Alticinae has the greatest amount of biodiversity among the Chrysomelidae, with 40,000 described species, only 290 of which have been analyzed cytogenetically. The majority of studies refer to conventional staining and few species have been analyzed or have responded to differential staining methods. The aim of the present study was to describe an 18S rDNA probe for Alticinae and the location of this cluster in species of the Omophoita genus. The fragment of approximately 750 bp obtained through a PCR (Polymerase Chain Reaction) amplification reaction with specific oligonucleotides to 18S rDNA was cloned and denominated pTZ_Ooct_18Sp and then submitted to automatic sequencing. The alignment of the sequences obtained through the sequencing of the clones generated a consensus sequence of 722 bp for Omophoita octoguttata with 98% homology with other species of Alticinae. The analysis of mitotic cells of O. octoguttata and Omophoita magniguttis submitted to fluorescent in situ hybridization (FISH) with the 18S rDNA probe revealed that the ribosomal genes are located in 6th pair. O. magniguttis also has a second labeled pair. Omophoita personata exhibited nucleolar organizer regions associated to one autosome pair. The analysis of meiotic cells submitted to FISH revealed one labeled bivalent in metaphase I in O. octoguttata and O. personata and in one chromosome in metaphase II in O. octoguttata. FISH data suggest a conserved pattern in the species analyzed and an apomorphy of O. magniguttis karyotype. The rDNA 18S probe could be considered an important marker to evidence the karyotypic differentiation, not observed with conventional methodologies, in species considered karyotypically conserved and uniform.  相似文献

Among the subfamilies of Chrysomelidae, Cassidinae sensu lato (s.l.) includes 6000 species distributed in 43 tribes. Approximately 100 of these species were cytogenetically analyzed and most of them presented 2n = 18 = 16 + Xyp, which was smaller than 2n = 20 = 18 + Xyp considered basal for Polyphaga. However, some groups of species presented maintenance of the basal diploid number and others showed increase in this number. Certain species of the latter group also exhibited variation in the type of sex chromosome system (SCS). Considering the recent taxonomic revision accomplished for the Cassidinae s.l. species, the existence of phylogenetic relationship for some species of this subfamily, the high diversity of species of this group in the Neotropical region, and the low number of Cassidinae s.l. species karyotyped so far, the aim of the present work was to establish the main mechanisms involved in the karyotype evolution of this subfamily through the study of seven species of the Brazilian fauna and overview of the cytogenetic data. The individuals were collected in southeast and south of Brazil. The chromosomal preparations obtained from embryo and testes of adult males were stained with Giemsa solution. The species Agroiconota inedita (2n = 42 = 40 + Xyp), Charidotella (s.str.) immaculata (2n = 22 = 20 + Xyp), Charidotella (s.str.) sexpunctata (2n = 22 = 20 + Xyp), and Stolas chalybaea (2n = 24 = 22 + Xyp) revealed diploid number higher than that established as basal for Polyphaga and biarmed chromosomes. The karyotype of Cteisella confusa, Deloyala cruciata, and Metriona elatior showed the chromosomal formulae 2n = 18 = 16 + Xyp considered modal for Cassidinae s.l. and biarmed chromosomes. The seven species exhibited easily identified sex chromosomes due to their size and/or morphology. The analysis of meiotic cells of all the species showed pachytenes with a positively heteropycnotic block probably corresponding to the sex chromosomes; diplotenes with a high number of bivalents with two chiasmata and sex chromosomes in a parachute configuration, and metaphases II that confirmed the chromosomal morphology, the type of SCS, and the regular segregation of all chromosomes. The data regarding to the number and morphology of the chromosomes, their behaviour during meiosis, and type of SCS were inedit for the majority of these species. In relation to the all Cassidinae s.l. species that presented SCS of the Xyp type, A. inedita was that with the highest diploid number. Furthermore, this work reported for the first time the cytogenetic information of representatives of the genera Cteisella and Metriona. Taking into account the phylogenetic and cytogenetic data of Cassidinae s.l. species, the karyotype differentiation of this group seems to have occurred from the basal karyotype of Polyphaga by decrease in the chromosome number and subsequent increase in this number. Pericentric inversion, centric fusion and fission seem to have been the main mechanisms that promoted the evolution of the autosomes. However, in the sex chromosome evolution, the mechanisms involved were centric fission and/or chromosomal translocation.  相似文献

