A simple method for quantitative analysis of aesculin and aesculetin in Cortex fraxini was developed using capillary zone electrophoresis (CZE). Berberin was employed as an internal standard, The running buffer was 6 mM Na(2)B(4)O(7) and 10 mM NaH(2)PO(4) (pH 6.70). The linear calibration range was 32-256 mug ml(-1) (r=0.9996) for aesculin and 23-230 mug ml(-1) (r=0.9993) for aesculetin. The contents of aeculin and aesculetin in C. fraxini were easily determined within 12 min the pKa values of aesculin and asculetin determined by CZE were 6.56 and 5.62, respectively. A simple method for estimation of the temperature inside the capillary during running was also proposed.  相似文献

An approach that combined green‐solvent methods of extraction with chromatographic chemical fingerprint and pattern recognition tools such as principal component analysis (PCA) was used to evaluate the quality of medicinal plants. Pressurized hot water extraction (PHWE) and microwave‐assisted extraction (MAE) were used and their extraction efficiencies to extract two bioactive compounds, namely stevioside (SV) and rebaudioside A (RA), from Stevia rebaudiana Bertoni (SB) under different cultivation conditions were compared. The proposed methods showed that SV and RA could be extracted from SB using pure water under optimized conditions. The extraction efficiency of the methods was observed to be higher or comparable to heating under reflux with water. The method precision (RSD, n = 6) was found to vary from 1.91 to 2.86% for the two different methods on different days. Compared to PHWE, MAE has higher extraction efficiency with shorter extraction time. MAE was also found to extract more chemical constituents and provide distinctive chemical fingerprints for quality control purposes. Thus, a combination of MAE with chromatographic chemical fingerprints and PCA provided a simple and rapid approach for the comparison and classification of medicinal plants from different growth conditions. Hence, the current work highlighted the importance of extraction method in chemical fingerprinting for the classification of medicinal plants from different cultivation conditions with the aid of pattern recognition tools used.  相似文献

High-speed counter-current chromatography (HSCCC) was successfully used for the isolation and purification of coumarin compounds from Cortex fraxinus, the Chinese herbal drug. n-Butanol-methanol-0.5% acetic acid (5:1.5:5, v/v) was used as the two-phase solvent system. 14.3 mg of fraxin, 26.5 mg of aesculin, 5.8 mg of fraxetin and 32.4 mg of aesculetin with the purity of 97.6, 99.5, 97.2 and 98.7%, respectively were obtained from 150 mg of crude extracts of C. fraxinus in a single run. The structures of the isolated compounds were identified by 1H NMR and 13C NMR.  相似文献

Ionic liquids (ILs) solutions as solvents were successfully applied in the microwave-assisted extraction (MAE) of polyphenolic compounds from medicinal plants. ILs, its concentration and MAE conditions were investigated in order to extract polyphenolic compounds effectively from Psidium guajava Linn. (P. guajava) leaves and Smilax china (S. china) tubers. The results obtained indicated that the anions and cations of ILs had influences on the extraction of polyphenolic compounds as well as the ILs with electron-rich aromatic π-system enhanced extraction ability. Under the optimized conditions, the extraction yields of the polyphenolic compounds were in the range of 79.5-93.8% with one-step extraction, and meanwhile the recoveries were in the range of 85.2-103% with relative standard deviations (R.S.D.s) lower than 5.6%. Compared to conventional extraction procedures, the results suggested that the proposed method was effective and alternative for the extraction of polyphenolic compounds from medicinal plants. In addition, the extraction mechanisms and the structures of samples before and after extraction were also investigated. ILs solutions as green solvents in the MAE of polyphenolic compounds from medicinal plant samples showed a great promising prospect.  相似文献

Our earlier work showed that the stability of the bioactive compounds gastrodin (GA) and vanillyl alcohol (VA) in Gastrodia elata Blume behaved differently with varying compositions of water-ethanol using pressurized liquid extraction (PLE) at room temperature. To have a better understanding of the extraction process of these thermally labile compounds under elevated temperature conditions, pressurized hot water extraction (PHWE) and microwave-assisted extraction (MAE) methods were proposed. PHWE and MAE showed that GA and VA could be extracted using pure water under optimized conditions of temperature and extraction time. The extraction efficiency of GA and VA by the proposed methods was found to be higher or comparable to heating under reflux using water. The marker compounds present in the plant extracts were determined by RP-HPLC. The optimized conditions were found to be different for the two proposed methods on extraction of GA and VA. The method precision (RSD, n=6) was found to vary from 0.92% to 3.36% for the two proposed methods on different days. Hence, PHWE and MAE methods were shown to be feasible alternatives for the extraction of thermally labile marker compounds present in medicinal plants.  相似文献

