Fischer Synthesis of 3-(N-Acylamino)-2-phenylindoles

Phenylhydrazones were obtained by the reaction of phenylhydrazine with -(N-acylamino)-acetophenones and were converted into 3-(N-acylamino)indoles by the Fischer cyclizaction.     

New derivatives of 3-aminoindole. Synthesis of 2-aryl(hetaryl)-3-(3,5-dimethyl-1-pyrazolyl)indoles

The interaction of arylhydrazines with ù-(3,5-dimethyl-1-pyrazolyl)acetophenones and acetyl(2-thiophene) leads to arylhydrazones, which are converted by Fischer cyclization into 2-aryl(thienyl)-3-(3,3-dimethyl-1-pyrazolyl)indoles with substituents in positions 1, 5, and 7.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 11, pp. 1662–1669, November, 2004.     

A Novel Synthesis of 1-Aminoalkanephosphonic Acid Derivatives from 1-(N-Acylamino)- alkyltriphenylphosphonium Salts

Efficient and convenient procedures for the α-amidoalkylation of trialkylphosphites with 1-(N-acylamino)alkyltriphenylphosphonium salts followed by a Michaelis–Arbuzov-type reaction to afford 1-(N-acylamino)alkanephosphonic acid esters have been developed. High yields and simple isolation and purification protocols are the main advantages of this method.     

2-Polyfluoroalkylchromones. 14. Synthesis of 4-chloro-3(5)-(2-hydroxyaryl)-5(3)-polyfluoroalkylpyrazoles

3-Chloro-2-polyfluoroalkyl- and 3-chloro-6-nitro-2-polyfluoroalkylchromones were synthesized. These compounds react with N₂H₄·2HCl on boiling in ethanol to form 4-chloro-3(5)-(2-hydroxyaryl)-5(3)-polyfluoroalkylpyrazoles.     

Synthesis of 4-Hydroxy- and 4-Amino-2-Methyl-3-(2-Methylindol-3-Yl)methylquinolines and Their 6- and 8-Substituted Derivatives

A method has been developed for the synthesis of substituted 4-hydroxy- and 4-amino-2-methyl-3-(2-methylindol-3-yl)methylquinolines by treating the corresponding 4-hydroxy(chloro)-2-methyl-3-(3-oxobutyl)quinolines with phenylhydrazine hydrochloride. It was found that nucleophilic substitution occurred in the case of the 4-chloroquinolines together with subsequent rearrangement to give the 4-amino derivatives. The thiosemicarbazones of the corresponding 4-hydroxy-2-methyl-3-(3-oxobutyl)quinolines were also obtained.__________Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 4, pp. 554–557, April, 2005.     

Synthesis of Gingerenone C and 5-Hydroxy-1-(4'-hydroxy- 3'-methoxyphenyl)-7-(4"-hydroxyphenyl)-3-heptanone

The two diarylheptanoids (E)-1-(4'-hydroxy-3'-methoxyphenyl)-7-(4"-hydroxyphenyl) hept-4-en-3-one 1 (Gingerenone C) and (±)-5-hydroxy-1-(4'-hydroxy-3'-methoxyphenyl)-7-(4"-hydroxyphenyl)-3-heptanone 2 were synthesized from vanillin 3 and 4-hydroxybenzaldehyde 9.     

3-(2-Quinolyl)- and 3-(5-carbethoxyfuryl-2)coumarins

3-(2-Quinolyl)- and 3-(5-carbethoxyfuryl-2)coumarins were prepared by reaction of substituted salicylaldehydes and hetarylacetonitriles. Synthesis of Conjugated Systems Based on 3-(5-Methylfur-2-yl)-2-(3-oxobutyl)benzofurans

Methods were developed for the synthesis of 3-butyl-2-(3-oxobutenyl)benzofurans and 3-(3-furylbenzofuran-2-yl)acrylic acids on the basis of 3-furyl-2-(3-oxobutyl)benzofurans.     