Male mitotic and meiotic chromosomes of two species of the genus Deltochilum (Scarabaeidae) were analyzed through conventional staining, C-banding, base-specific fluorochromes, silver nitrate staining (AgNO3) and FISH (45S rDNA). The two species possessed karyotypes with 2n = 14, neo-XY and meta-submetacentric chromosomes. The analysis of constitutive heterochromatin (CH) revealed mainly diphasic chromosomes in the two species, showing heterochromatic long arms. Silver nitrate staining labeled the blocks corresponding to CH in D. (Deltohyboma) aff morbillosum while in D. (Deltohyboma) calcaratum, AgNO3 staining revealed only the CH blocks of the diphasic autosomes. The fluorochrome staining revealed in D. (D.) calcaratum the diphasic autosomes and the sex chromosomes with CMA3+ blocks, and in D. (D.) aff morbillosum, the GC-rich sequences were restricted to the terminal regions of the long arms of the pairs 1 and 2 and the X. The FISH revealed 45S rDNA sites in two autosomic pairs and in the X chromosome. The analyses performed allowed for the identification of cytogenetic markers and the discussion of possible chromosome rearrangements that have been involved in the karyotypic differentiation of these species mainly related to the repetitive genome.  相似文献

Potassium isopropyl xanthate, (CH3)2CHOC(S)SK, reacts with methyl chloroformiate ClC(O)OCH3 to yield (methoxycarbonyl) (2‐propoxythiocarbonyl) sulfide, (CH3)2CHOC(S)SC(O)OCH3. This novel xanthogen formate was characterized by 1H and 13C{1H} NMR spectroscopy, mass spectrometry and IR and Raman spectroscopy. The structure of a single crystal of (CH3)2CHOC(S)SC(O)OCH3 was determined by X‐ray diffraction analysis at 173 K. The conformational properties have been studied by liquid IR and Raman spectroscopy, matrix isolation spectroscopy together with photochemical studies and quantum chemical calculations (HF and B3LYP methods with the 6‐31+G* basis set). The analysis of the IR spectrum of liquid (CH3)2CHOC(S)SC(O)OCH3 suggests the presence of two conformers in equilibrium at room temperature. However, in the photochemical matrix study, an equilibrium of three conformers was detected. These forms were further characterized by theoretical calculations. Different photolysis products, such as CH3OC(O)SCH(CH3)2, OCS, CO, CO2 and CS2, were identified by matrix spectroscopy. The IR absorptions of CH3OC(O)SCH(CH3)2, for which literature data are scarce, were analysed in the light of the results of appropriate theoretical calculations. The frequency dependence of the freezing temperatureT f(ν) is determined for the dilute spin glass systems (La, Gd)B6 and (Y, Gd)Al2 in the frequency range 10–1,000 Hz. While for (La, Gd)B6,T f(ν) is found to be weak, for (Y, Gd)Al2 T f(ν) is even stronger than for the previously studied system (La, Gd)Al2. Both, measurements of the temperature dependence of the susceptibility nearT f and calculations of the RKKY pair interaction, suggest that this difference is correlated with a different sign of the nearest-neighbor interaction, which appears to be antiferromagnetic for (La, Gd)B6 and ferromagnetic for (Y, Gd)Al2 as well as (La, Gd)Al2.  相似文献

Aromatic hydrocarbons doped with K have been shown to be potential high-temperature superconductors. To investigate the doping effects of trivalent metals(Al, Ga, and In) that have a smaller radii than K, we studied the crystal structure, stability,charge transfer, band structure, and superconductivity of trivalent metal-doped phenanthrene via first-principles calculations.Doping with Al/Ga/In considerably differs from doping with K and cannot be simply regarded as a linear developmental change in the structural and electronic characteristics caused by a change in the valence electron numbers. Al/Ga/In atoms are difficult to dope into the intralayer region, and the charge transfer is close to zero, which is far less than the effect of K doping. We found that the metallization of the Al/Ga/In-doped system originates from the formation of gap states instead of charge transfer. The weak superconductivity obtained in the Al/Ga/In-doped system is also different from the K-doped system. These results are helpful in terms of understanding the structure and superconductivity of metal-doped aromatic superconductors.  相似文献

