RNA‐cleaving DNAzymes are useful tools for intracellular metal‐ion sensing and gene regulation. Incorporating stimuli‐responsive modifications into these DNAzymes enables their activities to be spatiotemporally and chemically controlled for more precise applications. Despite the successful development of many caged DNAzymes for light‐induced activation, DNAzymes that can be intracellularly activated by chemical inputs of biological importance, such as reactive oxygen species (ROS), are still scarce. ROS like hydrogen peroxide (H2O2) and hypochlorite (HClO) are critical mediators of oxidative stress‐related cell signaling and dysregulation including activation of immune system as well as progression of diseases and aging. Herein, we report ROS‐activable DNAzymes by introducing phenylboronate and phosphorothioate modifications to the Zn2+‐dependent 8–17 DNAzyme. These ROS‐activable DNAzymes were orthogonally activated by H2O2 and HClO inside live human and mouse cells.  相似文献

Graphitic carbon nitride (g‐C3N4) has been used as photosensitizer to generate reactive oxygen species (ROS) for photodynamic therapy (PDT). However, its therapeutic efficiency was far from satisfactory. One of the major obstacles was the overexpression of glutathione (GSH) in cancer cells, which could diminish the amount of generated ROS before their arrival at the target site. Herein, we report that the integration of Cu2+ and g‐C3N4 nanosheets (Cu2+–g‐C3N4) led to enhanced light‐triggered ROS generation as well as the depletion of intracellular GSH levels. Consequently, the ROS generated under light irradiation could be consumed less by reduced GSH, and efficiency was improved. Importantly, redox‐active species Cu+–g‐C3N4 could catalyze the reduction of molecular oxygen to the superoxide anion or hydrogen peroxide to the hydroxyl radical, both of which facilitated the generation of ROS. This synergy of improved ROS generation and GSH depletion could enhance the efficiency of PDT for cancer therapy.  相似文献

Photodynamic therapy (PDT) is widely used to treat diverse diseases, but its dependence on oxygen to produce cytotoxic reactive oxygen species (ROS) diminishes the therapeutic effect in a hypoxic environment, such as solid tumors. Herein, we developed a ROS‐producing hybrid nanoparticle‐based photosensitizer capable of maintaining high levels of ROS under both normoxic and hypoxic conditions. Conjugation of a ruthenium complex (N3) to a TiO2 nanoparticle afforded TiO2‐N3. Upon exposure of TiO2‐N3 to light, the N3 injected electrons into TiO2 to produce three‐ and four‐fold more hydroxyl radicals and hydrogen peroxide, respectively, than TiO2 at 160 mmHg. TiO2‐N3 maintained three‐fold higher hydroxyl radicals than TiO2 under hypoxic conditions via N3‐facilitated electron–hole reduction of adsorbed water molecules. The incorporation of N3 transformed TiO2 from a dual type I and II PDT agent to a predominantly type I photosensitizer, irrespective of the oxygen content.  相似文献

A straightforward aqueous synthesis of MoO3?x nanoparticles at room temperature was developed by using (NH4)6Mo7O24?4 H2O and MoCl5 as precursors in the absence of reductants, inert gas, and organic solvents. SEM and TEM images indicate the as‐prepared products are nanoparticles with diameters of 90–180 nm. The diffuse reflectance UV‐visible‐near‐IR spectra of the samples indicate localized surface plasmon resonance (LSPR) properties generated by the introduction of oxygen vacancies. Owing to its strong plasmonic absorption in the visible‐light and near‐infrared region, such nanostructures exhibit an enhancement of activity toward visible‐light catalytic hydrogen generation. MoO3?x nanoparticles synthesized with a molar ratio of MoVI/MoV 1:1 show the highest yield of H2 evolution. The cycling catalytic performance has been investigated to indicate the structural and chemical stability of the as‐prepared plasmonic MoO3?x nanoparticles, which reveals its potential application in visible‐light catalytic hydrogen production.  相似文献

