We assessed the bioactivity of G. kola seeds on Streptococcus pyogenes, Staphylococcus aureus, Plesiomonas shigelloides and Salmonella typhimurium. The crude ethyl acetate, ethanol, methanol, acetone and aqueous extracts were screened by the agar-well diffusion method and their activities were further determined by Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) assays. The extracts were fractionated by Thin Layer Chromatography (TLC). Bioautography was used to assess the activity of the possible classes of compounds present in the more active extracts. Column chromatography was used to purify the active compounds from the mixture, while GC-MS was used to identify the phytocomponents of the fractions. The inhibition zone diameters of the extracts ranged from 0-24 ± 1.1 mm, while MIC and MBC values ranged between 0.04-1.25 mg/mL and 0.081-2.5 mg/mL, respectively. The chloroform/ethyl acetate/formic acid (CEF) solvent system separated more active compounds. The MIC of the fractions ranged between 0.0006-2.5 mg/mL. CEF 3 (F3), CEF 11 (F11) and CEF 12 (F12) revealed the presence of high levels of linoleic acid, 1,2-benzenedicarboxylic acid and 2,3-dihydro-3,5-dihydroxy-6-methyl ester, respectively. The results obtained from this study justify the use of this plant in traditional medicine and provide leads which could be further exploited for the development of new and potent antimicrobials.  相似文献

The increasing numbers of cases of antibiotic resistance among pathogenic bacteria such as Vibrio species poses a major problem to the food and aquaculture industries, as most antibiotics are no longer effective in controlling pathogenic bacteria affecting these industries. Therefore, this study was carried out to assess the antibacterial potentials of crude aqueous and n-hexane extracts of the husk of Cocos nucifera against some selected Vibrio species and other bacterial pathogens including those normally implicated in food and wound infections. The crude extracts were screened against forty-five strains of Vibrio pathogens and twenty-five other bacteria isolates made up of ten Gram positive and fifteen Gram negative bacteria. The aqueous extract was active against 17 of the tested bacterial and 37 of the Vibrio isolates; while the n-hexane extract showed antimicrobial activity against 21 of the test bacteria and 38 of the test Vibrio species. The minimum inhibitory concentrations (MICs) of the aqueous and n-hexane extracts against the susceptible bacteria ranged between 0.6-5.0 mg/mL and 0.3-5.0 mg/mL respectively, while the time kill study result for the aqueous extract ranged between 0.12 Log?? and 4.2 Log?? cfu/mL after 8 hours interaction in 1 x MIC and 2 x MIC. For the n-hexane extract, the log reduction ranged between 0.56 Log?? and 6.4 Log?? cfu/mL after 8 hours interaction in 1 x MIC and 2 x MIC. This study revealed the huge potential of C. nucifera extracts as alternative therapies against microbial infections.  相似文献

The formulation of Garcinia kola seeds into tablet dosage form and evaluation of some physical properties of the tablets are presented. A chemical assay was conducted on the dry, powdered seeds as well as the crude aqueous extract of the seeds. The dry powdered seeds contain 0.003% of flavonoids while the crude extract contained 0.007% of flavonoids based on rutin used as the standard. The powdered material (50 mg) and crude extract (10 mg) were formulated into tablets using the wet granulation method. Named binders were evaluated in these formulations. The various tablet parameters were evaluated, namely: weight variation, thickness and diameter, hardness, friability, disintegration time, dissolution profile and content uniformity. The results indicated that the tablets had good disintegration time, dissolution and hardness/friability profiles. Tablets formulated with starch had the best disintegration properties but were consequently very friable. Tablets formulated from 10 mg of the crude extract needed a larger proportion of diluents, which affected the tablet properties.  相似文献