Deep eutectic solvents (DESs) are considered as efficient and green solvents for the extraction of bioactive compounds from medicinal plants. In this work, a novel method of DES-based ultrasound-assisted extraction of bioactive compounds from Baphicacanthus cusia leaves (BCL) was established. Systematic screening and the morphology of the original and treated BCL were observed with scanning electron microscopy to determine the extraction efficiency of different solvents. The extraction conditions were optimized by Box–Behnken design (BBD) tests and the optimal extraction conditions were as follows: lactic acid/L-menthol ratio of 5: 2 (mol/mol), solid–liquid ratio of 80.0 mL/g and temperature of 60.5 °C. The extraction yields of tryptanthrin, indigo and indirubin reached 0.356, 1.744 and 0.562 mg/g, respectively. The results of a 2,2-diphenyl-1-picrylhydrazy (DPPH) radical scavenging activity test indicated the feasibility of DESs in the extraction of bioactive compounds. This study indicated that L-menthol/lactic acid was a green and efficient solvent for the extraction of bioactive compounds from BCL, and DES-based ultrasound-assisted extraction could be used as an effective application strategy for the extraction of bioactive compounds from medicinal plants.  相似文献

In this paper, the application of 1-n-butyl -3-methylimidazolium-based ionic liquids aqueous solutions as solvents in the microwave-assisted extraction (MAE) technique was first developed for the extraction of trans-resveratrol from Rhizma Polygoni Cuspidati. 1-butyl-3-methylimidazolium bromide ([bmim]Br) solution was selected as solvent; the conditions of MAE including the size of sample, liquid/solid ratio, extraction temperature and time were optimized by means of an orthogonal design L(9)(3(4)). Under optimized conditions, the extraction yield value of trans-resveratrol was 92.8% in a one-step extraction. The recovery was in the range 93.7-103.2% with relative standard deviation lower than 3.0% by the proposed procedure. The method was applied to extract trans-resveratrol from several different region samples. On the basis of extraction solvent and time, the proposed extraction technique was a green, rapid and alternative technique to extract and analyze trans-resveratrol in Rhizma Polygoni Cuspidati samples.  相似文献

A consecutive preparation method based upon accelerated solvent extraction (ASE) coupled with high‐speed counter‐current chromatography (HSCCC) was presented and aesculin was obtained from Cortex fraxinus. The extraction condition of ASE was optimized with response surface methodology; some significant parameters such as the solvent system and its stability, the amount of loading sample in HSCCC were also investigated. The original sample was first extracted with methanol at 105°C and 104 bar for 7 min using ASE, then the extracts were consecutively introduced into the HSCCC system and separated and purified with the same ethyl acetate/n‐butanol/water (7:3:10, v/v/v) solvent system for five times without further exchange and equilibrium. About 3.1 ± 0.2 mg/g in each time and total of 15.4 mg/g aesculin with purity over 95% was isolated from Cortex fraxinus. The results demonstrated that the consecutive preparation method was time and solvent saving and high throughput, it was suitable for isolation of aesculin from Cortex fraxinus, and also has good potential on the separation and purification of effective compounds from natural product.  相似文献

Flavonoid glycosides are a significant group of compounds found in Ginkgo biloba leaves, but the long extraction procedures in existing methods are a challenging problem. In this work, a microwave-assisted extraction (MAE) method has been developed for extracting bioactive compounds from G. biloba. Several variables were optimized, such as extracting solvent, microwave power, and extraction time that can potentially affect the extraction yield. The total phenolic content, antioxidant activity (using DPPH, ABTS and FRAP assays) and flavonoid glycosides of different extracts using RP-HPLC were assessed. The antioxidant capacity was found to be highest with MAE using 60% aq. ethanol as extracting solvent and microwave power of 120W for 20 min.  相似文献

