We describe a method for detecting DNA methylation. It is based on direct oxidation of DNA bases at a glassy carbon electrode (GCE) modified with film of a multiwalled carbon nanotube-β-cyclodextrin composite. This nano-structured film causes a strong enhancement on the oxidation current of DNA bases due to its large effective surface area and extraordinary electronic properties. Well-defined peaks were obtained as a result of electro-oxidation of guanine (at 0.67 V), adenine (at 0.92 V), thymine (at 1.11 V), cytosine (at 1.26 V), and 5-methylcytosine (at 1.13 V; all data vs. saturated calomel electrode (SCE)). The potential difference between 5-methylcytosine and cytosine (130 mV) is large enough to enable reliable simultaneous determination and analysis. The interference by thymine can be eliminated by following the principle of complementary pairing between purine and pyrimidine bases in DNA. The modified electrode was successfully applied to the evaluation of 5-methylcytosine in a fish sperm DNA, the methylation level of cytosine was found to be 7.47 %, and the analysis process took less than 1 h.  相似文献

An electrochemical sensor was developed based on gold nanoparticles incorporated in mesoporous MFI zeolite for the determination of purine bases. Au nanoparticles (AuNPs) were incorporated into the mesoporous MFI zeolite (AuNPs/m‐MFI) by post‐grafting reaction. The composite materials were characterized by transmission electron microscopy (TEM), X‐ray photoelectron spectroscopy (XPS) and electrochemical methods. Au nanoparticles with a size of 5‐20 nm are uniformly dispersed in the pores of mesoporous MFI zeolite. And the morphology of MFI zeolite can be perfectly kept after pore expansion and Au nanoparticles incorporation. The electrocatalytic oxidation of purine bases (guanine and adenine in DNA) is investigated by cyclic voltammetry (CV) and differential pulse voltammetry (DPV). The surface‐confined Au nanoparticles provide the good catalytic activity for oxidation of purine bases. The simultaneous detection of guanine and adenine can be achieved at AuNPs/m‐MFI composites modified glassy carbon electrode (GCE). The electrochemical sensor based on AuNPs/m‐MFI exhibits wide linear range of 0.5–500 μM and 0.8–500 μM with detection limit of 0.25 and 0.29 μM for guanine and adenine, respectively. Moreover, the electrochemical sensor is applied to evaluation of guanine and adenine in herring sperm DNA samples with satisfactory results.  相似文献

A nano-material carboxylic acid functionalized graphene (graphene-COOH) was prepared and used to construct a novel biosensor for the simultaneous detection of adenine and guanine. The direct electrooxidation behaviors of adenine and guanine on the graphene-COOH modified glassy carbon electrode (graphene-COOH/GCE) were carefully investigated by cyclic voltammetry and differential pulse voltammetry. The results indicated that both adenine and guanine showed the increase of the oxidation peak currents with the negative shift of the oxidation peak potentials in contrast to that on the bare glassy carbon electrode. The electrochemical parameters of adenine and guanine on the graphene-COOH/GCE were calculated and a simple and reliable electroanalytical method was developed for the detection of adenine and guanine, respectively. The modified electrode exhibited good behaviors in the simultaneous detection of adenine and guanine with the peak separation as 0.334V. The detection limit for individual determination of guanine and adenine was 5.0×10(-8)M and 2.5×10(-8)M (S/N=3), respectively. Furthermore, the measurements of thermally denatured single-stranded DNA were carried out and the value of (G+C)/(A+T) of single-stranded DNA was calculated as 0.80. The biosensor exhibited some advantages, such as simplicity, rapidity, high sensitivity, good reproducibility and long-term stability.  相似文献