Coordination compounds of copper have been invoked as major actors in processes involving the reduction of molecular oxygen, mostly with the generation of radical species the assignment for which has, so far, not been fully addressed. In the present work, we have carried out studies in solution and on surfaces to gain insights into the nature of the radical oxygen species (ROS) generated by a copper(II) coordination compound containing a thioether clip‐phen derivative, 1,3‐bis(1,10‐phenanthrolin‐2‐yloxy)‐N‐(4‐(methylthio)benzylidene)propan‐2‐amine (2CP‐Bz‐SMe), enabling its adsorption/immobilization to gold surfaces. Whereas surface plasmon resonance (SPR) and electrochemistry of the adsorbed complex indicated the formation of a dimeric CuI intermediate containing molecular oxygen as a bridging ligand, scanning electrochemical microscopy (SECM) and nuclease assays pointed to the generation of a ROS species. Electron paramagnetic resonance (EPR) data reinforced such conclusions, indicating that radical production was dependent on the amount of oxygen and H2O2, thus pointing to a mechanism involving a Fenton‐like reaction that results in the production of OH..  相似文献

Design of green, safe, and sustainable process for the synthesis of hydrogen peroxide (H2O2) is a very important subject. Early reported processes, however, require hydrogen (H2) and palladium‐based catalysts. Herein we propose a photocatalytic process for H2O2 synthesis driven by metal‐free catalysts with earth‐abundant water and molecular oxygen (O2) as resources under sunlight irradiation (λ>400 nm). We use graphitic carbon nitride (g‐C3N4) containing electron‐deficient aromatic diimide units as catalysts. Incorporating the diimide units positively shifts the valence‐band potential of the catalysts, while maintaining sufficient conduction‐band potential for O2 reduction. Visible light irradiation of the catalysts in pure water with O2 successfully produces H2O2 by oxidation of water by the photoformed valence‐band holes and selective two‐electron reduction of O2 by the conduction band electrons.  相似文献

Excessive reactive oxygen species (ROS) can oxidatively damage DNA to cause severe biological consequences. In the study, a natural flavonoid, myricitrin (myricetin‐3‐O‐α‐L‐rhamnopyranoside), was found to have a protective effect against hydroxyl‐induced DNA damage (IC50 159.86 ± 54.24 μg/mL). To investigate the mechanism, it was determined by various antioxidant assays. The results revealed that myricitrin could effectively scavenge ·OH, ·O2?, DPPH· (1,1‐diphenyl‐2‐picrylhydrazyl radical), and ABTS+· (2,2′‐Azino‐bis(3‐ethylbenzothiazoline‐6‐sulfonic acid) radicals (IC50 values were respectively 69.71 ± 5.93, 69.71 ± 5.93, 25.34 ± 2.14, and 1.71 ± 0.09 μg/mL), and bind Cu2+ (IC50 27.33 ± 2.36 μg/mL). Based on the mechanistic analysis, it can be concluded that: (i) myricitrin can effectively protect against hydroxyl‐induced DNA oxidative damage via ROS scavenging and deoxynucleotide radicals repairing approaches. Both approaches can be attributed to its antioxidant. From a structure‐activity relationship viewpoint, its antioxidant ability can be attributed to the ortho‐dihydroxyl moiety, and ultimately to the stability of its oxidized form ortho‐benzoquinone; (ii) its ROS scavenging is mediated via metal‐chelating, and direct radical‐scavenging which is through donating hydrogen (H·) and electron (e); and (iii) its protective effect against DNA oxidative damage may be primarily responsible for the pharmacological effects, and offers promise as a new therapeutic reagent for diseases from DNA oxidative damage.  相似文献

