A novel bare-eye based one-step signal amplified semi-quantitative immunochromatographic assay (SAS-ICA) was developed for detection of the pesticide imidacloprid. This method was based on competitive immunoreactions. Signal amplification was achieved by dual labeling of the test lines (TLs) on the strip using high affinity nanogold-biotinylated anti-imidacloprid mAb (BAb) and nanogold-streptavidin (Sa) probes. The relative color intensities of three TLs (TL-I, TL-II and TL-III) on a nitrocellulose (NC) membrane were used for direct visual analysis of the SAS-ICA strips, and could be used for semi-quantitation of analyte concentrations by observing what TLs disappeared in the amplification zone. Under optimized conditions, the following imidacloprid concentration ranges would be detected by visual examination of the SAS-ICA strip: 0–5 ng mL−1 (negative samples), and 5–25 ng mL−1, 25–250 ng mL−1, 250–1000 ng mL−1 and >1000 ng mL−1 (positive samples). The sensitivity (the visual detection limit (VDL) of TL-III) and semi-quantitative analytical capacity (when TL-III disappeared completely) of the SAS-ICA strip were 10-fold and 160-fold higher than those of traditional ICA, respectively. The developed SAS-ICA strip was applied to the analysis of spiked and authentic contaminated Chinese cabbage samples in the laboratory and under field conditions, and the results were validated by high-performance liquid chromatography (HPLC). This process could be adopted as a potential generous technique for all ICA-based detection methods.  相似文献

A one-step immunochromatographic assay (ICA) was developed for the detection of seven kinds of cephems in milk. Polyclonal antibodies (PcAb) with group-specific to cephems were raised in rabbits after immunization with cephalexin-keyhole limpet hemocyanin (KLH) conjugate. The specificity of anti-sera was determined by indirect competitive enzyme-linked immunosorbent assay (icELISA), and the 50% inhibitions (IC50) of cephalexin and cefadroxil were obtained at 1.5 ng mL−1; IC50 of cefatiofur, cefapirin, cefazolin, cefalothin and cefotaxine were 4, 3.7, 3.2, 4.5 and 5 ng mL−1, respectively. The PcAb against cephems were conjugated to colloidal gold particles as the detection reagent for ICA strips to test for cephems. This method achieved semi-quantitative detection of cephems in <5 min, with high sensitivity to cephalexin and cefadroxil (both 0.5 ng mL−1). At the same time, cefatiofur, cefapirin, cefazolin, cefalothin and cefotaxine were detected at <100 ng mL−1 in spiked processed-milk samples. This method was compared with an enzyme-linked immunosorbent assay by testing 40 milk samples, and the positive samples were validated by a high-performance liquid chromatographic method, with an agreement rate of 100% for both comparisons. In conclusion, the method was rapid and accurate for the multi-residue detection of cephems in milk.  相似文献

It is critical to develop a cost-effective quantitative/semiquantitative assay for rapid diagnosis and on-site detection of toxic or harmful substances. Here, a naked-eye based semiquantitative immunochromatographic strip (NSI-strip) was developed, on which three test lines (TLs, TL-I, TL-II and TL-III) were dispensed on a nitrocellulose membrane to form the test zone. Similar as the traditional strip assay for small molecule, the NSI-strip assay was also based on the competitive theory, difference was that the analyte competed three times with the capture reagent for the limited number of antibody binding sites. After the assay, the number of TLs developed in the test zone was inversely proportional to the analyte concentration, thus analyte content levels could be determined by observing the appeared number of TLs. Taking aflatoxin B1 as the model analyte, visual detection limit of the NSI-strip was 0.06 ng mL−1 and threshold concentrations for TL-I–III were 0.125, 0.5, and 2.0 ng mL−1, respectively. Therefore, according to the appeared number of TLs, the following concentration ranges would be detectable by visual examination: 0–0.06 ng mL−1 (negative samples), and 0.06–0.125 ng mL−1, 0.125–0.5 ng mL−1, 0.5–2.0 ng mL−1 and >2.0 ng mL−1 (positive samples). That was to say, compared to traditional strips the NSI-strip could offer more parameter information of the target analyte content. In this way, the NSI-strip improved the qualitative presence/absence detection of traditional strips by measuring the content (range) of target analytes semiquantitatively.  相似文献

