Diaryliodonium salts containing the 2-thienyl group as an example of an electron-rich heteroaromatic moiety proved to be very potent precursors for the nucleophilic, regioselective no-carrier-added (nca) radiofluorination of various arenes. It even allowed the nucleophilic introduction of nca [18F]fluoride into electron-rich arene compounds in one step. The influences of the substitution pattern, of counteranions, and of different reaction conditions were studied. Effects of counterions could be explained by the influence of solvent on ion pair separation of precursor salts. Different aryl(2-thienyl)iodonium salts were used as precursors, where the homoaromatic group systematically varied from bearing electron-deficient to electron-rich substituents. Relative rates of exchange kinetics correlated linearly with Hammett constants of the appropriate substituents confirming a nucleophilic aromatic substitution reaction of high reactivity and low selectivity. 相似文献

2,5-Dibutoxy-4-(α-thienyl)-Δ3-dihydropyran was obtained by bromoalkoxylation at the double bond and dehydrobromination of 2-butoxy-4-(α-thienyl)-Δ5-dihydropyran. Summary The reactions of Cu(NO3)2 · 3 H2O with 2- (2-thienyl)-1-(2-thienylmethyl)benzimidazole (L) and of CuCl2 · 2H2O with 2-(2-thienyl)benzimidazole (L) have been carried out. The crystal structure of two isolated complexes,(1) and(2), have been determined by single-crystal x-ray diffraction methods. Crystals of(1) are monoclinic, space groupP21
/c, witha=9.884(4),b=9.892(4),c=18.732(8) Å, =114.89(2)°,Z=2;R=0.068 for 1909 observed reflections. Crystals of (2) are orthorhombic, space groupPbcn, witha=14.835(1),b=8.193(1),c=20.493(1) Å,Z=4;R=0.086 for 415 observed reflections. In the complex (1) the copper co-ordination is tetragonal (compressed octahedral) and involves a nitrogen atom of the organic ligand [Cu-N 1.970(6) Å] and two oxygen atoms of the nitrate group [Cu-O 2.318(7) and 2.246(9) Å] which appears to coordinate nearly symmetrically to the metal; however it has unusually large thermal motion which suggests a fluxional behaviour. In the complex (2) the co-ordination is square planar and involves an imidazole nitrogen [Cu-N 1.95(3) Å] and a chlorine atom [Cu-Cl 2.27(1) Å]. A long interaction Cu-S=3.37(2) Å completes the co-ordination polyhedron to a very elongated distorted tetragonal bipyramid. In both compounds a thiophene ring is disordered. 相似文献

As degradation product of Antineoplaston A10 in vivo, phenylacetyl glutamine showed antitumor activities. According to literatures, we designed and radiosynthesized a phenylacetyl glutamine derivative, which was achieved under a mild reaction condition. Evaluations in vitro and in vivo were performed on tumor bearing mice. Excitingly, the radiochemical purity of (S)-2-((S)-2-(4-(3-fluoropropyl)benzamido)-3-phenylpropanamido)pentanedioic acid ([18F]FBPPA) was 98%, and besides the best radiochemical yield was up to 46%. T/Bl (Tumor/Blood) and T/M (Tumor/Muscle) ratios of [18F]FBPPA at 60 min post injection were 2.33 and 3.51. Meanwhile, it showed satisfied stability in vitro and in vivo, compared with 2-[18F]fluorodeoxyglucose ([18F]FDG). Although [18F]FBPPA deserved further studies to make optimizations on its structure, the results revealed it might become a potential PET imaging agent for detecting tumors. 相似文献

Condensation of 1-amino-2-hydroxynaphthalene with thenoyl chloride in 1-methyl-2-pyrrolidinone medium afforded 2-(2-thienyl)naphtho[1,2-d]oxazole. The latter was brought into electrophilic substitution reactions like nitration, bromination, sulfonation, formylation, and acylation. d-glucosamine at concentration of certain range could kill tumor cells without influencing normal cells. There are also some reports on the antitumor activity of d-glucosamine and its derivatives in murine models. It was therefore postulated that d-glucosamine might have the potential to invade tumor cells. We designed and radiosynthesized a glucosamine derivative, N-(2-[18F]fluoro-4-nitrobenzoyl)glucosamine ([18F]FNBG([18F]7)). Evaluations in vitro and in vivo were performed on tumor bearing mice. Excitingly, the radiochemical purity of [18F]FNBG([18F]7) was 99%, and besides the best radiochemical yield was up to 35%. The best T/Bl (Tumor/Blood) and T/M (Tumor/Muscle) ratios of [18F]FNBG([18F]7) were 4.40 and 4.84. Although [18F]FNBG([18F]7) deserved further studies, the results revealed it might become a potential PET imaging agent for detecting tumors. 相似文献

