以3.0 μm无孔单分散亲水性交联聚甲基丙烯酸环氧丙酯树脂为基质,与三嗪染料活性蓝F3GA(Cibacron Blue F3GA)反应,制备出 一种固定化染料聚合物高效亲和色谱填料。考察了应用该填料时流动相中的盐离子浓度、有机溶剂及流速等对牛血清白蛋白(BSA)和 溶菌酶(Lys)保留行为的影响。实验结果表明,该染料亲和填料具有良好的色谱性能。利用前沿色谱法测定了染料与溶菌酶之间的表观 解离常数为5.26×10-5 mol/L。使用该填料成功地从鸡蛋清和小牛血清中分别分离纯化了Lys和BSA,十二烷基硫酸钠-聚丙烯酰胺凝胶 电泳(SDS-PAGE)分析显示Lys和BSA的纯度分别为95%和92%。  相似文献

Hydroxyethyl methacrylate and hydroxypropyl methacrylate (both having extremely high solubilities in water) were polymerized in aqueous medium to obtain the respective polymer latices with a solid content as high as 10 wt.-%. The initial state of the polymerization is in solution rather than in dispersion, and the polymer product is sparingly soluble in the aqueous phase. The polymerization was carefully controlled to avoid forming hydrogel by using an oil soluble initiator and a mixture of sodium dodecyl sulfate and poly(vinyl alcohol). Solubilities of both monomers and polymers in water were also investigated.  相似文献

采用反相悬浮交联法制备壳聚糖微球,对微球进行羟丙基氯化及氨基化,并偶联色素配体Cibacron Blue F3GA,得到一种新型染料亲和吸附剂.以牛血清白蛋白(BSA)为目标蛋白,考察了该染料亲和吸附剂的吸附性能,发现其对BSA有较高的吸附量(95.2mg/g),吸附行为满足Langmuir吸附等温式.负载牛血清白蛋白的微球容易洗脱,洗脱率高达99%.  相似文献

Surface grafting of a polymerizable monomer onto Au was used to produce nanometer-scale planar hydrogel films with controllable volume. A self-assembled monolayer of 11-mercaptoundecanoic acid on a planar Au surface was activated through water-soluble carbodiimide and N-hyroxysuccinimide followed by reaction with 2-aminomethacrylate to produce a methacrylate-terminated surface layer, which readily polymerized under UV radiation in the presence of hydroxyethyl methacrylate monomer, ethylene glycol dimethacrylate cross-linker, and a photoinitiator. The reaction steps were characterized by external reflection mode Fourier transform IR spectroscopy. Under controlled UV exposure, thin (3 nm < d < 10 nm) hydrogel films were obtained from 1:1 ethanol/H(2)O. Surface plasmon resonance measurements were used to characterize both the synthesis of the hydrogel and the potential-induced volume changes. The nanometer-scale hydrogels thus produced undergo reproducible changes in thickness, when a potential is applied across the film. The adsorption of poly(ethylene glycol) (PEG) and ammonium poly(methacrylate) (APMA) onto alumina has been examined both individually and in combination. The adsorption density of APMA was found to be higher than that of PEG onto alumina. The adsorption isotherms of PEG and APMA for alumina exhibited a Langmuirian behavior. The adsorption density of PEG was significantly reduced in the presence of APMA, but the reverse was not true. About 60% desorption of PEG from alumina was achieved, while in the case of APMA the amount desorbed was only 10% in the pH range of 3-6. The zeta potential values of alumina were decreased and the isoelectric point (i.e.p.) values were shifted toward acidic pH values, proportional to the concentration of APMA added. However, such changes in the electrokinetic behavior were not observed by the addition of PEG. The dispersion behavior of alumina in the combined presence of PEG and APMA essentially followed the trends obtained for the alumina-APMA system, corroborating the electrokinetic measurements. Coprecipitation tests confirmed complexation between aluminum species and APMA in the bulk solution, but not with PEG. The interaction between alumina and PEG is primarily governed by hydrogen-bonding forces, while both hydrogen bonding and chemical interaction are involved in the case of the alumina-APMA system. FTIR spectroscopic studies provided evidence in support of the interaction mechanisms proposed.  相似文献

Hydrophilic three-dimensional polymer networks (hydrogels) were prepared from hydroxyethyl methacrylate (HEMA) and the copolymer HEMA and glycerol monomethacrylate (GMMA). The equilibrium water content of the hydrogels in water was investigated as a function of the initial dilution of the polymerization mixture, the type of solvent, and the hydrophilicity of the polymer. The initial dilution was found to have a decisive effect on the swelling or deswelling of hydrogels after the completion of the gel formation. With relatively less hydrophilic hydrogels, there is a critical initial dilution to produce the gel which does not swell or deswell in water after the gel formation. This “isovolumic” initial dilution shifts toward a higher dilution as the hydrophilicity of the hydrogels increases; however, when hydrophilicity of the polymer rises above a certain point, gels always swell in water. Permeability of oxygen through hydrogels was also studied.  相似文献

