Electrochemical CV and SWV studies were performed with double stranded DNA from salmon testes (dsDNA) and single stranded DNAs, containing 25 nucleotides (ssDNA) directly adsorbed at polycrystalline Au electrodes. A distinct oxidation peak at +730 mV (SWV, scan rate 0.248 V s?1) or at +730 – +780 mV (CV, scan rate from 0.3 to 1 V s?1) was obtained with DNA‐modified Au electrodes after a time‐dependent prepolarization step at a positive potential value, i.e., at +500 mV (vs. Ag|AgCl), performed with the DNA‐modified Au electrodes dipped in a blank buffer solution. No electrochemical activity was detected when ssDNA, containing no guanines, was used for adsorptive modification of the Au electrodes. Electrochemical impedance measurements registered a possible reorganization of the adsorbed DNA layer in the course of the prepolarization, accompanied by decreasing in‐phase impedance. The results enable us to relate the oxidation process observed at the DNA‐modified Au electrodes with the oxidation of guanine residues in DNA.  相似文献

An electrochemical hybridization biosensor based on peptide nucleic acid (PNA) probes with a label‐free protocol is described. The detection of PNA‐DNA and DNA‐DNA hybridizations were accomplished based on the oxidation signal of guanine by using differential pulse voltammetry (DPV) at carbon paste electrode (CPE). It was observed that the oxidation signals of guanine obtained from the PNA and DNA probe modified CPEs were higher than those obtained from the PNA‐DNA and DNA‐DNA hybrid modified CPEs due to the accessible unbound guanine bases. The detection of hybridization between PNA probe and point mutation containing DNA target sequences was clearly observed due to the difference of the oxidation signals of guanine bases, because the point mutation was guanine nearly at the middle of the sequence. The effect of the DNA target concentration on the hybridization signal was also observed. The PNA probe was also challenged with excessive and equal amount of noncomplementary DNA and also mixtures of point mutation and target DNA.  相似文献

In this paper, nano‐gold modified carbon paste electrode (NGMCPE) was employed to develop an electrochemical DNA hybridization biosensor. The proposed sensor was made up by immobilization of 15‐mer single stranded oligonucleotide probe for detection of target DNA. Hybridization detection relies on the alternation in guanine oxidation signal following hybridization of the probe with complementary genomic DNA. The guanine oxidation was monitored using differential pulse voltammetry (DPV). Different factors such as activation potential, activation time and probe immobilization conditions were optimized. The selectivity of the sensor was investigated by non‐complementary oligonucleotides. Diagnostic performance of the biosensor was described and the detection limit was found 1.9 × 10?13 M at the NGMCPE surface. All of the investigations were performed in both CPE and NGMCPE and finally their results were compared.  相似文献

A chitosan modified carbon paste electrode (ChiCPE) based DNA biosensor for the recognition of calf thymus double stranded DNA (dsDNA), single stranded DNA (ssDNA) and hybridization detection between complementary DNA oligonucleotides is presented. DNA and oligonucleotides were electrostatically attached by using chitosan onto CPE. The amino groups of chitosan formed a strong complex with the phosphate backbone of DNA. The immobilized probe could selectively hybridize with the target DNA to form hybrid on the CPE surface. The detection of hybridization was observed by using the label‐free and label based protocols. The oxidation signals of guanine and adenine greatly decreased when a hybrid was formed on the ChiCPE surface. The changes in the peak currents of methylene blue (MB), an electroactive label, were observed upon hybridization of probe with target. The signals of MB were investigated at dsDNA modified ChiCPE and ssDNA modified ChiCPE and the increased peak currents were observed, in respect to the order of electrodes. The hybridization of peptide nucleic acid (PNA) probes with the DNA target sequences at ChiCPE was also investigated. Performance characteristics of the sensor were described, along with future prospects.  相似文献

