The development of a rapid method for the determination of perchlorate in rain and drinking waters is presented. In the optimised method, an on-line preconcentration technique was employed utilising a 10 mm × 4.6 mm Phenomenex Onyx monolithic guard cartridge coated with (N-dodecyl-N,N-dimethylammonio)undecanoate for selective preconcentration, with subsequent elution into a fixed volume injection loop (‘heart-cut’ of the concentrator column eluate) and separation using an IonPac AS16 (250 mm × 2 mm) anion exchange column and a potassium hydroxide concentration gradient. Off-line optimisation studies showed that the coated monolith displayed near quantitative recovery up to 50 μg/L perchlorate level from standards prepared in reagent water. On-line preconcentration of perchlorate obtained detection limits down to 56 ng/L in reagent water, between 70 and 80 ng/L in rainwater samples and 2.5 μg/L in non-pretreated drinking water. After an additional sample sulphate/carbonate removal step, low ng/L perchlorate concentrations could also be observed in drinking water. The complete on-line method exhibited reproducibility for n = 10 replicate runs of R.S.D. ≤ 3% for peak height/area and R.S.D. = 0.08% for retention time. The optimised method, of 20 min total duration, was applied to the determination of perchlorate by standard addition in 10 rainwater samples and one drinking water sample. Concentrations of perchlorate present ranged from below the detection limit for four rainwater samples, with another three samples showing perchlorate present at between 70 and 100 ng/L, and one sample showing perchlorate present at 2.8 μg/L. Levels of 1.1 μg/L in the drinking water sample were also recorded.  相似文献

Mutagenic and carcinogenic heterocyclic amines (HCAs) are formed during heating of various proteinaceous foods, but human exposure to HCAs has not yet been elucidated in detail. To assess long-term exposure to HCAs, we developed a simple and sensitive method for measuring HCAs in hair by automated on-line in-tube solid-phase microextraction (SPME) coupled with liquid chromatography–tandem mass spectrometry (LC–MS/MS). Using a Zorbax Eclipse XDB-C8 column, 16 HCAs were analyzed within 15 min. The optimum in-tube SPME conditions were 20 draw/eject cycles of 40 μL sample at a flow rate of 200 μL min−1 using a Supel-Q PLOT capillary column as an extraction device. The extracted HCAs were easily desorbed from the column by passage of the mobile phase, with no carryover observed. This in-tube SPME LC–MS/MS method showed good linearity for HCAs in the range of 10–2000 pg mL−1, with correlation coefficients above 0.9989 (n = 18), using stable isotope-labeled HCA internal standards. The detection limits (S/N = 3) of 14 HCAs except for MeAαC and Glu-P-1 were 0.10–0.79 pg mL−1. This method was successfully utilized to analyze 14 HCAs in hair samples without any interference peaks, with quantitative limits (S/N = 10) of about 0.17–1.32 pg mg−1 hair. Using this method, we evaluated the exposure to HCAs in cigarette smoke and the suitability of using hair HCAs as exposure biomarkers.  相似文献

A new method for sensitive and fast screening of melamine (MEL) in milk products was developed with a low-cost disposable microfluidic device coupled with ultraviolet (UV) detection. This method avoided the need of sample pretreatment prior to the separation process, thus was simple and green. Due to the advantages of the device and fracture sampling technique, milk sample could be directly sampled through the fracture to achieve baseline separation from amino acids, and proteins in the sample did not interfere with the detection. Using 20 mM phosphate running buffer (pH 9.0), a sampling time of 3 s at +180 V and a separation voltage of +1800 V (240 V/cm), this method could detect MEL in milk within 75 s. At the detection wavelength of 202 nm, the linear range for MEL was from 1.0 to 100 μg mL−1 with a detection limit of 0.23 μg mL−1 (S/N = 3). The novel protocol had been successfully used to screen MEL in milk samples with recovery more than 82%. The environmentally friendly methodology for pretreatment-free sensitive screening of MEL provided promising applications in monitoring the quality of different foods.  相似文献

