3-Methyl-6H-[1,2,4]triazolo[4′,3′: 4,5] [1,3,4]thiadiazolo[2,3-b]quinazolin-6-one (6) has been synthesized by the condensation of isatoic anhydride (1) with 4-amino-5-mercapto-3-methyl-[1,2,4]triazole (2) and final cyclisation of the intermediate3 with POCl3 and PCl3. Alternatively6 could also be synthesized by the condensation of 3-amino-2-mercapto-3H-quinazolin-4-one (7) withN-carbethoxy hydrazine in presence of hydrochloric acid and final cyclisation of the intermediate8 with acetic acid. The structures have been confirmed on the basis of IR, PMR and analytical results.  相似文献

The key 3-(2-oxo-2H-chromen-3-yl)-2-oxo-2H,5H-pyrano[3,2-c]chromen-5-yl acetates 3 were synthesized in high yields by cyclocondensation of 4-oxo-4H-chromen-3-carbaldehydes 1 with coumarin-3-acetic acids 2 under mild conditions. The reaction pathway involves aldol condensation and subsequent intramolecular lactonization to afford 2-oxo-2H,5H-pyrano[3,2-c]chromene skeleton 3. Further treatment of acetates 3 with alcohols, water or nitrogen containing compounds led to 5-alkoxy-, 5-hydroxy- or 5-acylamino-2H,5H-pyrano[3,2-c]chromen-2-ones 4-6 via nucleophilic substitution of acetyloxy group at C-5. Acetates and hydroxyl derivatives 3 and 5 undergo facile rearrangement in an acid medium yielding 5-hydroxypyrano[2,3-b]chromen-2(10aH)-ones 7. Twelve prepared compounds were evaluated on their antineoplastic activities on 60 human tumour cell line panels in NCI USA. The obtained biological results confirmed that 3-(2-oxo-2H-chromen-3-yl)-2H,5H-pyrano[3,2-c]chromen-2-one represents a new leading skeleton suitable for further antitumour activity study.  相似文献

A series of tetrazolomethylbenzo[d][1,2,3]triazole derivatives (214) have been synthesized and evaluated as antimicrobial agents from 1H-benzo[d][1,2,3]triazole (1) as starting material. The reaction of benzotriazole 1 with chloroacetonitrile afforded 2-(1H-benzo[d][1,2,3]-triazol-1-yl)acetonitrile 2, which was reacted with sodium azide to give tetrazole derivative 3. Esterification of benzotriazole 1 with ethyl bromoacetate in the presence of anhydrous potassium carbonate afforded ester 4, which was treated with hydrazine hydrate to afford the corresponding hydrazide 5. Reaction of 3 with 2,3,4,6-tetra-O-acetyl-α-d-glucopyranosyl bromide afforded the nitro-glycoside derivative 6, which was deacetylated using methanolic ammonia to deprotected nitroglycoside 7. The hydrazide 5 was reacted with 4,5,6,7-tetrachlorophthalic anhydride or 1,2,4,5-benzenetetracarboxylic dianhydride in refluxing glacial acetic acid to give the corresponding imides 8 and 9, respectively. Also, the hydrazide 5 was reacted with carbon disulphide in ethanol to give potassium salt 10, which was reacted with hydrazine hydrate to afford aminotriazole derivative 11. The latter compound was reacted with carbon disulphide to afford thiadiazole derivative 12, which was treated with 2,3,4,6-tetra-O-acetyl-α-d-glucopyranosyl bromide to give the thioglycoside derivative 13. Deacetylation of the thioglycoside 13 using methanolic ammonia solution at room temperature afforded the deprotected thioglycoside 14. The antimicrobial screening of some synthesized compounds showed that many of these compounds have good antimicrobial activities comparable to streptomycin and fusidic acid as reference drugs.  相似文献