The family Ariidae comprises approximately 130 catfish species on both warm-temperate and tropical continental shelves around the world. The systematics of the group is problematic, with several misidentification problems. In order to better understand the evolutionary relationships in the family, the present study used a cytogenetic approach to characterize two populations of Genidens genidens and two populations of Aspistor luniscutis from the southern coast of Brazil using conventional techniques and fluorescent in situ hybridization with 18S rDNA probes. The two species had the same diploid number (2n = 56), high fundamental numbers and similar banding patterns, thereby corroborating the karyotypic homogeneity proposed for the group. Single nucleolus organizer regions (NORs) were found in the genus Genidens and multiple NORs were found in Aspistor, which are considered an important cytotaxonomic marker for this genus. Karyotypic evolution trends were hypothesized, providing a better understanding of the karyotype diversity and chromosome evolution processes.  相似文献

The present work deals with the removal of Brilliant Green dye from wastewater using a poly(acrylic acid) hydrogel composite (PAA-K hydrogel) prepared by incorporation of kaoline clay. The composite has been synthesized using ultrasound assisted polymerization process as well as the conventional process, with an objective of showing the better effectiveness of ultrasound assisted synthesis. It has been observed that the hydrogel prepared by ultrasound assisted polymerization process showed better results. The optimum conditions for the removal of dye are pH of 7, temperature of 35 °C, initial dye concentration of 30 mg/L and hydrogel loading of 1 g. The extent of removal of dye increased with an increase in the contact time and initial dye concentration. A pseudo-second-order kinetic model has been developed to explain the adsorption kinetics of dye on the PAA-K hydrogel. Thermodynamic and kinetic parameters indicate that the adsorption process is spontaneous in nature and the PAA-K hydrogel prepared by ultrasound process is a promising adsorbent compared to conventional process. The obtained adsorption data has also been fitted into commonly used adsorption isotherms and it has been found that Freundlich as well as Langmuir adsorption isotherm models fits well to the experimental results.  相似文献

The (3He, t) reaction populating 0+ and 2+ states in 58, 60Cu and 46, 48V which are isobaric analogue states (IAS) of the 0+ ground states and 2+ first excited states in 58, 60Ni and 46, 48Ti have been studied at an incident 3He energy of 24.6 MeV. Triton spectra were measured for the targets 46,48Ti, natNi and 58Ni and angular distributions for the 0+ and 2+ IAS of 46, 48Ti and 58, 60Ni determined. The data were obtained using a magnetic spectrometer and position-sensitive detectors. The results have been analysed using DWBA theory. The 0+ → 0+ transitions to analogue states are described quite well using a microscopic form factor derived from a nucleon-nucleon interaction. However, with a Gaussian form, the m.s. radius of this interaction is only limited to the region 0–9 fm2. Comparisons with data at other incident energies indicate that the strength of the effective interaction is strongly energy dependent. The Coulomb energies and (3He, t) angular distributions of the states assigned as the 2+ analogues in 48V and 58,60Cu are not described well by the models investigated. The 46V 2+ IAS angular distribution is reproduced by a microscopic calculation, however. The ratios of the 0+ → 2+ IAS to the 0+ → 0+ IAS transitions are used to deduce a quadrupole deformation for the valence neutrons. The difference in the quadrupole deformations of the matter and proton distributions, as determined by other means, is found to be correlated with those of the valence neutrons. Several transitions to non-analogue states are also investigated.  相似文献

Independent component analysis (ICA) is an approach for decomposing fMRI data into spatially independent maps and time courses. We have recently proposed a method for ICA of multisubject data; in the current paper, an extension is proposed for allowing ICA group comparisons. This method is applied to data from experiments designed to stimulate visual cortex, motor cortex or both visual and motor cortices. Several intergroup and intragroup metrics are proposed for assessing the utility of the components for comparisons of group ICA data. The proposed method may prove to be useful in answering questions requiring multigroup comparisons when a flexible modeling approach is desired.  相似文献

A new approach to the quantitative elemental analysis of alloys by means of laser-induced breakdown spectroscopy (LIBS) is proposed. The disproportion between the element stoichiometry and spectral intensities is attributed to selective evaporation of components during the heating-melting-evaporation stage. The proposed correction to plasma spectra with account for the Prokhorov-Bunkin melt transparency wave ensures a good agreement between the relative intensities of LIBS analytical lines [(nm): Cu, 511; Zn, 472; Sn, 286; Pb, 406] and the alloy stoichiometry for five samples of four-component bronze measured in various regimes of plasma excitation and signal detection. A criterion is formulated to select the analytical lines, for which the concentration of elements is proportional to the constant of the process and the intensity of the corrected lines.  相似文献

