Synthesis of 1,3,4-Oxadiazoles and 4,5-Dihydro-l,2,4-triazines from 3-Dimethylamino-2,2-dimethyl-2 Hazirine and Carbohydrazides 3-Dimethylamino-2, 2-dimethyl-2 H-azirine ( 1 ) reacts with aromatic carbohydrazides to give 2-(1-amino-1-methylethyl)-5-aryl-1, 3, 4-oxadiazoles ( 7 , 10 , 11 ). With ethyl carbazate the azirine 1 forms the aminoester 15 , which is easily cyclized to the 4, 5-dihydro-1, 2, 4-triazin-3 (2H)-one 16 . From the reaction of 1 with oxamohydrazide ( 17 ) and oxalodihydrazide 19 the 4, 5-dihydro-1, 2, 4-triazin-3-carboxamide 18 and the symmetric compound 20 , respectively, have been isolated. Reactions supporting the structures of the new compounds are described.  相似文献

3-Alkyl-1-benzoxepin-5-one derivatives and 2-alkyl-1,4-naphtoquinones from 2-acylaryl propargyl ethers. It was found that 3-alkyl-1-benzoxepin-5(2H)-ones of type B can be synthesized by treating 2-acylaryl propargyl ethers of type A with sodium methylsulfinyl methide (NaMSM, dimesyl sodium) (Scheme 13). Oxepinone derivatives of type B undergo ring contraction with base (also NaMSM) to yield the quinol derivatives C which, oxidize (during work-up), if R2 = H, to the 1,4-naphthoquinones D (Scheme 13). The propargyl ethers used are listed in Scheme 1. The naphthalene derivatives 1 and 3 give oxepinones (E- 9 and a mixture of 14/15 respectively), whereas the expected oxepinone from 2 is transformed directly into the quinone 11 (Scheme 2, 3 and 5). Isomerizations of 2-acetylphenyl propargyl ethers ( 4, 5 and 6 ) (Schemes 6, 7 and 8) are less successful because of side reactions. If however the acetyl group is replaced by a propionyl or substituted propionyl group (as in ethers 7 and 8 ) oxepinones are obtained again in good yield (Scheme 9). The mechanistic pathway for the transformation of naphthyl propargyl ethers (and phenyl derivatives) under influence of NaMSM is shown in Scheme 10. The base-catalysed conversion of 4-phenyl-l-benzoxepin-5(2H)-one,benzo[f]furo[2,3-c](10 H)-oxepin-4-oncsand 3-methoxy-G,11- dihydro-dibenzo[b, e]loxepin-11-oneinto thc corresponding quinones has been reported [13] [20] [21]. The conversion of 2-acylaryl propargyl others via the isolable benzoxepin-5-one derivativcs or directly into the specifically substituted 1,4-naphthoquinone derivatives is of synthetic interest.  相似文献

Dipolar 1:1 Adducts from the Reaction of 3-Amino-2H-azirines with 1,3,4-Oxadiazol- and 1,3,4-Thiadiazol-2(3H)-ones 3-Amino-2H-azirines 1 react with 5-(trifluoromethyl)-1,3,4-oxadiazol-2(3H)-one ( 2 ) as well as with different 5-substituted 1,3,4-thiadiazol-2(3H)-ones ( 5a–e ) in 2-propanol at room temperature to give dipolar 1:1 adducts of type 3 and 6 , respectively, in reasonable-to-good yields (Schemes 3 and 6, Tab. 1 and 2). The structure of two of these dipolar adducts, 6a and 6e , which are formally donor-acceptor-stabilized azomethin imines, have been elucidated by X-ray crystallography (Figs. 1-4). In the reaction of 2 and sterically crowded 3-amino-2H-azirines 1c–e with a 2-propyl and 2-propenyl substituent, respectively, at C(2), a 4,5-dihydro-1,2,4-triazin-3(2H)-one of type 4 is formed as minor product (Scheme 3 and Table 1). Independent syntheses of these products proved the structure of 4 . Several reaction mechanisms for the formation of compounds 3 and 4 are discussed, the most likely ones are described in Scheme 4: reaction of 2 as an NH-acidic compound leads, via a bicyclic zwitterion of type A , to 3 as well as to 4 . In the latter reaction, a ring-expanded intermediate B is most probable.  相似文献