A series of new unsymmetrical 2,5-disubstituted 1,3,4-oxadiazoles (3ad) has been synthesized to evaluate their antibacterial and thermal properties. All compounds have been tested for their in vitro antibacterial activity against two Gram-positive bacteria namely, Staphylococcus aureus and Bacillus subtilis. Among the tested compounds, compound 2-(4-chlorophenyl)-5-(thiophen-2-yl)-1,3,4-oxadiazole (3c) has been found to be most potent member having minimum inhibitory concentration. Thermal stability and melting point of compounds have been studied by TG and DSC analysis in air atmosphere at heating rate of 10 °C min?1. Thermal degradation kinetics of the most potent antibacterial compound 3c has been carried out by multiple heating rate model free kinetic methods namely, Ozawa–Flynn–Wall, modified Coats-Redfern, and Kissinger.  相似文献

10-Nitro-6.11-dihydro-dibenz[b,e]oxepine-11-one (12) was obtained both from 2-nitro-6-phenoxymethyl-benzoic acid (8) by dehydration and from 2-nitro-6-phenoxymethyl-benzoyl chloride (10) by Friedel—Crafts-reaction.12 was reduced to the amino compound15 which yielded 10-aminoacetylamino-6.11-dihydro-dibenz[b, e]oxepine-11-one (19) on reaction with chloroacetyl chloride and subsequent treatment with ammonia. The cyclization of19 yielded the title compound (21). The behaviour of the ketons12 and15 against NH2OH·HCl was investigated.  相似文献

A new series of di- and tri-organotin(IV) compounds with the general formula R4?n SnL n , where R?=?Me (1,2), Et (3), n-Bu (4,5), n-Oct (6), Ph (7) and L?=?(E)-3-(4-methoxyphenyl)-2-(4-chlorophenyl)-2-propenoate, were synthesized by reaction of silver salt of ligand or ligand acid with diorganotin dichloride/oxide and triorganotin chloride in 2:1 and 1:1 molar ratio, respectively. These compounds were characterized by elemental analyses, FT-IR, multinuclear (1H, 13C, 119Sn) NMR and mass spectrometry. The spectroscopic results revealed that all the diorganotin(IV) compounds possess trigonal bipyramidal structures in solution and octahedral geometry in the solid state around the tin atom. A linear polymeric trigonal bipyramidal structure in the solid state and a tetrahedral environment around the tin atom in non-coordinating solvents has been proposed for the triorganotin(IV) compounds. All synthesized compounds were tested in vitro against a number of microorganisms to assess their biocidal activity. These studies revealed that ligand acid and some of its organotin compounds show promising activity against different strains of bacteria and fungi but lowered than reference drugs.  相似文献

Three coumarin derivatives, 7-hydroxy-3-(2-pyridyl)coumarin (HPC), 7-(4-(5-(4-tert-butylphenyl)-1,3,4-oxadiazol-2-yl)benzyloxy)-3-(2-pyridyl)coumarin (Ox-PC), and 7-(4-(9H-carbazol-9-yl)butoxy)-3-(2-pyridyl)coumarin (Cz-PC), were synthesized and characterized by elemental analysis, 1H NMR, FT-IR, and UV?Cvis absorption spectra. The fluorescence behaviors of the compounds in methanol solutions and solid states were investigated. HPC exhibits weak green emission, whereas Cz-PC and Ox-PC show strong blue emissions in dilute solutions.  相似文献

Because of the well-known chemotherapeutic activity of pyrimidoquinolines, a new series of the title compounds were synthesized and biologically screened for their antimicrobial and anti-oxidant activities. The intermediate compound, 2-amino-1-cyclohexyl-4-(3,4-dimethoxyphenyl)-1,4,5,6,7,8-hexahydroquinoline-3-carbonitrile (2), was prepared and allowed to react with oxalyl chloride to give the corresponding strategic starting material, 10-cyclohexyl-5-(3,4-dimethoxyphenyl)-4-oxo-3,4,5,6,7,8,9,10-octahydropyrimido[4,5-b]quinoline-2-carbonyl chloride 3. Compound 3 underwent diverse reactions with amines to give the corresponding carboxylic amides 4, with alcohols to give the carboxylic esters 5 and with hydrazine to give the carbohydrazide 8. Compound 8 reacted with aldoses to give the corresponding polyhydroxy(-)alkyl Schiff bases 9, which upon reaction with thioglycolic acid afforded the thiazolidinone-C-acyclic nucleosides 10. Several other pyrimidoquinolines incorporated to oxadiazole, iminothiazolidinones and thiadiazoles were synthesized starting from the 2-carbohydrazide derivative 8. Most of the newly prepared derivatives showed considerable antimicrobial activity against Gram +ve and Gram ?ve bacteria and anti-oxidant activity.  相似文献