The present study demonstrated microbial corrosion protection of MS 1010 on cooling tower water using plant-based inhibitors derived from methanolic extraction of dry mangrove leaves (R.mangle and A.marina), and its assessment of antibacterial activity against corrosive bacteria (B.megaterium) was investigated. FT-IR and GC–MS analyzed the inhibitors component and corrosion behavior of MS 1010 on cooling water, with and without inhibitors were analyzed by EIS and Tafel studies. GC–MS spectra confirmed the presence of Myo-Inositol, 4-C-methyl and chromene as major constituents presented on the R.mangle whereas Lupeol, trifluoroacetate and beta-amyrin compounds were found on the A.marina. In the cooling water, these two inhibitors demonstrated outstanding antibacterial activity and controlled biofilm growth. As plant-based inhibitors were used in cooling water systems, EIS data showed a significant increase in Rct value when compared to the control system. Tafel plot indicates inhibitors have mixed inhibitory effects and for the systems with and without inhibitors, the icorr value was 1.5649A and 2.0875A, respectively. At the optimal dose of 25 ppm, the inhibitory efficiency of MERM and MEAM was 81% and 80%, respectively. The overall discussion reveals that inhibitor substances can be absorbed on the metal surface and then act as a dual role in inhibiting corrosive bacterial growth and barrier to the corrosion process in the cooling water system.  相似文献

Shigellosis is an important cause of worldwide morbidity and mortality among young children and old people for which treatment with antimicrobial agents is limited. Hence, the need for curative potentials obtainable from medicinal plants becomes inevitable. This study was carried out to assess the antibacterial potentials of crude methanolic extract of the stem bark of Acacia mearnsii against some selected bacteria of clinical importance in shigellosis. The bacteria were inhibited by the extract to produce concentration dependent inhibition zones. The extract exhibited a varied degree of antibacterial activity against all the tested isolates. The MIC values for Gram negative (0.0391-0.3125) mg/mL and those of Gram positive bacteria (0.0781-0.625) mg/mL indicated that the Gram negative bacteria were more inhibited by the extract than the Gram positive bacteria. Average log reduction in viable cell count in time-kill assay ranged between -2.456 Log?? to 2.230 Log?? cfu/mL after 4 h of interaction, and between -2.921 Log?? and 1.447 Log?? cfu/mL after 8 h interaction in 1× MIC and 2× MIC of the extract. The study provided scientific justification for the use of the crude methanolic extract from the stem bark of A. mearnsii in shigellosis. The degree of the antibacterial activity indicated that the crude extract is a potential source of bioactive compounds that could be useful for the development of new antimicrobial agents capable of decreasing the burden of drug resistance and cost of management of diseases of clinical and public health importance in South Africa.  相似文献

The complexation of In(III) and U(VI) with thiodipropionic acid has been investigated polarographically in water and water-methanol solutions at 30 +/- 0.1 degrees . All the chelates belong to polaro-graphically reversible systems. With indium(III), complexes with metal to ligand ratios of 1:1, 1:2, 1:3 and 1:4 are found at pH 4.8. Uranium(VI) is found to form three successive complexes with metal to ligand ratios of 1:1,1:2 and 1:3 in 0.1M HCl, with 0.1M KCl as supporting electrolyte.  相似文献

As part of an International Cooperative Biodiversity Groups (ICBG) program to study Jordan's biodiversity, the relative levels of antioxidant activity and the total phenolic content of aqueous and methanolic extracts of a total of 95 plant species, all of Jordanian origin and those collected at random, have been measured. The total phenolic content of aqueous and methanolic extracts of the investigated plant species ranged from 4.4 to 78.3 mg and from 2.1 to 52.8 mg gallic acid equivalents g(-1) dry weight, respectively, while the total antioxidant capacity ranged from 20.0 to 916.7 and from 15.1 to 915.6 micromol Trolox equivalents g(-1) dry weight, respectively. Based on this collection, approximately 5% of assayed plants showed high levels of antioxidant activity. There was a significant linear correlation between antioxidant activity and total phenolic content for aqueous and methanolic extracts, suggesting that phenolic compounds were the predominant antioxidant components in the investigated plant species. Interestingly, a few of the collected plants had high-antioxidant activity yet "low" phenolic content includes Ceratonia siliqua and Viscum cruciatum. These plants may serve as sources of antioxidants with new chemotypes.  相似文献

