A novel approach for the synthesis of enamine derivatives of N-protected 3-acyltetramic acids is described. The synthetic procedure relies on α-C-acylation of β-enamino amides with N-protected α-amino acids and subsequent cyclisation of the obtained intermediates in refluxing TFA. The tetramic derivatives are obtained with very good enantiopurity (e.r. ≥95:5). Ring-enlarged analogues (piperidine-2,4-diones) can also be obtained from β-amino acids.  相似文献

We have examined whether the allyl halide cross-metathesis reaction tolerates α-alkoxy amide groups. Ruthenium-based catalysts I-III did not catalyze the cross-metathesis of allyl halides in the presence of an α-alkoxy N,N-dimethylamide group to any appreciable extent, but the reaction could tolerate either a bulky N,N-diisopropylamide or Weinreb amide group. In particular, the Grubbs-Hoveyda-Blechert 2nd generation catalyst (III) efficiently catalyzed the cross-metathesis of allyl halides with olefins bearing a Weinreb amide group.  相似文献

Successive treatment of N-acylsultams 3 with sodium hexamethyldisilazide, 1-chloro-1-nitrosocyclohexane ( 1 ), and aq. HCl gave diastereoisomerically pure, crystalline N-hydroxyamino-acid derivatives 5 . These were converted into various amino acids 7 , N-hydroxyamino acids 8 , and an N-Boc-amino acid 9 . (S, S)-Isoleucine ( 17 ) and (S, S)-2-acetamido-3-phenylbutyric acid ( 23 ) were obtained from N-crotonoylsultam 15 via 1,4-addition of an organomagnesium or organocopper reagent followed by enolate ‘amination’ with 1 .  相似文献

Enantiomerically pure 1-methylene and 1-oxo derivatives (compounds 4 and 10, respectively) of compounds 1 were obtained and studied as precursors of N-acyliminium ions. 1-Substituted-1-hydroxyderivatives, obtained by regioselective syn-addition of organometallics to compounds 10 gave the desired species under acid treatment while compounds 4 did not. N-Phenethyl substituted N-acyliminium ions gave isoquino[1′,2′:3,4]pyrazino[2,1-b]quinazoline-8,11-diones through a Pictet–Spengler-type cyclisation.  相似文献

We synthesized porphyrinylamide 4b and its N-methylated derivative 5b. Direct N-methylation of porphyrinylamides 4 proved unsuccessful, so 5b was obtained via N-methylation of 5-aminoporphyrin 10 with potassium hexamethyldisilazide. The secondary amide 4b exists in trans-amide form, while N-methylated amide 5b exists in the cis-amide form, both in solution and in the crystal. Thus, N-methylation of the amide bond of 4b resulted in trans to cis conformational alteration.  相似文献

A versatile route for the modular synthesis of (R)-3-hydroxyalkanoic acids, constituents of the naturally biodegradable poly(3-hydroxyalkanoate) polymers, and its application to the synthesis of (R)-3-hydroxydecanoic acid is described. Key steps include a microwave-assisted catalytic transfer hydrogenation and a facile microwave-assisted hydrolysis of an N-methoxy-N-methyl (Weinreb) amide, which enhances the practicality of this protecting group for carboxylic acids.  相似文献

The parallel iterative solution-phase synthesis of 5-amino-1-aryl-[1,2,4]triazolo[1,5-a]pyridine-7-carboxylic acid amide derivatives is described. The key intermediate 2,6-bis-aminopyridine-4-carboxylic acid methyl ester was synthesised in a two step procedure in 64% overall yield and elaborated to a variety of triazolopyridine-5-carboxylic acid methyl ester by selective pyridine-N-amination, condensation of the adduct with a wide selection of aldehydes and subsequent cyclisation and oxidation. The desired esters were obtained in yields up to 70%. The final transformation to the amide derivatives was accomplished by application of carefully optimised reaction conditions thus giving access to a library of total 500 triazolopyridine amide derivatives. Iterative synthetic cycles (12-48 library members each) allowing for maximal flexibility in chemistry and maximal efficiency in in vitro biological activity optimisation guided by molecular modelling efforts constitute a synergistic procedure for rapid lead optimisation.  相似文献

A method of convergent synthesis of novel 4,6-unsubstituted 5-acyl-2-aminodihydropyrimidines 7 is developed. The synthetic intermediate of 7, 4,6-unsubstituted 2-aminodihydropyrimidines 9 having a Weinreb amide at the 5-position, is prepared by the sequential Staudinger/aza-Wittig/cyclization reactions of (E)-tert-butyl{3-azido-2-[methoxy(methyl)carbamoyl]allyl}carbamate (E)-10. The transformation of the Weinreb amide of 9 to an acyl group proceeds smoothly by a substitution reaction using aryllithiums or alkyllithiums in the presence of a catalytic amount of BF3 etherate, affording 7 in good to high yield. The N-protecting group of 7 can be easily removed to obtain N-unsubstituted 2-amino-5-acyldihydropyrimidines 8, and the derivatives are observed as a single isomer in 1H NMR spectroscopy. All dihydropyrimidines in this study were hitherto unavailable and difficult to synthesize by conventional methods.  相似文献

