A novel analytical method for determination of total amount of chondroitin sulfate (CS) based on its conversion to desulfated chondro-disaccharide via an enzyme-catalyzed reaction, was developed. Using the in-capillary enzyme reaction, the method was also applied to the successful construction of an on-line analytical system. Within this system, electrophoretic migration was used to mix zones containing the enzyme mixture (chondroitinase ABC, chondro-4-sulfatase, chondro-6-sulfatase and 2-o-sulfatase) and the substrate (CS). The reaction was then allowed to proceed in the presence of a weak electric field and, finally, the product (desulfated chondro-disaccharide) of enzyme reaction migrated to the detector under the influence of an applied electric field. A polyvinyl alcohol-coated capillary was used to reduce protein adsorption. Desulfated chondro-disaccharide was successfully migrated toward the anode in 10 mM Tris-acetate buffer (pH 7.0) under reversed polarity and detected at 232 nm. The established method was validated and demonstrated to be applicable in the determination of total amount of CS in a commercial ophthalmic solution. No interference from the formulation excipients was observed. Good linearity was obtained, with correlation coefficients above 0.999. Recoveries and precisions ranged from 100.0 to 100.5%, and from 0.2 to 0.6% of the relative standard deviation, respectively. Good agreement was obtained between the established method and traditional photometric method based on carbazole reaction. In this study, application of the method to disaccharide compositional analysis was also performed.  相似文献

A new procedure was developed for the simultaneous determination of water- and fat-soluble vitamins by microemulsion electrokinetic chromatography. Using microemulsions based on sodium dodecyl sulfate, Brij 35, 1-butanol, and heptane, 10 vitamins were separated (4 water-soluble and 6 fat-soluble) within 35 min. The efficiency of the separation was 800000 theoretical plates (effective capillary length, 40 cm). To enhance the selectivity, 2-propanol was used as a hydrophobizing addition into the microemulsion. The procedure was used for analyzing a kaolin-based vitamin premix. The use of microemulsion as an agent for extracting vitamins from the support considerably simplifies the sample preparation.  相似文献

采用更简便的流动相体系,建立了高效液相色谱法同时测定多维元素片中9种水溶性维生素的快速分析方法。以酸水解与离心的方法处理样品,用C8柱分离,流动相A为0.1%三氟乙酸的水溶液,B为甲醇,梯度洗脱,二极管阵列检测器(DAD)检测。28min内实现了9种水溶性维生素的同时分离测定。各维生素线性关系、精密度、回收率均良好。并使用美国国家标准技术研究院(NIST)的SRM3280多维元素片标准物质对方法进行了确认,运用此法测定了市售多维元素片中的水溶性维生素含量。该法可作为维生素片剂中水溶性维生素分离测定的质控方法。  相似文献

HPLC stability-indicating method has been developed for the simultaneous determination of some water-soluble vitamins (ascorbic acid, thiamine hydrochloride, riboflavin-5'-phosphate sodium, pyridoxine hydrochloride, nicotinamide, D(+)-panthenol) and two preservatives (methylparaben and sodium benzoate) in multivitamin syrup preparation. Water-soluble vitamins, preservatives and their degradants were separated on Zorbax SB-Aq (C(18)) (250 mm x 4.6 mm, 5 microm) column at an ambient temperature. Combined isocratic and gradient elution was performed with a mobile phase consisting of 0.0125 M hexane-1-sulfonic acid sodium salt in 0.1% (m/v) o-phosphoric acid, pH 2.4-2.5 (solvent A) and acetonitrile (solvent B) at the flow-rate 1 ml min(-1). Starting with solvent A an isocratic elution was performed for 15 min, then the composition was changed to 85% of A and 15% of B during the next 20 min and it was constant for 5 min, then the composition was changed to 70% of A and 30% of B during next 15 min and it was constant for 5 min and finally was changed to 100% of A as at the beginning of the elution. Detection was performed with diode array detector at 210, 230 and 254 nm. Multivitamin syrup preparation was subjected to stress testing (forced degradation) in order to demonstrate that degradants from the vitamins, preservatives and/or product excipients do not interfere with the quantification of vitamins and preservatives. Typical validation characteristics: selectivity, accuracy, precision, linearity, range, limit of quantification and limit of detection were evaluated for vitamins and preservatives.  相似文献

We have applied a quick and convenient method for determining water-soluble vitamins excreted in human urine. We found that the Sep-Pak C(18) cartridge was useful for preconcentration and recovery of water-soluble vitamins in urine with minimized loss of vitamins. The recovery of vitamins was well over 90%. The separation was carried out by gradient elution with 90/10 (v/v%) methanol/water with 0.1% trifluoroacetic acid (TFA) and water with 0.1% TFA on a muBondapak C(18) column. The separation was completed within 15 min. We measured concentrations of water-soluble vitamins excreted in urine after swallowing an overdose of vitamin pills on purpose, and found that the concentration of each vitamin increased rapidly to the maximum in 2-3 h and decreased swiftly.  相似文献

