Based on the existing reports on the bioactive isatin derivatives, a number of Schiff bases were synthesized by reacting 5-substituted isatins with bioactive amines/hydrazides and their structures were confirmed using spectroscopic methods such as NMR, IR and mass spectrometry. Furthermore, Nbenzylation of isatin followed by the Schiff base formation furnished a new series of compounds(11a–13c) which allowed the analysis of the effect of isatin N-substitution on the bioactivity of the resulting compounds. The antibacterial activity of the synthesized derivatives was evaluated using a microtiter plate method on a series of gram positive and gram negative bacterial strains. Compounds 2d, 3b, 5c and 6a were among the most potent derivatives against Pseudomonas aeruginosa(MIC = 6.25 μg/m L).Analysis of the structure–activity relationship showed that the incorporation of(thio)urea-based Schiff bases lead to more potent derivatives with a broader spectrum of antibacterial activity. In addition,highly lipophilic compounds such as 11a–12c did not show any measurable antibacterial activity, which implies that an optimal lipophilicity might be an important requirement for the antibacterial activity of the studied isatins. Finally, the finding that hydantoin derivatives of N-benzylisatins(13a–13c) still exhibit some antibacterial activity prompted us to consider exploring the bioactivity of more diverse derivatives of isatin-aminohydantoin Schiff bases(compounds 1a–1d) in our future studies.  相似文献

In this study, ZnFe2O4 nanoparticles (NPs) were prepared by a hydro/solvothermal method, and their structures were characterized by infrared, scanning electron microscopy, energy-dispersive X-ray spectroscopy and X-ray diffraction analyses. These ZnFe2O4 NPs were used in the synthesis of new bis-Schiff bases by condensation of hydroxyl-substituted aldehydes with various aromatic diamines in aqueous media. The antimicrobial activities of the synthesized bis-Schiff bases were tested against gram-negative as well as gram-positive bacteria. The MTT method was used to assess the anticancer activity of the bis-Schiff bases towards human lung and breast cancer cell lines. The results indicated that these compounds could be considered as new anti-tumor candidates.  相似文献

With the aim of developing potential antimicrobials, a series of novel Ciprofloxacin methylene isatin derivatives incorporating different aromatic aldehydes were synthesized and characterized by FTIR, 1H NMR, Mass spectroscopy and bases of elemental analysis. In addition, the in vitro antibacterial and antifungal properties were tested against some human pathogenic microorganisms by employing the disc diffusion technique. A majority of compounds were showing activity against several of the microorganisms. The relationship between the functional group variation and the biological activity of the evaluated compounds is discussed. From comparisons of the compounds, 3c was determined to be the most active compound.  相似文献

Condensation of aromatic primary bis-amines with isatin (1H-indole-2,3-dione) and 5-flouroisatin occurred cleanly and efficiently in a water suspension medium without using any organic solvent or acid catalyst. The corresponding bis-Schiff bases were obtained in good yields and were easily isolated by filtration. Their structures were confirmed by (1)H-NMR, (13)C-NMR, IR and mass spectra.  相似文献

A new series of di-spirooxindole analogs, engrafted with oxindole and cyclohexanone moieties, were synthesized. Initially, azomethine ylides were generated via reaction of the substituted isatins 3a–f (isatin, 3a, 6-chloroisatin, 3b, 5-fluoroisatin, 3c, 5-nitroisatin, 3d, 5-methoxyisatin, 3e, and 5-methylisatin, 3f, and (2S)-octahydro-1H-indole-2-carboxylic acid 2, in situ azomethine ylides reacted with the cyclohexanone based-chalcone 1a–f to afford the target di-spirooxindole compounds 4a–n. This one-pot method provided diverse structurally complex molecules, with biologically relevant spirocycles in a good yields. All synthesized di-spirooxindole analogs, engrafted with oxindole and cyclohexanone moieties, were evaluated for their anticancer activity against four cancer cell lines, including prostate PC3, cervical HeLa, and breast (MCF-7, and MDA-MB231) cancer cell lines. The cytotoxicity of these di-spirooxindole analogs was also examined against human fibroblast BJ cell lines, and they appeared to be non-cytotoxic. Compound 4b was identified as the most active member of this series against prostate cancer cell line PC3 (IC50 = 3.7 ± 1.0 µM). The cyclohexanone engrafted di-spirooxindole analogs 4a and 4l (IC50 = 7.1 ± 0.2, and 7.2 ± 0.5 µM, respectively) were active against HeLa cancer cells, whereas NO2 substituted isatin ring and meta-fluoro-substituted (2E,6E)-2,6-dibenzylidenecyclohexanone containing 4i (IC50 = 7.63 ± 0.08 µM) appeared to be a promising agent against the triple negative breast cancer MDA-MB231 cell line. To explore the plausible mechanism of anticancer activity of di-spirooxindole analogs, molecular docking studies were investigated which suggested that spirooxindole analogs potentially inhibit the activity of MDM2.  相似文献

