Plots of the retention factor against mobile phase composition were used to organize a varied group of solutes into three categories according to their retention mechanism on an octadecylsiloxane-bonded silica stationary phase HyPURITY C18 with methanol-water and acetonitrile-water mobile phase compositions containing 10-70% (v/v) organic solvent. The solutes in category 1 could be fit to a general retention model, Eq. (2), and exhibited normal retention behavior for the full composition range. The solutes in category 2 exhibited normal retention behavior at high organic solvent composition with a discontinuity at low organic solvent compositions. The solutes in category 3 exhibited a pronounced step or plateau in the middle region of the retention plots with a retention mechanism similar to category 1 solutes at mobile phase compositions after the discontinuity and a different retention mechanism before the discontinuity. Selecting solutes and appropriate composition ranges from the three categories where a single retention mechanism was operative allowed modeling of the experimental retention factors using the solvation parameter model. These models were then used to predict retention factors for solutes not included in the models. The overwhelming number of residual values [log k (experimental) - log k (model predicted)] were negative and could be explained by contributions from steric repulsion, defined as the inability of the solute to insert itself fully into the stationary phase because of its bulkiness (i.e., volume and/or shape). Steric repulsion is shown to strongly depend on the mobile phase composition and was more significant for mobile phases with a low volume fraction of organic solvent in general and for mobile phases containing methanol rather than acetonitrile. For mobile phases containing less than about 20 % (v/v) organic solvent the mobile phase was unable to completely wet the stationary phase resulting in a significant change in the phase ratio and for acetonitrile (but less so methanol) changes in the solvation environment indicated by a discontinuity in the system maps.  相似文献

Steric repulsion resulted in a systematic reduction in retention compared with predicted values for the fully inserted solute. The bonding density of the stationary phase; the type and composition of the mobile phase; and the size, conformation, type and number of functional groups on the solute are shown to affect the contribution of steric repulsion to the retention mechanism.  相似文献

描述了熊果酸、齐墩果酸色谱保留行为与流动相pH和有机溶剂体积分数之间的函数关系,并求解了各自模型中的9个参数,得到在流动相中熊果酸的电离常数pKa=4.8145,齐墩果酸电离常数pKa=4.9317。文章还对模型外推过程中存在的误差进行了校正,并运用校正后的模型预测熊果酸的容量因子,实验值接近预测值,误差小于5%。最后采用熊果酸和齐墩果酸的容量因子与流动相pH和有机溶剂组成之间的函数关系,进行了两者在同一流动相中分离条件的优化,获得了最佳的分离条件:流动相中甲醇体积分数为85.2%,pH为6.50。  相似文献

This paper describes the results of the evaluation of an alternative solvation parameter model for ionizable compounds. The new model is described as Log(k) = Int + rR2 + spi2(H) + asigmaalpha2(H) + bsigmabeta2(H) + mVx + U/(1 + V10 (+/-(pH-Pk))). The first six terms are the usual solvation parameter equation for neutral solutes, and the last term represents the contribution to retention from the ionization of solutes. Retention data obtained for 30 solutes in acetonitrile/aqueous buffer mobile phases are used to evaluate the capability of the function using different pH/pK scales. Because the function is not linear, nonlinear least-squares analysis is used to perform the data processing. It is concluded that the model function describes similarly the retention of ionizable compounds to the literature model without the need to accurately measure the mobile phase pH and solute's pK. Accordingly, the function simplifies the application of linear solvation energy relationships (LSERs) to ionizable compounds, and allows us to easily predict their retention for chromatographic optimization.  相似文献

The aim of this work is to optimise the proportion of the organic modifier and the pH of the mobile phase, in order to separate a series of peptide hormones with therapeutic interest in the molecular mass range from 500 to 6000. The composition of the mobile phase was optimised by establishing relationships between retention parameters and either the scale of solvent polarity, or the Kamlet–Taft multiparameter solvent scale of the eluent, using linear solvation energy relationships. Likewise, linear correlations between the chromatographic retention and Reichardt’s ENT parameter were obtained. These relationships allowed an important reduction of the experimental retention data needed for developing a given separation. In addition, a model describing the effect of the correctly measured pH of the mobile phase on retention in LC was established and tested for the series of selected peptides using an octadecylsilica column. The proposed equations permit the prediction of the optimum pH and also permit the determination of the acidity constants of the peptides in the hydro-organic mixtures using a minimum number of measurements.  相似文献

