New approaches for the synthesis of some heterocyclic compounds, such as the pyridopyrimidodiazepine derivative 3 , pyrazolopyrido[1,2‐a]pyrimidine derivative 4 , tetrazolo[1.5‐a][1,8]naphthyridine derivative 9 , pyrazolylpyrido[1,2‐a]pyrimidine derivatives 10a , 10b , 12 , pyrrolopyrido[1,2‐a]pyrimidine derivatives 14a , 14b , 14c , 14d , and 16a , 16b , starting from 2‐chloro‐4H‐4‐oxo‐pyrido[1,2‐a]pyrimidine ( 1 ), are described.  相似文献

The interaction of condensed nitrogen-containing bridge systems with chloral has been studied and the high sensitivity of the reaction to the -excess of the initial heteroaromatic system has been established. It has been shown that chloral is a convenient formylating agent for systems with a moderate -excess —imidazo[l,2-a]imidazole, 9H-imidazo[1,2-a]benzimidazole, imidazo[1,2-a]pyridine, imidazo[1,2-a]naphtho[2,3-d]imidazole. Heterocycles with a high -excess (indolizine, pyrrolo[1,2-a]benzimidazole) form cyanine dyes under the action of chloral. Systems with a lowered -excess (1H-imidazo[1,2-a]benzimidazole, imidazo[1,2-a]quinoline, imidazo[2,1-a]isoquinoline, imidazo[1,2-a]perimidine, imidazo[5,1-b]benzoxazole, imidazo[1,2-a]benzothiazole, and imidazo[1,2-a]pyrimidine) do not react with chloral in a neutral medium. A cyclopropyl quinone methide is formed by elimination of a leaving group from an appropriately functionalized hydroquinone. The presence of a carbon spacer results in the formation of a fused ring rather than the classic methide species. Discussed herein is cyclopropyl quinone methide formation from a pyrido[1,2-a]indole ring system. Both nucleophilic and electrophilic attack on the fused cyclopropane ring results in pyrido[1,2-a]indole and azepino[1,2-a]indole products. The stereoelectronic effect plays less a role in the relatively flexible pyrido[1,2-a]indole system compared to its role in the pyrrolo[1,2-a]-indole system. A 13C label on the fused cyclopropane ring permitted the rapid identification of complex rearrangement products observed in this study.  相似文献

Phenanthro[a] phenazine, a new heterocyclic ring structure of both biological and physical interest, has been prepared by coupling 1,2-phenylenediamine with triphenylene-1,2-diquinone derivatives in high yields. 2,3,6,7,10-Penta-alkoxyphenanthro[a] phenazines are mesogenic over a broad temperature range. These novel unsymmetrical, coloured, polar, heteroaromatic discotic mesogens are potential candidates for the study of conducting, photoconducting, electroluminescence, photorefrective, etc. properties.  相似文献

Due to their diverse range of biological activities, imidazoheterocycles are recognized as privileged structures making these structural motifs attractive targets for library preparation. We report herein the synthesis of a sizable collection of imidazo[1,2- a]heterocycle scaffolds well-suited for divergent library preparation by virtue of amine functional handles with diverse positioning and connectivities. Partial reduction of imidazo[1,2- a]pyrazines to the tetrahydroimidazo[1,2- a]pyrazines and regiospecific Mannich-type bond formation at the C-3 of imidazo[1,2- a]pyridine under mild conditions achieved additional topological and connective diversity within the scaffold collection. Subsequent parallel reaction of the functionalized imidazoheterocycles with polystyrene-tetrafluorophenol esters and sulfonates produced a 7500 compound library in high purity.  相似文献

The fused heterocyclic compounds 2 : imidazo [1,2-a] pyridine 2a-c and pyrido [1,2-a] pyrimidine 2d were obtained from the reaction of a -cinnamoyl ketene dibenzylthio acetals 1 with diamine. When a -cinnamoyl -a '-benzoyl ketene N, N-acetals 3a-b were treated by t-BuONa/t-BuOH solution, 8- benzoyl-pyrido[1,2-a] pyrimidine 4 was produced.  相似文献

Compounds containing a novel tetracyclic ring system, a hybrid of tetrahydropyrrolo[2,3-b]indole and tetrahydroimidazo[1,2-a]indole, are synthesized via an acid-mediated stereospecific domino tautomerization of a tryptophan-derived alpha-amino nitrile. The synthesis and characterization of the extended thieno[3,4-b]pyrazine analogues acenaphtho[1,2-b]thieno[3,4-e]pyrazine (3a), 3,4-dibromoacenaphtho[1,2-b]thieno[3,4-e]pyrazine (3b), 3-octylacenaphtho[1,2-b]thieno[3,4-e]pyrazine (3c), dibenzo[f,h]thieno[3,4-b]quinoxaline (4), and thieno[3',4':5,6]pyrazino[2,3-f][1,10]phenanthroline (5) are reported. Comparison of structural, electrochemical, and photophysical properties to those of simple thieno[3,4-b]pyrazines are provided in order to provide structure-function relationships within this series of compounds.  相似文献