Plant extracts are one of the best possible sources of bioactive molecules, and are being used globally as an antioxidants and natural antimicrobial compounds. In current study, Schefflera vinosa leaves extract was prepared through Soxhlet extraction procedure using methanol and chloroform as solvents. The extract was investigated for total antioxidant, phenolic and flavonoid contents, free radical scavenging and antimicrobial activities. The free radical scavenging activities were evaluated through 2,2- diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis-3-ethylbenzotiazolin-6-sulfonic acid (ABTS) and Ferric-reducing/ antioxidant power (FRAP) assay. The antimicrobial activity of extract was determined through poisoned food method. The methanolic extract has exhibited high antioxidant, phenolic, and flavonoid activities compared to chloroform extract. Similarly, free radical scavenging activities (ABTS, DPPH and FRAP) were higher in methanolic extract. Further, Fourier-Transform Infrared Spectroscopy (FTIR) used to determine the functional group and Gas chromatography-mass spectrometry (GC–MS) to elucidate volatile composition of the crude extract. Different functional group like N-H, O-H, C-O, C-N, C-H, C=O, C≡C and C-O-H presence indicate the existence of many metabolites in the extracts. GC–MS study identified 61 compounds and subsequently, these molecules were screened virtually using DockThor. Furthermore, antimicrobial study was confirmed against rice pathogens like Magnaporthe oryzae (M. oryzae) and Xanthomonas oryzae pv. oryzae (Xoo). Molecular docking study further suggested that phytomolecules (3-Isopropoxy-1,1,1,7,7,7-hexamethyl-3,5,5-tris (trimethylsiloxy) tetrasiloxane, and 2-Methoxy-5-methylthiophene) targets Histone Deacetylase (HDAC) of M. oryzae and Peptide Deformylase (PDF) of Xoo, which could inhibit their growth. Hence, this study indicated that Schefflera vinosa extracts could be an important ingredient as an antioxidant as well as antimicrobial agent against rice pathogens.  相似文献

Roasting of Coffea arabica L. seeds gives rise to chemical reactions that produce more than 800 compounds, some being responsible for the desired organoleptic properties for which the beverage called “coffee” is known. In the industry, the “roasting profile,” that is, the times and temperatures applied, is key to influence the composition of roasted coffee beans and the flavour of the beverage made from them. The impact of roasting on the chemical composition of coffee has been the subject of numerous studies, including by nuclear magnetic resonance (NMR) spectroscopy. However, the roasting equipment and profiles applied in these studies are often far from real industrial conditions. In this work, the effects of two critical technological parameters of the roasting process, namely, the “development time” (the period of time after the “first crack,” a characteristic noise due to seed disruption) and the final roasting temperature on coffee extracts, were investigated. Seeds were roasted at pilot scale according to 13 industrial roasting profiles and extracted in D2O. The extracts were analysed by 1H NMR experiments. The NMR spectra were compared using (a) quantitative analysis of main signals by successive orders of magnitude and (b) chemometric tools (principal component analysis, partial least squares and sparse-orthogonal partial least squares analysis). This allowed to identify compounds, which may serve as markers of roasting and showed that changes in chemical composition can be detected even for slight change in final temperature (~1°C) or in total roasting time (~25 s).  相似文献

Allelopathic effects of corn (Zea mays) extracts was studied, against seed germination and seedling growth of Phalaris minor, Helianthus annuus, Triticumaestivum, Sorghum halepense, Z. mays. Bioassay results showed that aqueous extracts of corn root and shoot, markedly affected seed germination, and other parameters compared with related controls. Preliminary phytochemical screening revealed the presence of various phytochemicals such as tannins, phlobatannins, flavonoids, terpenoids and alkaloids in both roots and shoot aqueous extracts. However, saponins were only present in the shoot aqueous extract, while in shoot ethanol extracts, only terpenoids and alkaloids were detected. Additionally, total polyphenolic (TPC) content in aqueous extracts of corn root and shoot, plus ethanol extracts of corn shoot were determined using an Ultraviolet–visible spectroscopy. Results revealed TPC content of the corn shoot aqueous extract showed the highest yield, compared to other extracts. These findings suggest that phytochemicals present in Z. mays extracts may contribute to allelopathy effect.  相似文献