Summary The solvation parameter model is used to characterize the retention properties of a cyanopropylsiloxanebonded, silica-based sorbent with methanol, acetonitrile, tetrahydrofuran, and isopropanol in water as mobile phases. The system constants over the composition range 1 to 50 % (v/v) organic solvent indicate that retention occurs because of the relative ease of cavity formation in the solvated stationary phase compared to the same process in the predominantly aqueous mobile phase as well as from more favorable stationary phase interactions with solutes containing π- and n-electrons. The capacity of the solute for dipole-type interactions is not important whereas all hydrogen-bond-type interactions result in reduced retention. Graphing the system constants as a function of mobile phase composition provides a simple mechanism for interpreting the change in capacity of the chromatographic system for retention in terms of changes in the relative weighting of fundamental intermolecular interactions. A comparison is also made with the retention properties of an octadecylsiloxane-bonded, silica-based sorbent with 30 % (v/v) methanol in water as the mobile phase and the extraction characteristics of a porous polymer sorbent with 1 % (v/v) methanol, acetonitrile, tetrahydrofuran, and isopropanol in water as the sample processing solvent. Changes in sorbent selectivity due to selective uptake of the processing solvent are much smaller for the cyanopropylsiloxane-bonded sorbent than the results found for a porous polymer sorbent.  相似文献

An extension of the treatment adopted in a recent paper [P. Nikitas, A. Pappa-Louisi, P. Agrafiotou, J. Chromatogr. A 946 (2002) 33] was used to derive expressions describing the variation of solute retention k with composition in ternary reversed phase liquid chromatography, RP-LC, solvent systems. The equation of the partition model obtained in this way for a ternary mobile phase was identical to that previously derived using the solubility parameter concept. This equation as well as two new expressions of In k versus organic modifiers content were tested in a variety of ternary solvent systems in order to examine the possibility of predicting retention behavior of solutes under ternary solvent mixture elution conditions from known retention characteristics in binary mobile phases. It was demonstrated the superiority of both new equations derived in this paper to that previously proposed and applied to date in ternary solvent mixtures.  相似文献

Summary The chromatographic properties of an alkylphosphonate-modified magnesia-zirconia composite stationary phase have been investigated by reversed-phase high-performance liquid chromatography with basic compounds as probes. The influence of organic modifier composition and mobile phase pH was studied. The new stationary phase, similar to a silica-based reversed-phase stationary phase, has hydrophobic properties, but greater pH stability. Use of the phase results in more symmetric peaks for basic compounds. A possible mechanism of retention of basic solutes on the new stationary phase is discussed. The chromatographic behavior of the basic solutes depends mainly on hydrophobic interactions between the solutes and the hydrophobic moiety of the stationary phase. Br?nsted acidic and basic sites on the surface of the new stationary phase play an important role in the retention of ionized solutes by ion-exchange interaction. Promising separations of some basic compounds have been achieved by use of methanolic TRIS buffer, pH 10.0, as the mobile phase.  相似文献

The influence of pH and solvent composition of acetonitrile-water mobile phases on the retention of acids and bases on a polymeric stationary phase is studied. Very good relationships between retention and mobile phase pH are obtained if the pH is measured in the proper pH scale. The fit of retention to pH for a particular solvent composition provides the pKa values of the equilibria between the different acid-base species and the retention parameters of these species at this solvent composition. Several models are tested that relate these parameters to solvent composition and properties in order to propose a general model to predict retention for any mobile phase pH and composition.  相似文献

The influence of temperature on the retention behavior of epirubicin and its analogues on high purity silica with reversed-phase solvents has been systematically investigated. It was found that temperature effects on retention are highly dependent on the type and concentration of organic modifier, as well as the pH of the mobile phase. In organic-rich mobile phases, the type of organic modifier plays an important role. With an aprotic solvent as modifier, retention times show anomalous increases with elevated temperature. At the same time, both efficiency and resolution are significantly improved but this is not the situation with a protic solvent as modifier. In addition, temperature shows different effects on retention time and selectivity when the pH is changed, and temperature-dependent selectivity reversal is found at higher pHs. In aqueous-rich mobile phases, regardless of the nature of the organic solvent and pH, retention of solutes drops as temperature is raised. It seems that the effect of temperature on chromatographic behavior of the solutes on bare silica using mobile phases containing various organic modifiers or pHs, results from a number of different retention mechanisms.  相似文献

