A comparative investigation of the noncovalent interaction of the platinum(II) polypyridine complexes [Pt(dipy)(n-Rpy)2]2+ and [Pt(4,4'-Me2dipy)(2-Rpy)2]2+ (dipy = 2,2'-dipyridine; Me = CH3; n = 2-4; R = H or CH3) with double-helical DNA (calf thymus) and RNA [poly(A).poly(U)] has been conducted. With the exception of [Pt(dipy)(2-Mepy)2]2+, all of the complexes interact strongly, by intercalation, with both nucleic acids giving rise to large changes in the electronic spectra and induced circular dichroism signals; in addition, viscosity experiments on rodlike DNA and RNA show that both biopolymers elongate upon interaction with the complexes. The binding constant values, KB, determined at 25 degrees C, indicate that, at 0.101 M ionic strength, the affinity for poly(A).poly(U) is strongly dependent on the complexes nature, while for DNA it is leveled off. [Pt(dipy)(2-Mepy)2]2+ binds to DNA but does not interact appreciably with poly(A).poly(U). 相似文献

The interaction with DNA of the platinum(II) square planar complexes [Pt(N-N)(py)(2)](2+) (N-N = 1,10-phenanthroline (phen), dipyrido[3,2-d:2',3'-f]quinoxaline (dpq), dipyrido[3,2-a:2',3'-c]phenazine (dppz), benzodipyrido[b:3,2-h:2'3'-f]phenazine (bdppz)) has been investigated by means of absorption, circular and linear dichroism spectroscopy, DNA melting, and viscosity. In the presence of excess [DNA] all the complexes intercalate to the double helix. For those with the most extended phenanthrolines the binding mode depends on the [DNA]/[complex] ratio (q); at low q values the substances bind externally to DNA probably self-aggregating along the double helix. When the DNA concentration is large enough, the aggregate breaks up and the complex intercalates within the nucleobases. The complexes self-aggregate, without added DNA, in the presence of a large salt concentration. 相似文献

We describe novel optically active double helices consisting of complementary strands stabilized by amidinium-carboxylate salt bridges. The m-terphenyl groups of each strand are joined by trans-Pt(II) acetylide complexes with pendant PPh(3) ligands as the surrogate linker, which converts to cis counterparts by a ligand exchange reaction with cis-1,2-bis(diphenylphosphino)ethylene, resulting in the formation of double helices with different structures. Subsequent iodine-promoted reductive elimination on the Pt(II) atoms generates the fully organic, enantiomerically pure double helices. [structure: see text] 相似文献

Novel platinum(II) square planar coordination complexes, in which two heteroaromatic ligands are held by the metal in an unusual L-shaped geometry orthogonal to each other, have been synthesized, and their interaction with DNA was investigated with absorption and linear dichroism spectroscopy. As a rule, the ligand that is coplanar with the coordination square of Pt is found to be oriented perpendicular relative to the DNA helix axis when bound, suggestive of its intercalation between the base pairs of DNA. However, when this coplanar ligand is replaced by two pyridines, the opposite ligand, orthogonal to the coordination square, is instead preferentially intercalated. This behavior shows that these new complexes do indeed show some properties of true ambintercalators, i.e., compounds that can bind by intercalation of either of two distinct aromatic moieties. 相似文献