To further explore an efficient modified route for the shift from an antibacterial fluoroquinolone to an antitumor one,mono-Schiff bases 6a-6h related to ciprofloxacin C3 carbonylhydrazone and bis-Schiff bases 4a-4h corresponding to C3/C7 carbonylhydrazone/hydrazone attached on a skeleton of ciprofloquinolone were designed and synthesized,and their in vitro antitumor activity against CHO,HL60,L1210 cells and antibacterial activity against Staphylococcus aureus and Escherichia coli were also reported.  相似文献

ABSTRACT: BACKGROUND: Cancer is one of the most dreaded diseases and it is a leading cause of mankind death worldwide. Recent reports documented a remarkable antiproliferative activity of isatin nucleus against various cancer cell lines. The current work describes the antiproliferative activity of Schiff bases of combinatorial mixtures of the isatin derivatives M1-M22 as well as the individual compounds 1-11(A-K) of these combinatorial mixtures. RESULTS: The designed combinatorial library composed from eleven hydrazides A-K and eleven isatin derivatives 1-11 has been synthesized to formally generate 22 mixtures, M1-M22 of 121 Schiff bases, and their antiproliferative activity against K562 chronic myelogenous leukemia cells was evaluated. The indexed method of analysis of the prepared library was applied to elucidate the active components in the tested mixtures M1-M22. The predictions from the crossing procedure was validated through evaluation of the antiproliferative activity of individual compounds 1-11(A-K) of the library. Individual compounds 1-11(A-K) were also evaluated against the non-tumorigenic MCF-12A cell line to investigate their selectivity. A pharmacophore model was developed to further optimize the antiproliferative activity among this series of compounds. CONCLUSIONS: Variable antiproliferative activity was revealed with the investigated mixtures M1-M22 and the individual compounds 1-11(A-K). Most of the tested mixtures and several individual Schiff bases displayed high potency with IC50 values in the low micromolar range. A considerable selectivity of some individual compounds to the tumorigenic K562 cell line compared with the non-tumorigenic MCF-12A cell line was observed as indicated by their selectivity index (SI).  相似文献

用2-苯基-1,2,3-三唑基-4-甲醛、喹喔啉基-2-甲醛和2-(4-溴苯基)-1,2,3-三唑基-4-甲醛为原料, 在冰醋酸的条件下分别与二胺反应, 合成了一系列新的双席夫碱5a~5d, 6a~6d和7a~7d. 目标化合物的结构经元素分析, IR, 1H NMR, MS, X射线确证. 同时测定了化合物6c的晶体结构. 其属于单斜晶系, C2/c空间群, 晶胞参数 a=1.7670(4) nm, b=0.48001(10) nm, c=1.1751(2) nm, α=90.00°, β=94.42(3)°, γ=90.00°, V=0.9937(4) nm3, Dc=1.399 g/cm3, Z=2, F(000)=436, μ=0.089 mm-1, R=0.0413, wR=0.1067. 初步生物活性筛选结果表明, 目标化合物6b在Vero细胞上有抗疱疹病毒I型活性.  相似文献