The Gelfand-Graev horospheric approach to the Fourier transform on SO(n,1) is confined to Class I representations; we extend their method to the regular representation of the group SL(2,R). The role of the cone is played by the manifold of 2×2 real singular matrices, upon which the representation and decomposition theory is much simplified. The discrete series is connected with the appearance of associated homogeneous functions on this manifold; in the inversion formula expressing ?(g) in terms of its components this is related to the presence of double poles in the measure of the continuous series. The measurement of Soret coefficients in liquids is not easy and usually not very precise because the resulting concentration gradient is small and moreover can be perturbed by undesired convection currents. In order to suppress, or to drastically reduce these convection currents, the use of a porous medium is sometimes suggested. The question arises as to whether the Soret coefficient is the same in free fluid and in porous medium. This is the aim of this paper. To this end, for a given liquid mixture, the time evolution of the vertical concentration gradient is experimentally measured in the same thermodiffusion cell filled first with the free liquid and next with a porous medium followed by saturation by the liquid mixture. Both the isothermal diffusion (Fick) coefficient and the Soret coefficient can be deduced, providing that a correct working equation is used. The proposed equation results from integration of the general mass conservation equation with realistic boundary conditions (zero mass flux at the boundaries) and some simplifying assumptions rendering this equation more tractable than the one proposed some decades ago by Bierlein (J.A. Bierlein, J. Chem. Phys. 23, 10 (1955)). The method is applied here to an electrolytic solution (CuSO4, 0.25 M) at a mean temperature of 37°C. The Soret coefficients in free and porous medium (zircon microspheres in the range of 250- 315 . 10-6m) may be considered to be equal ( ST = 13.2±0.5 . 10-3 K-1) and the tortuosity factors for the packed medium are the same relative to thermodiffusion and Fick coefficients ( = 1.51±0.02).  相似文献

Laser-activation-modification of semiconductor surfaces (LAMSS) was carried out on silicon with a series of 1-alkenes. These laser spots were studied by time of flight secondary ion mass spectrometry (ToF-SIMS). The resulting spectra were analyzed using the multivariate curve resolution (MCR) method within the Automated eXpert Spectral Image Analysis (AXSIA) toolkit, and also by MCR and cluster analysis using commercially available toolboxes for Matlab: the PLS_Toolbox and the MIA_Toolbox, respectively. AXSIA based MCR generally finds three components for the spectral images: one for the background and two for the laser-activated spots, for both the positive and negative ion images. The negative ion component spectra from the spots show increased carbon and hydrogen signals compared to oxygen. They also show reduced chlorine and fluorine (contamination) peaks. In order to compare AXSIA-MCR results from different images, the AXSIA component spectra of different spots were further analyzed by principal components analysis (PCA). PCA of all of the negative ion components shows that component 1 is chemically distinct from components 2 and 3. PCA of all of the positive ion components yields the same result. The loadings plots of this PCA analysis confirm that component 1 generally contains fragments expected from the substrate, while components 2 and 3 contain fragments expected from an overlayer composed of alkyl chains in the spots. A comparison of the two MCR analyses suggests that roughly the same information can be obtained from AXSIA, which is not commercially available, and the PLS_Toolbox. Cluster analysis of the data also clearly separates the spots from the backgrounds. A key finding from these analyses is that the degree of surface functionalization in a LAMSS spot appears to decrease radially from the center of the spot. Finally, a comparison of atomic force microscopy (AFM) of the spots versus the AXSIA analysis of the ToF-SIMS data produced another important result, which is that the surface morphology is only weakly correlated with the LAMSS chemistry.  相似文献

We present a comparative scanning tunnelling microscopy (STM) study of two features on the Si(0 0 1) surface with a single dangling bond. One feature is the Si-P heterodimer—a single surface phosphorus atom substituted for one Si atom of a Si-Si dimer. The other feature is the Si-Si-H hemihydride—a single hydrogen atom adsorbed to one Si atom of a Si-Si dimer. Previous STM studies of both surface species have reported a nearly identical appearance in STM which has hampered an experimental distinction between them to date. Using voltage-dependent STM we are able to distinguish and identify both heterodimer and hemihydride on the Si(0 0 1) surface. This work is particularly relevant for the fabrication of atomic-scale Si:P devices by STM lithography on the hydrogen terminated Si(0 0 1):H surface, where it is important to monitor the distribution of single P dopants in the surface. Based on the experimental identification, we study the lateral P diffusion out of nanoscale reservoirs prepared by STM lithography.  相似文献