We describe the design, synthesis, and biological activities of 5-chloro-2-(substituted phenyl)benzo[d]thiazole derivatives as novel tyrosinase inhibitors. Among them, 4-(5-chloro-2,3-dihydrobenzo[d]thiazol-2-yl)-2,6-dimethoxyphenol (MHY884) and 2-bromo-4-(5-chloro-benzo[d]thiazol-2-yl)phenol (MHY966) showed inhibitory activity higher than or similar to kojic acid, against mushroom tyrosinase. Therefore, we carried out kinetic studies on the two compounds with potent tyrosinase inhibitory effects. Kinetic analysis of tyrosinase inhibition revealed that all of these compounds are competitive inhibitors. MHY884 and MHY966 effectively inhibited tyrosinase activity and reduced melanin levels in B16 cells treated with ??-melanocyte stimulating hormone (??-MSH). These data strongly suggest that the newly synthesized compounds MHY884 and MHY966 could suppress production of melanin via inhibition of tyrosinase activity.  相似文献

A number of novel and synthetically important pyridyl/naphthyl/(diphenyl)methylseleno substituted alkanoic acids (20-25) have been synthesized using an efficient and operationally simple strategy. Starting substrates, ethyl pyridyl/naphthyl/(diphenyl)methylseleno substituted alkanoates (8-13) were easily prepared by treatment of ethyl chloroalkanoates 7(a-c) with nucleophilic selenium reagent RSeNa+, generated from the cleavage of dipyridyl/dinaphthyl/bis(diphenylmethyl) diselenide (1-6) with sodium borohydride in ethanol. The ethyl pyridyl/naphthyl/(diphenyl)methylseleno substituted alkanoates (8-13) on basic hydrolysis and subsequent acidification afford pyridyl/naphthyl/(diphenyl) methylseleno substituted alkanoic acids (20-25) in excellent yields. These selenoalkanoates (8-13) and selenoalkanoic acids (20-25) have been characterized by elemental analysis and various spectroscopic techniques viz. NMR (1H, 13C and 77Se), IR and mass spectrometry. The molecular structure of 2-pyridylselenoethanoic acid (20a), 2-naphthylselenoethanoic acid (23a) and 2-(diphenyl)methylselenoethanoic acid (24a) has also been established with the help of single crystal X-ray analysis.  相似文献

Heteroaryl substituted analogs of antirhnoviral (A), was prepared by a convergent approach. 3-Nitrophenyl-5- bromooromethylisoxazoles 5a–b were synthesized by [3+2] cycloaddition of 3-(benzoyloxy)-propyne 2 to in situ generated arylnitrile oxides followed by deprotection of cycloadducts 3a–b and bromination of the resulting alcohols 4a–b. Coupling of 3- nitrophenyl-5-bromooromethylisoxazoles (5a–b) with 4-[5-(2-alkyl-2H-tetrazolyl)]phenols (6a–d) in N-methylpyrrolidinone under mild conditions afforded a new series of 2-alkyl-5-{4-[1-(3-nitrophenyl-5-isoxazolyl)methyloxy]phenylr}-2H-tetrazoles (7a–h) in high yields. The structures of the synthesized compounds were confirmed by their 1H NMR, Mass spectral, and Elemental Analysis data.  相似文献