RP-HPLC determination of water-soluble vitamins in honey   总被引:4,自引:0,他引:4  
The assessment and validation of reliable analytical methods for the determination of vitamins in sugar-based matrices (e.g. honey) are still scarcely explored fields of research. This study proposes and fully validates a simple and fast RP-HPLC method for the simultaneous determination of five water-soluble vitamins (vitamin B2, riboflavin; vitamin B3, nicotinic acid; vitamin B5, pantothenic acid; vitamin B9, folic acid; and vitamin C, ascorbic acid) in honey. The method provides low detection and quantification limits, very good linearity in a large concentration interval, very good precision, and the absence of any bias. It has been successfully applied to 28 honey samples (mainly from Sardinia, Italy) of 12 different botanical origins. While the overall amount of the analytes in the samples is quite low (always below 40 mg kg−1), we have observed a marked dependence of some of their concentrations (i.e. vitamin B3 and vitamin B5) and the botanical origin of the honey. This insight might lead to important characterization features for this food item.  相似文献

Two simple and sensitive spectrofluorimetric methods were developed for determination of three water-soluble vitamins (B1, B2, and B6) in mixtures in the presence of cyanocobalamin. The first one was for thiamine determination, which depends on the oxidation of thiamine HCl to thiochrome by iodine in an alkaline medium. The method was applied accurately to determine thiamine in binary, ternary, and quaternary mixtures with pyridoxine HCl, riboflavin, and cyanocobalamin without interference. In the second method, riboflavin and pyridoxine HCl were determined fluorimetrically in acetate buffer, pH 6. The three water-soluble vitamins (B1, B2, and B6) were determined spectrofluorimetrically in binary, ternary, and quaternary mixtures in the presence of cyanocobalamin. All variables were studied in order to optimize the reaction conditions. Linear relationship was obeyed for all studied vitamins by the proposed methods at their corresponding lambda(exc) or lambda(em). The linear calibration curves were obtained from 10 to 500 ng/mL; the correlation ranged from 0.9991 to 0.9999. The suggested procedures were applied to the analysis of the investigated vitamins in their laboratory-prepared mixtures and pharmaceutical dosage forms from different manufacturers. The RSD range was 0.46-1.02%, which indicates good precision. No interference was observed from common pharmaceutical additives. Good recoveries (97.6 +/- 0.7-101.2 +/- 0.8%) were obtained. Statistical comparison of the results with reported methods shows excellent agreement and indicates no significant difference in accuracy and precision.  相似文献

Summary A method for semiquantitative determination of water-soluble vitamins such as thiamin hydrochloride, riboflavin, pyridoxine, choline chloride,p-aminobenzoic acid, cyanocobalamine, nicotinic acid and ascorbic acid is described. It involves making circular spots or rings on chromatoplates using circular thin-layer chromatographic apparatus. The method is based on sensitivity and specificity of spot reactions together with comparison of the color intensity of a circular spot with that of sensitivity standard. The accuracy of the method is ± 5%. The usefulness of the procedure has been illustrated by determining the water-soluble vitamins from multi-vitamin tablets and injections.
Zusammenfassung Eine Methode zur halbquantitativen Bestimmung wasserlöslicher Vitamine wie Thiaminhydrochlorid, Riboflavin, Pyridoxin, Cholinchlorid,p-Aminobenzoesäure, Cyanokobalamin, Nikotinsäure und Ascorbinsäure wurde beschrieben. Sie beruht auf der Herstellung von Kreisflecken oder Ringen im Wege der Dünnschichtchromatographie. Empfindliche und spezifische Reaktionen dienen zum Vergleich der Farbintensität mit Standardlösungen. Die Genauigkeit beträgt etwa ±5%. Das Verfahren wurde an Multivitamintabletten und Injektionslösungen geprüft.

A rapid, simple and sensitive method based on liquid chromatography/tandem mass spectrometry (LC/MS/MS) with an electrospray ionization (ESI) source for the simultaneous analysis of fourteen water-soluble vitamins (B1, B2, two B3 vitamers, B5, five B6 vitamers, B8, B9, B12 and C) in various food matrices, i.e. maize flour, green and golden kiwi and tomato pulp, is presented here. Analytes were separated by ion-suppression reversed-phase liquid chromatography in less than 10 min and detected in positive ion mode. Sensitivity and specificity of this method allowed two important results to be achieved: (i) limits of detection of the analytes at ng g(-1) levels (except for vitamin C); (ii) development of a rapid sample treatment that minimizes analyte exposition to light, air and heat, eliminating any step of extract concentration. Analyte recovery depended on the type of matrix. In particular, recovery of the analytes in maize flour was > or =70%, with the exception of vitamin C, pyridoxal-5'-phosphate and vitamin B9 (ca 40%); with tomato pulp, recovery was > or =64%, except for vitamin C (41%); with kiwi, recovery was > or =73%, except for nicotinamide (ca. 30%).  相似文献