A method of hollow fiber (HF) liquid phase microextraction (LPME) combined with gas chromatography (GC)-flame ionization detection (FID) was developed for the simultaneous quantification of trace amphetamine (AP), methamphetamine (MA), methylenedioxyamphetamine (MDA), methylenedioxymethamphetamine (MDMA), caffeine and ketamine (KT) in drug abuser urine samples. The factors affecting on the extraction of six target analytes by HF-LPME were investigated and optimized, and the subsequent analytical performance evaluation and real sample analysis were performed by the extraction of six target analytes in sample solution containing 30% NaCl (pH 12.5) for 20 min with extraction temperature of 30 °C and stirring rate of 1000 rpm. Under such optimal conditions, the limits of detection (LODs, S/N = 3) for the six target analytes were ranged from 8 μg/L (AP, KT) to 82 μg/L (MDA), with the enrichment factors (EFs) of 5-227 folds, and the relative standard deviations (RSDs, n = 7) were in the range of 6.9-14.1%. The correlation coefficients of the calibration for the six target analytes over the dynamic linear range were higher than 0.9958. The application feasibility of HF-LPME-GC-FID in illegal drug monitoring was demonstrated by analyzing drug abuser urine samples, and the recoveries of target drugs for the spiked sample ranging from 75.2% to 119.3% indicated an excellent anti-interference capability of the developed method. The proposed method is simple, effective, sensitive and low-cost, and provides a much more accurate and sensitive detection platform over the conventional analytical techniques (such as immunological assay) for drug abuse analysis.  相似文献

We describe a sensitive and rapid method to assay urinary cotinine levels among non-smokers using liquid chromatography-electrospray ionization tandem mass spectrometry (LC-ESI/MS/MS) and its application in studies assessing exposures to second hand smoke (SHS). Cotinine was initially extracted from 1 ml of urine with methylene chloride by using a liquid-liquid extraction Chem Elut™ column. The extracted sample was further separated by using a BetaBasic C18 column (1 mm × 150 mm, 3 μm) with isocratic elution (60:40 acetonitrile and 5 mM ammonium acetate at pH 5), and then examined using a triple quadrupole mass spectrometer with an electrospray ionization (ESI) source in multiple-reaction-monitoring (MRM) mode. The elution of cotinine from the LC column took approximately 2.3 min and the detection of cotinine by ESI/MS/MS provided a limit of detection (LOD) of 0.3 ng/ml. The ESI/MS/MS detection was able to easily distinguish between cotinine and nicotine. This method, validated using a cotinine concentration range from 0.8 to 102.4 ng/ml, was successfully applied in a cross-sectional study examining differences in levels and sources of second hand smoke (SHS) exposure among non-smokers. Self-reported measures of SHS exposure were significantly associated with urinary cotinine levels. This urinary cotinine assay using LC-ESI/MS/MS provides a robust, high throughput and very sensitive method for the evaluation of SHS exposure for use in epidemiologic and clinical research studies.  相似文献

The radium-226 (t1/2 = 1622 years) content of highly alkaline well water collected from the United Arab Emirates (UAE) was measured by double focusing sector-field inductively coupled plasma-mass spectrometry (SF-ICP-MS) after separation of the radium from other alkaline earth elements using a newly developed procedure. The results were comparable with those obtained by α-spectrometry for samples with concentrations ranging from 6.75 to 459 pg/L (0.25 to 17 Bq/L). Instrumental sensitivity on matrix-free samples was compared for two sample introduction systems, i.e. an Apex-Q high sensitivity system and a concentric nebulizer. A 12-fold improvement in sensitivity (instrumental detection limit = 1.5 pg/L or 55 mBq/L) was found when the Apex-Q system was used. Two chromatographic methods were tested for the sequential separation of the alkaline earth elements contained in the well water samples in order to reduce matrix and polyatomic interference effects. Optimal elution parameters were determined and used for the separation and pre-concentration of Ra-226 in those samples. A method detection limit of 0.189 pg/L (7 mBq/L), which corresponds to a mass of 0.38 fg of Ra-226 in the sample, was achieved. Only 2 mL of sample is necessary when a combination of 50 W-X8 and Sr*Spec resin, which are reusable, are utilized for the separation. This new analytical protocol significantly reduces sample preparation time resulting in a throughput rate of approximately 20 samples in only 8 h; faster than the other published extraction procedures.  相似文献