Hydroxyapatite (HA) has many applications in medicine as a biocompatible and bioactive biomaterial. Numerous studies have shown that modification of the HA surface can improve its biological and chemical properties. However, little is known about the surface properties of modified materials. In this paper the influence of organic polymers: polyethylene glycol (PEG) and polyhydroxyethyl methacrylate (pHEMA) on the surface properties and surface chemistry of hydroxyapatite (HA) is presented. The surface properties of modified HA were characterised by the FT-IR, XPS, BET, and zeta potential measurements. Specific surface area was determined by BET. Infrared and XPS spectra confirmed the presence of PEG and pHEMA on the surface of HA. The BET N2 adsorption revealed slight changes in the HA surface chemistry after grafting modification. The surface chemical properties of the HA were considered to be based on the zeta potential. The decrease in zeta potential results in the increasing stability of the modified material and also in the reduction of bacterial adhesion. The reaction for surface modification of HA is proposed and described.  相似文献

The present study examined poly(2‐hydroxyethyl methacrylate) (PHEMA)‐based hydrogels that have been extensively used in biomedical applications, including contact lens. In this research, we aimed to reduce adsorption of protein components from tears and bacterial deposition by surface modification of the hydrogel with different functional groups that included carboxylic acid, primary amine, and quaternary ammonium. The PHEMA was treated with a solution of sulfuric acid for partial hydrolysis of the HEMA ester groups to induce acid groups on the surface of the hydrogel. Carboxylic acid groups of the modified PHEMA were converted to primary amine and quaternary ammonium groups via carbodiimide chemistry. The surface physical and chemical properties of different samples were investigated by atomic force microscopy and X‐ray photoelectron spectroscopy, respectively. We conducted the bicinchoninic acid assay to evaluate protein deposition from artificial tear fluid on samples. Antibacterial properties of the modified hydrogels were investigated with a culture of Staphylococcus aureus, one of the major causes of eye infections. Our data showed that positively charged amine and ammonium groups efficiently resisted protein adsorption and bacterial deposition compared to alcohol and carboxylic acid groups.  相似文献

Polyethersulfone (PES) membranes are prevalent in the field of water treatment owing to their exceptional separation efficiency, robust mechanical properties, and resistance to chemical degradation. Nevertheless, these membranes are prone to fouling, resulting in a decrease in both flux and ultrafiltration efficiency. In the present study, PES membranes are blended with poly (3-Sulfopropyl Methacrylate) (PSPMA) in various weight percentages (0%–3%) to improve their antifouling and ultrafiltration properties. The physicochemical properties of the blended membranes, including surface morphology, contact angle, hydrophilicity and surface energy are evaluated. The findings indicate that incorporation PSPMA results in an enhancement of the hydrophilic properties and surface charge of the PES membranes, assessed by employing Bovine Serum Albumin (BSA) as a representative protein. Modified blended membranes display greater Flux Recovery Ratio (FRR%) and exhibit superior fouling resistance. Under the same experimental conditions (0.2 MPa applied pressure), a pure water flux of 154.18 L·m−2·h−1 for PES/PSPMA membrane found substantially greater than pure PES membrane (103.52 L·m−2·h−1) along with Total Fouling Ratio (TFR) of 36% and 64.9% respectively. Exceptional antimicrobial efficacy for modified membranes is revealed against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) using disc diffusion technique rendering them well-suited for water treatment applications.  相似文献

Modification of capillary electrophoresis (CE) capillaries by poly(hydroxyethyl methacrylate) (poly(HEMA), poly(diethylene glycol monomethacrylate) (poly(DEGMA) and poly(triethylene glycol monomethacrylate) (poly(TEGMA), was studied. Methods based on physical adsorption of the modifier and on its chemical binding were compared on the basis of the electroosmotic flow (EOF) reproducibility, the EOF dependence on the pH, the symmetry of the peak of positively charged tyramine, the stability of the coating and the separation of standard and milk proteins in the modified capillaries. Reproducible coatings were obtained by chemical binding of the polymers to the capillary walls and by coating with a solution of a polymer, as also demonstrated by the atomic force microscopy.  相似文献

