Free fatty acids (FFAs), major cellular metabolites, play an important role during tumor pathogenesis. Enhanced de novo fatty acid synthesis in tissues is a characteristic feature of cancer. Therefore, measurement of FFA concentration in biological samples is beneficial for cancer research and clinical diagnosis. Herein, a rapid, stable, and sensitive detection methodology was established to simultaneously quantify 22 FFAs using high-performance liquid chromatography/electrospray ionization tandem mass spectrometry (HPLC/ESI–MS/MS). The HPLC–MS/MS system was run in negative ion mode for 15 min using multiple reaction monitoring. The lipids were extracted from colon tissues of colon cancer patients and then injected into the HPLC–MS/MS system for analysis. Colon samples were analyzed by inter-day repeatability and intra-day repeatability, with less than 5 % deviation for most fatty acids. This approach is successful to determine low picogram concentrations of each FFA molecule using milligrams of tissue, and provides a promising method for FFA microanalysis in clinical samples. 相似文献

A rapid and sensitive liquid chromatography–electrospray ionization mass spectrometry method was developed for the determination of aesculin in rat plasma. The analyses were chromatographed on a Zorbax Extend-C18 analytical column (150 × 2.1 mm I.D., 5 µm) with 30:70 ( v/v) methanol–0.1% formic acid as mobile phase. Detection was performed by triple-quadrupole tandem mass spectrometry in multi-reaction-monitoring mode with an electrospray ionization source. The method was validated for accuracy and precision, and linearity in the two matrices was good. The assay was linear in the range 12.5–1,800 ng mL ?1. The lower limit of quantification of aesculin (LLOQ) was 12.5 ng mL ?1. The recovery of aesculin and tinidazole (IS) were well above 85%. The within- and between-batch accuracy was 100–104% and 97–109%, respectively. There were no stability-related problems in the procedure for the analysis of aesculin. The method was successfully used in a preclinical study of the pharmacokinetics of aesculin in rats. 相似文献

A rapid and sensitive liquid chromatographic–tandem mass spectrometric method has been developed and validated for the estimation of sarpogrelate in human plasma. Sarpogrelate was extracted from human plasma by solid-phase extraction. Temocapril was used as the internal standard. Heated electron spray ionization mass spectrometry was performed on a TSQ Quantum Ultra MS system. The LC column was a Hypurity C 18 and the mobile phase was 2 mM ammonium formate (pH 3.00 ± 0.05):acetonitrile (30:70 v/ v). A flow rate of 0.250 mL min ?1 was used. The quantitative analyses were carried out in the positive ion and full scan mode over the mass range m/ z 60–500. The capillary, vaporiser temperatures were 325 and 200 °C respectively. The sheath gas pressure, spray voltage, collision energy and tube lense were 40, 3,500 V, 19 V, 198 V, respectively, and the mass spectra of the drugs were recorded by total ion monitoring. Retention times and characteristic mass fragments were recorded and the chosen diagnostic mass fragments were monitored in the mass chromatography mode. Signal intensities of each of the mass fragments: m/ z 477 [M + H] + for temocapril, m/ z 430 [M + H] + for sarpogrelate, were used for quantification. The calibration curves (the ratio between the peak areas as signal intensities of the drug analyzed and that of the internal standard (temocapril: m/ z 477 [M + H] +) vs. the concentration of drug) exhibited linearity over the concentration range 5.00–2,500.00 ng mL ?1 human plasma. The recovery and the accuracy were calculated by comparing the peak areas as the signal intensities of each mass fragment for the drug in spiked samples after solid-phase extraction from human plasma to the peak area as the signal intensity of the mass fragment of internal standard sample. The method involves a rapid solid phase extraction from plasma, simple isocratic chromatography conditions and mass spectrometric detection that enables detection up to picogram levels with a total run time of 3.0 min only. The method was validated over the range of 5.0–2,500.0 ng mL ?1. The absolute recoveries for sarpogrelate (93.72%) and IS (91.42%) achieved from spiked plasma samples were consistent and reproducible. 相似文献

