-, and -cyclodextrin and heptakis-2,6-di-O-methyl--cyclodextrin enhance the nitrosation rate of l-ephedrine if the nitrosation assay procedure (NAP test) is applied. During this reaction with -cyclodextrin a solid inclusion compound of -cyclodextrin andN-nitrosoephedrine precipitates. Solubilities and stabilities of inclusion compounds of the cyclodextrins with ephedrine and nitrosoephedrine, respectively, explain especially the catalytic effects of some cyclodextrins on ephedrine.Part of the PhD thesis of V. Inclusion complexation of all-trans-retinol, retinal and retinoic acid with -cyclodextrin (-CD) and heptakis(2,6-O-dimethyl)--cyclodextrin (DM--CD) were investigated by means of UV-vis spectroscopy. The association constants (K a) obtained for vitamin A with DM--CD is greater than with -CD. On the other hand, for the same host compoundK a values of retinol, retinal and retinoic acid are very close to each other.  相似文献

The 1 : 2 chlorhexidine : -cyclodextrin(Cx : CD) complex was prepared and characterised using X-ray crystallography, infrared spectroscopy, thermal analysis and nuclearmagnetic resonance. The minimum inhibitory concentration (MIC50) of the chlorhexidine : -cyclodextrin inclusion compoundagainst Streptococcus mutans, Eubacterium Lentum, Fusobacterium nucleatum, Bacteroides fragilis andActinomices actinomycetemcomitans was determined. The binding of psychopharmaceutical drugs by -cyclodextrin (-CD) changed their effective electrophoretic mobilities. Based on this phenomenon the interactions between some psychopharmaceutical drugs and -cyclodextrin were studied and the binding constants of -CD with tiapride, imipramine, clomipramine, amitriptyline, trifluoperazine, perphenazine and carbamazepine were determined by affinity capillary electrophoresis. The presence of -CD improved the separation of these psychopharmaceutical drugs. Organic modifier was used as another kind of additive to be added in the running buffer to improve further the separation. 70 mM pH 7.2 phosphate buffer solution containing 12 mM -cyclodextrin and 15% (v/v) acetone was selected as optimal running buffer for separation by capillary electrophoresis.  相似文献

The asymmetric Michael addition of aromatic thiols to 2-cyclohexenone and maleic acid esters has been carried out by utilizing their crystalline cyclodextrin complexes suspended in water. The best chiral induction, 30% enantiomeric excess (ee), was achieved in combinations of 2-cyclohexenone and octyl maleate with the crystalline -cyclodextrin complex of benzenethiol (method A) to afford (S)-3-phenylthiocyclohexanone and (S)-octyl-2-phenylthiosuccinate, respectively, whereas the reaction of benzenethiol with 2-cyclohexenone included in -cyclodextrin (method B) inversely induced the chiral recognition to give the (R)-adduct with 4–9% ee.  相似文献

The interaction of six nonionic surfactants -[4-(1,1,3,3-tetra-methylbutyl)phenyl]--hydroxypoly(oxy-1,2-ethanediyl) with hydroxypropyl--cyclodextrin (HPCD) and dimethyl--cyclodextrin (DIMEB) was studied by reversed-phase thin-layer chromatography in the presence and absence of sodium chloride. Each surfactant formed complexes with both cyclodextrin derivatives; however, the strength of interaction varied considerably. DIMEB formed more stable inclusion complexes with the surfactants than did HPCD. A longer ethyleneoxide chain decreased the strength of interaction, whereas sodium chloride exerted a negligible impact. Principal component analysis indicated that both the hydrophobicity and the specific hydrophobic surface are of the surfactant influenced the complex formation indicating the hydrophobic character of the interaction.Dedicated to Professor József Szejtli.  相似文献

The enantiomeric separation of 12 chiral dihydrobenzofurans is reported on derivatized -cyclodextrin stationary phases using high performance liquid chromatography. The hydroxypropyl -cyclodextrin chiral stationary phase (CSP) with acetonitrile/water mobile phases was the most effective combination as it baseline separated 9 of the 12 compounds. The acetyl -cyclodextrin and 2,3-dimethyl -cyclodextrin CSPs were also effective in the reverse phase mode. The native -cyclodextrin was far less effective than the non-aromatic derivatized CSPs. The aromatic functionalized CSPs showed no selectivity in the normal phase mode. Structural characteristics, such as substituent polarity and ring location, were important in the observed enantioselectivity.  相似文献

The photo-oxidation reaction of phenothiazine has been studied in the presence of-cyclodextrin (-CD) and 2-hydroxypropyl-cyclodextrin (HP -CD). The influence of these organized media on the formation of the oxidation photoproduct upon UV irradiation has been investigated. Phenothiazine forms an inclusion complex with the cyclodextrins. The stoichiometry and formation constant of the complex formed with 2-hydroxypropyl -CD have been calculated using the changes of the fluorescence emission signal and of the absorbance of the drug upon inclusion. An increase of the fluorescence intensity of the photogenerated product is attained when it becomes included inside the cyclodextrin cavity.  相似文献

