The absolute configuration of rohitukine, isolated from the stem-bark of Dysoxylum binectariferum, was determined to be 5,7-dihydroxy-2-methyl-8-[4-(3S, 4R-3-hydroxy-1-methyl)-piperidinyl]-4H-l-benzopyran-4-one by X-ray crystallographic analysis on the crystal of,4-bromobenzoyl derivatives of rohitukine. At the same time, the modified Mosher method was proved to be unsuitable for determining the absolute configuration of C-3‘ position in rohitukine.  相似文献

Enantiomers of β-lactams bearing aryl, furyl or styryl substituents in the 4-position were chromatographically separated by means of high-performance liquid chromatography on chiral column packed with amino acid-derived chiral stationary phase. Separation factors are generally modest. To improve further the resolution of enantiomers, the rings of these β-lactams were opened with octanol in acidic conditions and converted into N-3,5- dinitrobenzoyl ester derivatives of the resulting β-amino acids. Enantiomers of these derivatives are efficiently separated on an amide-derived chiral stationary phase. The chromatographic separations enable accurate determination of optical purity of the chiral β- lactams, prepared from homochiral ester enolate-imine condensation. The absolute configuration of the major enantiomer of the β-amino acid derivatives was determined from elution order on a chiral column.  相似文献

采用毛细管电泳(CE)手性分离方法对8种不同取代基的芳香醇对映体样品的光学纯度进行分析。通过对手性选择剂β-CD-NH3Cl的浓度、背景电解质的浓度、pH值及操作电压等影响因素的研究,优化了分离条件,成功地对8种芳香醇对映体样品进行了手性拆分;同时,对8种对映体样品进行了光学纯度检查,并与HPLC测定结果进行比较。结果表明,3批样品中对映体过量(e.e.%)测定值与HPLC法的测定结果相一致。该方法简单、准确,可用于该8种不同取代基芳香醇的手性拆分和e.e.%值的测定。  相似文献

An LC method was developed and validated for the enantioselective separation and enantiomeric impurity quantitation of atenolol. Separation of the atenolol enantiomers on the Chirobiotic V2 (150 mm × 4.6 mm, 5 μm) column was best achieved using a ternary mobile phase of methanol–acetonitrile-triethylamine acetate 0.5% (w/v), pH 4.5 in a ratio of (45:50:5; v/v/v). Good resolution value of R s  = 3 was obtained at a flow rate of 1 mL min?1 within a total run time of less than 40 min. Peak identification was achieved using the standard reference of individual enantiomers. The peak of the impurity was eluted in front of the peak of the main enantiomer. Detection was performed by UV at 226 nm. Within and between day’s repeatabilities for both retention time and peak area were investigated at three concentration levels and found to be low. The method was also found to be efficient for the determination of atenolol enantiomeric impurity. An impurity quantitation level of (R)-atenolol down to 0.08% relative to the main enantiomer (S)-atenolol was found possible.  相似文献

Polyhydroxy enyne compound (+)-(1'S, 2R, 3S, 5S, 6S)-5,6-dimethoxy-5, 6-dimethyl- 2-(1'-hydroxylpropyl-2-ne)-3-vinyl-l,4-dioxane has been synthesized from D-(-)-tartaric acid. A new chiral center was established by nucleophilic addition with 87% de. The modified Mosher's method was employed to confirm the absolute configuration of 17, which assigned the S-configuration at the new chiral center.  相似文献

Chiral spirocyclic compounds have attracted the attention of scholars and scientists owing to their potential applications in the pharmaceutical industry as either active pharmaceutical ingredients, catalysts in synthesizing active enantiomers, or as surface modifiers on silica particles to resolve entantiomers. In this study, five spiro compounds of 3,9-diphenyl-2,4,8,10-tetraoxaspiro[5.5]-undecane(1), 3,9-(4-methoxyphenyl)-2,4,8,10-tetraoxaspiro [5.5] -undecane(2), 3,9-(4-methylphenyl)-2,4,8,10-tetraoxaspiro [5.5] -undecane(3), 4,4'-(2,4,8,10-tetraoxaspiro[5.5]undecane-3,9-diyl)dibenzoic acid(4) and 3,9-di(4-formyl-phenyl)-2,4,8,10-tetraoxa-spiro[5.5]-undecane(5) were synthesized by grinding pentaerythritol with benzaldehyde, 4-methoxybenzaldehyde, 4-methylbenzaldehyde, 4-carboxybenzaldehyde or terephthalaldehyde monoacetal in the presence of InI3r3 under solvent-free conditions. A normal phase HPLC method was successfully developed to resolve entantiomers of compounds 1--5 on a chiral column. Specific optical rotation of R or S entantiomers(1) was determined and the corresponding configurations were proposed based on Lowe's rule.  相似文献

The conversion of the four aminoindanol stereoisomers with salicylaldehyde to the corresponding Schiff base allows analysis of the dr based on a change in the UV maximum at 420 nm that is very different for the homo- and heterochiral diastereomers and of the concentration of the sample using a hypsochromic shift of another absorption band around 340 nm that is independent of the analyte stereochemistry. Subsequent in situ formation of CuII assemblies in the absence and presence of base enables quantification of the er values for each diastereomeric pair by CD analysis. Applying a linear programming method and a parameter sweep algorithm, we determined the concentration and relative amounts of each of the four stereoisomers in 20 samples of vastly different stereoisomeric compositions with an averaged absolute percent error of 1.7 %.  相似文献

Two pentameric foldamers, Q5 and Q5C-S , containing a C−F bond were synthesized based on quinoline oligamide foldamers for the measurement of enantiomeric excess and for the determination of absolute configuration of chiral amines, diamines, amino alcohols, and α-amino acid esters. Chiral induction of Q5 was triggered in situ when the chiral analytes reacted with the C−F bond in Q5 by a N-nucleophilic substitution reaction, leading to a linear correlation between the CD amplitude at the region of quinoline chromophores and the ee values of the chiral analytes, which can be used for the ee determination of chiral analytes. Furthermore, the CD intensity of Q5C-S containing a chiral motif at its C-terminus enhances via remote, favorable chiral communication when the chiral induction was triggered in situ by chiral analytes at the N-terminus matches the original helicity of Q5C-S , but decreases via remote, conflicted chiral communication when the chiral induction is triggered in situ by chiral molecules at the N-terminus mismatches the original one. The system can thus be used for determination of the absolute configuration of chiral analytes, given that the chirality of the chiral motif at the C-terminus of Q5C-S is known.  相似文献

A new method for the quantitative determination of the enantiomers of salsolinol, an endogenous neurotoxin, was reported. Enantiomeric separation of salsolinol was obtained by capillary zone electrophoresis using hydroxypropyl-β-cyclodextrin (HP-β-CD) as chiral selector. Coupled with UV detection, this separation was applied to the determination of R,S-salsolinol in dried banana chips, and both R- and S-salsolinol were found at 40 ktg/g level.  相似文献