3-(Naphthalen-1-ylmethylene)-5-phenylfuran-2(3H)-one 1 was prepared and converted into a variety of heterocyclic systems of synthetic and biological importance. Benzylamine was reacted with furanone 1 to afford compounds 2 and 3 according to the reaction conditions. Butanamide 2 was reacted with thionyl chloride or thiourea to give derivatives 4 and 5, respectively. Compound 3 was reacted with ethyl cyanoacetate to give the corresponding pyrrolopyridine derivative 6. Treatment of 1 with hydrazine hydrate afforded compounds 7 and 8 according to the reaction conditions. Also, compound 1 was reacted with phenyl hydrazine, hydroxyl amine, malononitrile or thiourea to give compounds 912, respectively. Cyclization of 7 with ethoxymethylene-malononitrile, ethyl-(ethoxymethylene)cyanoacetate, carbon disulphide or acetylacetone afforded the corresponding compounds 1316, respectively. Condensation of 7 with p-nitrobenzaldehyde gave the corresponding hydrazone 17, which was treated with thioglycolic acid or chloroacetyl chloride to give compounds 18 and 19, respectively. Also, most of the prepared products were tested for anti-avian influenza virus and revealed promising antiviral activity against H5N1 virus [A/Chicken/Egypt/1/2006 (H5N1)] by determination of both TC 50 and ED 50 and confirmed by plaque reduction assay on MDCK cells. Compounds 7, 8, 11, 12 and 13 showed the highest effect compared with the other tested compounds.  相似文献

4-Alkylaminopyridinethiones · HCl (1 · HCl) react with bis-trichlorethylmalonate (3) predominantly to 5-alkylamino-4H-thiopyrano [2,3-b]pyridine-4-ones (6). With alcohols in the presence of acids at 25°C6 undergoes an alcoholysis to the corresponding alkyl-3-(2-thioxo-3-pyridyl)propionates (9). On heating in dilute alkali6 is hydrolysed via 4-alkylamino-2-thioxopyridyl-propylketones (11) to the tautomers, 4-hydroxy-2-thioxopyridylpropylketone (12 A) and 2-thioxo-3-(1-hydroxybutenyl)-4-piperidon (12 B), resp. On refluxing with alkali the ethyl-pyridylpropionate9 a is cyclisized to the 1-alkyl-1,6-naphthyridine-2(1H)-one (4 a), but boiling in ethanolic acid hydrolyses9 a via the pyridylpropionic acid10 to 4-alkyl-aminopyridylpropylketone (11 a). The latter can be transformed via the tautomers12 A,B and 2-methylthio-3-pyridylpropylketone (13) to the 4-hydroxy-3-butyrylpyridone (14 A) and its tautomer, 3-(1-hydroxy-butenyl)-piperidine-2,4-diones (14 B) resp. The structure of14 A,B is established by reaction of 4-isopropylamino-2(1H)-pyridone (2) with butanoylchloride to the 4-isopropylamino-3-butyrypyridone (15) and hydrolysis of15 to the tautomers14 A,B.  相似文献

2H-Imidazole-4(3H)-thiones (a), available from methyl alkyl and methyl aryl ketones with sulfur and ammonia, react via their corresponding N-sodium compounds or in presence of tert. amines with alkyl and aryl carboxylic acid chlorides to give the corresponding intensely coloured (orange to violett) cryst. 3-acyl-2H-imidazole-4(3H)-thiones4 a-q and6–26. With dicarboxylic acid dichlorides the colourless cryst. N,N′-diacyl-bis-3-imidazoline-5-thiones5 a-d and27–32 are obtained. With carbamic acid chlorides and chloroformic acid esters the corresponding urea (33–35) and urethane derivatives36, 37 are formed. In an analogous way 2H-imidazol-4(3H)-ones react with acid chlorides to 3-acyl-2-imidazol-4(3H)-ones (44–50), which can also be obtained by treating the corresponding 3-acyl-2H-imidazole-4(3H)-thione with KMnO4.  相似文献