2-(2-Thienyl)-1(3)H-imidazo{cm[4,5-f]}quinoline was synthesized by the Weidenhagen reaction of quinoline-5,6-diamine with thiophene-2-carbaldehyde. Its methylation in the system KOH-DMSO gave isomeric 1-methyl-2-(2-thienyl)-1H- and 3-methyl-2-(2-thienyl)-3H-imidazo{cm[4,5-f]}quinolines, the latter being the major product. The 3-methyl derivative was subjected to electrophilic substitution reactions (bromination, nitration, formylation, acylation, sulfonation). 2-[(18)F]Fluoro-L-phenylalanine and 2-[(18)F]fluoro-L-tyrosine have been developed as promising radiopharmaceuticals for molecular imaging using positron emission tomography (PET). However, the lack of a convenient radiosynthetic pathway has limited their practical use. In this work a new three-step nucleophilic synthesis of these compounds starting from [(18)F]fluoride is described. Corresponding precursors (1a and 1b) were (18)F-fluorinated by isotopic exchange, followed by the removal of an activating formyl group with Rh(PPh(3))(3)Cl and subsequent hydrolysis of protecting groups in acidic medium. All reactions were carried out using both conventional and microwave heating. Conventional heated reactions yielded the desired products 2-[(18)F]Fphe and 2-[(18)F]Ftyr in 43% and 49% whereas radiochemical yields of 34% and 43%, respectively, were obtained when they were heated by microwaves. Under optimized conditions the enantiomeric purity was ≥94% for both radiopharmaceuticals. 相似文献

Taking advantages of nuclear analytical techniques (NATs) with non-destruction, multielement capability, small and estimable uncertainties over a wide range of sample sizes, the sampling behavior of multielements for a home-made natural matrix material was studied with sample sizes ranging from several hundred mg down to tenths ng, namely nine orders of magnitude, by a combination of three NATs, neutron activation analysis (NAA), proton induced X-ray emission (PIXE) and synchrotron radiation X-ray flurescence (SR-XRF), in an effort to explore a procedure for the development of certified reference materials (CRMs) suitable for quality control of microanalysis. For accurately weighable sample sizes (>1?mg), sampling uncertainties for 13 elements were found to be less than 1% by INAA. For sample sizes unable to be accurately weighed (<1?mg), PIXE and SR-XRF were used, respectively. Sampling uncertainties were found to be less than 1% at sample sizes of tenth mg level for seven elements, and less than 10% on ng levels for three elements. Considering these three elements have satisfied homogeneity (sampling uncertainty less than 10%) at ng sample size level, any one of them can be served as a ??relative balance?? in sampling behavior characterization of multielements on sample size levels larger than ng (e.g., ??g level). On this basis, sampling uncertainties for nine elements were found to be less than 10% on ??g sample size level by INAA. The results indicate that the matrix is eligible as a candidate of CRMs suitable for quality control of solid sampling microanalysis. 相似文献

2-Methylthiophene (1) was treated at 0°C with liquid bromine to form 3,5-dibromo-2-methylthiophene (2) which reacted with tributyl borate to give 2-methyl-3-bromo-5-boronate thiophene (3) at −78°C. Treatment of 3 with 3,4-difluorobrombenzene gave 2-methyl-3-bromo-5-(3,4-difluorophenyl)thiophene (4). Finally, a novel photochromic dithienylethene compound, 1,2-bis [2-methyl-5(3,4-difluorophenyl)-3-thienyl]perfluorocyclopentene
(DT-1), was synthesized by the reaction of 4 with perfluorocyclopentene at −78°C. The compound (DT-1) was characterized by IR, NMR, MS, elemental analysis and its photochromic behavior was also discussed.

A potential dopamine D4 receptor ligand, 1-(4-(4-(4-fluorobenzyl)-1-piperazinyl)butyl)indolin-2-one (4) was synthesized through a four-step process and its affinity and selectivity for dopamine D2-like receptors was determined through in vitro receptor binding assay. [18F]4 was prepared using a one-pot two-step method with total radiochemical yield 21.2 % (decay-corrected). The molar radioactivity was around 135 GBq/μmol and the radiochemical purity was greater than 95.5 %. The partition coefficient (Log P) of [18F]4 was determined to be 2.10 ± 0.30 through octanol experiment. The in vivo biodistribution and the competitive distribution of [18F]4 in rat exposed that the tracer passes through blood–brain-barrier (BBB) and may specifically bind to D4 receptor. Metabolite analysis revealed that there was no metabolism of [18F]4 in brain. Conclusively, these preliminary results demonstrated that [18F]4 shows promises as a radioligand for the in vivo study of dopamine D4 receptor. 相似文献