Preparation of Enantiomerically Pure 4,4,4-Trifluoro-3-hydroxy-butanoic Acid Derivatives, Branched in the 2- or 3-Position, from 6-Trifluoromethyl-1,3-dioxan- and -dioxin-4-ones Enantiomerically pure 3-hydroxy-3-trifluoromethyl-propionic acid and esters, substituted in the 2- or 3-position, are prepared (13 examples) from (R)- or (S)-4,4,4-trifluoro-3-hydroxy-butanoic acid. Key intermediates are the 2-t-butyl-6-trifluoromethyl-1,3-dioxan- and -dioxin-4-ones. The Li enolate of the cis-dioxanone is generated with t-BuLi and reacts with electrophiles (alkyl halides, aldehydes, imines, nitroolefins, Br2, I2) with predominant formation of trans,trans-2,5,6-trisubstituted dioxanones (9 examples). Elimination of HBr from the 5-Br-substituted dioxanone gives the (R)- or (S)-dioxinone, a chiral derivative of 4,4,4-trifluoro-3-oxo-butanoic acid (trifluoro-acetoacetate). Michael additions of cuprates or of CuCl-doped Grignard reagents to the dioxinone produce 6,6-disubstituted dioxanones (10 examples) bearing a CF3 group in the 6-position. In most cases this addition is highly diastereoselective, with the new substituent winding up in the trans position. There are, however, surprising exceptions, such as the product formed with benzylmagnesium chloride which is an abnormal adduct with a p-quinoid structure ( 26 ) and with the newly introduced group in the cis position with respect to the t-Bu group. The structures of four trisubstituted dioxanones bearing CF3 groups are determined by X-ray crystal structure analysis (Figure 1, Table 1), one of them including the absolute configuration (by anomalous diffraction). Besides the well-known sofa, a twist-boat conformation of dioxanones appears to be favorable. The solution conformations of the different types of CF3-substituted dioxanones are derived from Nuclear Overhauser NMR measurements and compared with the crystal structures (Figure 3).  相似文献

The simple LCAO MO method is used to calculate π-electron configurations of 1,2-dithiol-3-thione and its 4- and 5-phenyl derivatives. In these compounds the heterocyclic ring has an aromatic character, but the S-S bond is almost excluded from conjugation. The phenyl group in 4-phenyl-1,2-dithiol-3-thione is an electron acceptor, whereas in the 5 isomer it is an electron donor. The exocyclic sulfur atom has considerable negative charge, while the two endocyclic ones are positively charged. The dipole moments of 4- and 5-phenyl-1,2-dithiol-3-thione are determined, and the values found are compared with the calculated ones.  相似文献

Transition Metal Complexes with Anions of 1-substituted Tetrazolinones and -thiones and of 1,3-Thiazolin-2-thione The preparation of a series of anionic and phosphine containing complexes with the ambivalent anions of 1-R-tetrazolin-5-ones and -5-thiones, and of 1,3-thiazolin-2-thione is reported. According to the i.r. and 1H n.m.r. spectra the S-heterocycles are bonded through the exocyclic sulfur atom to the metal; in the oxotetrazolinate compounds coordination occurs via the N2 or N4 ring atoms. The copper and silver complexes [(Ph3P)MX]2 (X = anionic heterocycle) have been found to be dimeric.  相似文献

1, 5, 6, 7-Tetrahydro-2H-[1, 4]diazepin-5, 7-diones from Malonimides and 3-Dimethylamino-2, 2-dimethyl-2H-azirine Reaction of the aminoazirine 1 with malonimides of type 7 in 2-propanol at room temperature leads to the 1,4-diazepine derivatives of type 9 (Scheme 3). The structure of 6, 6-diethyl-3-dimethylamino-2,2-dimethyl-1,5,6, 7-tetrahydro-2H- [1,4] diazepin-5, 7-dione ( 9a ) has been proved by single crystal X-ray analysis (Chapter 4). Reduction of the 7-membered heterocycle 9a with sodium borohydride yields the perhydro-[1,4]diazepin-5, 7-dione 10 , while 9a in ethanol at 60° undergoes a ring contraction to the 4 H-imidazole derivative 11a (Scheme 4): Mechanisms of these two reactions are discussed in comparison with previously reported reactions (Chapter 5).  相似文献

The pyrano-cyclisation of 4-hydroxy-(1-methyl)-1H-quinoline-2-one with 3-chlor-3-methylbut-1-yne, supposed to proceed via an ether or vinyl intermediate, is studied by alkylating different 4-hydroxyquinolines. The nine derivatives characterized favour the ether-pathway (Späth type chromene synthesis).

The title substances with R2=H (type1) were synthesized by reductive desulfurization of corresponding 2-aryl-thieno-[2,3-d]pyrimidin-3(3H)-ones and 2-aryl-[1]benzothieno[2,3-d]-pyrimidin-4(3H)-ones (both:A), those with R2=CH3 (type2) by cyclization of -alkyl-acetoacetates with benzamidines. Some derivatives of1 and2 were also prepared (type3). In some cases Raney-Ni desulfurization ofA gave 2-cyclohexylproducts (type4).