Hybrid silica films containing cation‐exchange polyelectrolytes were designed and used to modify a glassy carbon electrode (GCE) for voltammetric determination of purine bases. Hybrid silica‐polyelectrolyte films synthesised in the presence of adenine as structure directed component have demonstrated enhanced sorption capacity to purine base. The anodic peaks of adenine and guanine at a hybrid film‐modified GCE were observed at +1.55 and +1.1 V, respectively, in phosphate buffer solution at pH 3.5. Oxidation currents of adenine and guanine were proportional to their concentration in the range of 0.02–0.50 mM, with a detection limit of 0.015 mM. The developed method was used to determine adenine in adenosine triphosphate and purine bases in hydrolyzed solutions of DNA and demonstrated good metrological characteristics.  相似文献

The fabrication of poly(2,6-pyridinedicarboxylic acid)/MWNTs modified glass electrode(PPDA/MWNTs/GCE) was proposed and used for individual or simultaneous determination of guanine and adenine.The performances of the PPDA/MWNTs/GCE were characterized with cyclic voltammetry(CV).The modified electrode exhibited enhanced electrocatalytic behavior and good stability for the detection of guanine and adenine.Differential pulse voltammetry(DPV) was used to determine the concentration of guanine,adenine.The detection limit(S/N = 3) for guanine and adenine was 0.045μmol/L and 0.05μmol/L,respectively.The electrochemical method for the measurement of guanine and adenine in calf thymus DNA was also developed with this modified electrode and the result was satisfactory.  相似文献

A large mesoporous carbon modified glassy carbon electrode (LMC/GCE) was prepared. The morphology and structure of the LMC were characterized. The LMC/GCE was used to investigate the electrochemical behaviors of metabolites of purine nucleotide, uric acid (UA), xanthine (XA) and hypoxanthine (HX). The LMC/GCE exhibited high electrocatalytic activity towards the three compounds when compared with those obtained at the GCE. Furthermore, the LMC/GCE realized simultaneous determination of UA, XA and HX at a physiological pH of 7.0 with wide linear range and low detection limit. The electrocatalytic activity of the LMC/GCE towards guanine (G) and adenine (A) was also investigated.  相似文献

A simple, rapid and accurate method for the simultaneous determination of four purine and pyrimidine bases (cytosine, 5-methylcytosine, adenine and N6-methyladenine) has been developed. The quantitative determination of these bases was accomplished by ion chromatography (IC) with direct conductivity detection (CD) based on their ionization in acidic medium without chemical suppression. The recovery of cytosine, 5-methylcytosine, and adenine in calf thymus DNA was more than 98% (n=3) and the relative standard deviation (RSD, n=5) less than 2.4%. In a single chromatographic run, the four bases could be separated and determined in less than 10 min. The detection limits were found to be 0.05 microg/mL for cytosine, 0.08 microg/mL for 5-methylcytosine, 0.07 microg/mL for adenine, and 0.07 microg/mL for N6-methyladenine. Linear ranges were 0.2-95.1 microg/mL for cytosine (r2=0.9996), 0.3-196.6 microg/mL for 5-methylcytosine (r2=0.9994), 0.3-105.5 microg/mL for adenine (r2=0.9998), and 0.3-159.1 microg/mL for N6-methyladenine (r2=0.9999). With the proposed method, purine and pyrimidine bases could be successfully detected in calf thymus DNA. We also determined these bases in calf thymus DNA using RP-HPLC. Compared to RP-HPLC, the IC method offers advantages such as high selectivity and simple mobile phase.  相似文献

A copper nano particles and cerium (IV) oxide modified carbon nanotube based composite on glassy carbon electrode (Cu−CeO2/MWCNT/GCE) was fabricated for simultaneous determination of guanine and adenine. The surface morphology, chemistry and conductance of the prepared electrodes were characterized by scanning electron microscopy (SEM), energy dispersion X‐ray (EDX), X‐Ray photoelectron spectroscopy (XPS) and electrochemical impedance spectroscopy (EIS). The Cu−CeO2/MWCNT/GCE improved electrochemical behaviour of guanine and adenine compared to other electrodes. The modified electrode was also used for individual and simultaneous determination of guanine and adenine. Under optimized conditions, the calibration curves were obtained linearly in the range of 0.20 to 6.00 μM for the guanine and 0.10 to 8.0 μM for the adenine by differential pulse voltammetry. The limits of detection of guanine and adenine were calculated as 0.128 and 0.062 μM, respectively. Interferences studies were also performed in the presence of inorganic and organic compounds. Moreover, the determination of guanine and adenine contents were carried out in a calf thymus DNA sample by the developed method with satisfactory results.  相似文献

