Crystal and molecular structures of (3Z)-(±)-4-(2′-hydroxypropyl)amino-and (3Z)-4-(2′-hydroxyethyl)amino-pent-3-en-2-ones

An X-ray study of (3Z)-(±)-4-(2′-hydroxypropyl)amino-and (3Z)-4-(2′-hydroxyethyl)amino-pent-3-en-2-ones is reported. The bond lengths inside the H ring are equalized due to the classical N-H...O hydrogen bond between the carbonyl oxygen and the amino group. In the 4-(2′-hydroxyethyl)amino-pent-3-en-2-one crystal, due to the classical N-H...O bonds, infinite zigzag chains are formed along the 0b axis and arranged into a layered structure due to the weak C-H...O interactions. In (±)-4-(2′-hydroxypropyl)aminopent-3-en-2-one crystal, however, centrosymmetric dimers are formed, which are then linked by weak C-H...O intermolecular interactions to form a layered structure along the a0b plane.

反相高效液相色谱检测2-(3-硝基-4-联苯基)丙腈和2-(4-氨基-3-硝基苯基)丙腈

氟比洛芬是一种高效低毒的消炎镇痛药物,该产品属国内空白,主要依赖进口.我们设计了以苯乙腈为起始原料合成氟比洛芬的新工艺路线,该路线中形成的中间体2-(3-硝基-4-联苯基)丙腈是导致最终产品收率及产品纯度的关键因素.对原料2-(4-氨基-3-硝基苯基)丙腈及中间体的高效液相色谱保留行为的研究文献尚未见报道.本文用反相高效液相色谱研究了两种化合物的保留行为,优选出了最佳分离条件,对指导实际生产具有一定意义.     

Synthesis of 4-(4-Methylsulfonylphenyl)-3-phenyl-2(3H)-thiazole Thione Derivatives as New Potential COX-2 Inhibitors

Synthesis of novel 4-（4-methylsulfonylphenyl）-3-phenyl-2（3H）-thiazole thione derivatives with functionalized diarylheterocycle pharmacophore as potential COX-2 inhibitors was described. The title compounds were synthesized by cyclocondensation of corresponding dithiocarbamate and 2-bromo-1-（4-methylsulfonylphenyl）ethanone, followed by dehydration with H2SO4. All of the target compounds were characterized by ^1H NMR, IR and mass spectral data.     

Structures, energetics, and aromaticities of the tetrasilacyclobutadiene dianion and related compounds: (Si4H4)(2-), (Si4H4)(2-)·2Li+, [Si4(SiH3)4](2-)·2Li+, [Si4(SiH3)4](2-)·2Na+, and [Si4(SiH3)4](2-)·2K+

In light of the important recent synthesis of a stable tetrasilacyclobutadiene dianion compound by Sekiguchi and co-workers and the absence of theoretical studies, ab initio methods have been used to investigate this dianion and a number of related species. These theoretical methods predict multiple minima for each compound, and most minima contain folded and bicyclic silicon rings. For (Si(4)H(4))(2-), (Si(4)H(4))(2-)·2Li(+), [Si(4)(SiH(3))(4)](2-)·2Li(+), [Si(4)(SiH(3))(4)](2-)·2Na(+), and [Si(4)(SiH(3))(4)](2-)·2K(+), respectively, the energetically lowest-lying structures are designated A-3 (C(2v) symmetry), B-8 (C(1) symmetry), C-1 (C(2) symmetry), D-1 (C(2) symmetry), and E-1 (C(2h) symmetry). None of these structures satisfies both the ring planarity and the cyclic bond equalization criteria of aromaticity. However, all of the representative NICS values of these lowest-lying structures are negative, indicating some aromatic character. Especially, structures C-1 and D-1 of C(2) symmetry effectively satisfy the criteria of aromaticity due to the slightly trapezoidal silicon rings, which are nearly planar with nearly equal bond lengths. SiH(3) substitution for hydrogen in (Si(4)H(4))(2-)·2Li(+) significantly reduces the degree of aromaticity, as reflected in the substantially smaller NICS absolute values for [Si(4)(SiH(3))(4)](2-)·2Li(+) than those of (Si(4)H(4))(2-) and (Si(4)H(4))(2-)·2Li(+). The aromaticity is further weakened in [Si(4)(SiH(3))(4)](2-)·2Na(+) and [Si(4)(SiH(3))(4)](2-)·2K(+) by replacing lithium with the sodium and potassium cations.     