The complex [Pt(C2H4)(PPh3)2] reacts with Pb2Ph6 to give cis-[PtPh(Pb2Ph5)(PPh3)2]; this decomposes in solution to cis-[PtPh(PbPh3)(PPh3)2], which may also be obtained from the ethylene complex and PbPh4. Lead compounds PbPhMe3 and PbPh3Br also give products of insertion into PbPh bonds, but PbMe3Cl gives cis- and trans-[PtCl(PbMe3)(PPh3)2]. The complex trans-[Pt(PbPh3)2(PEt3)2] reacts with 1,2-bis(diphenylphosphino)ethane (DPPE) to give [Pt(PbPh3)2(DPPE)] which readily decomposes in dichloromethane in presence of PEt3 to give [Pt(PbPh3)(PEt3)(DPPE)]Cl and [PtPh(PEt3)(DPPE)]Cl. The complex trans-[PtCl(PbPh3)(PEt3)2] was detected in the products of reactions between trans-[PtCl2(PEt3)2] and trans-[Pt(PbPh3)2(PEt3)2] or less than 2 moles of LiPbPh3; it was not detected in the mixture after treatment of trans -[Pt(PbPh3)2(PEt3)2] with HCl. In contrast to an earlier report, we were unable to detect lead-containing complexes in the products of the reaction between trans-[PtHCl(PPh3)2] and Ph3PbNO3. Summary As an approach to systems containing methionine residues, 3-acetyl-4-hydroxy-6-methyl-2H-pyran-2-one (HDh, dehydroacetic acid) was treated with L-methionine (MetH) or L-methionine methylester (MetM). By condensation at the acyl group and transfer of the phenolic hydrogen on the nitrogen atom, the related ligands DhMetH and DhMetM, were isolated, and form complexes of formula [MX2(L)2](M = Pd or Pt, L = DhMetM, X = Cl, Br or I; L = DhMetH, X = Cl or Br) and [MI2(DhMetH)] with palladium and platinum dihalides. The reaction of the DhMetK carboxylate with MCl2 in various media is discussed. Ligands and complexes were characterized by i.r. and n.m.r. (1H and13C) spectroscopy and, in some cases, by thermogravimetric measurements. The ligands behave as monodentate sulphur donors, the 12 complexes showing atrans geometry except for [PtCl2(DhMetH)2], which is probably a mixture ofcis andtrans isomers. 相似文献

A series of cyclometallated phenylpyridine platinum(II) complexes have been synthesised with a systematic variation in both the phenylpyridine and the ancillary ligand. Oxidation of one of the cyclometallated species leads to a number of isomeric platinum(IV) complexes, all of which eventually isomerize to a single compound. The route to these new compounds has been demonstrated to involve an initial slow oxidation followed by a rapid C-H activation to give doubly cyclometallated complexes. The solid state structures of a number of both the platinum(II) and the platinum(IV) species have been solved; many of the structures exhibited extended interactions that result in complex three dimensional packing. 相似文献

Molecular mechanics and dynamics calculations have been used in conjunction with experimental data to study the effects of amine ligand bulk on the formation of both guanine and methionine complexes with platinum diamine compounds. The AMBER force field has been supplemented with previous modifications [Yao; et al. Inorg. Chem. 1994, 33, 6061-6077. Cerasino; et al. Inorg. Chem. 1997, 36, 6070-6079] and has been further modified to include parameters for platinum bound to the sulfur atom of methionine. Molecular mechanics calculations with this modified AMBER force field have suggested that a platinum complex with two sulfur-bound methionine ligands and a bulky diamine ligand (N,N,N',N'-tetramethylethylenediamine, Me(4)en) would have severe interligand clashes; such interligand clashes are less pronounced in bis(9-ethylguanine) complexes. Consistent with these observations, NMR studies with [Pt(Me(4)en)(D(2)O)(2)](2+) have indicated that guanine 5'-monophosphate reacts in a 2:1 guanine:platinum ratio while both methionine and N-acetylmethionine react with only a 1:1 stoichiometry. Methionine forms a chelate via the sulfur and nitrogen atoms whereas N-acetylmethionine forms a chelate via the sulfur and oxygen atoms. The oxygen of the latter chelate can be displaced by the addition of guanosine 5'-monophosphate, although complete displacement of the N-acetylmethionine was not observed. 相似文献