Dopamine-melanin colloidal nanospheres (Dpa-melanin CNDs)–graphene composites-modified glassy carbon electrode (GCE) was prepared by a simple procedure and then successfully used to simultaneously determine guanine and adenine. Scanning electron microscopy (SEM) images and transmission electron microscopy (TEM) were used to characterize the morphology of the Dpa-melanin CNSs–graphene composite. Cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) were used to characterize the electrode modifying process. Differential pulse voltammetry (DPV) was used to study the electrocatalytic activity toward the electrochemical oxidation of guanine and adenine. The modified electrode exhibited enhanced electrocatalytic behavior and good stability for the simultaneous determination of guanine and adenine compared with bare GCE. The electrochemical biosensor exhibited wide linear range of 0.5 to 150 μM with detection limit of 0.05 and 0.03 μM for guanine and adenine detection (S/N?=?3), respectively. Furthermore, the biosensor showed high sensitivity, good selectivity, good reproducibility, and long-term stability to guanine and adenine detection. At the same time, the fabricated electrode was successfully applied for the determination of guanine and adenine in denatured DNA samples with satisfying results. These results demonstrated that Dpa-melanin CNSs–graphene composite was a promising substrate for the development of high-performance electrochemical biosensor.  相似文献

An inorganic film of ruthenium hexachloroplatinate (RuPtCl6), deposited on the surface of a glassy carbon electrode, exhibits electrocatalytic activity in the oxidation of purine bases, such as adenine and guanine. Appropriate working conditions are found for fabricating a polymer film on the surface of glassy carbon and for recording the maximum electrocatalytic effect for the modified electrode. A method is developed for the selective voltammetric determination of guanine and adenine in their simultaneous presence on an electrode modified by a RuPtCl6 film. A procedure is proposed for the amperometric detection of purine bases with this modified electrode under the conditions of flow-injection analysis. The dependence of the analytical signal on the concentration of guanine and adenine is linear up to 5 × 10?6 and 5 × 10?7 M in the stationary mode and to 5 × 10?7 and 5 × 10?8 M under flow conditions, respectively. The proposed method was tested in the analysis of calf thymus DNA for the concentrations of guanine and adenine.  相似文献

Highly boron-doped diamond (BDD) electrode, have been examined for simultaneous detection of purine and pyrimidine bases in mild acidic media by using HPLC with amperometric detection. Cyclic voltammetry at as-deposited (AD) and anodically oxidized (AO) BDD were used to study the electrochemistry and to optimize the condition for HPLC applications. At AO BDD electrode, due to its higher overpotential of oxygen evolution reaction, well-defined anodic peaks were observed for the oxidation of purine and pyrimidine bases in acid medium, whereas at AD BDD the oxidation peak of thymine was overlapped with the anodic current of oxygen evolution. The chromatograms of adenine, guanine, cytosine, thymine and 5-methylcytosine mixture were well resolved by using a silica-based column and a solution of 5% acetonitrile in 100 mM ammonium acetate buffer (pH 4.25) as the mobile phase. The detection was carried out at AO BDD electrode at an applied potential of 1.6 V versus Ag/AgCl. Linear calibration curves were obtained within the concentration range from 0.1 to 10 μM with the limits of detection (S/N = 3) ranging from 26.3 to 162.1 nM, resulting in an order of magnitude higher sensitivities than those at conventional electrodes. HPLC analysis with diamond amperometric detector was successfully applied for determination of 5-methylcytosine in real DNA samples with high reproducibility. No deactivation of the electrode was found during cyclic voltammetric and HPLC measurements, indicating the high stability for analysis of biological samples.  相似文献