A novel ultra-performance liquid chromatography electrospray ionization tandem triple quadrupole mass spectrometry method for the simultaneous determination of four water-soluble vitamins, including vitamin B5 (VB5), vitamin B8 (VB8), vitamin B9 (VB9), and vitamin B12 (VB12) in fortified infant foods is developed and validated. A reverse phase UPLC separation system consisting of a Waters ACQUITY UPLC BEH C-18 column (2.1 mm x 100 mm i.d., 1.7 microm) and a binary gradient acetonitrile-water mobile phase is applied for the separation of the four water-soluble vitamins. Formic acid is spiked into the mobile phase to enhance the ionization efficiency. Tandem MS-MS analysis is performed in multi-reaction monitoring mode (MRM). Product-ion traces at m/z 220.1 --> 89.9 for VB5, 245.1 --> 227.1 for VB8, 442.3 --> 295.2 for VB9, and 678.9 --> 147.0 for VB12 are used for quantitation of the corresponding vitamins, and traces at m/z 455.5 --> 308.0 are used for methotrexate (internal standard). Limits of quantitation (LOQs) are 0.016, 0.090, 0.020, and 0.019 microg/L for VB5, VB8, VB9, and VB12, respectively. Intra- and inter-day precisions for the determination of the four vitamins are better than 6.84% and 12.26% in relative standard deviations, and recoveries for the four vitamins are in the range of 86.0~101.5%. The developed approach is applied for the determination of the trace amounts of the vitamins in fortified milk powers and fortified rice powers.  相似文献

采用微乳液相色谱法同时分离7种水溶性维生素(V_(B1)、V_(B2)、V_(B6)、VB_(12)、叶酸、烟酰胺和VC)。考察了微乳流动相体系中表面活性剂、油相、助表面活性剂的种类以及流动相的pH值、柱温等对水溶性维生素分离的影响。优化后微乳体系的组成为:十二烷基硫酸钠(SDS)/聚氧乙烯月桂醇醚(Brij35)/正丁醇/乙酸乙酯/水(质量比为2∶60∶66∶8∶864)。色谱柱为Agilent TC C18(250 mm×4.6 mm,5μm),柱温为30℃,检测波长为254 nm,流速为0.5mL/min。7种水溶性维生素在20 min内达到基线分离。在4~36 mg/L范围内,7种水溶性维生素的质量浓度与峰面积的相关系数均大于0.999 1。不同添加水平下,V_(B1)、V_(B2)、V_(B6)、VC和烟酰胺的平均回收率为93.9%~102.9%。该方法可用于食品和药品中的多种水溶性维生素的分离、鉴别及快速测定。  相似文献

建立了一种新的微乳体系,并成功地应用于微乳液相色谱法(MELC)快速分析脂溶性维生素VA、VD2、VD3和VE。通过对影响分离选择性的主要因素进行考察,得到最佳微乳体系组成为98%(v/v)(50 g/L十二烷基硫酸钠(SDS)-10%(质量分数)正丁醇-1.0%(质量分数)正辛烷-84%水(质量分数))-2%(v/v)乙腈。该微乳体系中,表面活性剂类型和浓度、油相正辛烷的含量、有机添加剂乙腈对脂溶性维生素的分离起到了重要的作用。以Venusil ASB C18色谱柱(150 mm×4.6 mm, 5 μm)为分离柱,流速为0.7 mL/min,检测波长为265 nm,柱温为40 ℃, VA、VD2、VD3和VE在20 min内达到基线分离。4种脂溶性维生素的保留时间和峰面积的相对标准偏差(RSD) (n=5)分别小于2.3%和3.0%; VA、VD2、VD3和VE的线性范围分别为22.0~88.0 mg/L、20.2~81.0 mg/L、24.3~97.2 mg/L和125.0~500.0 mg/L,相应的线性相关系数r2分别为0.9996、0.9994、0.9998、0.9998;检出限(S/N=3)分别为0.37、0.34、0.41和2.12 mg/L。本方法已成功应用于多维元素片(21)中VA与VE的测定,结果令人满意。  相似文献

A comparative study of different surfactants, such as sodium dodecyl sulfate and bis(2-ethylhexyl) sodium sulfosuccinate (AOT), as pseudostationary phases was conducted in order to find the best conditions for the separation of the water- and fat-soluble vitamins by electrokinetic capillary chromatography. Separation was accomplished with AOT in a water-acetonitrile solution in the presence of boric acid-sodium borate buffer. A study of different variables was performed in order to obtain the best resolution and quantification of the vitamins: the variables explored were the pH and concentration of the buffer used, the percentage of acetonitrile, the concentration of surfactant and the applied voltage. Calibration curves and precision data were obtained for each analyte. Finally, a method to analyze these substances in pharmaceutical preparations is proposed.  相似文献