A novel immunochromatographic assay (ICA) was proposed for rapid and multiple assay of β2-agonists, by utilizing ractopamine (RAC) and salbutamol (SAL) as the models. Owing to the introduction of chemiluminescent (CL) approach, the proposed protocol shows much higher sensitivity. In this work, the described ICA was based on a competitive format, and horseradish peroxidase-tagged antibodies were used as highly sensitive CL probes. Quantitative analysis of β2-agonists was achieved by recording the CL signals of the probes captured on the two test zones of the nitrocellulose membrane. Under the optimum conditions, RAC and SAL could be detected within the linear ranges of 0.50–40 and 0.10–50 ng mL−1, with the detection limits of 0.20 and 0.040 ng mL−1 (S/N = 3), respectively. The whole process for multianalyte immunoassay of RAC and SAL can be completed within 20 min. Furthermore, the test strip was validated with spiked swine urine samples and the results showed that this method was reliable in measuring β2-agonists in swine urine. This CL-based multianalyte test strip shows a series of advantages such as high sensitivity, ideal selectivity, simple manipulation, high assay efficiency and low cost. Thus, it opens up new pathway for rapid screening and field analysis, and shows a promising prospect in food safety.  相似文献

A visual strip has been developed for sensing iron in different aqueous samples like natural water and fruit juices. The sensor has been synthesized by UV-radiation induced graft polymerization of acrylamide monomer in microporous poly(propylene) base. For physical immobilization of iron selective reagent, the in situ polymerization of acrylamide has been carried out in the presence of 1,10-phenanthroline. The loaded strip on interaction with Fe(II) in aqueous solution turned into orange red color and the intensity of the color was found to be directly proportional to the amount of Fe(II) in the aqueous sample. The minimal sensor response with naked eye was found for 50 ng mL−1 of Fe in 15 min of interaction. However, as low as 20 ng mL−1 Fe could be quantified using a spectrophotometer. The detection limit calculated using the 3s/S criteria, where ‘s’ is the standard deviation of the absorbance of blank reagent loaded strip and ‘S’ is the slope of the linear calibration plot, was 1.0 ng mL−1. The strip was applied to measure Fe in a variety of samples such as ground water and fruit juices.  相似文献

In this paper a novel solid phase extraction method to determine Co(II) and Ni(II) using silica gel-polyethylene glycol (Silica-PEG) as a new adsorbent is described. The method is based on the adsorption of cobalt and nickel ions in alkaline media on polyethylene glycol-silica gel in a mini-column, elution with nitric acid and determination by flame atomic absorption spectrometry. The adsorption conditions such as NaOH concentration, sample volume and amount of adsorbent were optimized in order to achieve highest sensitivity. The calibration graph was linear in the range of 0.5-200.0 ng mL−1 for Co(II) and 2.0-100.0 ng mL−1 for Ni(II) in the initial solution. The limit of detection based on 3Sb was 0.37 ng mL−1 for Co(II) and 0.71 ng mL−1 for Ni(II). The relative standard deviations (R.S.D.) for ten replicate measurements of 40 ng mL−1 of Co(II), and Ni(II) were 3.24 and 3.13%, respectively. The method was applied to determine Co(II) and Ni(II) in black tea, rice flour, sesame seeds, tap water and river water samples.  相似文献

This paper presents the development of a new flow-injection system combined with solid-surface fluorescence detection for the determination of the widely used fungicide thiabendazole. Nylon powder was probed as a novel solid support for building the optosensor. The method is based on the on-line immobilization of thiabendazole onto nylon in a continuous flow system, followed by the measurement of its native fluorescence. Aqueous samples are directly injected in a water carrier, resulting in a very simple and economical method. The analytical figures of merit obtained using 1500 μL of sample and 75% methanol (v/v) as eluting solution were: linear calibration range from 8 to 120 ng mL−1 (the lowest value corresponds to the quantitation limit), relative standard deviation, 0.9% (n = 5) at a level of 64 ng mL−1, limit of detection calculated according to 1995 IUPAC recommendations is to 2.8 ng mL−1, and sampling rate of 14 samples h−1. The potential interference from other agrochemicals, metal ions and common anions, and the viability of determining thiabendazole in real water samples were also evaluated.  相似文献