We report the synthesis and characterization of N,N-bis[(7-dimethylamino)phenothiazin-5-ium-3-yl]-4,4-ethylenedipiperidine diiodide (3), consisting of two photosensitizing phenothiazinium rings attached to a central ethylenedipiperidine linker. At all time points (10, 30, 60 min) and all wavelengths (676, 700, 710 nm) tested, photocleavage of pUC19 plasmid DNA (22 degrees C and pH 7.0) was markedly enhanced by 1 microM of 3 in comparison to 1 microM of the parent phenothiazine methylene blue (MB). At concentrations of phenothiazine ranging from 5 to 0.5 microM, the photocleavage levels produced by compound 3 were consistently higher than the cleavage produced using approximately twice the amount of MB (e.g., 710 nm irradiation of 5 microM of 3 and 10 microM of MB cleaved the plasmid DNA in 93% and 71% yields, respectively). Scavenger assays provided evidence for the involvement of singlet oxygen and, to a lesser extent, hydroxyl radicals in DNA damage. Analysis of photocleavage products at nucleotide resolution revealed that direct strand breaks and alkaline-labile lesions occurred predominantly at guanine bases. While compound 3 and MB were both shown to stabilize duplex DNA, the DeltaTm values of calf thymus (CT) and C. perfringens DNAs were approximately three fold higher in the presence of compound 3. Finally, viscometric data indicated that CT DNA interacts with compound 3 and MB by a combination of groove binding and monofunctional intercalation, and with compound 3 by a third, bisintercalative binding mode.  相似文献

A DNA probe that was based on methylene blue (MB) imprinted polyvinyl pyridine polymer (MIP) modified carbon paste electrodes were developed for the first time for electrochemical monitoring of DNA. Probes were built up by adsorbing MB onto modified electrodes prior to DNA immobilization. It was shown that DNA strongly immobilizes on MIP modified electrodes when MB was adsorbed in advance of DNA immobilization. The performance of the MB imprinted polymer modified carbon paste electrodes (MIP‐CPE) to rebind the template molecule (MB) were compared to those of control polymer modified (non‐imprinted polymer NIP‐CPE) and bare (CPE) electrodes. Electrochemical signal resulting from the oxidation of guanine moiety of the immobilized probe DNA was high enough on the constructed platform, implicating that probes of this kind could be favorably used for DNA analysis. These probes exhibited high selectivity for its complementary DNA sequences (target). HBV‐DNA hybridization was studied to evaluate the selectivity of the probes for complementary, non‐complementary and mismatch sequences. The detection limit of the probe for the target DNA was 8.72 µg/mL (1.38 µM), which was better than those attained by some earlier DNA sensor studies.  相似文献

Multi‐walled carbon nanotubes (MWNTs) were used as nanowires, which combined DNA molecules to a carbon paste electrode (CPE). The attachment of MWNT on the electrode surface was controlled by a hybridization assay between adenine and thymine containing oligonucleotides. The appearance of guanine oxidation signal after hybridization with target DNA greatly simplified the specific sequence DNA detection mechanism. Combination of sidewall‐ and end‐functionalization of MWNT provided a significant enhancement in the voltammetric signal of guanine oxidation in comparison with the signals obtained from only end‐oxidized MWNT modified CPE and a bare CPE. A control experiment involving adenine containing polynucleotide (poly(A)) instead of adenine probe modified MWNT was performed. The effect of target and noncomplementary DNA concentration on the guanine signal was also monitored. Discrimination against single‐base mismatch and noncomplementary DNA was achieved by surfactant containing washing solution. The promising conductivity of carbon nanotubes, and the creation of a larger surface area for DNA immobilization by sidewall‐ and end‐oxidation of MWNT provided a detection limit down to 10 pg/mL, which is compatible with the demand of the genetic tests.  相似文献

An electrochemical hybridization biosensor based on the intrinsic oxidation signals of nucleic acids and proteins has been designed, that makes use of the unique binding event between Escherichia coli single-strand binding protein (SSB) and single-stranded DNA (ssDNA). The voltammetric signal from guanine oxidation significantly decreased upon binding of SSB to single-stranded oligonucleotides (probe), anchored on a single-walled carbon nanotube (SWCNT) -modified screen-printed carbon electrode (SPE). Simultaneously, oxidation of the tyrosine (Tyr) and tryptophan (Trp) residues of the SSB protein increased upon binding of the SSB protein to ssDNA and ss-oligonucleotides. After the hybridization, SSB did not bind to the double helix form, and the guanine signal could be observed along with the disappearance of the oxidation signal of the protein. The amplification of intrinsic guanine and protein oxidation signals by SWCNT, and a washing step with sodium dodecylsulfate, enabled the specific detection of a point mutation. Monitoring the changes in the guanine and protein signals upon hybridization greatly simplified the detection procedure. The detection limit of 0.15 g/ml target DNA can be applied to genetic assays. To the best of our knowledge, this is the first work that utilizes the monitoring of SSB–DNA interactions on a solid transducer for the electrochemical detection of DNA hybridization by using intrinsic oxidation signals.  相似文献