Liquid–liquid–liquid microextraction (LLLME) with directly suspended droplet in high-performance liquid chromatography (HPLC) has been applied as a new, rapid and easy method for the determination of 3-nitroaniline in environmental water samples. The target compound was extracted from the aqueous sample solution (donor phase, pH 13) into an organic phase and then was back-extracted into a directly suspended droplet of an acidic aqueous solution (acceptor phase, pH 0.3). In this method, without using a microsyringe as supporting device, an aqueous large droplet is freely suspended at the top-center position of an immiscible organic solvent, which is laid over the aqueous sample solution while being agitated. Then, the droplet was withdrawn into the microsyringe and directly was injected into the HPLC system with UV detection at 227 nm. Up to 148-fold enrichment of the analyte could be obtained under the optimal conditions [i.e. donor phase: 0.1 M sodium hydroxide solution (4.5 mL); organic phase: o-xylene/1-octanol (90:10, v/v; 250 μL); acceptor phase: 0.5 M hydrochloric acid and 500 mM 18-crown-6 ether (6 μL); extraction time: 60 s; back-extraction time: 6 min and stirring rate: 600 rpm]. The limit of detection was 1 μg/L (n = 7) and the relative standard deviation (RSD, n = 5) was 4.9 at S/N = 3. The calibration graph was linear in the range of 5–1500 μg/L with r = 0.9983. All experiments were carried out at room temperature (22 ± 0.5 °C).  相似文献

A new approach using single-drop microextraction (SDME) and gas chromatography–mass spectrometry for the determination of six haloacetic acids (HAAs) in water samples was presented. n-Octanol was used as extractant and derivatization reagent. HAAs were derivatized both simultaneously during the extraction in the solvent microdrop, and after extraction, inside a glass microvial (1.1 mm I.D.). Trifluoroacetic anhydride (TFAA) was used as the reaction catalyst. The influence of catalyst amount, derivatization time and temperature on the yield of the in-microvial derivatization was investigated. Derivatization reaction was performed using 1.2 μL of TFAA at 100 °C for 20 min. Extraction was performed using 1.8 μL of n-octanol containing TFAA (10%, v/v). Experimental parameters, such as, exposure time, sample pH and extraction temperature were controlled and optimized. Analytical parameters such as linearity, precision and limit of detection were also evaluated. The proposed method was proved to be a suitable analytical procedure for HAAs in water with limits of detection 0.1–1.2 μg/L. The relative recoveries range from 82.5 to 97.6% for all the target analytes. Precision values were from 5.1 to 8.5% (as intra-day relative standard deviation, RSD) and 8.8–12.3% (as inter-day RSD).  相似文献

We developed a sensitive and useful method for the determination of five fluoroquinolones (FQs), enoxacin, ofloxacin, ciprofloxacin, norfloxacin, and lomefloxacin in environmental waters, using a fully automated method consisting of in-tube solid-phase microextraction (SPME) coupled with liquid chromatography-tandem mass spectrometry (LC/MS/MS). These compounds were analysed within 7 min by high-performance liquid chromatography (HPLC) using a CAPCELL PAK C8 column and aqueous ammonium formate (pH 3.0, 5 mM)/acetonitrile (85/15, v/v) at a flow rate of 0.2 mL/min. Electrospray ionization conditions in the positive ion mode were optimized for MS/MS detection. In order to optimize the extraction of FQs, several in-tube SPME parameters were examined. The optimum in-tube SPME conditions were 20 draw/eject cycles of 40 μL of sample at a flow-rate of 150 μL/min, using a Carboxen 1010 PLOT capillary column as an extraction device. The extracted compounds were easily desorbed from the capillary by passage of the mobile phase. Using the in-tube SPME LC/MS/MS method, good linearity of the calibration curve (r ≥ 0.997) was obtained in the concentration range from 0.1 to 10 ng/mL for all compounds examined. The limits of detection (S/N = 3) of the five FQs ranged from 7 to 29 pg/mL. The in-tube SPME method showed 60-94-fold higher sensitivity than the direct injection method (5 μL injection). This method was applied successfully to the analysis of environmental water samples without any other pretreatment and interference peaks. Several surface waters and wastewaters were collected from the area around Asahi River, and ofloxacin was detected in wastewater samples of a sewage treatment plant and other two hospitals at 17.5-186.2 pg/mL. The recoveries of FQs spiked into river water were above 81% for a 0.1 or 0.2 ng/mL spiking concentration, and the relative standard deviations were below 1.9-8.6%.  相似文献