1,3‐Bis(ethylamino)‐2‐nitrobenzene, C10H15N3O2, (I), and 1,3‐bis(n‐octylamino)‐2‐nitrobenzene, C22H39N3O2, (II), are the first structurally characterized 1,3‐bis(n‐alkylamino)‐2‐nitrobenzenes. Both molecules are bisected though the nitro N atom and the 2‐C and 5‐C atoms of the ring by twofold rotation axes. Both display intramolecular N—H...O hydrogen bonds between the amine and nitro groups, but no intermolecular hydrogen bonding. The nearly planar molecules pack into flat layers ca 3.4 Å apart that interact by hydrophobic interactions involving the n‐alkyl groups rather than by π–π interactions between the rings. The intra‐ and intermolecular interactions in these molecules are of interest in understanding the physical properties of polymers made from them. Upon heating in the presence of anhydrous potassium carbonate in dimethylacetamide, (I) and (II) cyclize with formal loss of hydrogen peroxide to form substituted benzimidazoles. Thus, 4‐ethylamino‐2‐methyl‐1H‐benzimidazole, C10H13N3, (III), was obtained from (I) under these reaction conditions. Compound (III) contains two independent molecules with no imposed internal symmetry. The molecules are linked into chains via N—H...N hydrogen bonds involving the imidazole rings, while the ethylamino groups do not participate in any hydrogen bonding. This is the first reported structure of a benzimidazole derivative with 4‐amino and 2‐alkyl substituents.  相似文献

The condensation polymerization of isophthalodihydrazide and diphenyl isophthalate affords poly(isophthaloylhydrazine‐1,2‐diyl). High‐molecular‐weight poly(5‐tert‐butylisophthaloylhydrazine‐1,2‐diyl) is prepared by the polycondensation of 5‐tert‐butylisophthalodihydrazide and bis(4‐nitrophenyl) 5‐tert‐butylisophthalate in NMP at 100 °C. Td of the poly(diacylhydrazine) is observed above 300 °C. No Tg is observed below Td. The high‐molecular‐weight poly(diacylhydrazine) exhibits a film‐forming ability. The poly(diacylhydrazine) decomposes on treatment with an oxidant such as sodium hypochlorite solution to obtain the corresponding carboxylic acid and nitrogen. However, poly(diacylhydrazine) was stable to oxygen and hydrogen peroxide even in the presence of transition metal ions. © 2008 Wiley Periodicals, Inc. The C‐terminal octapeptide of cholecystokinin (CCK8) includes some easily oxidizable amino acids. The oxidation of CCK8 by reactive oxygen species (ROS) such as hydrogen peroxide (H2O2) and hydroxyl radicals (OH?) was investigated using reversed‐phase high performance liquid chromatography (RP‐HPLC) and subsequent electrospray ionization mass spectrometry. The mechanism of oxidation of CCK8 in the H2O2 system differed from that of CCK8 in the Fenton system, in which OH? are produced. In the H2O2 system, 28Met and 31Met were oxidized to methionine sulfoxide, and no further oxidation or degradation/hydrolysis occurred. On the other hand, in the Fenton system, 28Met and 31Met residues were oxidized to methionine sulfone via the formation of methionine sulfoxide. In addition, the oxidized product was observed at the Trp residue but not at the Tyr residue, and small peptide fragments from CCK8 were observed in the Fenton system. From these results, it was concluded that 28Met and 31Met residues of CCK8 are susceptible to oxidation by ROS. Copyright © 2009 John Wiley & Sons, Ltd.  相似文献

The non‐aqueous Li–air (O2) battery has attracted intensive interest because it can potentially store far more energy than today′s batteries. Presently Li–O2 batteries suffer from parasitic reactions owing to impurities, found in almost all non‐aqueous electrolytes. Impurities include residual protons and protic compounds that can react with oxygen species, such as the superoxide (O2?), a reactive, one‐electron reduction product of oxygen. To avoid the parasitic reactions, it is crucial to have a fundamental understanding of the conditions under which reactive oxygen species are generated in non‐aqueous electrolytes. Herein we report an in situ spectroscopic study of oxygen reduction on gold in a dimethyl sulfoxide electrolyte containing phenol as a proton source. It is shown directly that O2?, not HO2, is the first stable intermediate during the oxygen reduction process to hydrogen peroxide. The unusual stability of O2? is explained using density functional theory (DFT) calculations.  相似文献