The aim of this study was to evaluate the efficiency of microwave-assisted extraction (MAE) for the analysis of organophosphorus (OP) and pyrethroid (PYR) pesticides in green onions by GC/MS. We optimized MAE extraction solvent, temperature, and time by using a certified reference material. As a result, the concentrations of two OP and two PYR target pesticides obtained by MAE with acetonitrile at 110 degrees C for 10 min were in good agreement with certified concentrations and comparable to the results by homogenization used as reference extraction technique. When the recovery test, performed by spiking the target pesticides into blank samples (5.0 g), was carried out with our optimized MAE conditions, mean recoveries of 16 OP and 10 PYR pesticides were 72-108% for a 1.0 pg spiking level and 70-119% for a 0.2 microg level. These results were acceptable according to the validation guideline for testing method of agricultural chemicals in food by Ministry of Health, Labour, and Welfare in Japan. The results suggested that MAE can be used for the analysis of OP and PYR pesticides in green onions.  相似文献

The brewing properties of coffee products are defined by the chemical composition in the bean, including sugars and polyols. Some factors, such as coffee species and roasting, may affect the level of these compounds in the bean. A new analytical microwave-assisted extraction (MAE) method has been developed to extract sugars and polyols from the coffee bean. The studied extraction conditions for the MAE were temperature (30–80 °C), solvent composition (0–50% ethanol in water), and solvent-to-sample ratio (10:1–30:1 mL solvent per g sample). A Box-Behnken design was applied to study the effect of extraction variables, and subsequently, the influential variables were optimized by response surface methodology (RSM). In addition to the main effect of the solvent-to-sample ratio, all quadratic effects significantly influenced (p < 0.05) the recovery of sugars and polyols from the coffee beans. RSM suggested the optimized MAE conditions: temperature 52 °C, ethanol concentration in water 18.5%, and solvent-to-sample ratio 17:1. Under the optimum condition, a kinetics study confirmed that 15 min showed high precision and accuracy of the developed method. Ultimately, a real sample application of the developed MAE revealed that the new method successfully described the composition of sugars and polyols in regular and peaberry coffee beans. Additionally, the method also effectively characterized the green and roasted Arabica and Robusta coffee beans.  相似文献

Summary A capillary zone electrophoresis method for identification and determination of aesculin and aesculetin has been established using borate-phosphate buffer containing 30% ethanol with on-column UV detection. A detailed investigation of the influence of changes in borate concentration, pH, applied voltage, temperature and organic modifier was then carried out. For both aesculin and aesculetin, a linear plot of migration time (MT) against borate concentration was obtained, and ln[measured peak area (MA)] and lnMT both gave linear plots against ln(applied voltage) with correlation coefficient r>0.999, which also resulted in a linear correlation between MA and MT (r≥0.9998) under varied voltage. Ethanol as organic modifier to the background electrolytes helped in separating aesculin and aesculetin from other components in ash barks. The reproducibility with relative standard deviation in MT and in normalized peak area(NA) and linearity based on NA against concentration were evaluated. Finally, the method was successfully applied to monitor the quality of different ash barks and to compare the effect of sample preparation on content of bioactive components in ash bark. Results indicate that CZE promises to be applicable to quality control of traditional Chinese medicines containing aesculin and aesculetin.  相似文献

A multiresidue method developed for the analysis of atrazine and its principal conversion products, deisopropylatrazine (DIA), deethylatrazine (DEA) and hydroxyatrazine (HA), in soil is presented. The method is based on the microwave-assisted extraction (MAE) of soil with aqueous methanol followed by solid-phase extraction (SPE) of the extracts and subsequent analysis by LC–UV with a diode array detector. MAE operational parameters (extraction solvent, extractant volume) were optimized with respect to extraction efficiency of the target compounds from soils with 2.5% organic matter (OM) content. Recoveries above 80% were obtained for all solutes. Soil OM content did not affect analyte recoveries. Recoveries from fresh and aged residues, the latter weathered under cold storage conditions, were not statistically different. Finally, MAE was found to be superior in terms of extraction efficiency, sample throughput, and solvent consumption to conventional flask-shaking extraction.  相似文献