1-(4-取代苯基)-4-苯基-5-(2-羟基苄基)氨基-1,2,3-三唑类衍生物的合成及抑菌活性

将邻羟苯基引入1,2,3-三唑结构中, 设计合成了10个1-(4-取代苯基)-4-苯基-5-取代-1,2,3-三唑类衍生物. 首先, 以对位取代的芳胺为原料, 经重氮化、叠氮化、闭环和缩合反应制得1-(4-取代苯基)-4-苯基-5-水杨醛亚胺-1,2,3-三唑类衍生物(3a~3e), 再用硼氢化钠还原制得1-(4-取代苯基)-4-苯基-5-(2-羟基苄基)氨基-1,2,3-三唑类衍生物(4a~4e). 目标化合物的结构经核磁、IR及元素分析确认. 抑菌活性测试表明, 当质量浓度为0.1 mg/L时, 除化合物3e和4e外, 所有化合物对白色念球菌的抑菌率均达95%以上, 对大肠杆菌的抑菌率达85%以上, 具有强抑菌活性, 表明该类化合物在抗菌药物开发方面有重要应用价值.     

1-(4-硝基苯)-3-(3-甲基吡啶)-三氮烯合成及其与铜的显色反应

报道了新显色剂 1-(4-硝基苯)-3-(3-甲基吡啶)-三氮烯(NPMPDT)的合成及其与铜的显色反应.在OP的存在下,pH 10.5的Na2B4O7-NaOH缓冲溶液中,该试剂能与铜发生显色反应,铜与NPMPDT形成摩尔比为2∶1型的黄色配合物,在460 nm处有一最大正吸收,在535 nm 处有一最大负吸收.以460 nm为参比波长,535 nm为测量波长进行双波长测定,其表观摩尔吸光系数ε＝2.18×105 L·mol-1·cm-1,铜的质量浓度在0～0.48 μg/mL 范围内符合比尔定律,用本法测定大米和面粉中微量的铜,结果满意.     

3-取代硫基-5-(1-羟基苯基)-4H-1,2,4-三唑类化合物的合成及抑菌活性

依据邻羟基二苯醚及三唑类化合物的抗菌特性及生物活性叠加原理, 将邻羟苯基和1,2,4-三唑分子片断有机结合, 设计合成了12个新型3-取代硫基-5-(1-羟基苯基)-4H-1,2,4-三唑类化合物. 首先, 水杨酸甲酯与水合肼反应生成水杨酰肼, 水杨酰肼再与硫氰酸铵和盐酸反应, 生成5-(1-羟基苯基)-4H-1,2,4-三唑-3-硫酮(3), 最后在碱性条件下化合物3与取代苯乙酮、氯苄和碘甲烷发生烷基化反应生成目标化合物, 化合物结构经 1H NMR及IR等表征确认. 抑菌测试结果表明, 当化合物质量分数为0.01%时, 目标化合物对白色念珠菌和大肠杆菌的抑菌率高达90%, 具有强抑菌活性; 对金黄色葡萄球菌的抑菌率高达80%, 具有一定的抑菌活性.

Synthesis of propyl O-(α-L-rhamnopyranosyl)-(1→3)-[2,4-di-O-(2s-methylbutyryl)-α-L-rhamnopyranosyl]-(1→2)=(3-O-acetyl-β-D-glucopyranosyl)-(1→2)-βp-D-fucopyranoside,the tetrasaccharide moiety of Tricolorin A

Propyl O-(α-L-rhamnopyranosyl)-(1→3)-[2,4di-O-(2s-methylbutyryl)-α-L-rhamnopyranosyl]-(1→2)-(3-O-acetyl-β-glucopyranosyl)-(1→2)-β-D-fucopyranoside (1), the tetrasaccharide moiety of Ricolorin A, was synthesized in total 23 steps with a longest linear sequence of 10 steps, and overall yield of 3.7% from D-Glucose. The isomerization of the dioxolane-type benzylidene in the prance of NIS/AgOTf was observed. Tetrasaccharide 1 exhibited no activity against the cultured P388 cell as Tricolorin A did.