Preparation of a polypyrrole nanofiber (PPyNF) modified pencil graphite (PG) electrode and its usage in the electrochemical DNA biosensors was investigated. The electrodes (PPyNF/PG1 and PPyNF/PG2) were prepared from a solution containing 0.1 M pyrrole, 0.1 M Na2CO3 and 0.1 M LiCIO4 by using potentiostatic and potentiodynamic methods. PPyNF/PG2 electrodes which were prepared by potentiostatic procedure showed higher responses for the oxidation of ds‐DNA than the PPyNF/PG1 electrodes prepared by potentiodynamic methods. Immobilization of the ds‐DNA on PG and PPyNF/PG surfaces was performed at a constant potential, +0.5 V, for 300 s in 0.5 M ABS (pH 4.8) containing 15 μg mL?1 ds‐DNA and 20 mM LiCIO4. The oxidation peak potentials of the ds‐DNA bases, guanine and adenine, were shifted to more cathodic values by using PPyNF/PG electrodes. The oxidation signal of the guanine base of ds‐DNA was decreased in the presence of methylene blue.  相似文献

An electrochemical DNA sensor was constructed using single‐walled carbon nanotubes (SWNTs) attached to a self‐assembled monolayer of 11‐amino‐1‐undecanethiol on a gold surface. The voltammetric peak of methylene blue (MB), which interacts with the DNA guanine bases specifically, was used to follow the DNA hybridization process. After DNA hybridization with its complementary DNA strand, the MB electrochemical signal response decreased and the change in MB signal response was used as the basis for the electrochemical sensing of DNA hybridization. The as described DNA sensor demonstrated to have good stability, selectivity, a linear response over the DNA concentration range from 100 to 1,000 nM and a limit of detection of 7.24 nM.  相似文献

Damage of salmon sperm double strand ss dsDNA in solution or immobilized on screen‐printed carbon electrode (SPCE) induced by incubation of DNA with the antineoplastic alkylating agent busulfan (BUS) at various conditions was detected for the first time by simple electrochemical methods. Chemical changes in DNA bases can be detected through the altered electroactivity of the DNA. Electrochemical voltammetric sensing of damage caused by BUS to dsDNA in solution was monitored by the appearance of peaks diagnostic of the oxidation of guanine and adenine. Moreover, crystal violet, which interacts with the DNA immobilized on SPCEs, was used as an effective electroactive indicator, in combination with cyclic voltammetry and differential pulse voltammetry techniques to monitor the cross‐links or damage to DNA. The interaction between BUS and DNA were determined by the changes in the voltammetric peak of crystal violet. The effects of various conditions upon the crystal violet signal were investigated.  相似文献

The interaction of cyclophosphamide (CP) with calf thymus double-stranded DNA (dsDNA) and thermally denatured single-stranded DNA (ssDNA) immobilized at the carbon paste (CPE) and pencil graphite electrodes (PGE), was studied electrochemically based on oxidation signals of guanine and adenine using differential pulse voltammetry (DPV).As a result of the interaction of CP with DNA, the voltammetric signals of guanine and adenine increased in the case of dsDNA while a slight increase was observed in ssDNA. The effect of experimental parameters such as the interaction time between CP and DNA forms and the concentration of CP, were studied using DPV with CPE and PGE. Additionally, reproducibility and detection limits were determined using both electrodes. A comparison of the analytical performance between CPE and PGE was done. Our results showed that these two different DNA biosensors could be used for the sensitive, rapid and cost effective detection of CP itself as well as of CP-DNA interaction.Furthermore, the interaction of CP with dsDNA and ssDNA was studied in solution and at the electrode surface by means of alternating current voltammetry (ACV) in 0.3 M NaCl and 50 mM sodium phosphate buffer (pH 8.5) supporting electrolyte, using a hanging mercury drop electrode (HMDE) as working electrode. The conclusions of this study were mainly based on tensammetric peaks I (at −1.183 V) and II (−1.419 V) of DNA. This study involved the interaction of CP with surface-confined and solution phase DNA where experimental parameters, such as the concentration of CP and the interaction time, were studied. By increasing the concentration of CP, an increase of peak II was observed in both ds and ssDNA, while an increase of peak I was observed only in the case of dsDNA. An overall conclusion of the study using HMDE was that the interaction of CP with surface-confined DNA significantly differed from that with solution phase DNA. The increase of peaks I and II was lower in the case of interaction of CP with surface-confined DNA, probably due to steric positioning of DNA at the electrode surface.  相似文献