A fast, economic and sensitive chemiluminescence (CL) method has been developed for the analysis of cetrizine hydrochloride (CET) in pharmaceutical formulations and in biological fluids. The CL method is based on the oxidation of tris(2,2′-bipyridyl)ruthenium(II) (Ru (bipy)32+) by peroxydisulphate in a two-chip device. Up to 180 samples can be analysed per hour, consuming only minute quantities of reagents. Three instrumental setups were tested to find the most economical, sensitive and high throughput setup. In the first setup, a continuous flow of sample and CL reagents was used, whereas in the second setup, a fixed volume (2 μL) of (Ru (bipy)32+) was introduced into a continuous infusion of peroxydisulphate and the sample. In the third design, a fixed volume of sample (2 μL) was injected while the CL reagents were continuously infused. Compared to the first setup, a 200% signal enhancement was observed in the third setup. Various parameters that influence the CL signal intensity, including pH, flow rates and reagent concentrations, were optimized. A linear response was observed over the range of 50 μg L−1 to 6400 μg L−1 (R2 = 0.9959) with RSD values of 1.1% (n = 15) for 1000 μg L−1. The detection limit was found to be 15 μg L−1 (S/N = 3). The amount of consumed sample was only 2 μL, from which the detected amount of CET was found to be 6.5 × 10−14 mol. This procedure was successfully applied to the analysis of CET in pharmaceutical formulations and biological fluids.  相似文献

A simple, sensitive LC-ESI-MS method was optimized for quantitative analysis of octahydro-1,3,5,7-tetranitro-1,3,5,7-tetrazocine (HMX) in environmental samples. Under negative ionization mode, HMX can form adduct ions with various organic acids and salts, including acetic acid, formic acid, propionic acid, ammonium nitrate, ammonium chloride, sodium nitrite, and sodium nitrate. Acetic acid was chosen as additive and the ion, [M + CH3COO] with m/z = 355 was used for selective ion monitoring (SIM) in this study. Good sensitivity was achieved with low acetic acid concentration in the mobile phase and relatively low capillary temperature. The method detection limit was 0.78 pg for HMX in standard solution. Linearity (R2 > 0.9998) was obtained at low concentrations (0.5-50 μg/L). This method has been used to determine HMX concentrations in water samples and lizard egg samples from an animal exposure study.  相似文献

A flow injection method has been developed for the determination of uranium in seawater combining the on-line preconcentration with spectrophotometric detection. An aliquot (10 mL) of the seawater sample adjusted to pH 5.5 was injected into the analytical system and uranium was adsorbed on the column packed with styrene-divinylbenzene copolymer resin (Bio-Beads SM-2) modified with dodecylamidoxime which showed high selectivity to uranium. Uranium was then eluted with 0.01 M hydrochloric acid and detected spectrophotometrically after the reaction with Chlorophosphonazo III. Interference from calcium and strontium was masked with cyclohexanediaminetetraacetic acid added to the chromogenic reagent solution. The sample throughput, the detection limit (3σ), and the preconcentration factor were 23 per hour, 0.13 μg/L, and 20, respectively, when the sample injection volume was kept at 10 mL. The precision at the 2 μg/L level was less than 4% (RSD). The proposed method was applied to the determination of uranium in the seawater samples collected off the Boso peninsula, Japan and the uranium concentration was found to be ca. 3 μg/L, which is close to the literature data. The yield of the recovery test ranged from 95% to 99%.  相似文献