A simple, rapid, specific and sensitive liquid chromatography–tandem mass spectrometric method has been developed and validated for the simultaneous estimation of alfuzosin and dutasteride in human plasma. Both alfuzosin and dutasteride were extracted from human plasma by solid-phase extraction using terazosin and finasteride as the internal standards for alfuzosin and dutasteride, respectively. Chromatographic separation of analytes and their respective internal standards was carried out using a Hypurity C18 (50 × 4.6 mm i.d., 5 μm particle size) column followed by detection using an applied biosystems API 5000 mass spectrometer with a UPLC as the front end. The method involves a rapid solid phase extraction from plasma, simple isocratic chromatographic conditions and mass spectrometric detection in the positive ionization mode using multiple reactions monitoring that enables detection down to low nanogram levels with a total run time of 2.5 min only. The method was validated over a range of 0.25–20.0 ng mL ?1 for alfuzosin and 0.1–10.0 ng mL ?1 for dutasteride. The absolute recoveries for alfuzosin (65.57%), dutasteride (103.82%), terazosin (69.38%) and finasteride (102.25%) achieved from spiked plasma samples were consistent and reproducible. Acceptable precision and accuracy were obtained for concentrations over the standard curve ranges. Due to the short run time of 2.5 min it was possible to analyze a throughput of more than 180 human plasma samples per day. The validated method can be successfully used to analyze human plasma samples for application in pharmacokinetic, bioavailabilty or bioequivalence studies. As an example the application of this validated method to a bioequivalence study is also illustrated. 相似文献

The purpose of this article was to develop a rapid and robust LC–MS–MS method for quantifying shikonin and deoxyshikonin simultaneously in rat plasma using emodin as internal standard. The LC system consisted of an Agilent ZORBAX SB-C18 (1.8 μm, 250 × 4.6 mm, 20 °C) column. Elution with an isocratic mobile phase consisted of methanol/10 mM ammonium acetate in water/acetonitrile containing 0.05% formic acid (45:10:45, v/v/v) at a flow rate of 0.8 mL min ?1 yielded sharp, high-resolved peaks within 12 min. The lower limits of quantitation were 0.5 ng mL ?1 for shikonin, and 8 ng mL ?1 for deoxyshikonin. Correlation coefficient ( r) values for the linear range of two analytes were greater than 0.99. Assay precision was <13% and accuracy was 87–99%. This newly developed method was used to the pharmacokinetic studies of the shikonin analogues in rats after intravenous administration ( n = 4). 相似文献

Liquid chromatography–ion trap mass spectrometry was employed to investigate the metabolism of linarin in rats. Identification and structural elucidation of the metabolites were performed by comparing the differences in molecular masses, retention times, and full scan MS n spectra between linarin and its metabolites. Six metabolites (acacetin, apigenin, acacetin glucuronide, apigenin glucuronide, acacetin sulfate, apigenin sulfate) were detected in rat urine after oral administration of linarin at the dose of 50 mg kg ?1. Furthermore, a selective and sensitive liquid chromatography–triple quadruple mass spectrometry assay was developed and validated for the simultaneous determination of linarin and acacetin (the major metabolite of linarin) in rat urine. Chromatographic separation was carried out on a C 18 column, and mass spectrometric detection was performed using a triple-quadrupole mass spectrometer coupled with an electrospray ionization source in the positive ion mode. Quantitation of linarin and acacetin was performed using selected reaction monitoring of precursor–product ion transitions at m/z 593 → 285 for linarin, 285 → 242 for acacetin, and 303 → 153 for hesperitin (internal standard), respectively. The assay exhibited good linearity ( r > 0.9900) for both linarin and acacetin. The intra- and inter-day precisions were <13.4 % and the accuracy was between ?8.1 and 3.1 %. A new method based on high-performance liquid chromatography coupled with electrospray ionization tandem mass spectrometry has been developed for simultaneous analysis of 27 components (eighteen diterpenoids, six phenolic acids, and three flavonoids) of Isodon serra, a famous traditional Chinese medicine. Separation on a C 18 column was achieved by gradient elution with water and methanol both containing 0.1% formic acid. Identification and quantification of the analytes were achieved by use of a hybrid quadrupole linear ion-trap mass spectrometer. Multiple-reaction monitoring (MRM) was used for quantification, with switching of electrospray ion source polarity between positive and negative modes in the same chromatographic run. An information-dependent acquisition (IDA) method was used to trigger product ion scans above the MRM signal threshold so that the 27 compounds could be identified by use of enhanced product-ion scans (EPI). The method was fully validated (for linearity, precision, accuracy, and limits of detection and quantification). The results indicated that this simple method was rapid, specific, and reliable. The method was successfully applied to analysis of 45 batches of Isodon serra samples from different sources, and quantification of the compounds in Isodon serra was achieved. 相似文献