4-甲氧基-N-(4-(3-吗啉基丙氧基)苯基)-3-(4-(吡啶-3-基)嘧啶-2-基氨基)苯甲酰胺与人血清白蛋白的相互作用研究

应用分子对接模拟技术结合荧光猝灭法、同步荧光法、三维荧光法和紫外-可见光谱法,模拟生理条件(p H 7.4),在298 K和310 K温度下,研究了人血清白蛋白(HSA)与4-甲氧基-N-(4-(3-吗啉基丙氧基)苯基)-3-(4-(吡啶-3-基)嘧啶-2-基氨基)苯甲酰胺(1z)之间的相互作用。发现1z的加入引起了HSA内源荧光的静态猝灭,蛋白构象发生改变。理论研究与实验结果相结合研究了蛋白与药物的作用机理,从而为进一步寻找治疗慢性粒细胞白血病的药物小分子提供了实验依据。

以酸酐及有机惰性溶剂作混合溶剂，少量叔胺等作催化剂，2-(4-氯苯基)-3-甲基丁酸能在比较温和的条件下基本完全消旋，收率接近90％。     

新型磷酸钒孔道结构化合物(H3NCH2CH2NH3)3- [ (VO)4(PO4)2 (HPO4)4]的水热 合成及结构表征

以有机分子乙二胺作为模板剂合成了新型磷酸钒孔道化合物(H3NCH2CH2NH3)3^-[(VO)4(PO4)2(HPO4)4,并通过X射线单晶衍射实验进行了结构表征,晶体学数据为：C2/c,a=1.8505(9)nm,b=0.7089(4)nm,c=2.3304(10)nm,β=96.43(3)°,V-3.038(3)nm^3,Z=8,R=0.067,Rw^b=0.1635,该化合物具有非常独特和规整的二维孔道骨架结构,进一步的晶体化学研究表明该化合物为一新的VPO物相。

2-(4-甲硫苯氧基)-3-(4-甲苯基)-4H-咪唑啉-4-酮衍生物的合成及杀菌活性(Ⅱ)

用烯基膦亚胺与4-甲苯基异氰酸酯、对甲硫苯酚的串联aza-Wittig反应合成了2-(4-甲硫基苯氧基)-3-(4-甲苯基)-4H-咪唑啉-4-酮衍生物.探讨了反应进行的条件和产物的波谱性质,提出了可能的环化反应机理.生成的环化产物均为新的化合物,其结构经元素分析、IR、1H NMR和MS确证.并探讨了所合成的新型杂环化合物的生物活性,结果表明部分化合物表现出良好的抑菌活性,其中3f的活性最好,在质量分数为5×10-5浓度时,对稻瘟菌和苹果轮纹菌的抑制率均达到100%.

4-(N-乙酰氨基)-3-(4-芳基噻唑-2-氨基)苯甲酸乙酯的合成与生物活性

以3-氨基-4-乙酰氨基苯甲酸乙酯为原料设计合成了18种4-(N-乙酰氨基)-3-(4-芳基噻唑-2-基)-苯甲酸乙酯新化合物.化合物结构经质谱,元素分析,1H NMR和13C NMR确证.生物活性实验结果表明,化合物3d,3h,3p(40μg/mL)对神经氨酸酶(NA)的抑制率分别为36.02%,33.40%,42.05%;化合物3g,3h(500mg/L)对纹枯病菌的抑制率为50%.