A novel, highly selective and sensitive paper-based colorimetric sensor for trace determination of copper (Cu2+) ions was developed. The measurement is based on the catalytic etching of silver nanoplates (AgNPls) by thiosulfate (S2O32−). Upon the addition of Cu2+ to the ammonium buffer at pH 11, the absorption peak intensity of AuNPls/S2O32− at 522 nm decreased and the pinkish violet AuNPls became clear in color as visible to the naked eye. This assay provides highly sensitive and selective detection of Cu2+ over other metal ions (K+, Cr3+, Cd2+, Zn2+, As3+, Mn2+, Co2+, Pb2+, Al3+, Ni2+, Fe3+, Mg2+, Hg2+ and Bi3+). A paper-based colorimetric sensor was then developed for the simple and rapid determination of Cu2+ using the catalytic etching of AgNPls. Under optimized conditions, the modified AgNPls coated at the test zone of the devices immediately changes in color in the presence of Cu2+. The limit of detection (LOD) was found to be 1.0 ng mL−1 by visual detection. For semi-quantitative measurement with image processing, the method detected Cu2+ in the range of 0.5–200 ng mL−1(R2 = 0.9974) with an LOD of 0.3 ng mL−1. The proposed method was successfully applied to detect Cu2+ in the wide range of real samples including water, food, and blood. The results were in good agreement according to a paired t-test with results from inductively coupled plasma-optical emission spectrometry (ICP-OES).  相似文献

A new method for separation/preconcentration of trace amounts of Cr, Cu and Pb in environmental samples by magnetic solid-phase extraction (SPE) with Bismuthiol-II-immobilized magnetic nanoparticles and their determination by ICP-OES has been developed. The separation of the target analytes from the aqueous solution containing the target analytes and Bismuthiol-II-immobilized magnetic nanoparticles was simply achieved by applying external magnetic field. Optimal experimental conditions including pH, sample volume, eluent concentration and volume and co-existing ions have been studied and established. Under the optimal experimental conditions, the detection limits for Cr, Cu and Pb with enrichment factors of 96, 95 and 87 were found to be 0.043, 0.058 and 0.085 ng mL−1 and their relative standard deviations (R.S.D.s) were 3.5%, 4.6% and 3.7% (n = 5, C = 2 ng mL−1), respectively. The method was validated with certified reference material (GBW50009-88) of environmental water sample and the analytical results coincided well with the certified values. Furthermore, the method was successfully applied to the determination of target analytes in river and lake water samples. Compared with established methods, the proposed method is characterized with high enrichment factor, fast separation and low detection limits.  相似文献

A novel cloud point phase separation of cationic surfactant, Aliquat-336 and capabilities of its reactive solubilizing sites for selective extraction of chromium species at ultra trace levels was examined in natural water. The phase separation behavior of Aliquat-336 is studied with various additives. The nonionic surfactant, Triton X-114 was found to induce the cloud point phase separation of Aliquat-336. The separation of anionic Cr(VI) was enabled by the formation of ion associate with quaternary ammonium head group of Aliquat-336 at pH 2, and the recovery of Cr(VI) and Cr(III) were 101.4 ± 1.4% and 2.2 ± 0.4%, respectively at 0.5-1 ng mL−1, Total Cr was pre-concentrated as Cr-APDC species using the hydrophobic tail group at pH 6.5. The Cr(III) concentration was obtained by subtracting Cr(VI) from total Cr. The recovery of total Cr was 99.5 ± 1.2%. Parameters affecting extraction were assessed. The procedure was applied to NIST 1643c and NIST 1643d waters, and the sum of individual species obtained was compared with the certified chromium values. The method was also applied to various natural waters with limits of detection and pre-concentration factor of 0.010 and 0.025 ng mL−1; 10 and 10, respectively, for Cr(VI) and Cr(III)-APDC using ICP-MS operated in DRC mode.  相似文献

