the flow rates were 60 μL s−1 for the aspiration into the holding coil and 100 μL s−1 for detection. Calibration curve relating the intensity of CL (peak height of the transient CL signal) to concentration of I was curvilinear (second order polynomial) for 0.1–50 μM I (r = 0.9997; n = 9) with rectilinear section in the range 0.1–10 μM I (r = 0.9995; n = 5). The limit of detection (3σ) was 0.05 μM I. Repeatability of peak heights (R.S.D., n = 10) ranged between 2.4% (0.5 μM I) and 2.0% (7 μM I). Sample throughput was 180 h−1. The method was applied to determination of 1 to 5% of I in semisolid dosage forms (gels and ointments). The results compared well with those of UV spectrophotometric method.  相似文献

An electrochemiluminescence (ECL) sensor with good long-term stability and fast response time has been developed. The sensor was based on the immobilization of tris(2,2′-bipyridyl)ruthenium(II) (Ru(bpy)32+) into the Eastman-AQ55D–silica composite thin films on a glassy carbon electrode. The ECL and electrochemistry of Ru(bpy)32+ immobilized in the composite thin films have been investigated, and the modified electrode was used for the ECL detection of oxalate, tripropylamine (TPA) and chlorpromazine (CPZ) in a flow injection analysis system and showed high sensitivity. Because of the strong electrostatic interaction and low hydrophobicity of Eastman-AQ55D, the sensor showed no loss of response over 2 months of dry storage. In use, the electrode showed only a 5% decrease in response over 100 potential cycles. The detection limit was 1 μmol l−1 for oxalate and 0.1 μmol l−1 for both TPA and CPZ (S/N=3), respectively. The linear range extended from 50 μmol l−1 to 5 mmol l−1 for oxalate, from 20 μmol l−1 to 1 mmol l−1 for TPA, and from 1 μmol l−1 to 200 μmol l−1 for CPZ.  相似文献

A sensitive and selective luminescence quenching method is developed and used for manual and flow injection analysis (FIA) of chromium(VI) by reaction with [Ru(bpy)3]2+. The emission peak of ruthenium(II) at 595 nm is linearly decreased as a function of Cr(VI) concentration. This permits determination of chromium(VI) ion over the concentration range 0.1-20 μg ml−1 with a detection limit of 33 ng ml−1. The quenching process is due to an electron transfer from the luminescent [Ru(bpy)3]2+ complex ion to Cr(VI) resulting in the formation of the non-luminescent [Ru(bpy)3]3+ complex ion. Selectivity for Cr(VI) over many anions and transition, alkali and alkaline earth metal cations is demonstrated. High concentration levels of sulphate, chloride, borate, acetate, phosphate, nitrate, cyanide, Pb2+, Zn2+, Hg2+, Cu2+, Cd2+, Ni2+ and Mn2+ ions are tolerated. The effects of solution pH and [Ru(bpy)3]2+ reagent concentration are examined and the reaction conditions are optimized. Validation of the method according to the quality assurance standards show suitability of the proposed method for use in the quality control assessment of Cr(VI) in complex matrices without prior treatment. The method is successfully applied to determine chromium(VI) in electroplating baths using flow injection analysis. Results with a mean standard deviation of ±0.6% are obtained which compare fairly well with data obtained using atomic absorption spectrometry.  相似文献