A series of novel moxifloxacin methylene and ethylene isatin derivatives with remarkable improvement in lipophilicity, compared to the parent moxifloxacin, was designed, synthesized and characterized by 1H NMR, MS and HRMS. These derivatives were initially evaluated for their in vitro antimycobacterial activity against M. smegmatis CMCC 93202. Compounds 3a―3f, 5a, 5f and 5j were chosen for the further evaluation of their in vitro activity against Mycobacterium tuberculosis(MTB) H37Rv ATCC 27294 and MDR-MTB 09710. All the target compounds[ minimum inhibitory concentration(MIC): 0.39―>16 μg/mL] were far more active than rifampin(MIC: 2.0―>256 μg/mL), but less active than moxifloxacin(MIC: 0.1―1.0 μg/mL) against the three tested strains. The most active compounds 3a and 3c were found to be 2―64 fold more potent than isoniazid and rifampin against M. smegmatis CMCC 93202, 2 fold more potent than rifampin against MTB H37Rv ATCC 27294, and 16―>64 fold more potent than ethambutol, isoniazid and rifampin against MDR-MTB 09710.  相似文献

The reaction of 5-(3,4-dimethoxyphenyl)-1,3,4-oxadiazole-2(3H)-thione 3 with formaldehyde solution and primary aromatic amines or 1-substituted piperazines, in ethanol at room temperature yielded the corresponding N-Mannich bases 3-arylaminomethyl-5-(3,4-dimethoxyphenyl)-1,3,4-oxadiazole-2(3H)-thiones 4a–l or 3-[(4-substituted piperazin-1-yl)methyl]-5-(3,4-dimethoxyphenyl)-1,3,4-oxadiazole-2(3H)-thiones 5a–d, respectively. The in vitro inhibitory activity of compounds 4a–l and 5a–d was assessed against pathogenic Gram-positive, Gram-negative bacteria, and the yeast-like pathogenic fungus Candida albicans. The piperazinomethyl derivatives 5c and 5d displayed broad-spectrum antibacterial activities the minimal inhibitory concentration (MIC) 0.5–8 μg/mL) and compounds 4j, 4l, 5a, and 5b showed potent activity against the tested Gram-positive bacteria. In addition, the anti-proliferative activity of the compounds was evaluated against prostate cancer (PC3), human colorectal cancer (HCT-116), human hepatocellular carcinoma (HePG-2), human epithelioid carcinoma (HeLa), and human breast cancer (MCF7) cell lines. The optimum anti-proliferative activity was attained by compounds 4l, 5a, 5c, and 5d.  相似文献

Summary Partial displacement of the symmetrical bis-Schiff bases (1)–(4) of ethylenediamine and salicyladehyde, o-hydroxyacetophenone, acetylacetone or benzoylacetone by dehydroacetic acid have led to the formation and isolation of the unsymmetrical bis-Schiff base ligands (6)–(9). Attempted partial displacements of the symmetrical bis-Schiff base (5) of dehydroacetic acid or (1)–(4) by each of the other four carbonyl compounds have been unsuccessful. A simple, efficient, ecofriendly, and cost-effective method has been developed for the synthesis of 16 spiropyrazolo pyrimidine derivatives (5a–5p) by a four-component, one-pot reaction of pyrimidine-2,4,6(1H,3H,5H)-trione, 3-oxo-3-phenylpropanenitrile, hydrazine, and isatins (4a–4f) by using nanocopper ferrite catalyst (20?mol%) in water with excellent yields (73–91%). The isatin with electron-withdrawing groups gave products in high yields (5h and 5p). The present methodology offers an environmentally benign, cost-effective, high-yielding method with recyclable catalyst. The drug likeness or “drugability” of all the synthesized compounds were tested through rule of five (RO5) parameters. Compounds 5f, 5h, 5n, and 5p have shown one RO5 violation each. The compounds were screened for their antibacterial activity against human pathogenic bacteria wherein two of the synthesized compounds were found to possess significant antibacterial activity. Compounds showing RO5 violations had negligible antibacterial activity.  相似文献