The effect of -cyclodextrin (-CD) and hydroxypropyl--cyclodextrin (HP--CD) upon the photochemically-induced fluorescence (PIF) properties of four phenylurea herbicides, including linuron, diuron, isoproturon and neburon has been studied. Photochemical conversion of these nonfluorescent herbicides into strongly fluorescent photoproducts was shown to occur in -CD and HP--CD aqueous media. The influence of pH, UV irradiation time and photoproduct stability on the fluorescence intensity was also investigated. In addition, the stoichiometry and formation constants of the complexes formed between herbicides and -cyclodextrin (-CD) or 2-hydroxypropyl--cyclodextrin (HP--CD) were determined. The formation constant values, ranging from 184 ± 40 to 1498 ± 245 M-1, were calculated by applying the iterative nonlinear regression (NLR) approach to the PIF data. Linear calibrations graphs were established in the interval 1–12 g/mL, for diuron, linuron and neburon. The IUPAC limits of detection ranged between 580 and 700 ng/mL, according to the compound. Application to the analysis of phenylurea herbicides in spiked river water was also described.  相似文献

Inclusion complex formation of piromidic acid (PA) with dimethyl--cyclodextrin (DM--CD) in aqueous solution and in the solid state was confirmed by the solubility method, differential scanning calorimetry (DSC) and proton nuclear magnetic resonance (1H-NMR) spectroscopy. The apparent stability constant,K c , of the complex was estimated to be 244 M–1. The stoichiometry of the complex was given as the ratio 1:2 of PA to DM--CD. In an effort to modify the solubility and dissolution rate of the contraceptive steroid, 19-norprogesterone in order to improve its bioavailability, the cyclodextrin complexation approach was chosen. In solution, the complex formation with -cyclodextrin (-CD), hydroxyethyl -cyclodextrin (HE--CD) and hydroxypropyl -cyclodextrin (HP--CD) was confirmed by using solubility, UV, IR and 1H-NMR spectrophotometric techniques. The phase solubility diagrams were categorized as AL-type. The complexing affinity of the CDs investigated were ranked as follows: -CD > HP--CD > HE--CD. The complexation thermodynamic parameters were obtained from the temperature dependence of the dissociation constants. In the solid state, differential scanning calorimetery (DSC) and optical microscopy methods were utilized to characterize the complexes. Dissolution studies showed that such molecularly encapsulated forms offered a marked improvement in the dissolution rate compared to the parent drug.  相似文献

Sodium ion activities were determined potentiometrically in sodium dodecylsulfate and sodium dodecanoate solutions containing-cyclodextrin. Whereas sodium ion did not bind with-cyclodextrin, its activity decreased noticeably when the 1:1-cyclodextrin/surfactant inclusion complex was formed in solution. When the anionic guest was methyl red or methyl orange, no sodium ion binding occurred. When the guest was the cationic dodecylammonium ion in chloride solutions containing-cyclodextrin, pronounced chloride ion binding to the 1:1 inclusion complex occurred.  相似文献

The disposition of cholesterol inside the -cyclodextrin cavity(-CD) was deduced from oxidation of cholesterol secondary alcoholgroups by Ca(OCl)2 and H2O2 in thepyridine–acetic acid system. The amount of cholest-4-ene-3-one formedwas found to be proportional to the concentration of -cyclodextrin,resulting in 56.1% of ketone. The oxidation rate was enhanced by-cyclodextrin and its methyl, polymer and 1 : 1copper(II)–-cyclodextrin derivatives. Detailed investigationsinvolving UV-visible, 13C- and 1H-NMR(T1, 1D NOE and ROESY) spectroscopic studies were carried out.A binding constant value of 15,385 ± 1500 M-2 wasobtained for the 2 : 1heptakis-2,6-di-O-methyl--cyclodextrin(DM-CD) : cholesterolcomplex in chloroform from UV studies. Proton and solid state13C-CP MAS spectra of the -CD–cholesterol mixtureshowed large magnitude shifts for the protons from the wider end of the-CD cavity as well as those of ring A and ring B of cholesterol. Both1D NOE and ROESY measurements indicated the proximity between ring A andring B protons of cholesterol and the wider end protons of -CD andDM-CD. Besides, analysis of c,i and tau;m from T1measurements showed not only a lowering of rotational motions but a value of 0.016–0.048 for some of the cholesterol protons, typical of aweak complex. Based on these studies, a probable structure for the 2 : 1complex involving two molecules of -CD/DM-CD was proposed withportions of ring A and ring B being present inside the wider end of the-CD/DM-CD cavity and ring D and the side chain attached atposition 17, projecting into the wider end of the secondCD/DM-CD molecule.  相似文献

