Chemical force titrations-measurements of the adhesive interaction between a pair of suitably chemically modified atomic force microscopy (AFM) tip and sample surfaces as a function of pH-have been carried out for various combinations of silanol, amine, carboxylic acid, and sulfonic acid functional groups on both tip and sample. The primary surface material studied was poly(dimethylsiloxane) (PDMS). Surface modification was carried out using a plasma oxidation process to form silanol sites; further modification with amine or sulfonic acid sites was carried out by reaction of the silanol sites with the appropriate trialkoxysilane derivative. AFM tips were also modified using trialkoxysilane compounds. In the cases of tip/sample combinations with the same functional group on each, surface pK(1/2) values could be determined. In several "mixed" tip/sample combinations, a peak appeared in the titration curve midway between the surface pK(1/2) values of the tip and sample, consistent with an ionic H-bonding model for the interactions. The amine/sulfonic acid pair showed more complex behavior; the amine-terminated tip/sulfonic acid-terminated PDMS surface force titration curve consisted of two peaks centered at pH 4 and pH 8. Reversing the tip/sample pair resulted in the peak positions being shifted upward by 1.0 pH unit. The peak appearing at lower pH is assigned to electrostatic interactions between the two oppositely charged surfaces, whereas the higher pH peak is believed to arise due to ionic H-bonding interactions. AFM images show the effects on surface patterning of amine- and sulfonic acid-modified PDMS surfaces that have undergone two different oxidation methods (air plasma oxidation and Tesla coil oxidation). The surface morphologies of freshly prepared and 24 h aged air plasma oxidized PDMS are also discussed in this study.  相似文献

The concept of force titration was firstly proposed based on the technique of Chemical Force Microscopy (CFM). Self-Assembled Monolayer (SAM) on substrate surface can be titrated with buffer solutions at a nanometer scale by measuring the adhesion force between the SAM-modified substrate and probe tip. The plot of adhesion force vs pH value was termed as force titration curve. As an example, the titration behavior of w-mercaptoundecanoic acid monolayer on gold has been studied. It was found that there is a big hump around pH 5~6 in its force titration curve. Taking the contact angle titration result together, an interaction model for the monolayer was suggested from the chemical hysteresis point of view.  相似文献

Poly(methylmethacrylate) (PMMA) is a versatile polymer that displays desirable properties for development of cheap and disposable microfluidic devices for sensing biomolecular interactions. Atomic force microscopy (AFM) and chemical force titrations were used to determine the efficacy of surface modifications made to accommodate protein-substrate linkage. AFM images show the effects on surface morphology of carboxylated-, amine-, hCG antigen- and anti-hCG antibody-modified PMMA substrates. Confocal microscopy was used to determine the fluorescent intensity of labeled antibody species on the PMMA substrate, confirming the success of surface antigen/antibody immobilization. Surface pK(1/2) value for carboxylic acid and amine species grafted on PMMA were determined. When carboxylic acid or amine-terminated tips were titrated against PMMA samples terminated with the hCG antigen and anti-hCG antibody, peaks appeared in the force titration curve consistent with the pI range of the antigen or antibody species. Strong adhesive forces were present at pH values above 7.0 when the antigen was present on the PMMA substrate, and these were attributed to hydrophobic interactions between the antigen and the alkane "linker" chain attaching the amine or carboxylate group to the AFM tip. Such hydrophobic interactions were not observed with the carboxylic acid or amine/antibody combinations suggesting that the surface-linked antibody was more resistant to denaturation under higher pH. The results demonstrated the feasibility of using AFM approaches for interrogating protein grafting strategies in the fabrication of PMMA-based microsystems.  相似文献