A new fluorogenic method for the selective and sensitive determination of chromium(VI) in acidic water using rhodamine B hydrazide was developed. This method was based on the oxidation of non-fluorescent rhodamine B hydrazide by potassium dichromate in acidic aqueous conditions to give rhodamine B, which was highly fluorescent, as a product. With the optimum condition described, the fluorescence enhancement at 585 nm was linearly related to the concentration of chromium(VI) in the range of 5.0 × 10−8 to 2.0 × 10−6 mol L−1 (2.60-104 ng mL−1) with a correlation coefficient of R2 = 0.9993 (n = 18) and a detection limit of 5.5 × 10−9 mol L−1 (0.29 ng mL−1). The R.S.D. was 2.2% (n = 5). The proposed method was also applied to the determination of chromium(VI) in drinking water, river water and synthetic samples.  相似文献

A method based on cloud point extraction was developed to determine four kinds of estrogens: estriol (E3), estradiol (E2), estrone (E1), and progesterone (P) in water by high performance liquid chromatography separation and ultraviolet detection (HPLC-UV). The non-ionic surfactant Triton X-114 was chosen as extractant solvent. The parameters affecting extraction efficiency, such as concentrations of Triton X-114 and Na2SO4, equilibration temperature, equilibration time and centrifugation time were evaluated and optimized. Under the optimum conditions, preconcentration factors of 99 for E3, 73 for E2, 152 for E1 and 86 for P were obtained for 10 mL water sample. The detection of limitation was 0.23 ng mL−1 for E3, 0.32 ng mL−1 for E2, 0.25 ng mL−1 for E1 and 5.0 ng mL−1 for P. The proposed method was successfully applied to the determination of trace amount of estrogens in wastewater treatment plant (WWTP) effluent water and exposure water with 10 ng mL−1 E2 for toxicological study in our lab. For the case of WWTP effluent water samples, no estrogen was found. The accuracy of the proposed method was tested by recovery measurements of spiked samples and good recoveries of 81.2-99.5% were obtained.  相似文献

A rapid and sensitive method for the determination of carbendazim (methyl benzimidazole-2-ylcarbamate, MBC) and thiabendazole (TBZ) in water and soil samples was developed by using dispersive liquid-liquid microextraction (DLLME) coupled with high performance liquid chromatography with fluorescence detection. The water samples were directly used for the DLLME extraction. For soil samples, the target analytes were first extracted by 0.1 mol L−1 HCl. Then, the pH of the extract was adjusted to 7.0 with 2 mol L−1 NaOH before the DLLME extraction. In the DLLME extraction method, chloroform (CHCl3) was used as extraction solvent and tetrahydrofuran (THF) as dispersive solvent. Under the optimum conditions, the enrichment factors for MBC and TBZ were ranged between 149 and 210, and the extraction recoveries were between 50.8 and 70.9%, respectively. The linearity of the method was obtained in the range of 5-800 ng mL−1 for water sample analysis, and 10-1000 ng g−1 for soil samples, respectively. The correlation coefficients (r) ranged from 0.9987 to 0.9997. The limits of detection were 0.5-1.0 ng mL−1 for water samples, and 1.0-1.6 ng g−1 for soil samples. The relative standard deviations (RSDs) varied from 3.5 to 6.8% (n = 5). The recoveries of the method for MBC and TBZ from water samples at spiking levels of 5 and 20 ng mL−1 were 84.0-94.0% and 86.0-92.5%, respectively. The recoveries for soil samples at spiking levels of 10 and 100 ng g−1 varied between 82.0 and 93.4%.  相似文献

A simple and rapid headspace solid-phase microextraction (HS-SPME) based method is presented for the simultaneous determination of atrazine and ametryn in soil and water samples by ion mobility spectrometry (IMS). A dodecylsulfate-doped polypyrrole (PPy-DS), synthesized by electrochemical method, was applied as a laboratory-made fiber for SPME. The HS-SPME system was designed with a cooling device on the upper part of the sample vial and a circulating water bath for adjusting the sample temperature. The extraction properties of the fiber to spiked soil and water samples with atrazine and ametryn were examined, using a HS-SPME device and thermal desorption in injection port of IMS. Parameters affecting the extraction efficiency such as the volume of water added to the soil, pH effect, extraction time, extraction temperature, salt effect, desorption time, and desorption temperature were investigated. The HS-SPME-IMS method with PPy-DS fiber, provided good repeatability (RSDs < 10 %), simplicity, good sensitivity and short analysis times for spiked soil (200 ng g−1) and water samples (100 and 200 ng mL−1). The calibration graphs were linear in the range of 200-4000 ng g−1 and 50-2800 ng mL−1 for soil and water respectively (R2 > 0.99). Detection limits for atrazine and ametryn were 37 ng g−1 (soil) and 23 ng g−1 (soil) and 15 ng mL−1 (water) and 10 ng mL−1 (water), respectively. To evaluate the accuracy of the proposed method, atrazine and ametryn in the three kinds of soils and two well water samples were determined. Finally, comparing the HS-SPME results for extraction and determination of selected triazines using PPy-DS fiber with the other methods in literature shows that the proposed method has comparable detection limits and RSDs and good linear ranges.  相似文献