A rapid, sensitive and efficient liquid phase microextraction (LPME) method was developed to determine trace concentrations of some organophosphorus pesticides in water samples. This method combines liquid phase microextraction with gas chromatographic (GC) analysis in a simple and inexpensive apparatus involving very little organic solvent consumption. It involves exposing a floated drop of an organic solvent on the surface of aqueous solution in a sealed vial. Experimental parameters which control the performance of LPME such as type of organic solvent, organic solvent and sample volumes, sample stirring rate, sample solution temperature, salt addition and exposure time were investigated and optimized. Finally, the enrichment factor, dynamic linear range (DLR), limit of detection (LOD) and precision of the method were evaluated by the water samples spiked with organophosphorus pesticides. Using optimum extraction conditions, very low detection limits (0.01-0.04 μg L−1) and good linearities (0.9983 < r2 < 0.9999) were achieved. The LPME was performed for determination of organophosphorus pesticides in different types of natural water samples and acceptable recoveries (96-104%) and precisions (3.5 < R.S.D.% < 8.9) were obtained. The results suggested that the newly proposed LPME method is a rapid, accurate and effective sample preparation method and could be successfully applied for extraction and determination of organophosphorus pesticides in water samples.  相似文献

The paper describes a homemade ultrasonic microdialysis device coupled with capillary electrophoresis electrochemiluminescence (CE-ECL) for studying the interaction between human serum albumin (HSA) and trimetazidine dihydrochloride (TMZ). The time required for equilibrium by ultrasonic microdialysis was 45 min, which was far less than that by traditional dialysis (240 min). It took 80 min to achieve the required combination equilibrium by normal incubation and only 20 min by ultrasonic. Compared with traditional dialysis, the use of ultrasonic microdialysis simplified experimental procedures, shortened experimental time and saved consumption of sample. A simple, sensitive and selective determination of TMZ was developed using CE-ECL and the parameters that affected ECL intensity were optimized. Under the optimized conditions, the linear range of TMZ was from 0.075 to 80 μmol/L (r2 = 0.9974). The detection limit was 26 nmol/L with RSD of 2.8%. The number of binding sites and binding constant were 1.54 and 15.17 L/mol, respectively.  相似文献

A novel method for analysis of essential oil in Cuminum cyminum L. using simultaneous ultrasonic nebulization extraction and headspace single drop microextraction (UNE-HS-SDME) followed by gas chromatography-mass spectrometry was developed. Experimental parameters, including the kind of suspended solvent, microdrop volume, sample amount, extraction time, enrichment time and salt concentration were examined and optimized. Compared with hydrodistillation (HD), UNE-HS-SDME provides the advantages of a small amount of sample (50 mg), time-saving (20 min), simplicity, cheapness and low toxicity. In addition, UNE-HS-SDME also provided higher enrichment efficiency and sensitivity compared with stirring extraction (SE)-HS-SDME, ultrasonic assistant extraction (UAE) and UNE. Some constituents in the essential oil, were identified and the detection limits for β-pinene, p-cymene and γ-terpinene range from 6.67 pL L−1 to 14.8 pL L−1. The results indicated that the UNE-HS-SDME is simple and highly efficient extraction and enrichment technique.  相似文献

A simple and sensitive liquid chromatographic method is described for the analysis of γ-amino-n-butyric acid (GABA) in human urine. GABA is increased in the urine of cancer patients and could be used as a biomarker in the diagnosis and treatment of related patients. The method is based on derivatizing GABA with a fluorescent reagent (naproxen acyl chloride) for transforming the non-chromophoric GABA to a derivative with chromophoric and fluorophoric properties. The resulting derivative is highly responsive to a fluorimetric detector (λex = 230 nm, λem = 350 nm). The lower quantitation of the method is attainable at 100 nM GABA with a detection limit about 10 nM (S/N = 3 with 20 μL injected). Application of the method to the analysis of GABA in the urine of patients with ovarian and uterine cancer was demonstrated.  相似文献