Mesoporous and nonporous SiO(2) and Al(2)O(3) adsorbents were reacted with the fluoroquinolone carboxylic acid ofloxacin over a range of pH values (2-10) and initial concentrations (0.03-8 mM) to investigate the effects of adsorbent type and intraparticle mesopores on adsorption/desorption. Maximum ofloxacin adsorption to SiO(2) surfaces occurs slightly below the pK(a2) (pH 8.28) of the antibiotic and sorption diminishes rapidly at pH>pK(a2). For Al(2)O(3), maximum sorption is observed at pH values slightly higher than the adsorbent's point of zero net charge (p.z.n.c.) and less than midway between the pK(a) values of ofloxacin. The effects of pH on adsorption and ATR-FTIR spectra suggest that the zwitterionic compound adsorbs to SiO(2) solids through the protonated N(4) in the piperazinyl group and, possibly, a cation bridge; whereas the antibiotic sorbs to Al(2)O(3) solids through the ketone and carboxylate functional groups via a ligand exchange mechanism. Sorption edge and isotherm experiments show that ofloxacin exhibits a higher affinity for mesoporous SiO(2) and nonporous Al(2)O(3), relative to their counterparts. It is hypothesized that decreased ofloxacin sorption to mesoporous Al(2)O(3) occurs due to electrostatic repulsion within pore confines. In contrast, it appears that the environment within SiO(2) mesopores promotes sorption by inducing formation of ofloxacin-Ca complexes, thus increasing electrostatic attraction to SiO(2) surfaces.  相似文献

Atomic force microscopy (AFM) was used to measure single interaction forces between corrole (host) and phenol derivatives (guests) in aqueous media. A gold tip was modified with thiol derivatives of corrole via the Au–S covalent bond. Such a tip was used to measure adhesion forces with a planar gold substrate modified with thiol derivatives of phenol and ortho-nitrophenol in aqueous solutions. The mean force between the corrole and ortho-nitrophenol was higher than that between corrole and phenol, probably reflecting stronger hydrogen bond interaction in the former complex. In the presence of a supporting electrolyte (0.1 M K2SO4), the mean force increased, suggesting that electrostatic and π–π interactions play an essential role in the adhesion force. In addition, the adhesion force measured at pH 6.0 was larger than that at pH 10, reflecting the electrostatic repulsion at the higher pH. These behaviours are consistent with the potentiometric responses of a liquid membrane based on corrole to phenolic compounds. Also, the values of forces for the interaction between corrole and phenol derivatives showed the same tendency as energy calculated for these complexes. The Poisson method was used for the calculation of the single force of the chemical bond between the corrole host and the phenolic guests.  相似文献

Chemical force titrations-plots of the adhesive force between an atomic force microscope tip and sample as a function of pH-were acquired on alkyl monolayer-derivatized Si(111) surfaces. Gold-coated AFM tips modified with thioalkanoic acid self-assembled monolayers (SAM) were employed. Alkyl monolayer-derivatized Si(111) surfaces terminated with methyl, carboxyl, and amine groups were produced via hydrosilylation reactions between 1-alkene reagents and H-terminated silicon. The functionalized surfaces were characterized using standard surface science techniques (AFM, FTIR, and XPS). Titration of the methyl-terminated surface using the modified (carboxyl-terminated) atomic force microscope tip resulted in a small pH-independent hydrophobic interaction. Titration of the amine-terminated surface using the same tip resulted in the determination of a surface pKa of 5.8 for the amine from the pH value from the maximum in the force titration curve. A pK(1/2) of 4.3 was determined for the carboxyl-terminated Si(111) in a similar way. These results will be discussed in relation to the modified Si(111) surface chemistry and organic layer structure, as well as with respect to existing results on Au surfaces modified with SAMs bearing the same functional groups.  相似文献

The reversible desorption and adsorption of ethanethiol (ET) and hexadecane thiol (HDT) self-assembled monolayers (SAMs) on gold substrates are addressed with potential-dependent AFM force measurements where both tip and substrate potentials are controlled independently. For HDT-modified tip and substrate, the potential dependence of the force curve corresponds to the observed voltammetric features. The adhesion interaction between HDT-modified tip and substrate exhibits a large adhesion, whereas the adhesion is reduced to one-quarter of its original value after HDT on the substrate is removed. The presence of both attractive features on the approach curve and large adhesion on retraction after thiol desorption are ascribed to micelle formation from the desorbed, insoluble, thiols above the Au surface. For the ET-modified tip and substrate, the force curve evinces time-dependent recovery after the thiol adsorption peak which arises from the finite time of diffusion of the desorbed thiol back to the substrate. However, the force curves exhibit little potential dependence when the ET-desorbed tip is interacted with ET-modified substrate.  相似文献