In the present work a method for the determination of mercury at trace levels by energy dispersive X-ray fluorescence (EDXRF) is introduced. Mercury ions were concentrated on cation selective membranes that have been prepared on Mylar® thin film substrate, immobilized on plastic cups. The produced membranes were immersed in water solutions containing low concentrations of mercury. The membranes were left to equilibrate in 1000 mL of mercury solutions and were analyzed by EDXRF. The effects of various experimental parameters were examined. Minimum detection limits of pg mL−1 (ppt) (0.069 ng mL−1 for ASTM Type I water and 0.064 ng mL−1 for seawater) and good linearity were achieved.  相似文献

A new procedure was described with multiwalled carbon nanotubes as solid phase extraction packing material for the trace analysis of nicosulfuron, thifensulfuron and metsulfuron-methyl in water samples. The possible parameters influencing the enrichment were optimized and the optimal conditions were as followed: eluent, sample pH, flow rate and sample volume were acetonitrile containing 1% acetic acid, pH 3, 8 mL min−1 and 500 mL, respectively. Under the optimal chromatographic separation and SPE conditions, the linear range, detection limit (S/N = 3) and precision (R.S.D., n = 6) were 0.04-40 ng mL−1, 6.8 ng L−1 and 2.5% for nicosulfuron, 0.04-40 ng mL−1, 11.2 ng L−1 and 5.4% for thifensulfuron, 0.02-20 ng mL−1, 5.9 ng L−1, 2.1% for metsulfuron-methyl, respectively. The established method was well employed to determine nicosulfuron, thifensulfuron and metsulfuron-methyl in tap water, seawater, reservoir water and well water samples, and satisfactory results were obtained, the spiked recoveries in the range of 87.2-100.7%, 96.5-105.6% and 83.7-111.1% for them each, respectively.  相似文献

A novel in-capillary reduction and capillary electrophoretic (CE)-chemiluminescence (CL) method was developed for the sensitive and selective determination of chromium(III) and chromium(VI). The proposed method was based on the in-capillary reduction of Cr(VI) with acidic H2O2 to form Cr(III) using the zone-passing technique and chemiluminescence detection of Cr(III). The sample [Cr3+ and CrO42−], hydrochloric acid, and H2O2 (reductant) solution segments were injected for specified periods of time in this order from the anodic end of a capillary, followed by application of an appropriate running voltage between both ends. As both chromium species have opposite charges, Cr3+ migrates to the cathode while CrO42− ion, moving oppositely to the anode, reacts with acidic H2O2, resulted in formation of Cr3+. Based on the migration time difference of both Cr3+ ions, they were separated by zone electrophoresis. Running buffer was composed of 0.02 mol l−1 HAc-NaAc (pH 4.7) with 1×10−3 mol l−1 EDTA. Parameters affecting CE-CL separation and detection, such as reductant concentration, mixing mode of the analytes with CL reagent, CL reaction reagent pH and concentration, stability of luminol-hydrogen peroxide mixed solution were optimized. The limits of detection for chromium(III) and chromium(VI) (3σ) were 6×10−13 mol l−1 (mass concentration 12 zmol) and 8×10−12 mol l−1 (160 zmol), respectively. This method offered potential advantages of simplicity, sensitivity, selectivity and applicability to the determination of Cr(III) and Cr(VI) in environmental water.  相似文献