Methanolic extracts of Myrtus communis leaves from two Italian regions (Calabria and Sardinia) were processed to determine the content of myrtenol, linalool and eucalyptol. Among the Calabrian and Sardinian myrtle samples, linalool and eucalyptol chemotypes were prevalent. The extracts were also tested for antioxidant, antibacterial and antifungal activities. Myrtle leaves samples were dried and extracted through maceration. Partition chromatography was adopted to separate myrtenol, linalool and eucalyptol fractions. Analyses were performed through GC and GC-MS. Some of the samples showed a good scavenger activity evidenced by DPPH radical scavenging assay and beta-carotene bleaching test. Antibacterial and antifungal activities were generally weak. The phytochemical and biological characterization of all the extracts were determined with an aim to characterize the intra-specific biodiversity of myrtle populations.  相似文献

Single-column ion-chromatography (SCIC) was investigated as a routine, rapid, precise and selective analytical method for the determination of chromium(VI) in aqueous extracts of soil and sewage sludge. Chromatographic parameters were optimized for determination of Cr(VI), NO(-)(3) and SO(2-)(4). A low-capacity resin-based column was used for the separation and the anions were determined by conductometric detection. p-Hydroxybenzoic acid (5mM) at pH 8.5 was used as the eluent. The limit of detection, defined as S/N = 3, was 92 mug/l. The resolution between Cr(VI) and SO(2-)(4) was 2.8, the precision ranged from 0.9% for NO(-)(3) to 2.0% for Cr(VI) with a 500-mul injection. The SCIC results for Cr(VI) agreed closely with those obtained by inductively coupled argon-plasma emission and spectrophotometry.  相似文献

The antibacterial activity of essential oil and methanolic extract of Teucrium polium was determined against Pseudomonas aeruginosa, Pantoea agglomerans, Brenneria nigrifluens, Rhizobium radiobacter, Rhizobium vitis, Streptomyces scabies, Ralstonia solanacearum, Xanthomonas campestris and Pectobacterium cartovorum by disc diffusion method. Minimum inhibitory concentration and minimum bactericidal concentration were determined by using the serial dilution method. Chemical composition of essential oil and methanolic extract was determined by GC-MS and HPLC. α-Pinene (25.769%) and myrcene (12.507) were of the highest percentage in T. polium essential oil, and sinapic acid (15.553 mg/g) and eugenol (6.805 mg/g) were the major compounds in the methanolic extract. Our results indicate that both methanolic extract and essential oil did not show antibacterial activity against P. aeruginosa. Also the essential oil did not show antibacterial activity against P. cartovorum. In general, both methanolic extract and essential oil showed the same antibacterial activity against R. solanacearum, P. agglomerans, B. nigrifluens and S. scabies.  相似文献

Plant pathogenic bacteria and fungi negatively affect a large number of important fruit and vegetables during the growing season and throughout postharvest storage. Therefore, the current study focuses on the preparation of N-(benzyl)chitosan derivatives as antimicrobial agents to control these microorganisms. Chitosan was reacted with a set of aromatic aldehydes by reductive amination involving formation of the corresponding imines, followed by reduction with sodium borohydride to produce the N-(benzyl)chitosan derivatives. The end products were analyzed using 1H NMR spectroscopy and the degrees of substitution ranged from 0.12 to 0.29. The antibacterial activity was evaluated in vitro against the crown gall disease Agrobacterium tumefaciens (Family: Rhizobiaceae; Class: Alpha Proteobacteria) and the soft mold disease Erwinia carotovora (Family: Enterobacteriaceae; Class: Gamma Proteobacteria) by the nutrient agar dilution method. A higher activity of chitosan and its derivatives was obtained with N-(o-ethylbenzyl)chitosan with a MIC of 500 mg/L against E. carotovora, while N-(o,p-diethoxybenzyl)chitosan was the most active one against A. tumefaciens with a MIC of 1050 mg/L. In addition, the in vitro antifungal assessment against root rot disease Fusarium oxysporum (Family: Tuberculariaceae; Class: Deuteromycetes) and the leaf spots and blights disease Pythium debaryanum (Family: Pythiaceae; Class: Oomycetes) was tested by a mycelial radial growth technique. The data showed that N-(o,p-diethoxybenzyl)chitosan was the most active one with an EC50 of 400 and 468 mg/L for F. oxysporum and P. debaryanum, respectively. In addition, chitosan derivatives had a detrimental effect on spore germination for F. oxysporum. Most of these derivatives exhibited high inhibition percentage (>90%) of spore germination at 1000 mg/L.  相似文献