A nanostructured film electrode, a multi-wall carbon nanotubes (MWNT)-modified glassy carbon electrode (GCE), is described for the simultaneous determination of guanine and adenine. The properties of the MWNT-modified GCE were investigated by scanning electron microscopy (SEM) and cyclic voltammetry. The oxidation peak currents of guanine and adenine increased significantly at the MWNT-modified GCE in contrast to those at the bare GCE. The experimental parameters were optimized and a direct electrochemical method for the simultaneous determination of guanine and adenine was proposed. Using the MWNT-modified GCE, a sensitive and direct electrochemical technique for the measurement of native DNA was also developed, and the value of (G+C)/(A+T) of HCl-digested DNA was detected.  相似文献

Two kinds of TiO(2) nanobelts were prepared from commercial P-25 powders via an alkaline hydrothermal method with and without an acid etching process. The uncauterized nanobelts (TNs) exhibited a smooth surface, and mixed phases of anatase and TiO(2) (B), whereas the cauterized ones (CTNs) displayed a rough surface and a pure anatase structure. TNs and CTNs were then deposited onto a glassy carbon electrode (GCE) surface with a conductive adhesive (CA), and the resulting chemically modified electrodes exhibited electrocatalytic activities in the oxidation of nucleobases in a 0.1 M phosphate buffer solution (PBS) at pH 7.4. For guanine and adenine, well-defined oxidation peaks were observed in voltammetric measurements at about +0.62 and +0.89 V, respectively, at a potential sweep rate of 100 mV s(-1), whereas for cytosine, uracil and thymine, the voltammetric features were not obvious. The average surface coverages (Γ) of guanine and adenine on the CTNs/CA/GCE electrode were estimated to be 4.75 × 10(-10) and 7.44 × 10(-10) mol cm(-2), respectively, which were about twice those at the TNs/CA/GCE electrode. The enhanced activity of the CTN-based electrode towards purine nucleobase oxidation was ascribed to the large specific surface area and anatase structures with enhanced (001) facets of the CTN that facilitated adsorption of the analytes onto the electrode surface and charge transport through the electrode surface layer.  相似文献

 用高效液相色谱法测定了核酸水解的中间产物及最终产物 6种嘌呤、嘧啶碱基 ,探讨了色谱柱、流动相等对其分离的影响 ,确定了最佳色谱条件为 :HypersilBDS C18柱 ,乙腈 0 1mol/LKH2 PO4 (H3 PO4 调节 pH至4 0 5 ) (体积比为 2∶98)作流动相 ,紫外检测器在 2 6 0nm波长下检测。方法的精密度在 3%以内 ,回收率在 82 %~ 114%。方法应用于酵母核酸样品的测定中 ,取得了很好的结果。  相似文献

A glassy carbon electrode (GCE) was modified by electrochemically reduced graphene oxide (ERGO) for subsequent dsDNA immobilization. The interaction of cisplatin with dsDNA was studied at this modified electrode. Quantitative investigations were performed by adsorptive transfer stripping voltammetry (AdTSV) using differential pulse voltammetry (DPV). The morphology and structure of graphene oxide (GO) and ERGO modified GCEs (GO/GCE and ERGO/GCE, respectively) were characterized by UV‐vis, FT‐IR, Raman spectroscopy and cyclic voltammetry. Compared with the bare GCE and the GO/GCE, the ERGO/GCE exhibited excellent electrocatalytic activity towards the oxidation of dsDNA due to guanine and adenine groups, testified by high oxidation peak currents and decreased oxidation potentials. The interaction of micromolar concentrations of cisplatin with surface confined dsDNA was readily detected as inferred from the decrease of the voltammetric oxidation peaks of guanine and adenine. This trend was significantly greater at the ERGO/GCE compared to the GO/GCE. The interaction of cisplatin with dsDNA was also studied in solution phase by AdTSV with detection at the ERGO/GCE.  相似文献

