A simple, novel, and an efficient synthesis of α-hydroxy phosphonates (3a–d) and phosphinates (5a–f) has been accomplished from febuxostat aldehyde (1) with various dialkylphosphites (2) and dialkylphosphinates (4) in the presence of 1, 4-dimethylpiperazine as a strong base with good to excellent yields (78%–95%) is reported. The structures of the title compounds were established by elemental analysis and spectral data (IR, 1H, 13C and 31P NMR, and LC-MS). All the title compounds (3a–d) and (5a–f) were tested for their antimicrobial and antioxidant activities. Bioassay results reveal that a few of the title compounds exhibited promising antimicrobial and antioxidant activities.  相似文献

A new and highly flexible procedure is described for the synthesis of α-amino- and α-hydroxy phosphonates. In the presence of a catalytic amount of oxalic acid (10 mol %), trimethyl phosphite reacts with aldehydes or imines (generated in situ from an aldehyde and an amine) to yield the corresponding coupled products in good yield.  相似文献

6h-Aminophosphonates containing alicyclic, aromatic, and heterocyclic fragments were obtained in high yields by the Kabachnick—Fields reaction of the corresponding carbonyl compounds with 2-, 3- and 4-aminopyridines and diethyl phosphite using tetra- tert-butyl- phthalocyanine aluminum chloride as the catalyst. 2-, 3-, 4-Aminopyridines were found to vary in their reactivity with ketones and diethyl phosphite in the catalytic three-component one-pot process.  相似文献

An efficient and general two-step halogenation procedure to prepare diethyl α,α-bromofluorophosphonates was described, which included bromination by PPh3, 2,3-dichloro-5,6-dicyanobenzquinone (DDQ) and n-Bu4NBr, and electrophilic fluorination by N-fluorobisbenzenesulfonimide (NFSI). Abstract  

Schiff-base Kabachnik–Fields intermediates generated in situ from substituted pyrazole-4-carbaldehyde and 2-aminothiophene derivatives were trapped by dialkyl phosphites to produce the corresponding α-aminophosphonates in moderate yields. The latter products could be also obtained in excellent yields (≥75%) by directly applying the phosphorus reagents to the Schiff bases. Next, dialkyl phosphites were applied to one of the parent aldehydes to give the expected α-hydroxyphosphonate derivatives. Applying hexaalkyl triamidophosphites to the Schiff base in ethanol afforded methylphosphonic diamide derivatives, whereas ring attack on the pyrazole ring occurred when the same amidophosphites were applied to the parent aldehyde to give the corresponding alkylidenephosphorane ylides in an open structure form in good yields. Some of the new compounds exhibited considerable anti-inflammatory properties.  相似文献

An expeditious green synthetic approach was developed for the synthesis of α-aminophosphonates in good yields through one-pot three component reaction (Kabachnik-Fields reaction) of equimolar quantities of N-(4-amino-2-phenoxy phenyl)methanesulfonamide, diethylphosphite and various aldehydes under conventional as well as microwave irradiation methods. The newly synthesized compounds were characterized by NMR (31P, 1H, and 13C), Mass, IR and C, H, N analyses. The synthesized compounds were screened for their anti-inflammatory activity using rat paw edema method. Most of the compounds from the series showed good anti-inflammatory activity when compared with standard drug. Especially the compounds 5d bearing 4-hydroxy-3-nitrophenyl moiety, 5e bearing 3-bromo-4-fluorophenyl moiety, 5g incorporated with 2,4-dichlorophenyl moiety and 5f containing 4-chlorophenyl moiety exhibiting edema inhibition of 91.01% to 85.39% after 4 h of carrageenan injection while the other compounds displayed inhibition ≥75%.  相似文献

A series of new α-hydroxyphosphonate (5–14) have been synthesized in high yields by reaction of β-chloro-α,β-unsaturated aldehydes (14) and trialkyl phosphite in the presence of chlorotrimethylsilane at room temperature under solvent-free conditions. In addition, compounds (514) were reacted with acetic anhydride in DBU to give the new α-acetoxyphosphonate derivatives (1523). All these phosphonates (523) were obtained as mixtures of two (E and Z) isomers and fully characterized by multinuclear (1H, 13C, and 31P) NMR, IR spectroscopy and HRMS.  相似文献