The present study comprises the determination of some phenolic acids from the leaves and flowers of Alhagi maurorum by HPLC-DAD, confirmed by LC-MS-APCI. The antioxidant properties and measurements of the total phenolic contents of the extracts were assessed by 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging and Folin-Ciocalteu methods, respectively. It was found that the leaf extract had higher antioxidant potential (83.5%) than the flower extract (72.3%). The antioxidant properties and total phenolic contents of the leaves were higher than those of the flowers.  相似文献

The potentiometric and conductometric titration was used to study complexation between some metal ions and 3-(4-bromophenylazo)pentane-2,4-dione (L) in aqueous ethanol. The standard thermodynamic characteristics of the complex formation reaction were determined. The effective atomic charges in the tautomeric forms of L (azo enol, azo keto, hydrazo) were calculated by MO LCAO quantum-chemical method in the Hückel approximation.  相似文献

The GPC procedure for analyzing molecular mass characteristics of cardo poly(benzimidazoles) has been developed. In most cases, the reaction between 4,4′-oxydibenzene-1,2-diamine and 4,4′-(3-oxo-1,3-dihydroisobenzofuran-1,1-diyl)dibenzoic acid in Eaton’s reagent is accompanied by formation of a microgel. Depending on the synthesis conditions (temperature, duration of heating, and content of phosphorus pentoxide in the reaction mixture), polymers with both unimodal and bimodal molecular mass distributions can be prepared. Formation of the microgel fraction is observed for many representatives of poly(benzimidazoles) of various chemical structures. Based on the experimental evidence, the most probable pathway is suggested for the branching side reaction of poly(benzimidazoles) during their synthesis in Eaton’s reagent.  相似文献

Solanum ferrugineum Jacq. is a wild species that in previous analysis reported a significant antioxidant capacity. The aim of our research was to determine total phenolic content (TPC) and total flavonoid content (TFC), and the phenolic composition by HPLC-DAD and HPLC/ESI-MS/TOF of methanolic extracts of S. ferrugineum fruits, collected from Paredones, Jiquilpan, and Fray Dominguez, Pajuacarán in the Mexican state of Michoacán. TPC and TFC were determined by the spectrophotometric Folin–Ciocalteu reagent and the AlCl3 method, respectively. TPC in S. ferrugineum fruit [31.41?±?0.91?mg gallic acid equivalent/g dry tissue (DT)] was similar to those reported for Turkey berry (Solanum torvum Sw.) and eggplant (Solanum melongena L.) fruits. The TFC values of S. ferrugineum fruits (29.14?±?4.99?mg catechin equivalent /g DT) corresponded to 80.24% of the TPC. Eight phenolic compounds (PC) were identified by HPLC analysis. The main PC identified in S. ferrugineum fruits were chlorogenic acid, caffeic acid, p-coumaric acid, gallic acid, quercetin, and kaempferol. S. ferrugineum fruits could be used as a starting material for the extraction of high-value PC with potential applications.  相似文献

We use molecular dynamics simulations to study the stabilization of the hexameric, cagelike silicate with double three-ring structure in aqueous and methanolic solutions. We find that in purely aqueous environments its stabilization requires the presence of both tetramethylammonium and tetraethylammonium (TEA) cations and involves the formation of a stable TMA layer which leads to a water-silicate heteronetwork clathrate. We also find that TEA alone can facilely stabilize the hexamer when methanol cosolvent is added, in accordance with experiment. The mechanism of this stabilization, however, differs from that in purely aqueous environments. Because of the unique properties of water-methanol mixtures, the organosilicate complex does not participate in heteronetwork clathrates but resides in a large solvent cavity; that is, it is forced out of true solution.  相似文献
