Cross-coupling reactions have become indispensable tools for creating carbon-carbon (or heteroatom) bonds in organic synthesis. Like in other important transition metal catalyzed reactions, such as metathesis, addition, and polymerization, unsaturated compounds are usually employed as substrates for cross-coupling reactions. However during the past decade, a great deal of effort has been devoted to the use of alkyl halides as saturated compounds in cross-coupling reactions, which has resulted in significant progress in this undeveloped area by introducing new effective ligands. Many useful catalytic systems are now available for synthetic transformations based on C(sp(3))-C(sp(3)), C(sp(3))-C(sp(2)) and C(sp(3))-C(sp) bond formation as complementary methods to conventional C(sp(2))-C(sp(2)), C(sp(2))-C(sp) and C(sp)-C(sp) coupling. This tutorial review summarizes recent advances in cross-coupling reactions of alkyl halides and pseudohalides catalyzed by a palladium complex.  相似文献

The catalytic activity of a PCN-type palladium pincer complex is evaluated in the construction of C(sp(2))-C(sp(2)) and C(sp(2))-C(sp(3)) bonds by Suzuki-Miyaura cross-couplings employing nontypical substrates such as benzyl halides, alpha-haloenones, or alkylboronic acids as coupling partners. A method for the cross-coupling of alkyl electrophiles with various potassium aryl- and heteroaryltrifluoroborates has been developed. Nearly stoichiometric amounts of organoboron species could be employed to cross-couple a large variety of challenging heteroaryl nucleophiles. Several functional groups were tolerated on both the electrophilic and the nucleophilic partners. Chemoselective reactivity of C(sp(3))-Br bonds in the presence of C(sp(2))-Br bonds was achieved.  相似文献

A general protocol for direct transformation of unreactive C(sp(3))-H bonds to C(sp(3))-CN bonds has been developed. The C-H activation was effected by photoexcited benzophenone, and the generated carbon radical was subsequently trapped with tosyl cyanide to afford the corresponding nitrile in a highly efficient manner. The present methodology is widely applicable to versatile starting materials and, thus, serves as a powerful tool for selective one-carbon elongation for construction of architecturally complex molecules.  相似文献

Detailed investigation on the origin of the acidity of the alpha-protons of a set of the carbonyl molecules was carried out on the basis of properties of the localized molecular orbital. An anomalously high acidity of Meldrum's acid, as compared with those of dimedone and dimethyl malonate, is one of the well-known but unresolved issues. The well-localized sigma orbitals of the C-H bonds at the alpha-position of the carbonyl groups can be obtained with the reactive hybrid orbital (RHO) theory. We found that the energy levels of the unoccupied RHOs of the C-H moiety of Meldrum's acid and other carbonyl compounds showed a good linear correlation with the experimental deprotonation energies. This is probably because the deprotonation reaction to form the proposed naked anions in a polar solvent is a highly endothermic process, in which the thermodynamic energy differences between the neutral molecules and the corresponding anions approximately coincide with the activation energies. We also investigated the effect of the conformational change upon deprotonation on the electron-accepting energy level of the relevant C-H bonds of cyclic/acyclic and monocarbonyl/dicarbonyl compounds. A conformational change occurs in the cases of cyclic six-membered compounds, but its influence on the reactivity of the C-H bond is small. The acidity of dicarbonyl compounds, including Meldrum's acid, showed a good correlation with the deviations from the perpendicular position of the dihedral angles of the relevant C-H bond with respect to the adjacent carbonyl C=O bond. This angle parameter can be related to the magnitude of the in-phase orbital interaction between the sigma(CH) and pi(C)(=)(O) orbitals, which facilitate electron acceptance. These results indicated that the acidity of the alpha-proton of carbonyl compounds can be represented in terms of the electron-accepting orbital levels of the unoccupied RHO of the C-H moiety. All the linear relationships found in the present work strongly suggested that the acidity of Meldrum's acid, which is conventionally regarded as an anomaly, is consistent with those of the other carbonyl compounds.  相似文献

Although metal-catalyzed direct arylation reactions of non- or weakly acidic C-H bonds have recently received much attention, chemists have relied heavily on substrates with appropriately placed directing groups to steer reactivity. To date, examples of intermolecular arylation of unactivated C(sp(3))-H bonds in the absence of a directing group remain scarce. We report herein the first general, high-yielding, and scalable method for palladium-catalyzed C(sp(3))-H arylation of simple diarylmethane derivatives with aryl bromides at room temperature. This method facilitates access to a variety of sterically and electronically diverse hetero- and nonheteroaryl-containing triarylmethanes, a class of compounds with various applications and interesting biological activity. Key to the success of this approach is an in situ metalation of the substrate via C-H deprotonation under catalytic cross-coupling conditions, which is referred to as a deprotonative-cross-coupling process (DCCP). Base and catalyst identification were performed by high-throughput experimentation (HTE) and led to a unique base/catalyst combination [KN(SiMe(3))(2)/Pd-NiXantphos] that proved to efficiently promote the room-temperature DCCP of diarylmethanes. Additionally, the DCCP exhibits remarkable chemoselectivity in the presence of substrates that are known to undergo O-, N-, enolate-, and C(sp(2))-H arylation.  相似文献

The intramolecular Friedel-Crafts acylation of arenes with Meldrum's acid derivatives catalyzed by Sc(OTf)3 was reported as a mild and general entry into functionalized 1-indanones. Mechanistic investigations were undertaken to determine the rate-determining step in the acylation sequence using Meldrum's acid, as well as to examine the role of the Lewis acid catalyst. Enolizable Meldrum's acid derivatives react via an acyl ketene intermediate under thermal conditions, while quaternized Meldrum's acid derivatives are thermally stable and only act as effective Friedel-Crafts acylating agents in the presence of a Lewis acid catalyst. The acylation was postulated to proceed through direct acylation of a Lewis acid-activated carbonyl. In the catalytic Friedel-Crafts acylation of Meldrum's acids, triflic acid appeared to be the active catalytic species, with Sc(OTf)3 serving as a very mild and convenient reagent for its delivery.  相似文献

We report the efficient synthesis of alkyl ethers by the functionalization of unactivated sp(3)- and sp(2)-hybridized C-H bonds. In the Pd(OAc)(2)-catalyzed, PhI(OAc)(2)-mediated reaction system, picolinamide-protected amine substrates undergo facile alkoxylation at the γ or δ positions with a range of alcohols, including t-BuOH, to give alkoxylated products. This method features a relatively broad substrate scope for amines and alcohols, inexpensive reagents, and convenient operating conditions. This method highlights the emerging value of unactivated C-H bonds, particularly the C(sp(3))-H bond of methyl groups, as functional groups in organic synthesis.  相似文献