The World Health Organization issued a nitrosation procedure (NAP Test) which allows to carry out nitrosation under standard conditions. It has proved that the in vitro reaction rates of the fast nitrosatable drugs piperazine, cimetidine and ethambutol are not influenced by -, - and -cyclodextrin. On the contrary, -, -cyclodextrin and heptakis-2,6-di-O-methyl--cyclodextrin enhance the nitrosation of the slower nitrosatable 1-ephedrine and fencamfamine significantly. This possible reaction must be considered if nitrosatable drugs are formulated with cyclodextrins to be administered to human beings.  相似文献

The effects of -cyclodextrin (-CyD), heptakis(2,6-di-O-methyl)--cyclodextrin (DMCyD) and heptakis(2,3,6-tri-O-methyl)--cyclodextrin (TMCyD) ontrans-cis photoisomerization of 1-ethyl-4-(4-hydroxystyryl)pyridinium (POH) have been studied in aqueous solutions. The ratio of [cis]/[trans] for POH in the photostationary state at pH 8.54 was remarkably reduced by the presence of CyD or DMCyD. The reduction of the [cis]/[trans] ratio in the photostationary state was explained in terms of the shift of the equilibrium of POH + trans PO trans + H toward PO trans formation. The binding constants of CyD and DMCyD for PO trans were 2.00- and 1.36-fold larger than those for POH + trans , respectively. The binding constants of TMCyD for both species are much smaller than those of CyD and DMCyD. This result indicates that PO trans , which has a betain structure, forms stable complexes with CyD and DMCyD with its hydrophobic parts inside and the charged parts outside the CyD cavities.  相似文献

Neutral cyclosophoraoses (unbranched cyclic -1,2-d-glucans) produced by the Rhizo-bium meliloti 2011 were prepared by size exclusion and anion-exchange chromatographic techniques. The degree of polymerization (DP) of isolated cyclosophoraoses was determined by matrix associated laser desorption/ionization mass spectrometry (MALDI/MS) techniques. A family of purified neutral cyclosophoraoses (DP 17–27) was used as a host for the inclusion complexation with hardly soluble ergosterol. High performance liquid chromatographic (HPLC) analysis showed that it induced much enhanced solubility of ergosterol compared to -cyclodextrin. In order to understand the molecular basis of the complex forming ability of cyclosophoraoses, a Monte Carlo (MC) docking-minimization method was used for host-guest complex formation of cyclosophoraoses or -cyclodextrin with ergosterol. From the MC simulation we propose the `hand-shake' mechanism for complexation of cyclosophoraoses with ergosterol.  相似文献

Polygodial is a sesquiterpene drimane isolated from the genus Drimys, whichexhibits anti-asthmatic, anti-allergic, anti-inflammatory and antinociceptive effects.We have prepared a polygodial--cyclodextrin inclusion complex for furtherpharmacological studies. The inclusion complex was synthesized by co-precipitationand analyzed by Thermogravimetric Analysis, showing a decrease in the number ofwater molecules of hydration in relation to the native -cyclodextrin. DifferentialThermogravimetric Analysis indicated a peak corresponding to the evaporation ofpolygodial. With Differential Scanning Calorimetry, the melting peak to polygodial was not observed, however, there was an increase in the energy of vaporization of the water molecules in relation to native -cyclodextrin. Using a Scanning Electron Microscopy a clear difference in the morphology of crystals of the inclusion complex and native -cyclodextrin could be seen. The association constant between polygodial and -cyclodextrin, measured by UV spectroscopy was 1,006 M-1 at 37 °C, pH 7.0 and ionic strength 0.2 M, following stoichiometry 1:1.  相似文献

The influence of cyclodextrins (CDs) on the chemical stability of the contraceptive steroid progestin, ST1435, in aqueous solution has been studied using reversed phase high performance liquid chromatography. The effects of CD structure, temperature, and CD concentration on the rate of degradation were investigated. It was found that the drug degraded to different extents following a pseudo-first order reaction mechanism. The presence of the host molecules affected the degradation rate as a result of complexation which might result in protection of the labile moiety of the drug molecule against degradation. Hydroxypropyl--cyclodextrin (HP--CD) and hydroxyethyl--cyclodextrin (HE--CD) retarded the degradation in contrast to -cyclodextrin (-CD) which accelerated the steroid degradation. The stabilizing action of HP--CD is larger than that of HE--CD. The degradation rate increased upon increasing temperature and the Arrhenius equation is valid. Lineweaver-Burk equation analysis indicated that the steroid included inside the CD cavity degraded three times more slowly than did the free ST1435 in solution. This equation further supported the formation of a 1 : 1 inclusion complex between ST1435 and HP--CD with a stability constant of 934.5 M-1 at 65°C.  相似文献

Iomeglamic acid has been successfully applied for radioscopy of the gall-bladder since 1972. This pharmacon has the disadvantageous property that it is practically insoluble in water. Preparations containing the drug in different concentrations were made with-cyclodextrin by different methods, and their dissolution characteristics were determined. In the best situation the solubility increased ten-fold. The tablets were made from the most suitable complex, with-cyclodextrin. The conditions of tableting, the parameters and the dissolution of the tablets were tested.  相似文献