The cation complexation behavior of dibenzocrown ethers adsorbed on highly oriented pyrolytic graphite substrates was investigated by means of atomic force microscopy using probe tips modified chemically with ammonium ion by silane coupling. The specific adhesion force based on the intermolecular force between dibenzocrown ether and ammonium ion was observed via force curve measurements in ethanol at the interface between the substrate and tip. The observed specific force decreased in the presence of the alkali metal ion in solution, indicating that the cation in solution interferes with the complexation of the crown ethers adsorbed on the substrate with the ammonium ion immobilized on the tip. The blocking effect of metal ions in solution on the observed force depended on the sizes of both the blocking cation and crown ether ring, suggesting that the surface-adsorbed dibenzocrown ethers possess a selective cation-complexing ability similar to that in their bulk state and that the adhesion force measurements using cation-modified tips allow evaluation of the cation-complexing ability of crown ethers under cation-competitive conditions.  相似文献

The interactions of various polypeptides with individual carbon nanotubes (CNTs), both multiwall (MW) and single wall (SW), were investigated by atomic force microscopy (AFM). While adhesion forces arising from electrostatic attraction interactions between the protonated amine groups of polylysine and carboxylic groups on the acid-oxidized multi-wall carbon nanotubes (Ox-MWCNTs) dominate the interaction at a low pH, weaker adhesion forces via the hydrogen bonding between the neutral -NH2 groups of polylysine and -COO- groups of the Ox-MWCNTs were detected at a high pH. The adhesion force was further found to increase with the oxidation time for Ox-MWCNTs and to be negligible for oxidized single-wall carbon nanotubes (Ox-SWCNTs) because carboxylate groups were only attached onto the nanotube tips in the latter whereas onto both the nanotube tips and sidewall in the former. Furthermore, it was demonstrated that proteins containing aromatic moieties, such as polytryptophan, showed a stronger adhesion force with Ox-MWCNTs than that of polylysine because of the additional pi-pi stacking interaction between the polytryptophan chains and CNTs.  相似文献

The mechanism underlying the bioinertness of the self-assembled monolayers of oligo(ethylene glycol)-terminated alkanethiol (OEG-SAM) was investigated with protein adsorption experiments, platelet adhesion tests, and surface force measurements with an atomic force microscope (AFM). In this work, we performed systematic analysis with SAMs having various terminal groups (-OEG, -OH, -COOH, -NH(2), and -CH(3)). The results of the protein adsorption experiment by the quartz crystal microbalance (QCM) method suggested that having one EG unit and the neutrality of total charges of the terminal groups are essential for protein-resistance. In particular, QCM with energy dissipation analyses indicated that proteins absorb onto the OEG-SAM via a very weak interaction compared with other SAMs. Contrary to the protein resistance, at least three EG units as well as the charge neutrality of the SAM are found to be required for anti-platelet adhesion. When the identical SAMs were formed on both AFM probe and substrate, our force measurements revealed that only the OEG-SAMs possessing more than two EG units showed strong repulsion in the range of 4 to 6 nm. In addition, we found that the SAMs with other terminal groups did not exhibit such repulsion. The repulsion between OEG-SAMs was always observed independent of solution conditions [NaCl concentration (between 0 and 1 M) and pH (between 3 and 11)] and was not observed in solution mixed with ethanol, which disrupts the three-dimensional network of the water molecules. We therefore concluded that the repulsion originated from structured interfacial water molecules. Considering the correlation between the above results, we propose that the layer of the structured interfacial water with a thickness of 2 to 3 nm (half of the range of the repulsion observed in the surface force measurements) plays an important role in deterring proteins and platelets from adsorption or adhesion.  相似文献