三氮烯试剂是测定镉、汞、锌、镍、铜等金属离子的高灵敏显色试剂[1~3],其中含氮杂环的三氮烯试剂的显色效果更佳[4~6]。为改善试剂的分析性能,提高此类试剂的灵敏度和选择性,合成了未见报道的1-(4-硝基苯基)-3-(5-溴-吡啶)-三氮烯(NPBPDT),并对其与镍的显色反应进行了研究。结

报道了1-(4-硝基苯基)-3-(5-氯吡啶)三氮烯(NPCPDT)的合成及其与镉的显色反应研究。在Triton X-100的存在下,pH 11.5的Na2B4O7-NaOH缓冲溶液中,该试剂能与镉发生显色反应,镉与NPCPDT形成摩尔比为1∶3的黄色配合物,在440 nm处有一最大吸收,在535 nm处有一最大负吸收。以440 nm为参比波长,535 nm为测量波长进行双波长测定,表观摩尔吸光系数为2.68×105L.mol-1.cm-1,镉的质量浓度在0~0.48μg/mL范围内符合比尔定律。用拟定方法测定废水和环境水样中的微量镉。     

Synthesis of 3- and 5-amino-5-(3)-(pyrrol-2-yl)isoxazoles

5-Amino-3-(pyrrol-2-yl)isoxazoles were selectively prepared by the reaction of 2-(2,2-dicyano-1-ethylthioethenyl)pyrroles with hydroxylamine in methanol. Under analogous conditions, 2-(2-carbamoyl-2-cyano-1-ethylthioethenyl) pyrroles with hydroxylamine gave 5-aminoisoxazoles and their structural isomers, 3-aminoisoxazoles (3-5% yield). The latter were selectively prepared by reacting 2-(2-carbamoyl-2-cyano-1-ethylthioethenyl)pyrroles with hydroxylamine in the presence of aqueous NaOH and from the products of intramolecular cyclization of 2-(2-carbamoyl-2-cyano-1-ethylthioethenyl)pyrroles, 1-ethylthio-3-iminopyrrolizines and hydroxylamine.

Synthesis of （S）-2-（3-Arylacrylamido）-3-{4-[2-（5-methyl-2-phenyloxazol-4-yl）ethoxy]phenyl}propanoic Acids

The synthesis of （S）-2-（3-arylacrylamido）-3-{4-[2-（5-methyl-2-phenyloxazol-4-yl）etho- xy]phenyl}propanoic acids is described. Their structures were confirmed by ^1H-NMR.     

1-(4-硝基苯基)-3-(5-硝基-2-吡啶)-三氮烯的合成及其与汞(Ⅱ)的显色反应研究

合成了 1 -(4 -硝基苯基 ) -3 -(5-硝基 -2 -吡啶 ) -三氮烯 ,并研究了它与汞(Ⅱ )的显色反应。在TritonX -1 0 0的存在下 ,pH1 1 2的Na2 B4 O7-NaOH缓冲溶液中 ,试剂与汞生成 2∶1型桔黄色配合物 ,在 4 3 8nm和 52 5nm处分别有一正一负两个吸收波峰 ,用双峰双波长测定 ,其表观摩尔吸光系数为 2 1× 1 0 5L·mo1 1·cm 1,Hg2 +的浓度在 0～ 2 80 μg/L范围内符合比耳定律。成功地测定了人发和尿液中微量的汞。     

1-(4-硝基苯基)-3-(2-吡嗪)-三氮烯的合成及与镉(Ⅱ)的显色反应

报道了显色剂1-(4-硝基苯基)-3-(2-吡嗪)-三氮烯化合物的合成及其与镉(Ⅱ)的显色反应。在非离子表面活性剂Triton X-100存在下,pH 10.5的Na2B4O7-NaOH的缓冲介质中,该试剂能与镉(Ⅱ)与发生显色反应,形成摩尔比为4∶1的黄棕色配合物,配合物在波长453 nm处有最大吸收峰,表观摩尔吸光系数ε为6.90×104L.mol-1.cm-1,镉(Ⅱ)质量浓度在0~0.56μg/mL范围内遵守比尔定律。用拟定方法测不同废水中的镉(Ⅱ)。