A novel approach is proposed for the simultaneous optimization of mobile phase pH and gradient steepness in RP‐HPLC using artificial neural networks. By presetting the initial and final concentration of the organic solvent, a limited number of experiments with different gradient time and pH value of mobile phase are arranged in the two‐dimensional space of mobile phase parameters. The retention behavior of each solute is modeled using an individual artificial neural network. An “early stopping” strategy is adopted to ensure the predicting capability of neural networks. The trained neural networks can be used to predict the retention time of solutes under arbitrary mobile phase conditions in the optimization region. Finally, the optimal separation conditions can be found according to a global resolution function. The effectiveness of this method is validated by optimization of separation conditions for amino acids derivatised by a new fluorescent reagent.  相似文献

The isocratic retention of 67 widely-different solutes in reversed-phase liquid chromatography (RP-LC) has been investigated as a function of temperature and mobile phase composition (% B) for three different C18 columns. Similar studies were also carried out in a gradient mode, where temperature, gradient time and solvent type were varied. These results show that changes in retention with these conditions are similar for each of these three columns. This suggests that relative column selectivity as defined by experiments for one set of experimental conditions will be approximately applicable for other conditions, with the exception of changes in mobile phase pH-which can affect values of the column parameter C (a measure of silanol ionization). Column selectivity as a function of pH was explored for several columns.  相似文献

Molecular simulations of water/acetonitrile and water/methanol mobile phases in contact with a C(18) stationary phase were carried out to examine the molecular-level effects of mobile phase composition on structure and retention in reversed-phase liquid chromatography. The simulations indicate that increases in the fraction of organic modifier increase the amount of solvent penetration into the stationary phase and that this intercalated solvent increases chain alignment. This effect is slightly more apparent for acetonitrile containing solvents. The retention mechanism of alkane solutes showed contributions from both partitioning and adsorption. Despite changes in chain structure and solvation, the molecular mechanism of retention for alkane solutes was not affected by solvent composition. The mechanism of retention for alcohol solutes was primarily adsorption at the interface between the mobile and stationary phase, but there were also contributions from interactions with surface silanols. The interaction between the solute and surface silanols become very important at high concentrations of acetonitrile.  相似文献

Summary The influence of acetonitrile, methanol and isopropanol as retention selectivily modifiers in reversed phase liquid chromatography on a poly(styrene-divinylbenzene) macroporous polymer sorbent (PLRP-S) is evaluated using the solvation parameter model. Retention results from a combination of adsorption and partitioning and is influenced by the equilibrium absorption of organic solvent by the polymer from the mobile phase. The sorption of solutes is dominated by the ease of cavity formation in or on the solvated sorbent, with a small contribution from lone pair-lone pair electron interactions. All polar interactions, such as dipole-type and hydrogenbond formation, are more favorable in the mobile phase and reduce retention. Changes in the uptake of organic solvent from the mobile phase affect kinetic properties of the column such as band broadening and porosity as well as retention. The PLRP-S solvated sorbent is suitable for solid-phase extraction and is more retentive than typical silica-based, bonded phase sorbents for extraction from water. As a surrogate system for estimating solute lipophilicity and biological activity through retention-property correlations it provides a poor fit for hydrogen-bond acid solutes and is too dipolar/polarizable to fit some models.  相似文献

Peptides, quinolones, diuretics and peptide hormones are important substances of biomedical interest. In this work a model describing the effect of pH on retention in liquid chromatography (LC) is established and tested for these compounds using an octadecylsilica column. The suggested model uses the pH value measured in the hydro-organic mixture used as mobile phase instead of the pH value in water and takes into account the effect of the activity coefficients. The proposed equations permit the prediction of the pH optimum using a minimum number of measurements and also permit the determination of the acidity constants of the compounds considered in the medium used as mobile phase. Moreover, these equations can be combined with the previously derived equations, that relate the retention with the solvent composition of the mobile phase, to establish a general model that relates the elution behaviour of the solute with the significant mobile phase properties: composition, pH and ionic strength.  相似文献