A new colorimetric method is proposed for the determination of maleic acid. Among aliphatic dicarboxylic acids tested, maleic acid was found to be selectively extracted into nitrobenzene as the red ion-association complex (lambda(max) 516 nm) formed between the hydrogen maleate anion and the tris(1,10-phenanthroline)iron(II) cation. At least a 32-fold molar excess of tris(1,10-phenanthroline)iron(II) relative to maleic acid is needed and the optimal pH range is 3-5. A linear relationship is obtained over the concentration range 10(-5)-10(-4)M maleic acid. The relative standard deviation was 1.0 %. The colour intensity of the extract remains constant at room temperature for at least 24 hr. A large amount of fumaric acid, the trans-isomer of maleic add, is not extracted under the same conditions. This makes it possible to determine maleic acid in the presence of fumaric acid.  相似文献

An improved method for the preparation of the heme octapeptide acetyl-MP8, obtained by proteolysis of horse heart cytochrome c, is described. AcMP8 obeys Beer's law at pH 7.0 in aqueous solution up to a concentration of 3 x 10(-)(5) M. The self-association constant measured at 25 degrees C (log K(D) = 4.04) is an order of magnitude lower than that for MP8, reflecting the role of the N-acetyl protecting group in abolishing intermolecular coordination. However, AcMP8 does form pi-stacked dimers in aqueous solution with increasing ionic strength. A more weakly packed pi-pi dimer reaches a maximum abundance at approximately 3 M ionic strength, but a more tightly packed dimer is favored at &mgr; > 3 M. An equilibrium model based on charge neutralization by specific binding of Na(+) ions gives a total molecular charge of 3- for AcMP8 at pH 7.0 and a self-association constant log K(D) = 4.20. AcMP8 exhibits six spectroscopically active pH-dependent transitions. The Glu-21 c-terminal carboxylate binds to the heme iron at low pH (pK(a) = 2.1) but is substituted by His-18 (pK(a) = 3.12) as the pH increases. The two heme propanoic acid substituents ionize with pK(a)'s of 4.95 and 6.1. This is followed by ionization of iron-bound water with a pK(a) = 9.59, DeltaH = 48 +/- 1 kJ mol(-)(1), and DeltaS = -22 +/- 3 J K(-)(1) mol(-)(1). The electronic spectra indicate that AcMP8 is predominantly in the S = (5)/(2) state at pH 7.0, while the hydroxo complex at pH 10.5 corresponds to an equilibrium mixture of S = (5)/(2) and S = (1)/(2) states at 25 degrees C. In the final transition, His-18 ionizes to form the S = (1)/(2) histidinate complex with a pK(a) of 12.71. AcMP8 is relatively stable under alkaline conditions, dimerizing slowly at high pH (k = 2.59 +/- 0.14 M(-)(1) s(-)(1)) to form a high-spin &mgr;-oxo-bridged species. The pH-dependent behavior of AcMP8 in the presence of excess 3-cyanopyridine, however, is markedly different. At low pH, AcMP8 simultaneously binds the exogenous ligand and the Glu-21 c-terminal carboxylate with a pK(a) < 2. His-18 replaces the carboxylate ligand at higher pH (pK(a) = 2.60), and both heme propanoic acid groups ionize with a mean pK(a) = 5.10. Unlike AcMP8.OH(-), the axial histidine of the 3-CNPy complex ionizes at near neutral pH (pK(a) = 7.83), prior to being replaced by OH(-) (pK(a) = 10.13). The sixth transition in the AcMP8/3-CNPy system produces the bis(hydroxo) complex (pK(a) > 13).  相似文献