Electrochemically modified glassy carbon electrode (GCE) was used to study the electrochemical oxidation and detection of denatured single‐stranded (ss) DNA by means of adsorptive stripping voltammetry. The modification of GCE, by electrochemical oxidation at +1.75 V (vs.SCE) for 10 min and cyclic sweep between +0.3 V and ?1.3 V for 20 cycles in pH 5.0 phosphate buffer, results in 100‐fold improvement in sensitivity for ssDNA detection. We speculated that the modified GCE has a high affinity to single‐stranded DNA through hydrogen bond (specific static adsorption). Single‐stranded DNA can accumulate at the GCE surface at open circuit and produce a well‐defined oxidation peak corresponding to the guanine residues at about +0.80 V in pH 5.0 phosphate buffer, while the native DNA gives no signal under the same condition. The peak currents are proportional to the ssDNA concentration in the range of 0–18.0 μg mL?1. The detection limit of denatured ssDNA is ca. 0.2 μg mL?1 when the accumulation time is 8 min at open circuit. The accumulation mechanism of ssDNA on the modified GCE was discussed.  相似文献

The electrocatalytic oxidations of guanine, adenine, guanosine-5'-monophosphate(GMP) and ssDNA were performed in the presence of Fe(II) bis(2,2':6',2'-terpyridine) and Fe(II) tris(1,10-phenanthroline) complexes as homogeneous catalysts by cyclic voltammetric methods. The Fe(II/III) redox couple of these compounds is responsible for their catalytic properties. The electrocatalytic oxidation current of above substrates were developed from the anodic peak currents of Fe(II) bis(2,2':6',2'-terpyridine) and Fe(II) tris(1,10-phenanthroline) complexes at about +0.93 V and 0.97 V, respectively. The electrocatalytic oxidative properties of guanine by Fe(II) bis(2,2':6',2'-terpyridine) complex was measured by amperometry method using the rotating disk electrodes. Electropolymerization of Fe(II) tris(5-amino-1,10-phenanthroline) complex produced thin polymer films on gold and glassy carbon electrodes. The electrochemical quartz crystal microbalance (EQCM) and cyclic voltammetry were used to study the in situ growth of the polymer. The poly(FeII(5-NH(2)-1,10-phen)(3)) exhibited a good electrocatalytic oxidation towards guanine and also for the mixture of guanine and adenine too.  相似文献

We report the advantages of the adsorption and electrooxidation of oligonucleotides and calf‐thymus double stranded DNA (dsDNA) at glassy carbon electrodes (GCE) modified with a dispersion of multiwalled carbon nanotubes (MWCNT) in poly‐L ‐lysine (Plys) (GCE/MWCNT‐Plys). Important enhancement in the guanine oxidation signal was obtained by adsorptive stripping voltammetry (AdSV) due to a most favorable interaction between the negatively charged DNAs and the positively charged Plys that support the MWCNT. The layer of oligo(dG)11 immobilized at GCE/MWCNT‐Plys was successfully used for the selective detection of the hybridization event using oligo(dG)11/oligo(dC)11 as model.  相似文献

In this article, for the first time, the electrochemical properties of a novel pyridine derivative, 4‐(2‐(2‐hydroxybenzylidene) hydrazinyl)‐1‐(3‐phenylpropyl) pyridinium bromide (abbreviated as 4‐Pyri), and its interaction with double stranded DNA (dsDNA) was investigated. The interaction between candidate drug molecule (4‐Pyri) and dsDNA was analyzed by examining 4‐Pyri (+0.6 V and +0.8 V) and guanine (+1.0 V) oxidation signal changes with Differential Pulse Voltammetry (DPV) and Cyclic Voltammetry (CV). Electrochemical Impedance Spectroscopy (EIS) was used to show the resistance changes before and after the interaction between 4‐Pyri and dsDNA. We showed that after the interaction with 4‐Pyri, the oxidation currents of guanine decreased dramatically, whereas the intrinsic oxidation currents of 4‐Pyri dramatically increased. 4‐Pyri oxidation current differences before and after the interaction with dsDNA enabled us to determine such interaction separately from guanine oxidation signals. In addition, resistance differences were observed at before and after the interaction with each other that confirmed the possible interaction. In addition, toxicity effect (S%) value, which is an important parameter for electrochemical studies indicated 4‐Pyri's toxicity to dsDNA. Our results demonstrated that 4‐Pyri interacts with dsDNA, and could be used as a potential candidate drug molecule due to its remarkable impact on dsDNA.  相似文献