A series of neodymium(III) and samarium(III) complexes of type [Ln(L)Cl(H2O)3] have been synthesized with Schiff bases (LH2) derived from 3‐(phenyl/substituted phenyl)‐4‐amino‐5‐mercapto‐1,2,4‐triazoles and isatin. The structures of the complexes were established using elemental analysis, molar conductivities, magnetic moments, infrared, NMR (1H, 13C) and UV–visible spectra, X‐ray diffraction and mass spectrometry. The thermal behaviour of these compounds under non‐isothermal conditions was investigated using thermogravimetry and differential thermogravimetry. The intermediates obtained at the end of various thermal decomposition steps were identified from elemental analysis and infrared spectral studies. All the ligands and their complexes were also screened for their antibacterial activity against Staphylococcus aureus and Bacillus subtilis and antifungal activity against Aspergillus niger, Aspergillus flavus and Colletotrichum capsici. A novel series of isatin hybrids 5a-g was designed, synthesized, and characterized spectroscopically. The synthesized compounds were evaluated for their cytotoxic activity against the human breast cancer cell line (MCF-7) by in vitro MTT assay. Amongst the tested compounds, 5e compound bearing benzyl moiety at N4 piperazine was found to be the most active with the promising IC50 (12.47 µM). Moreover, the active compounds 5e and 5g were subjected to antitumor evaluation (in vivo) against Dalton’s ascitic lymphoma (DAL) cell line and the results suggested that the best active compound 5e can normalize the blood picture in comparison to the standard drug. An in silico molecular docking study using the crystal structure of Hsp90 protein described the role of significant protein–ligand interactions and revealed more insights into the binding mode. The drug-likeliness of the compounds was predicted based on Lipinski's rule of five and pharmacokinetic ADME parameters. Hence, the synthesized isatin hybrids could be novel starting point anticancer lead compounds demonstrating drug-like properties which can be explored further for anticancer drug discovery.  相似文献

Reactions of 3-hyrazino-5,6-diphenyl-1,2,4-triazine with various carbonyl compounds such as ethyl acetoacetate, acetylacetone, benzoin, isatin, phthalic anhydride, phenyl isocyanate and acetic anhydride were discussed. Its reactions with α, β unsaturated compounds such as arylidinemalononitrile, diethyl acetylenedicarboxylate, dibenzylidine hydrazine were studied. These reactions led to the formation of various triazine and fused-triazine derivatives. The antitumor activity of the synthesized compounds was tested against HePG2 and MCF-7 cell lines. Some of the tested compounds were most active, whereas other compounds exhibited little or no activity.  相似文献

Five seven- or eight-coordinate manganese complexes of hydrazone ligands have been prepared. Three seven-coordinate neutral Mn(II) complexes: [Mn(dapA2)]n (1), [Mn(dapB2)(H2O)2] (2), [Mn(dapS2)(H2O)2] (3) have been synthesized from the bis-Schiff bases of 2,6-diacetylpyridine: dap(AH)2, dap(BH)2 and dap(SH)2 (AH = anthraniloyl hydrazide, BH = benzoyl hydrazide, SH = salicyloyl hydrazide), respectively. Two eight-coordinate Mn(II) complexes: [Mn(dapS)2] (4) and [Mn(dapB)2].3H2O (5) have been synthesized from the mono-Schiff bases dapBH and dapSH, respectively. The complexes have been characterized by elemental analyses and by IR, UV-Vis., FAB mass, EI mass and EPR spectroscopy. The molecular structures of 1, 3.DMF and 4.DMF have been determined by single-crystal X-ray diffraction. The mono-Schiff bases are monoanionic and the bis-Schiff bases are dianionic. The octa-coordinated mono-Schiff base complex 4 adopts a dodecahedral geometry, while the hepta-coordinated bis-Schiff base complex 1 forms a one-dimensional linear polymeric chain. A weak antiferromagnetic exchange interaction (J=-0.15 cm(-1)) between the Mn(II) ions in is attributed to weak Mn...Mn interaction through the PhNH(2) moiety of the ligand, as indicated by extended-Hückel molecular orbital calculations. A good simulation of the EPR spectrum of a frozen solution (DMSO at 4 K) of compound 1 was obtained with g=2.0, D=0.1 cm(-1), E=0.01 cm(-1). The EPR spectrum of a powdered sample of compound 1 shows a large broadening of the signal, due in part, to the important zero-field splitting of the hepta-coordinated Mn(II) ion.  相似文献