Action of 2.4-(or 2.6-)xylenol on4 a resp.3 b yields 2-amino-6-[2(or 4)-hydroxy-3.5-dimethylphenyl]-4-methyl-(resp. 4-phenyl)-3.4.5.6-tetrahydro-1H-pyrimidiniumchlorides (8 a resp.8 b or9 a resp.9 b), which are transformed to the zwitterionic compounds10 a–11 b by aqu. NaOH.6 a reacts with 2.4-xylenol to give the triazaoxabenzanthraceniumchlorid12 a·HCl (prove for the structure given for6 a). A series of 5-(trifluoromethyl)-N-alkyl-1-(3-phenylisoquinoline-1-yl)-1H-pyrazole-4-carboxamides 4 has been effectively achieved in high yield and purity from the reaction of pyrazole carboxylic acid 2 with amines 3 in the presence of TBTU as a catalyst and diisopropyl ethylamine as a base in acetonitrile at room temperature.  相似文献

4,12- and 4,14-Dibromo-[2.2]metacyclophane (1 and2) were prepared by coupling of 1-bromo-2,4-bis(bromomethyl)-benzene with phenyllithium (isomeric ratio 1∶1). Lithiation of the bromides and subsequent carboxylation afforded the isomeric carboxylic acids3 and4. 4,12-Dimethyl-[2.2]metacyclophane (13) is also accessible with 90% stereoisomeric purity by reaction of 2,4-bis-(chloromethyl)toluene (20) with the Li-salt of 4-methyl-isophthalaldehyde bis(propylene thioacetal) (24) and subsequent desulphurization with Raney-Ni. In this context it was found that bromination of 1,2,4-trimethylbenzene affords 2,5- andnot (as stated in the literature) 2,4-bis(bromomethyl)toluene. The separation of isomers can be accomplished on a preparative scale by column chromatography and for analytical purposes by high pressure liquid and/or gas chromatography. Structural assignments are based on the different symmetries of 4,12- and 4,14-disubstituted [2.2]metacyclophanes belonging to point groupsS 2 andC 2, resp., which account for the different types of spin systems of the bridge protons in the1H-NMR-spectra.  相似文献

Hydrolysis of the 4-alkyliminothiopyrano[2,3-b]pyridinedioles (5) and 4-alkylaminothiopyrano[2,3-b]pyridones (6) resp. with 10% NaOH gives 5,7-dihydroxy-2H-thiopyrano[2,3-b]pyridine-4(3H)-one (7).7 can be obtained in better yield by reaction of 4-dimethylamino-2(1H)-pyridinethione (8) with bistrichlorphenylethylamlonate (2). Aminolysis of7 affords the two isomeric products5 and6. On treatment with hydrazines,7 reacts only to 4-hydrazonoderivatives5. By heating in bromobenzene5d is cyclisized to 1H-5,1,2,6-thiatriaza-acenaphthylen-7-ol (11). On methylation with methyljodide5,6 and7 furnish the 7-methoxyproducts13,14 and12. By heating in 20% NaOH7 is transformed into the 2-thioxo-3-pyridylmethylketone16 A and its tautomer, 2-mercapto-3-pyridylmethylketone16 B. The structures of5,6 and7 are discussed.  相似文献

In the present study, thermal stabilities of five new family of azocalix[4]arene mono ethyl ester derivatives, 4ae, were investigated using thermogravimetry, differential thermogravimetry, and differential thermal analysis methods. It was found that all compounds showed thermal stability up to 236 °C averagely. After this temperature, decomposition of compounds starts gradually. The decomposition routes of 4ac compounds are similar and occur with two stages. Ester alkyl groups decompose and remove from the structure in the first stage. Second stage corresponds to rest of structure decomposition. The decomposition routes of the 4de compounds are different from the decomposition routes of the 4ac compounds. These compounds include halogen, and decomposition reactions realize with three and four stages respectively.  相似文献

N,N′-Diisopropyl-substituted 1,3,4-oxadiazolidine3c, 1,3,4-thiadiazolidine6, 4,5-benzo-1,2,3,6-tetrahydropyridazine8, and new 3,4-dialkyl-1,3,4-oxadiazolidines9–14 were synthesized and studied. The configurational stability of the N atoms does not change on going from compound3c to itsS-analog6 and decreases in the case of pyridazine8. 3,4-Di-tert-alkyl-1,3,4-oxadiazolidines3d and11–14 were found to be promising objectes for optical resolution.  相似文献