A new kind of copolymeric hydrogel adsorbent containing hydrophilic groups that both provides swelling in water and chelates with uranyl ions was synthesized, and its adsorptive ability for recovering uranium from aqueous media was investigated. The uranyl adsorption capacities of poly(2‐hydroxyethyl methacrylate/maleic acid) hydrogels were determined with a polarographic technique to be 3.2–4.8 (mg UO/g dry gel) from a 15‐ppm uranyl nitrate solution at pH, 6 depending on the molar content of maleic acid in the hydrogel. Adsorption studies showed that other stimuli, the temperature, and the ionic strength of the solution also have important roles in the uranyl‐ion adsorption capacity of these hydrogels. © 2000 John Wiley & Sons, Inc. Lysozyme adsorption onto a dye ligand (Procion Red HE-3B) immobilized and Cu(II) incorporated poly(2-hydroxyethylmethacrylate) (pHEMA) membrane, were investigated. The membranes were prepared by UV initiated photopolymerization of HEMA in the presence of an initiator (α-α′-azoisobutyronitrile; AIBN). The amount of immobilized dye on the membrane was 112.2 μmol g−1. Lysozyme adsorption on to these membranes from aqueous solutions containing different amounts of lysozyme at different pH was investigated in batch system. Lysozyme adsorption capacity of the dye-ligand immobilized membrane was 45.6 mg g−1. Lysozyme adsorption capacity of the Cu(II) incorporated membranes (112.3 mg g−1) was greater than that of the Procion Red HE-3B immobilized membranes. The non-specific adsorption of lysozyme on the pHEMA membranes was 0.14 mg g−1. More than 97% of the adsorbed lysozyme were desorbed in 60 in the desorption medium containing 1.0 M KSCN at pH 8.0.  相似文献

The effects of adsorption of poly(lauryl methacrylate) (PLMA), a side‐chain crystalline polymer, on silica were investigated. Fourier transform infrared spectroscopy and differential scanning calorimetry (DSC) measurements were made on both bulk and adsorbed PLMA. The reversible heat flow rates were observed as a function of temperature and the degree of crystallinity of the samples determined based on the broad melting transitions of the side chains in the surface samples. It was found that adsorption caused a disruption of the side‐chain crystallinity primarily in the tightly bound layer of the polymer, but did not significantly affect its glass transition temperature. A change in the packing of the hydrophobic side chains, as a result of adsorption, was also observed for the tightly adsorbed polymer. These results indicated that PLMA side chains in proximity to the silica surface have different properties from those in bulk PLMA. © 2017 Wiley Periodicals, Inc. J. Polym. Sci., Part B: Polym. Nonspecific interaction is a key parameter affecting the efficiency of proteins, nucleic acids or cell separation. Currently, many approaches to introduce antifouling properties to materials have been developed. Among these, surface modification with polymer brushes plays a prominent role. The aim of this study was to synthesize new magnetic microspheres grafted with poly(N,N‐dimethylacrylamide) (PDMA) that resist nonspecific protein adsorption. Monodisperse macroporous poly(2‐hydroxyethyl methacrylate) (PHEMA) microspheres, 4 μm in size, were synthesized by a multiple swelling polymerization method. To render the microspheres magnetic, iron oxide was precipitated inside the microsphere pores. Functional carboxyl groups, introduced by the hydrolysis of the 2‐(methacryloyl)oxyethyl acetate (HEMA‐Ac) comonomer, were used to react with propargylamine, followed by coupling of a chain transfer agent via an azide‐alkyne click reaction. PDMA was grafted from the PHEMA microspheres using reversible addition‐fragmentation chain transfer polymerization (RAFT), resulting in surfaces with more than 81 wt % PDMA attached. The successful modification of the microspheres was confirmed by XPS. The free-radical polymerization of methyl methacrylate in the presence of chlorine-containing complexes of Fe(III) with 5,10,15,20-tetrakis(3′,5′-di-tert-butylphenyl)porphyrin and 5,10,15,20-tetrakis(3′-butoxyphenyl)porphyrin, as well as in the presence of the acetate complex of Co(III) 5,10,15,20-tetrakis(3′,5′-di-tert-butylphenyl)porphyrin, has been investigated. The kinetic features of the process and the molecular mass characteristics of polymers are studied, and a feasible polymerization mechanism is proposed.  相似文献

The present study is aimed to investigate the degree of crystallinity of poly(3-hydroxybutyrate) P(3HB) grafted with poly(2-aminoethyl methacrylate hydrochloride) (PAEMA) chains using WAXS, micro Raman, and FTIR spectroscopy. The samples were obtained by radiation induced graft polymerization of the monomer in the substrate using different solvents for comparison. The results of crystallinity are consistent with those obtained of lower crystallinity in grafting copolymer relative to the substrate P(3HB). The low crystallinity is directly related to the increase of the degree of grafting, meaning that although the P(3HB) amorphous region is grafted, the crystalline zone is also affected in some extent by the grafting process and the environment of the new molecule. Three different methods were surveyed to determine the variation of crystallinity degree with the grafting degree. It is shown that all methods provide linear relationships between these variables, but WAXS method was found more acceptable than the others (FTIR and Raman). A detailed characterization of the vibrational bands characteristic of amorphous and helical crystalline structure of the grafting copolymers are also highlighted.  相似文献