A versatile method for the solid-phase synthesis of imidazo[1,2-a]pyridine-based derivatives, imidazo[1,2-a]pyridine-8-carboxamides, has been developed. They were obtained by treatment of the amino group of the polymer-bound 2-aminonicotinate with different alpha-haloketones, followed by halogenation at the 3-position of the polymer-bound imidazo[1,2-a]pyridine. The derived polymer-bound imidazo[1,2-a]pyridines 5, 6, and 7 were finally cleaved from the solid-support with an excess of primary or secondary amines. The final crude products were purified from excess amines by solid-supported liquid-liquid extraction (SLE).  相似文献

Quantum chemistry calculations have been used to study the metal-free hydrogenation reactions of a variety of simple aromatic, heteroaromatic, and related linear conjugated systems. We find that the barrier for uncatalyzed 1,4-hydrogenation is always substantially lower (by approximately 200 kJ mol-1) than that for 1,2-hydrogenation, despite similar reaction enthalpies. The presence of hydrogen fluoride as a catalyst is found to decrease the 1,2-hydrogenation barriers but, in most cases, to slightly increase the 1,4-hydrogenation barriers when a constrained geometric arrangement is employed. These qualitative observations are consistent with orbital symmetry considerations, which show that both the uncatalyzed 1,4-hydrogenation and the catalyzed 1,2-hydrogenation are formally symmetry-allowed processes. An extreme example of the catalyzed 1,2-hydrogenation of benzene is provided by the involvement of a second molecule of hydrogen, which leads to a substantial lowering of the barrier. The effect of catalysis was further investigated by applying a selection of additional catalysts to the 1,2- and 1,4-hydrogenation of benzene. A decreasing barrier with increasing catalyst acidity is generally observed for the catalytic 1,2-hydrogenation, but the situation is more complex for catalytic 1,4-hydrogenation. For the uncatalyzed 1,4-hydrogenation of aromatic systems containing one or more nitrogen heteroatoms, the barriers for [C,C], [C,N], and [N,N] hydrogenations are individually related to the reaction enthalpies by the Bell-Evans-Polanyi principle. In addition, for a given reaction enthalpy, the barriers for [C,C] hydrogenation are generally lower than those for [C,N] or [N,N] hydrogenation. Finally, we find that the distortion experienced by the reactants in forming the transition structure represents a secondary factor that influences the reaction barrier. The correlation between these quantities allows the 1,4-hydrogenation barriers to be predicted from a ground-state property.  相似文献

The five-step synthesis of the new 4H-anthra[1,2-b]pyran derivative 1 is reported. The key steps in this approach included a Marschalk alkylation of 1,4-dihydroxyanthraquinone followed by a Baker-Venkataraman reaction and then an acid-catalyzed cyclization of ring A to form the 4H-anthra[1,2-b]pyran system. Two compounds, the 4H-anthra[1,2-b]pyran 1 and the anthraquinone derivative 6 were evaluated for antimicrobial activity and showed moderate antialgal, antifungal, and antibacterial activities.  相似文献

[formula: see text] The preparation of the first library of imidazo[1,2-a]pyridine derivatives on a solid support is described. A sulfone linker strategy was applied in the synthesis. Key steps involved in the solid-phase synthetic procedure include (i) alpha-haloketone resin formation by sulfinate-->sulfone alkylation, (ii) imidazo[1,2-a]pyridine ring formation by treatment with 2-aminopyridine, (iii) sulfone anion alkylation, and (iv) traceless product release by oxidation-elimination. When derivatives of 1,2,3,9a-tetrahydro-9H-imidazo[1,2-a]indol-2-one or 1-carbamoylmethyl-2-methylene-2,3-dihydroindole are reacted with lithium aluminum hydride, derivatives of 1,2,3,9a-tetrahydro-9H-imidazo[1,2-a]indole are formed. Under the same conditions 1-(N-phenylcarbamoylmethyl)-2-methylene-2,3-dihydroindole is not cyclized to an imidazo[1,2-a]indole. WHen treated with proton acids imidazo[1,2-a]indoles are converted to 3H-indolium salts. Derivatives of 2-thiono-5-oxo-2,3,4,5-tetrahydroindeno[1,2-d] pyrimidine and 5-oxo-1,2,4,5-tetrahydroindeno [1,2-d] [3,1] thiazine are formed in the cyclocondensation of 2-arylideneindan-1,3-diones with thiourea and N-monomethylthiourea, while only derivatives of indeno [1,2-d]-pyrimidine are formed in the reaction with N,N-di-methylthiourea. The recent research results suggest that a series of octahydro-pyrazino[ 1,2-a ]pyrazines are useful as antiallergic agents.[1] Gubert et al. [2] developed a route from 1,4-dibenzyl-2-ethoxycarbonylpiperazine for the synthesis of this ring system. Under their conditions, this ring system was obtained with a low overall yield in six steps. Optimization of synthetic route to this ring system is still desirable. In this paper, we presented an efficient synthesis of octahydropyrazino[1,2-a]pyrazine by a new and efficient method starting from 1,3-dichloro-2-propanol and N-tosylated diethylenetriamine. The structure of title compound was determined by spectroscopy and X-ray diffraction technique.  相似文献

A convenient method is proposed for the preparation of new derivatives of free heterocyclic bases of imidazo[1,2-a]pyridine, pyrido[1,2-a]benzimidazole, and [1,2,4]triazolo[4,3-a]pyridine series.