Anthranilic acid amide reacts with cyclic anhydrides to give the corresponding N-acyl derivatives at the amino group, while analogous reactions of o-aminobenzohydroxamic acid lead lead to formation of 3-hydroxy-quinazolin-4-ones under mild conditions. N-Acyl derivatives of anthranilic acid amide undergo intramolecular cyclization to imides on microwave irradiation or on melting, and their treatment with acetic anhydride in the presence of sodium acetate on heating yields quinazolin-4-ones.  相似文献

A new synthesis of the furo[3,2-c]quinolin-4(5H)-one heterocycle has been developed using a palladium-catalysed cyclisation of N-(2-iodophenyl)-N-methyl-3-furamide. By varying the catalyst, base and solvent, the yield of the cyclisation was optimised. It was found that the use of palladium oxide with potassium acetate in N,N-dimethylacetamide (DMA) with a small amount of tetrabutylammonium chloride gave the highest yield of 5-methylfuro[3,2-c]quinolin-4(5H)-one (9).  相似文献

(−)-N-Benzyl cytisine has been stereoselectively substituted in moderate to high yields on its carbon 6 (Csp3 α to the pyridone nitrogen). The reaction involved the in situ trapping of the carbanion formed by reaction of lithium diisopropyl amide (LDA) and its reaction with electrophiles (alkyl, allyl, benzyl halides, non-enolizable aldehydes, and Weinreb amide). In the absence of an electrophile or with its addition after the formation of the carbanion, a dimeric structure was isolated (yield: 42%) resulting from the 1,4-addition of the carbanion on the pyridone ring of another cytisine molecule. Deprotection of the benzyl group (Olofson's reagent) allowed the formation of 6-substituted derivatives of the natural product, cytisine, a potent agonist of nicotinic receptors of subtype α4β2.  相似文献

The first total synthesis of janadolide, a new cyclic polyketide-peptide hybrid possessing a tert-butyl group, is described. The synthesis of an unsaturated hydroxycarboxylic acid was effected via the lithiation of vinyl iodide followed by addition to a Weinreb amide with a tert-butyl group and stereoselective 1,2-reduction. The cyclic structure was constructed by macrolactamization at the amide bond between the proline moiety and fatty acid moiety.  相似文献

An investigation was conducted to determine whether the cross-metathesis (CM) of allyl halides tolerates amide groups. The results show that the ruthenium-based complexes IIII serve as poor catalysts for the CM of allyl halides with olefins that contain an N,N-dimethylamide group. In contrast, the Grubbs–Hoveyda–Blechert second generation catalyst (III) efficiently promotes these processes with olefins bearing a Weinreb amide group. Lastly, a reinvestigation of the ester group tolerance of the allyl halide CM with unsaturated esters demonstrated that III serves as an efficient catalyst for these reactions.  相似文献

The synthesis of bis-γ-amino acid dibenzobarrelene derivatives (9,10-bis-aminomethyl-11,12-bis-carboxy-dibenzobarrelene) is presented. Bromomethylation of anthracene followed by azide substitution gave 9,10-bis-azidomethylanthracene. Azide reduction, N-Boc protection, and Diels-Alder cycloaddition in DMAD furnished the protected 9,10-bi-aminomethyl-11,12-dicarboxy-dibenzobarrelene derivative, which was further converted into the bis-γ-amino acid methyl ester, the N-Boc-protected bis-γ-amino methyl amide, and a bis-γ-lactam. Monte Carlo simulations and X-ray analysis of the 9,10-substituted dibenzobarrelenes revealed an exposed hydrophobic surface surrounded by amino and carboxy groups.  相似文献

This report describes the synthesis of enantiomerically pure (S)- and (R)-α-methylserines on a multigram scale, starting from the Weinreb amide of 2-methyl-2-propenoic acid and using a stereodivergent synthetic route that involves a Sharpless asymmetric dihydroxylation reaction. As a synthetic application of these quaternary α-amino acids, they were used as starting materials in the synthesis of the well-known valuable homochiral (S)- and (R)-N-Boc-N,O-isopropylidene-α-methylserinal building blocks.  相似文献

A short, simple and inexpensive synthesis of several diversely substituted benzo[b]thiophenes and one naphthothiophene is described. The method involves introduction of methylsulfanyl group ortho- to the amide function of readily available N,N-diethylamides of aryl carboxylic acid by directed metalation. Thioindoxyls, obtained in high yields through side-chain deprotonation and cyclisation in one pot, are reduced to benzo[b]thiophene or napthothiophene.  相似文献

The synthesis of 1,2-dihydro-1-oxopyrrolo[1,2-d]-1,2,4-triazines was achieved by rearrangement of 2-pyrrolyloxadiazoles under alkaline conditions or by cyclisation of pyrrole N-ethoxymethylidene hydrazides. The cyclisation of the N-carbethoxy hydrazone of the pyrrole-2-carboxaldehyde gave the 3,4-dihydro-4-oxopyrrolo[1,2-d]-1,2,4-triazine. Electrophilic substitution reactions of the 1- and 4-pyrrolotriazinones were made either on the lactam nitrogen with methyl sulphate, benzyl chloride and monochloroacetic acid or on the pyrrole ring with bromine and nitric acid. The structure of the derivatives was determined by 1H nmr.  相似文献