A simple and sensitive method for measuring airborne particle-associated benz[a]anthracene-7,12-quinone (BaAQ) based on two-dimensional high-performance liquid chromatography (HPLC) with fluorescence detection was established. The system involves an ODS column for sample clean up, a 6-port switching valve, an ODS column for trapping the fraction containing BaAQ, an ODS column for sample separation, and a Pt-Rh catalytic column for reduction of BaAQ to a corresponding fluorescent compound. The accuracy of the assay, as applied to airborne particulate sample extracts spiked with known amounts of BaAQ was 94–108%. The detection limit was 97 fmol per injection (signal-to-noise ratio = 3), and the calibration range was from 1 to 100 pmol with excellent proportionality (R2 = 0.9995). BaAQ in airborne particles collected in Osaka, Japan every 3 h was successfully measured using the established analytical method with simple sample preparation steps: ultrasonic extraction in organic solvent and concentration under reduced pressure and/or nitrogen stream.  相似文献

A chemiluminescence one-shot sensor for hydrogen peroxide is described. It is prepared by immobilization of cobalt chloride and sodium lauryl sulphate in hydroxyethyl cellulose matrix cast on a microscope cover glass. Luminol, sodium phosphate and the sample are mixed before use and applied on the membrane by a micropipette. The calibration graph is linear in the range 20-1600 μg/L, and the detection limit of the method (3σ) is 9 μg/L. A relative standard deviation of 4.5% was obtained for 100 μg/L H2O2 (n = 11). The sensor has been applied successfully to the determination of hydrogen peroxide in rainwater.  相似文献

An electrochemical DNA detection method for the phosphinothricin acetyltransferase (PAT) gene sequence from the transgenetic plants was established by using a microplate hybridization assay with cadmium sulfide (CdS) nanoparticles as oligonucleotides label. The experiment included the following procedures. Firstly target PAT ssDNA sequences were immobilized on the polystyrene microplate by physical adsorption. Then CdS nanoparticle labeled oligonucleotide probes were added into the microplate and the hybridization reaction with target ssDNA sequences took place in the microplate. After washing the microplate for three times, certain amounts of HNO3 were added into the microplate to dissolve the CdS nanoparticles anchored on the hybrids and a solution containing Cd2+ ion was obtained. At last differential pulse anodic stripping voltammetry (DPASV) was used for the sensitive detection of released Cd2+ ion. Based on this principle a sensitive electrochemical method for the PAT gene sequences detection was established. The voltammetric currents of Cd2+ were in linear range with the target ssDNA concentration from 5.0 × 10− 13 to 1.0 × 10− 10 mol/L and the detection limit was estimated to be 8.9 × 10− 14 mol/L (3σ). The proposed method showed a good promise for the sensitive detection of specific gene sequences with good selectivity for the discrimination of the mismatched sequences.  相似文献

Laser-induced fluorescence (LIF) is a highly sensitive detection method for capillary electrophoresis (CE). However, it usually requires analyte to be derivatized, unless the wavelength of native fluorescence of analyte matches the laser's. That limits its application in drug analysis. In this work, we introduced a rapid, simple and sensitive method of nonaqueous capillary electrophoresis with laser-induced native fluorescence (NACE-LIF) detection for the analysis of chelerythrine and sanguinarine for the first time. As these two alkaloids have some native fluorescence, they were directly detected using a commercially available Ar+ laser without troublesome fluorescent derivatization. The fluorescence was enhanced by nonaqueous media. Compared with previously reported UV detection method, lower limit of detection (LOD) is achieved thanks to the high sensitivity of LIF detection (2.0 ng/mL for chelerythrine and 6.3 ng/mL for sanguinarine). Moreover, with NACE, the baseline separation of these alkaloids is finished within 3.5 min. This method is successfully applied to determine the contents of chelerythrine and sanguinarine in Macleaya cordata (Willd.) R. Br. and Chelidonium majus L.  相似文献