In photodynamic therapy (PDT), the level of reactive oxygen species (ROS) produced in the cell directly determines the therapeutic effect. Improvement in ROS concentration can be realized by reducing the glutathione (GSH) level or increasing the amount of photosensitizer. However, excessive amounts photosensitizer may cause side effects. Therefore, the development of photosensitizers that reduce GSH levels through synergistically improving ROS concentration in order to strengthen the efficacy of PDT for tumor is important. We report a nano‐metal–organic framework (CuII‐metalated nano‐MOF {CuL‐[AlOH]2}n (MOF‐2, H6L=mesotetrakis(4‐carboxylphenyl)porphyrin)) based on CuII as the active center for PDT. This MOF‐2 is readily taken up by breast cancer cells, and high levels of ROS are generated under light irradiation. Meanwhile, intracellular GSH is considerably decreased owing to absorption on MOF‐2; this synergistically increases ROS concentration and accelerates apoptosis, thereby enhancing the effect of PDT. Notably, based on the direct adsorption of GSH, MOF‐2 showed a comparable effect with the commercial antitumor drug camptothecin in a mouse breast cancer model. This work provides strong evidence for MOF‐2 as a promising new PDT candidate and anticancer drug.  相似文献

The title compound, C6H9BrN2O6P2, a micromolar inhibitor of the farnesyl pyrophosphate synthase, is a Z‐isomer zwitterion with one negative phospho­nate group and a protonated pyridine N atom. Two types of ribbons, both parallel to the a axis, formed by several centrosymmetrically related O—H⋯O and N—H⋯O hydrogen bonds are generated in the crystal structure. Thiopurine prodrugs are currently among the leading treatment options for leukemia, immunosuppression, and arthritis. Patients undergoing long‐term thiopurine treatment are at a higher risk of developing sunlight‐induced skin cancers than the general population. This side effect originates from the cellular metabolization of thiopurine prodrugs to form 6‐thio‐2′‐deoxyguanosine, which can absorb UVA radiation, populating its reactive triplet state and leading to oxidatively generated damage. However, the photo‐oxidation mechanism is not fully understood. In this contribution, the oxidation potential and the adiabatic triplet energy of 6‐thio‐2′‐deoxyguanosine are estimated computationally, whereas the intrinsic rate of triple‐state decay and the rate constant for triplet quenching by molecular oxygen are determined using time‐resolved spectroscopic techniques. A singlet oxygen quantum yield of 0.24 ± 0.02 is measured in aqueous solution (0.29 ± 0.02 in acetonitrile). Its magnitude correlates with the relatively low percentage of triplet‐O2 collision events that generate singlet oxygen (SΔ = 37%). This behavior is rationalized as being due to the exergonic driving force for electron transfer between the triplet state of 6‐thio‐2′‐deoxyguanosine and molecular oxygen (ΔGET = ?69.7 kJ mol?1), resulting in the formation of a charge‐transfer complex that favors nonradiative decay to the ground state over triplet energy transfer.  相似文献

The poly(m‐toluidine) film was prepared by using the repeated potential cycling technique in an acidic solution at the surface of carbon paste electrode. Then transition metal ions of Ni(II) were incorporated to the polymer by immersion of the modified electrode in a 0.2 M NiSO4, also the electrochemical characterization of this modified electrode exhibits stable redox behavior of the Ni(III)/Ni(II) couple. The electrocatalytic ability of Ni(II)/poly(m‐toluidine)/modified carbon paste electrode (Ni/PMT/MCPE) was demonstrated by electrocatalytic oxidation of hydrogen peroxide with cyclic voltammetry and chronoamperometry methods in the alkaline solution. The effects of scan rate and hydrogen peroxide concentration on the anodic peak height of hydrogen peroxide oxidation were also investigated. The catalytic oxidation peak current showed two linear ranges with different slopes dependent on the hydrogen peroxide concentration and the lower detection limit was 6.5 μM (S/N=3). The catalytic reaction rate constant, (kh), was calculated 5.5×102 M?1 s?1 by the data of chronoamperometry. This modified electrode has many advantages such as simple preparation procedure, good reproducibility and high catalytic activity toward the hydrogen peroxide oxidation. This method was also applied as a simple method for routine control and can be employed directly without any pretreatment or separation for analysis cosmetics products.  相似文献