Abstract

This work reports the development of a biosensor method for the label‐free detection of specific DNA sequences. In the initial phase, square wave voltammetry (SWV) was used in a comparative investigation into the electrochemical oxidation of purines (guanine and adenine) and DNA fragments at various electrode surfaces: carbon paste (CPE), glassy carbon electrode (GCE), and gold (AuE). Relative to the carbon electrodes, an approximate 4.0‐fold, 6.0‐fold, and 3.25‐fold increase in the anodic response was observed when guanine, adenine, and hydrolyzed DNA, respectively, were measured on the AuE. It was shown that the guanine and adenine bases could be successfully determined by use of SWV for a deoxyribonucleic acid sample following acid hydrolysis. This label‐free detection of hydrolyzed DNA on gold electrodes has significant advantages over methods using existing carbon electrode materials because of its higher sensitivity and the potential applicability of microfabrication techniques for the production of the requisite gold electrodes.

In another phase of development, the times and conditions for DNA hydrolysis and purine release were investigated. It was shown that under optimal conditions, trace levels of the purine bases could be readily detected following 20 min of hydrolysis at room temperature. The proposed method can be used to estimate the guanine and adenine contents in DNA with in a linear range of 5–30 ng ml?1.

Finally, when appropriate probe sequences were first adsorbed on the surface of the screen‐printed gold electrode (SPGE), this electrochemical biosensor could be used to specifically detect sequences from ss corona virus aviair following hybridization and hydrolysis reactions on the sensor surface. No enhancement of the voltammetric response was observed when the sensor was challenged with a non‐complementary DNA sequence.  相似文献

A simple method for simultaneous detection of guanine, adenine, thymine and cytosine was set up by using a bare glassy carbon electrode in acetate buffer solution of pH 4.5. The peak current responses of these four DNA bases in this supporting electrolyte were significantly increased comparing with those in phosphate buffer solution and Tris-HCl, moreover, the peak current values were linearly dependent on the concentration of four DNA bases, respectively. Individual and simultaneous determinations of four bases were performed by controlling certain experimental conditions, and broad linear ranges and low detection limits (S/N = 3) were obtained. The assay processes do not need any separation or pretreatment steps. In addition, this method showed good selectivity, reproducibility, and stability and can be used for determination of the four bases content in real DNA sample.  相似文献

A novel multiwalled nanotubes (MWNTs)/Cerium(III) 12 ‐ tungstophosphoric acid (CePW) nanocomposite film glassy carbon electrode was prepared in this paper. Electrochemical behaviors of the CePW/MWNTs modified electrode were investigated by electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). This modified electrode brought new capabilities for electrochemical devices by combining the advantages of carbon nanotubes, rare‐earth, and heteropoly‐acids. The results demonstrated that the CePW/MWNTs modified electrode exhibited enhanced electrocatalytic behavior and good stability for the detection of guanine and adenine in 0.1 M PBS (pH 7.0). The experimental parameters were optimized and a direct electrochemical method for the simultaneous determination of guanine and adenine was proposed. The detection limit (S/N=3) for guanine and adenine was 2.0×10?8 M and 3.0×10?8 M, respectively. Further, the acid‐denatured calf thymus DNA was also detected and the result was satisfied.  相似文献

Electrochemical detection of DNA is a highly important topic. Here we show that the electrochemical responses of one DNA base (guanine, adenine, cytosine or thymine), in terms of oxidation potential, current intensity, peak width and resolution can be highly influenced by the presence of other DNA bases at electrochemically reduced graphene oxide (ER‐GO) as well as standard glassy carbon electrode. We have observed that the effects were more significant for adenine base on ER‐GO and cytosine base on glassy carbon (GC) electrode. Differences in responses were generally low in a mixture of four different DNA bases but interestingly, deviations become significantly larger when only one or two other bases were present. Our findings are of paramount importance for future developments in DNA detection and analysis since individual DNA bases are not present in isolation in nature or in typical biosensing systems.  相似文献