A rapid, reliable and sensitive method has been developed to determine malic and citric acid in fruits and vegetables. The methodology is based on simple extraction with an aqueous solution of ethanol (80% v/v) and subsequent chromatographic analysis by liquid chromatography coupled to mass spectrometry. Electrospray ionization in negative mode was used. The best response for citric and malic acid was provided by molecular ions [M?H] ? at m/z 191 and 133 respectively. These ions were used for quantification, whereas other fragments were used as confirmation ions. Different variables involved in the separation and detection process, such as mobile phase, gradient profile and flow rate have been optimised. Linearity, repeatability, recovery and limits of quantification were evaluated. Good linearity was obtained up to 5,000 mg kg ?1. Recovery ranged from 90.0 to 104.6%, repeatability (expressed as RSD) was <8% for tested matrices, and limits of quantification were equal or lower than 65 mg kg ?1. In this work, an analytical multiresidue method using liquid chromatography tandem-mass spectrometry (LC–MS–MS) with triple quadrupole in selected reaction monitoring (SRM) mode for the simultaneous determination of 54 pesticides in vegetables (pepper and tomato) and citrus fruits (orange and lemon) has been developed. The procedure involves initial single phase extraction of sample with acetonitrile by agitation, followed by liquid–liquid partition aided by “salting out” process using NaCl. The average recovery by the LC–MS–MS method obtained for these compounds varied from 65.5 to 114.5% with a relative standard deviation between 2.3 and 8.3%. The method presents good linearity over the range assayed 10–500 μg L ?1 (except famoxadone 50–1,000 μg L ?1) and the detection limits for the pesticides studied varied from 0.03 to 14.9 μg kg ?1. The proposed method was used to determine pesticide levels in vegetables and citrus fruit samples from different experimental orchards and greenhouses from the Region of Murcia. 相似文献

The lower limit of quantification (LLOQ) was identifiable and reproducible at 0.01 ng mL ?1. A new, rapid, and efficient method, multiple reaction monitoring liquid chromatography–tandem mass spectrometry, has been developed for analysis of nanafrocin in foodstuffs of animal origin. The researchers used a C 18 stationary phase coupled with triple-quadrupole tandem mass spectrometry in negative-electrospray mode. The limits of detection (LOD) and quantification (LOQ) were 0.005 and 0.01 mg kg ?1, respectively, in the matrixes. Detector response was found to be a linear function of concentration over the range 0.005–0.1 mg kg ?1 in each matrix. Mean overall recovery ( n = 10) of nanafrocin varied from 71 to 101%. The results show that identification and quantification of nanafrocin residues in foodstuffs of animal origin can be successfully achieved by use of the proposed LC–MS–MS method. 相似文献