(1R,2R)-N1-2-amyl-1,2-diaminocyclohexane, which has an amyl substituent as compared with 1,2-diaminocyclohexane, was used as the carrier group to construct three platinum(II) complexes. MTT assay revealed that the complexes showed decent cytotoxicity against all of the four tested tumor cell lines with the IC50 values ranging from 1.08 to 253.36 μM. Particularly, the IC50 values of 2 against A549 and HCT-116 reached 3.32 and 1.08 μM, respectively, which were much lower than those of cisplatin and oxaliplatin. Flow cytometry demonstrated that 2 inhibited HepG2 cells proliferation and caused cytotoxicity by inducing apoptosis and arresting cells in the G2 phase. Furthermore, agarose gel electrophoresis showed that 2 had the ability to interact with DNA in a manner different from cisplatin and oxaliplatin, indicating the carrier ligand with an alkyl moiety had an influence on the action mode of the complex. 相似文献

Pd(II) and Pt(II) complexes with the anions of the model nucleobases 1-methylthymine (1-MethyH), 1-methyluracil (1-MeuraH), and 1-methylcytosine (1-MecytH) of the types [Pd(dmba)(mu-L)]2 [dmba = N,C-chelating 2-((dimethylamino)methyl)phenyl; L = 1-Methy, 1-Meura or 1-Mecyt] and [M(dmba)(L)(L')] [L = 1-Methy or 1-Meura; L' = PPh(3) (M = Pd or Pt), DMSO (M = Pt)] have been obtained. Palladium complexes of the types [Pd(C6F5)(N-N)(L)] [L = 1-Methy or 1-Meura; N-N = N,N,N',N'-tetramethylethylenediamine (tmeda), 2,2'-bipyridine (bpy), or 4,4'-dimethyl-2,2'-bipyridine (Me2bpy)] and [NBu4][Pd(C6F5)(1-Methy)2(H2O)] have also been prepared. The crystal structures of [Pd(dmba)(mu-1-Methy)]2, [Pd(dmba)(mu-1-Mecyt)]2.2CHCl3, [Pd(dmba)(1-Methy)(PPh3)].3CHCl3, [Pt(dmba)(1-Methy)(PPh3)], [Pd(tmeda)(C6F5)(1-Methy)], and [NBu4][Pd(C6F5)(1-Methy)2(H2O)].H2O have been established by X-ray diffraction. The DNA adduct formation of the new platinum complexes synthesized was followed by circular dichroism and electrophoretic mobility. Atomic force microscopy images of the modifications caused by the platinum complexes on plasmid DNA pBR322 were also obtained. Values of IC50 were also calculated for the new platinum complexes against the tumor cell line HL-60. A new series of DNA binding 5,10,15-tri(N-methyl-4-pyridiniumyl)porphyrin (TrisMPyP)-platinum(II) conjugates was synthesized, in which different spacer ligands were used for appropriate coordination to platinum(II) complexes. The compounds trans-[Pt(OCHO)R(PPh3)2] (R = C6Cl5; 2,3,4,6-C6HCl4; 2,3,4,5-C6HCl4; 2,5-C6H3Cl2) have been prepared by treatment of [PtIR(PPh3)2] with AgClO4 followed by reaction with NaOCHO in methanol. The cis isomers have been obtained by the direct reaction of HCO2H with compounds containing PtHg bonds. For these and the analogous compounds containing C6F5 ligands, the dependence of J(31P195Pt) on R has been studied, and the effects of cis-R shown to be in the opposite direction from those of trans-R ligands. 相似文献

The absorption spectra, emission spectra, and emission lifetimes of Pt(Phpy)2, Pt(Thpy)2, and Pt(Bhq)2 complexes (Phpy−, Thpy−, and Bhq− are the ortho C-deprotonated forms of 2-phenylpyridine, 2-(2-thienyl)-pyridine, and benzo(h)quinoline) have been studied and compared with those of the C-protonated neutral ligands. For all complexes examined the low-energy absorption bands in the near UV and visible region are assigned to metal-to-ligand charge-transfer transitions. The strong and structured luminescence emissions observed in the 500–600 nm region (lifetime in the microsecond range at 77 K) are assigned to metal-to-ligand charge-transfer excited states. 相似文献