A method for PEG‐based microwave‐assisted extraction (MAE) of flavonoid compounds from persimmon leaves has been successfully developed. The extraction efficiency of total flavonoid content was evaluated by the chromatographic peak areas of quercetin and kaempferol, which are two bioactive components typically found in persimmon leaves. The best combination of extraction parameters was obtained with response surface methodology. A microwave power of 525 W, liquid to solid ratio of 17:1 mL/g, and PEG aqueous solution concentration of 60% w/w were identified as the optimum parameters. Extraction dynamics analysis indicated that the quercetin, kaempferol, and total flavonoid contents were rising with increasing extraction time up to 20–25 min, from which point onwards they all decreased. Under the optimum conditions, quercetin, kaempferol, and total flavonoid contents obtained from the sample were 1.20 ± 0.05, 0.64 ± 0.11, and 16.90 ± 0.06 mg/g, respectively. Compared with ethanol‐based MAE, and ethanol‐based and PEG‐based ultrasonic‐assisted extractions, PEG‐based MAE had higher efficiency for the extraction of flavonoid compounds from persimmon leaves. Overall, PEG‐based MAE represents an efficient choice for the extraction of bioactive substances from traditional Chinese medicines.  相似文献

A microwave-assisted extraction (MAE) method has been developed and optimized for the extraction of six tricyclic antidepressants (TCAD; nordoxepin, nortriptyline, imipramine, amitriptyline, doxepin, dezypramine) from human serum. Optimal parameters of MAE (solvent and extraction temperature) for water solution of these drugs were defined. The microwave-assisted procedure developed was validated by extraction of serum samples at two concentration levels and then successfully applied to the analysis of reference material. Limit of quantification, precision and recovery were found for the studied compounds in the ranges 0.04-0.15 microg/mL, 1.57-4.34% (RSD) and 94-105%, respectively.  相似文献

A multi-residue method developed for the analysis of triazine herbicides, simazine, atrazine, propazine and prometryne, in sheep liver is presented. The method is based on microwave-assisted extraction (MAE) of sheep liver using methanol as extractant and analysis of extracts by high performance liquid chromatography (HPLC) and ultraviolet detection. MAE operational parameters, the solvent type and volume, extraction temperature and time, were optimized in detail with respect to extraction efficiency of the target compounds from sheep liver. The recoveries of the method at two different spiked levels were assessed by analyzing spiked liver samples and were found to be in the range from 90 to 102% with good precision (<11%).  相似文献

SPE is a commonly applied technique for preconcentration of pesticides from water samples. Microwave‐assisted extraction (MAE) technique is the extraction applied for preconcentration of different compounds from solid samples. SPE coupled with MAE is capable of preconcentrating these compounds from water samples too. This investigation was aimed at improving the efficiency of atrazine, alachlor, and α‐cypermethrin pesticide extraction from the spiked water samples applying SPE followed by MAE. In this way, MAE served for elution of pesticides from C18‐extraction disks with solvent heated by microwave energy. Various elution conditions were tested for their effects on the extraction efficiency of the SPE–MAE combined technique. Several parameters, such as elution solvent volume (mL), elution temperature (°C), and duration of elution (min), affect the extraction efficiency of the SPE–MAE coupled system and need to be optimized for the selected pesticides. In order to develop a mathematical model, 15 experiments were performed in the central composite design. The equation was then used to predict recoveries of the pesticides under specific experimental conditions. Optimization of microwave extraction was accomplished using the genetic algorithm approach. Best results were achieved using 20 mL of ethanol at 60°C. Optimal hold time was 5 min and 24 s. The SPE–MAE combination was also compared with the conventional SPE extraction technique with elution of a nonpolar or a moderately polar compound with nonpolar solvents.  相似文献

A simple and rapid microwave-assisted extraction (MAE) procedure was developed and optimized for two common color pigments, alizarin and purpurin, in various samples of Rubiaceae plants. Several variables that can potentially affect the extraction efficiency, namely temperature, methanol concentration in the extractant mixture, time, and solvent volume were optimized by means of a central composite design approach. The results suggest that temperature and methanol concentration in the solvent mixture are statistically the most significant factors. The separation and quantitative determination of the pigments was carried out in less than 6 min by a developed high-performance liquid chromatographic method with UV detection at 250 nm. Under optimum operating conditions, MAE showed significantly higher recoveries than those obtained by the conventional extraction methods (ultrasonic and reflux extraction), ranging from 84 to 94%. In addition, a drastic reduction of the extraction time (20 min versus 6 h) and solvent consumption (20 versus 100 mL) was achieved with a reproducibility (RSDs < 10%) comparable with that provided by the reflux extraction as a reference method.  相似文献