Two‐component self‐assembly is a promising approach to construct functional nanomaterials. Interaction of a flexible guanidiniocarbonyl pyrrole tetra‐cation ( 1 ) with naphthalene diimide dicarboxylic acid (NDIDC) in aqueous DMSO leads to the formation of supramolecular networks. First, the carboxylate groups of NDIDC bind to the guanidiniocarbonyl pyrrole cations of 1 in a 1:2 stoichiometry. Further π–π induced aggregation then leads to 3D networks, as established by dynamic light scattering studies (DLS), NMR, fluorescence titration, viscosity measurements, AFM, and TEM microscopy. Due to ion pairing, the resulting aggregates can be switched between the monomers and the aggregates reversibly using external stimuli like protonation or deprotonation. At high concentration, a stable colloidal solution is formed, which shows an extensive Tyndall effect. Increasing the concentrations even further leads to formation of a supramolecular gel.  相似文献

The flash photolysis kinetic spectra of the intermediate M(412) of bacteriorhodopsin were monitored during the process of acid titration. In the light-adapted state, the maximum peak amplitude of M(412) absorbance of bacteriorhodopsin decreased (pK(a)=3.40+/-0.05) as the pH decreased from 7.3 to 1.9. In the dark-adapted state, the maximum peak amplitude of M(412) absorbance of bacteriorhodopsin increased as the pH decreased from 6.9 to 4.1, and then decreased (pK(a)=2.85+/-0.05) as the pH dropped to 2.1. These different trends in the change in the maximum peak amplitude suggested that not only the transition of purple membrane to blue membrane had taken place in both light and dark-adapted states, but also the fraction of all-trans-bR had changed during the acid titration. The pH-dependent absorption changes at 640 nm of bacteriorhodopsin in both light- and dark-adapted states were also observed. The pK(a)-values of the purple-to-blue transition were 3.80+/-0.05 in light-adapted state and 3.40+/-0.05 in dark-adapted state, respectively. According to Balashov's method, the fraction of all-trans-bR was assayed as the pH decreased. All these results indicated that the purple-to-blue transition of light-adapted bacteriorhodopsin was accompanied by an all-trans to 13-cis retinal isomerization at acidic pH.  相似文献

Application of two complementary AFM measurements, force vs separation and adhesion force, reveals the combined effects of cation size and charge (valency) on the interaction between silica surfaces in three 1:1, three 2:1, and three 3:1 metal chloride aqueous solutions of different concentrations. The interaction between the silica surfaces in 1:1 and 2:1 salt solutions is fully accounted for by ion-independent van der Waals (vdW) attraction and electric double-layer repulsion modified by cation specific adsorption to the silica surfaces. The deduced ranking of mono- and divalent cation adsorption capacity (adsorbability) to silica, Mg(2+) < Ca(2+) < Na(+) < Sr(2+) < K(+) < Cs(+), follows cation bare size as well as cation solvation energy but does not correlate with hydrated ionic radius or with volume or surface ionic charge density. In the presence of 3:1 salts, the coarse phenomenology of the force between the silica surfaces as a function of salt concentration resembles that in 1:1 and 2:1 electrolytes. Nevertheless, two fundamental differences should be noticed. First, the attraction between the silica surfaces is too large to be attributed solely to vdW force, hence implying an additional attraction mechanism or gross modification of the conventional vdW attraction. Second, neutralization of the silica surfaces occurs at trivalent cation concentrations that are 3 orders of magnitude smaller than those characterizing surface neutralization by mono- and divalent cations. Consequently, when trivalent cations are added to our cation adsorbability series the correlation with bare ion size breaks down abruptly. The strong adsorbability of trivalent cations to silica contrasts straightforward expectations based on ranking of the cationic solvation energies, thus suggesting a different adsorption mechanism which is inoperative or weak for mono- and divalent cations.  相似文献