Two spectrophotometric methods, derivative and multivariate methods, were applied for the determination of binary, ternary, and quaternary mixtures of the water-soluble vitamins thiamine HCI (I), pyridoxine HCI (II), riboflavin (III), and cyanocobalamin (IV). The first method is divided into first derivative and first derivative of ratio spectra methods, and the second into classical least squares and principal components regression methods. Both methods are based on spectrophotometric measurements of the studied vitamins in 0.1 M HCl solution in the range of 200-500 nm for all components. The linear calibration curves were obtained from 2.5-90 microg/mL, and the correlation coefficients ranged from 0.9991 to 0.9999. These methods were applied for the analysis of the following mixtures: (I) and (II); (I), (II), and (III); (I), (II), and (IV); and (I), (II), (III), and (IV). The described methods were successfully applied for the determination of vitamin combinations in synthetic mixtures and dosage forms from different manufacturers. The recovery ranged from 96.1 +/- 1.2 to 101.2 +/- 1.0% for derivative methods and 97.0 +/- 0.5 to 101.9 +/- 1.3% for multivariate methods. The results of the developed methods were compared with those of reported methods, and gave good accuracy and precision.  相似文献

It is shown that fat-soluble vitamins A and E and water-soluble vitamins B (B1, B2, B6, and B12) can be separated by high-performance thin-layer chromatography using fractional elution. Glucose and sucrose are simultaneously determined by using a glucose-sensing enzyme electrode combined with a cell that contains immobilized invertase. The electrode current changes linearly with time for several minutes from ca. 1 min after the addition of a glucose-sucrose mixture. The concentration of sucrose (60 μM-6 mM) is determined from the rate of current change in the linear region, and that of glucose (5 μM-1 mM) is determined by extrapolating the straight current-time line to t=0.45 min and by measuring the intercept on the vertical (current) axis at t=0.45 min. The relative standard deviations are 1.8% for glucose and 3.7% for sucrose (n=10). More than 20 food samples can be analysed in 1 h.  相似文献

A spectrophotometric method is proposed for the simultaneous determination of vitamins A, D and E in multivitamin pharmaceutical preparations. This is based on multiple linear regression. Most vitamins are directly extracted from the preparations into n-hexane. Microen-capsulated vitamin A preparations require pretreatment of de-encapsulation before the vitamin is extracted. The wavelength range to be used for each preparation and the optimum spectral mode (absorbance or first-derivative) has been chosen in order to assure correct quantitation and avoid interferences from other absorbing species also extracted by n-hexane. The results obtained were validated by simultaneous HPLC analyses for accuracy and precision.  相似文献

建立了用高效液相色谱-二极管阵列检测器(HPLC-DAD)同时检测化妆品中9种水溶性着色剂(溶剂绿7、食品黄3、食品红17、酸性黄1、酸性红33、食品红4、食品红1、橙黄I、酸性橙7)的检测方法。不同种类的化妆品采用不同的样品前处理方法提取后,用Diamonsil C18色谱柱分离,以乙腈-磷酸二氢钾缓冲溶液(pH 6)为流动相进行梯度洗脱,检测波长为240 nm,15 min内可对9种目标物同时进行检测,且各化合物都达到基线分离。经测定,该方法的平均回收率(n = 9)为85.33%~100.2%,相对标准偏差(RSD)为3.68%~8.20%,检出限为0.01~0.1 mg/L。方法简单、快速,能有效地提取、分离和测定化妆品中9种水溶性着色剂。将该方法用于实际化妆品的检测,结果令人满意。  相似文献

A reversed-phase high-performance liquid chromatographic method is described for the simultaneous determination of vitamins A, D2, D3, E and K1, retinyl acetate, retinyl palmitate, tocopherol acetate, ergosterol and 7-dehydrocholesterol in milk and butter. Narrow-bore columns are recommended because this alternative provides a good separation and efficiency, plus greater economy and sensitivity. Detection limits for individual vitamins range from 0.14 to 6.9 ng. All vitamins are separated in less than 33 min. For the simultaneous determination of these vitamins and provitamins we use two sample pre-treatment methods, a liquid-liquid extraction with hexane or a solid-phase extraction with a C18 cartridge. Recovery studies show good results for all solutes (84-108% and 85-108% for milk and butter, respectively) and the intra-day coefficients of variations range from 1.6 to 4.5%. These methods permit the simple determination of fat-soluble vitamins using a small sample volume.  相似文献