Selective oxyfunctionalizations of inert C−H bonds can be achieved under mild conditions by using peroxygenases. This approach, however, suffers from the poor robustness of these enzymes in the presence of hydrogen peroxide as the stoichiometric oxidant. Herein, we demonstrate that inorganic photocatalysts such as gold–titanium dioxide efficiently provide H2O2 through the methanol‐driven reductive activation of ambient oxygen in amounts that ensure that the enzyme remains highly active and stable. Using this approach, the stereoselective hydroxylation of ethylbenzene to (R )‐1‐phenylethanol was achieved with high enantioselectivity (>98 % ee ) and excellent turnover numbers for the biocatalyst (>71 000).  相似文献

The selective oxidation of alkanes as a green process remains a challenging task because partial oxidation is easier to achieve with sacrificial oxidants, such as hydrogen peroxide, alkyl hydroperoxides or iodosylbenzene, than with molecular oxygen or air. Here, we report on a heterogeneous catalyst for n‐hexadecane oxidation comprised of the wheel shaped Cu20‐polyoxotungstate [Cu20Cl(OH)24(H2O)12(P8W48O184)]25? anchored on 3‐aminopropyltriethoxysilane (apts)‐modified SBA‐15. The catalysts were characterized by powder X‐ray diffraction (XRD), N2‐adsorption measurements and Fourier transform infrared reflectance (FT‐IR) spectroscopy. The heterogeneous Cu20‐polyanion system catalyzed the solvent‐free aerobic oxidation of n‐hexadecane to alcohols and ketones by using air as the oxidant under ambient conditions. The catalyst exhibits an exceptionally high turn over frequency (TOF) of 20 000 h?1 at 150 °C and is resistant to poisoning by CS2. Moreover, it can be easily recovered and reused by filtration without loss of its catalytic activity. Possible homogeneous contributions also have been examined and eliminated. Thus, this system can use air as oxidant, which, in combination with its good overall performance and poison tolerance, raises the prospect of this type of heterogeneous catalyst for practical applications.  相似文献

In this work, myoglobin (Mb) and sulfonated‐β‐cyclodextrin (S‐CD) were assembled into {S‐CD/Mb}n layer‐by‐layer films on solid substrates. In pH 7.0 buffers, the {S‐CD/Mb}n films assembled on electrodes showed a pair of well‐defined and nearly reversible CV peaks at about ?0.35 V vs. SCE. The stable CV response of {S‐CD/Mb}n films could be used to electrocatalyze reduction of oxygen and hydrogen peroxide in solution. For comparison, another modified β‐cyclodextrin, carboxyethyl‐β‐cyclodextrin (C‐CD), was also assembled with Mb into {C‐CD/Mb}n multilayer films. Tetrakis[heptadecafluorononyl] substituted phthalocyanine complexes were prepared by template synthesis from 4‐(heptadecafluorononyloxy)phthalonitrile with Co(CH3COO)·2H2O or PdCl2 in 2‐N, N‐dimethylaminoethanol. The corresponding phthalonitrile was obtained from heptadecafluorononan‐1‐ol and 4‐nitrophthalonitrile with K2CO3 in DMF at 50 °C. The structures of the compounds were characterized by elemental analysis, FTIR, UV–vis and MALDI‐TOF MS spectroscopic methods. Metallophthalocyanines are soluble in fluoroalkanes such as perfluoromethylcyclohexane (PFMCH). The complexes were tested as catalysts for benzyl alcohol oxidation with tert‐butylhydroperoxide (TBHP) in an organic–fluorous biphasic system (n‐hexane–PFMCH). The oxidation of benzyl alcohol was also tested with different oxidants, such as hydrogen peroxide, m‐chloroperoxybenzoic acid, molecular oxygen and oxone in n‐hexane–PFMCH. TBHP was found to be the best oxidant for benzyl alcohol oxidation since higher conversion and selectivity were observed when this oxidant was used. Copyright © 2008 John Wiley & Sons, Ltd.  相似文献