Four DNA electrochemical biosensors using four types of DNA (calf thymus ssDNA, calf thymus dsDNA, salmon testis ssDNA and salmon testis dsDNA) were constructed using graphite screen printed electrodes. These biosensors were exploited as analytical tool to detect polycyclic aromatic hydrocarbons-DNA interactions using benzo(a)anthracene and phenantrene as model analytes, the guanine oxidation peak variation being the signal revealing the interaction between PAHs and immobilized DNA. The salmon testis ssDNA biosensor resulted as the most promising device and was further evaluated for benzo(a)anthracene, fluorene, indeno(1,2,3-cd)pyrene, anthracene, and phenanthrene in 5–40 ng mL?1 solutions, and for benzo(a)pyrene (5–50 ng mL?1). A concentration dependent variation of the DNA guanine oxidation peak was observed for all compounds. The effect of benzo(a)pyrene ultraviolet (UV) activation on the benzo(a)pyrene (BaP)-DNA interaction was evaluated at concentration levels of 20 and 50 ng mL?1, and a 3.5- and 2.7-fold increases of the guanine oxidation peak was measured respectively. The salmon testis ssDNA biosensor was examined with PAHs contaminated samples of Mytilus galloprovincialis. Upon UV irradiation of three sample extracts exceeding the BaP maximum level, a positive variation of the DNA guanine oxidation was obtained. An average 2.4-fold increase of the guanine oxidation peak was detected demonstrating that the sensor can be used to detect toxic degradation products of PAHs.  相似文献

Cuprous oxide nanowhisker was prepared by using cetyltrimethyl ammonium bromide (CATB) as soft template, and was characterized by XRD and TEM methods. The electrochemical properties of nano‐Cu2O and nano‐Cu2O‐methylene blue (MB) modified electrode were studied. The experimental results indicate that nano‐Cu2O shows a couple of redox peaks corresponding to the redox of Cu(II)/Cu(I), the peak currents are linear to the scan rates which demonstrate that the electrochemical response of Cu2O is surface‐controlled. The composite nano‐Cu2O‐Nafion‐MB modified electrode shows a trend of decrease of peak currents corresponding to the Cu (II)/Cu (I). However, the electrocatalytic ability of nano‐Cu2O‐MB composite film to dopamine increases dramatically. At this composite electrode, dopamine shows a couple of quasireversible redox peaks with a peak separation of 106 mV, the peak current increases about 8 times and the oxidation peak potential decreases about 200 mV as compared to that at bare glassy carbon electrode. The peak currents change linearly with concentration of dopamine from 1×10?7 to 3.2×10?4 mol/L, the detection limit is 4.6×10?8 mol/L. The composite electrode can effectively eliminate the interference of ascorbic acid and has better stability and excellent reproducibility.  相似文献

PAMAM dendrimer/reduced graphene oxide nanocomposite modified pencil graphite electrode (PAMAM/RGO/PGE) was used to fabricate an electrochemical DNA biosensor for determination of Rituxan (RTX) at low concentrations, for the first time. The fabricated biosensor was characterized with FE‐SEM, EIS, and CV techniques. The ds‐DNA/PAMAM/RGO/PGE was used as a working electrode to study the interaction between the RTX and salmon sperm ds‐DNA by DPV technique. Because of the interaction between the drug and DNA leads to a decrease in the guanine oxidation peak current, it was used as an indicator for the determination of the RTX. Under the optimized experimental conditions, a wide linear relationship between RTX concentration and guanine signal was obtained within the range of 7.0 to 60.0 μmol L−1 and 60.0 to 300.0 μmol L−1 with a low detection limit (0.56 μmol L−1). To clarify the interaction mechanism between the RTX and the ds‐DNA, DPV and UV‐Vis measurements were used. The reproducibility, stability, and performance of the constructed biosensor was examined by quantitative measuring RTX in pharmaceutical and human serum samples with good precision (RSD; 2.0–6.0 %) and acceptable recoveries (100.04–101.95 %).  相似文献