Summary A sensitive and selective liquid chromatographic method coupled with electrospray ionization tandem mass spectrometry (LC–ESI–MS–MS) has been developed for quantification of felodipine in human and dog plasma. Compounds were separated on a 2.0 mm × 150 mm, 5.0 m particle, C 8 column with 1 m m ammonium acetate–acetonitrile, 20:80, pH 6.0, as mobile phase at a flow rate of 200 L min –1. Nifedipine was used as internal standard. Plasma samples were extracted with diethyl ether, the centrifuged upper layer was evaporated, the residue was reconstituted with mobile phase, and the reconstituted samples were injected. The analytical column lasted for at least 1000 injections. By use of multiple reaction monitoring (MRM) mode in MS–MS felodipine and nifedipine were detected without severe interference from the human or dog plasma matrix. Felodipine produced a protonated precursor ion ([M + H] +) at m/z 384 and a corresponding product ion at m/z 338. And internal standard (nifedipine) produced a protonated precursor ion ([M + H] +) at m/z 347 and a corresponding product ion at m/z 315. Detection of felodipine in human and dog plasma was accurate and precise, with a limit of quantification of 0.05 ng mL –1. The method has been successfully applied to preliminary pharmacokinetic study of felodipine in human and dog plasma. 相似文献

A sensitive, high-throughput and economic liquid chromatographic method for determination of fenofibric acid in human plasma was developed and validated by ultraviolet detection and tandem mass spectrometry, then applied in pharmacokinetic study to investigate Lipanthyl™ 200 mg MC bioavailability under food and fasting conditions. Fenofibric acid with 2-chloro fenofibric acid-d6 (internal standard) was extracted from 100 µL of human plasma by acetonitrile in a single extraction step. 25 and 2 µL from supernatant were injected onto ACE column, 50 mm, 5 micron with 4.6 mm inner diameter for LC–UV and 2.1 mm for LC–MS/MS, and both systems were eluted isocratically by water:methanol:formic acid (35:65:0.1, v/v/v), with a constant flow rate of 1 mL min−1. The established calibration curve was linear between 0.05–20 µg mL−1, and the within- and between-day precisions were all below 13 % in both LC–MS/MS and LC–UV systems during validation, and accuracies ranged between 91 and 112 %. Twenty-eight healthy adult subjects participated in this clinical study, and the pharmacokinetic parameters including coefficient of variation were calculated and discussed. A dramatic decrease in C

A sensitive LC–MS–MS method with electrospray ionization has been developed for analysis of mirtazapine in rat plasma. After addition of diazepam as internal standard, liquid–liquid extraction was used to produce a protein-free extract. Chromatographic separation was achieved on a 150 × 4.6 mm, 5 μm particle, ODS column with 84:16 ( v/ v) methanol–water containing 0.1% ammonium acetate and 0.01% glacial acetic acid as mobile phase. LC–MS–MS was performed in selected-ion-monitoring (SIM) mode using target fragment ions m/ z 195.09 for mirtazapine and m/ z 192.80 for the IS. Calibration plots were linear over the range of 0.516–618.8 ng mL ?1. The lower limit of quantification was 0.516 ng mL ?1. Intra-day and inter-day precision were better than 12.6 and 8.8%, respectively. Mean recovery of mirtazapine from plasma was in the range 87.41–90.06%; average recovery was 88.40% (RSD 3.95%). A simple and specific analytical method for the quantitative determination of shikimic acid from the methanol extract of the fruits of Illicium species and from various plant samples was developed. The LC–UV separation was achieved by reversed-phase chromatography on a C18 column with potassium dihydrogen phosphate and methanol as the mobile phase. In the LC–MS method, the separation was achieved by a C12 column using water and acetonitrile, both containing 0.1% acetic acid as the mobile phase. The methods were successfully used to study the percentage compositions of shikimic acid present in nine species of Illicium and various other plant samples. The detector response was linear with concentrations of shikimic acid in the range from 1.0–500.0 μg mL ?1 by LC–UV and 100–1000 ng mL ?1 by LC–MS. Mass spectrometry coupled with electrospray ionization interface is described for the identification of shikimic acid in various plant samples. This method involved the use of the [M-H] ? ions of shikimic acid at m/z 173.0455 (calculated mass) in the negative ion mode with extractive ion monitoring. 相似文献