An on-line flow injection (FI) preconcentration-electrothermal atomic absorption spectrometry (ETAAS) method is developed for trace determination of chromium in drinking water samples by sorption on a conical minicolumn packed with activated carbon (AC) at pH 5.0. The chromium was removed from the minicolumn with 1.0% (v/v) nitric acid. An enrichment factor (EF) of 35-fold for a sample volume of 10 ml was obtained. The detection limit (DL) value for the preconcentration method proposed was 3.0 ng l−1. The precision for 10 replicate determinations at the 0.5 μg l−1 Cr level was 4.0% relative standard deviation (R.S.D.), calculate with the peak heights obtained. The calibration graph using the preconcentration system for chromium was linear with a correlation coefficient of 0.9992 at levels near the detection limits up to at least 50 μg l−1. The method was successfully applied to the determination of Cr(III) and Cr(VI) in drinking water samples.  相似文献

Organophosphate triesters are common flame retardants used in a wide variety of consumer products from which they can migrate and pollute the indoor environment. Humans may thus be continuously exposed to several organophosphate triesters which might be a risk for human health. An analytical method based on direct injection of 5 μL urine into an ultra performance liquid chromatography system coupled to a time-of-flight mass spectrometry has been developed and validated to monitor exposure to organophosphate triesters through their respective dialkyl and diaryl phosphate metabolites (DAPs). The targeted analytes were: di-n-butyl phosphate (DNBP), diphenyl phosphate (DPHP), bis(2-butoxyethyl) phosphate (BBOEP), bis(2-chloroethyl) phosphate (BCEP), bis(1-chloro-2-propyl) phosphate (BCPP) and bis(1,3-dichloro-2-propyl) phosphate (BDCIPP). Separation was achieved in less than 3 min on a short column with narrow diameter and small particle size (50 mm × 2.1 mm × 1.7 μm). Different mobile phases were explored to obtain optimal sensitivity. Acetonitrile/water buffered with 5 mM of ammonium hydroxide/ammonium formate (pH 9.2) was the preferred mobile phase. Quantification of DAPs was performed using deuterated analogues as internal standards in synthetic urine (averaged DAP accuracy was 101%; RSD 3%). Low method limits of quantification (MLQ) were obtained for DNBP (0.40 ng mL−1), DPHP (0.10 ng mL−1), BDCIPP (0.40 ng mL−1) and BBOEP (0.60 ng mL−1), but not for the most polar DAPs, BCEP (∼12 ng mL−1) and BCPP (∼25 ng mL−1). The feasibility of the method was tested on 84 morning urine samples from 42 mother and child pairs. Only DPHP was found above the MLQ in the urine samples with geometric mean (GM) concentrations of 1.1 ng mL−1 and 0.57 ng mL−1 for mothers and children respectively. BDCIPP was however, detected above the method limit of detection (MLD) with GM of 0.13 ng mL−1 and 0.20 ng mL−1. While occasionally detected, the GM of DNBP and BBOEP were below MLD in both groups.  相似文献

A novel ultrasound-assisted surfactant-enhanced emulsification microextraction (UASEME) coupled with high performance liquid chromatography-diode array detection has been developed for the extraction and determination of six carbamate pesticides (metolcarb, carbofuran, carbaryl, pirimicarb, isoprocarb and diethofencarb) in water samples. In the UASEME technique, Tween 20 was used as emulsifier, and chlorobenzene and chloroform were used as dual extraction solvent without using any organic dispersive solvent that is normally required in the previously described common dispersive liquid–liquid microextraction method. Parameters that affect the extraction efficiency, such as the kind and volume of the extraction solvent, the type and concentration of the surfactant, ultrasound emulsification time and salt addition, were investigated and optimized for the method. Under the optimum conditions, the enrichment factors were in the range between 170 and 246. The limits of detection of the method were 0.1–0.3 ng mL−1 and the limits of quantification were between 0.3 and 0.9 ng mL−1, depending on the compounds. The linearity of the method was obtained in the range of 0.3–200 ng mL−1 for metolcarb, carbaryl, pirimicarb, and diethofencarb, 0.6–200 ng mL−1 for carbofuran, and 0.9–200 ng mL−1 for isoprocarb, with the correlation coefficients (r) ranging from 0.9982 to 0.9998. The relative standard deviations varied from 3.2 to 4.8% (n = 5). The recoveries of the method for the six carbamates from water samples at spiking levels of 1.0, 10.0, 50.0 and 100.0 ng mL−1 were ranged from 81.0 to 97.5%. The proposed UASEME technique has demonstrated to be simple, practical and environmentally friendly for the determination of carbamates residues in river, reservoir and well water samples.  相似文献