In this study photoinduced cation generation, based on the photochemical properties of malachite green (MG), was used for the surface design and in vitro photochemical control of cell adhesion and proliferation. The MG-derivatized surface was prepared by coating a photoreactive polymer as a substrate onto a poly(ethylene terephthalate) (PET) sheet. The photoreactive polymer was radical copolymer of styrene with the MG-derivatized monomer diphenyl(4-vinylphenyl)methane leucohydroxide (degree of substitution of MG unit: 12.4 mol%). Water contact angle measurements and X-ray photoelectron spectroscopy revealed high hydrophobicity and homogeneous distribution of the MG groups on the outermost surface of the coated film, respectively. When the coated film was exposed to ultraviolet light (UV) irradiation at wavelengths of 290-410 nm, a time-dependent color change of the film was observed from pale yellow, before irradiation, to green. These results indicated generation of cations on the film surface by photochemical cation generation of the MG groups, which was quantitatively characterized by force versus distance curves measurements in atomic force microscopic (AFM) observation using a carboxylated AFM tip. The seeding and culture of endothelial cells showed a marked reduction in adhesion on the nonirradiated coated film surface, whereas the UV-irradiated surface promoted cell adhesion and proliferation except for incubation in serum-free medium, which was similar to commercial tissue culture PET sheet. These observations may be due to adsorption of cell adhesive proteins, typified by fibronectin, in serum-containing medium onto the cationized photoreactive copolymer surface by electrostatic interactions.  相似文献

The photo‐induced association and dissociation of poly(sodium acrylate) containing a small amount of photoresponsive malachite green (MG) in aqueous solution were studied. It is known that MG dissociates into ion pairs under UV irradiation to produce the green triphenylmethyl cation. The cation thermally recombines with its counter anion to regenerate the colorless neutral compound. The random copolymer of acrylic acid with 0.05 mol % of MG monomer [P(A/MG0.05)] was soluble in aqueous 0.01 M NaCl at pH 12 as a unimer due to electrostatic repulsion between carboxylate pendent groups when the MG moieties were in the neutral form. On the other hand, these MG groups were converted to the cationic form on UV irradiation, leading to polymer aggregation driven by electrostatic interactions between the cationic MG and anionic carboxylate pendent groups. These aggregates could be dissociated by heating in the dark, as the cationic MG reverted to its neutral form, eliminating these attractive electrostatic interactions. The association and dissociation of the copolymer was monitored by dynamic light scattering (DLS). When the salt concentration in aqueous solutions of P(A/MG0.05) was increased from 0.01 to 0.5 M at pH 12, no aggregation was observed on UV irradiation because of ionic screening of the aforementioned electrostatic interactions. © 2011 Wiley Periodicals, Inc. Bacteria-metal interactions in aqueous solutions are important in biofilm formation, biofouling and biocorrosion problems in the natural environment and engineered systems. In this study, the adhesion forces of two anaerobes (Desulfovibrio desulfuricans and Desulfovibrio singaporenus) and an aerobe (Pseudomonas sp.) to stainless steel 316 in various aqueous systems were quantified using atomic force microscopy (AFM) with a cell probe. Results show that the nutrient and ionic strength of the solutions influence the bacteria-metal interactions. The bacteria-metal adhesion force was reduced in the presence of the nutrients in the solution, because a trace organic film was formed and thus decreased the metal surface wettability. Stronger ionic strength in the solution results in a larger bacteria-metal adhesion force, which is due to the stronger electrostatic attraction force between the positively charged metal surface and negatively charged bacterial surface. Solution pH also influences the interaction between the bacterial cells and the metal surface; the bacteria-metal adhesion force reached its highest value when the pH of the solution was near the isoelectric point of the bacteria, i.e. at the zero point charge. The adhesion forces at pH 9 were higher than at pH 7 due to the increase in the attraction between Fe ions and negative carboxylate groups.  相似文献

Abstract

Values for the ionization constants and the heats of ionization of phytic acid (inositol hexaphosphate) have been determined by simultaneous potentiometric and thermal titration in a titration microcalorimeter. The potentiometric results are in substantial agreement with those found by discontinuous titration; the heat liberated from pH 10.4 to 6.8 is essentially that found using a batch microcalorimeter. Ionization of phytic acid may be generally characterized as having five groups similar to pK of phosphoric acid, two groups similar to pK2 of phosphoric acid and 4 groups similar to pK4 of pyrophosphoric acid. A single group has pK ~ 5 with ΔHi ~ -1.1 kcal/mol.  相似文献
