Efforts towards the first total synthesis of (−)-oxazolomycin B and (+)-oxazolomycin C from the intermediate of our previous synthesis of (+)-neoxazolomycin are reported. The syntheses were achieved in a longest linear sequence of 25 steps from the amino acid serine in 3.6 and 2.7 % overall yields, respectively. The efficiency of our approach is derived from silyl triflate-mediated reductive oxazolidine ring-opening and Fürstner's Ru-catalyzed hydrosilylation and protodesilylation reactions. The obtained spectra and optical rotations were in good agreement with those of natural products, thus confirming the structures.  相似文献

Three tetraorganodistannoxanes containing silicon were synthesized and their catalytic properties in esterification and acetalization were observed via the reactions of acetic acid with isoamyl alcohol and butyraldehyde with glycol. The factors affecting the reaction, such as the catalyst dosage, reaction time and solvent, etc. were discussed. The results show that the three tetraorganodistannoxanes displayed similar good catalytic activities compared to dichlorotetrabutyldistannoxane in esterification and acetalization. When the dosage of [ClBu2SnOSn-(CH2SiMe3)2Cl]2 was 1.5% based on the mass of reactant, the yield of isoamyl acetate was 91.8% and the yield of butyraldehyde glycol acetal 94.2%. The different alkyl and bridging groups on Sn sites in the structure of tetraorganodistannoxanes showed some influence on the catalytic activities of these compounds. __________ Translated from Chinese Journal of Applied Chemistry, 2007, 24 (11): 1265–1272  相似文献

The proof of the pudding : The first asymmetric total synthesis of the marine tetracyclic oxasqualenoid (+)‐omaezakianol features a convergent olefin cross‐metathesis between a monotetrahydrofuran fragment and a triepoxy alkene, and cascade oxacyclizations of a triepoxy alcohol to form the right‐hand three ether rings. The total synthesis proved the absolute configuration of (+)‐omaezakianol to be that shown.

Here we report the synthesis of all four stereoisomers of mefloquine. Mefloquine (Lariam) is an important anti‐malaria drug that is applied as a racemate of the erythro form. However, the (?)‐isomer induces psychosis, while the (+)‐enantiomer does not have this undesired side effect. There are six syntheses of which five lead to the wrong enantiomer without the authors of these syntheses noting that they had synthesized the wrong compound. At the same time physical chemistry investigations had assigned the absolute configuration correctly and the last enantioselective synthesis that took these results into account delivered the correct absolute configuration. Since various synthetic approaches failed to provide the correct stereoisomers in previous syntheses, we submit here a synthetic approach with a domino Sonogashira‐6π‐electrocyclisation as key step that confirmed synthetically the correct absolute configuration of all four isomers.  相似文献

Three new cytotoxic 10,13-cyclotrichothecane-derived macrolides, myrothecines A-C (1-3), were characterized from the extracts of two Myrothecium roridum strains, IFB-E009 and IFB-E012, isolated as endophytic fungi found on the traditional Chinese medicinal plants Trachelospermum jasminoides and Artemisia annua, respectively. The absolute configuration of myrothecines A-C was elucidated by a combination of spectral techniques (UV, IR, MS, circular dichroism (CD), (1)H and (13)C NMR, DEPT, (1)H-(1)H COSY, NOESY, HMQC, and HMBC spectrascopic analyses), Mosher's ester analysis, and single-crystal X-ray diffraction. The absolute configuration of the reported bioactive analogue, mytoxin B was established by correlating its spectral data with that of known absolute configurational structures. Furthermore, the significance in endophytism (or symbiosis) and biocatabolism, highlighted by production of those macrolides by the endophytic strains, is discussed in brief.  相似文献

Recent studies of the synthesis, structures, spectroscopic properties, and reactions of a series of isolable metallylenes (R2E:, E = Si (1), Ge (2), and Sn (3); R2 = 1,1,4,4-tetrakis(trimethylsilyl)butane-1,4-diyl) are summarized. Because these group-14 metallylenes bear the same helmet-like ligand, a straightforward discussion of the element-dependence of the intrinsic properties of the group-14 element divalent compounds is possible. All these metallylenes were monomeric both in solution and in the solid state, indicating the effective steric protection by the ligand against dimerization. A small sigma-pi conjugation between C-Si(substituent) sigma orbitals and the vacant npz orbitals of divalent atoms in R2E: exists and the extent decreases in the order E = Si > Ge > Sn, as evidenced by UV-vis and NMR spectroscopies and X-ray crystallography. However, the extent of the sigma-pi conjugation in metallylenes 1-3 was much smaller than the electron-donating effects of neighboring nitrogen atoms in known stable cyclic diamino-substituted metallylenes, and hence metallylenes 1-3 are regarded as the least electronically perturbed. Comparative studies of the unique reactions among these metallylenes are also discussed.  相似文献

In the last ten years, the study and the search for new multiferroic materials have been a major challenge due to their potential applications in electronic technology. In this way, bismuth‐containing perovskites (BiMO3), and particularly those in which the metal M position is occupied by a magnetically active cation, have been extensively investigated as possible multiferroic materials. From the point of view of synthesis, only a few of the possible bismuth‐containing perovskites can be prepared by conventional methods but at high pressures. Herein, the preparation of one of these potential multiferroic systems, the solid solution xBiMnO3‐(1?x)PbTiO3 by mechanosynthesis is reported. Note that this synthetic method allows the oxides with high x values, and more particularly the BiMnO3 phase, to be obtained as nanocrystalline phases, in a single step and at room temperature without the application of external pressure. These results confirm that, in the case of Bi perovskites, mechanosynthesis is a good alternative to high‐pressure synthesis. These materials have been studied from the point of view of their structural characteristics by precession electron diffraction and magnetic property measurements.  相似文献

Diorcinols and related prenylated diaryl ethers were reported to exhibit activity against methicillin-resistant clinical isolates of Staphylococcus aureus (MRSA). Within these lines, we report the first total synthesis of diorcinol D, I, J, the proposed structure of verticilatin and recently isolated antibacterial diaryl ether by using an efficient and highly divergent synthetic strategy. These total syntheses furnish the diaryl ethers in only five to seven steps employing a Pd-catalyzed diaryl ether coupling as the key step. The total synthesis led to the structural revision of the natural product verticilatin, which has been isolated from a plant pathogenic fungus. Furthermore, these structures were tested in order to determine their antibacterial activities against different MRSA strains as well as further Gram-positive and -negative bacteria.  相似文献

Presilphiperfolanols constitute a family of biosynthetically important sesquiterpenes which can rearrange to diverse sesquiterpenoid skeletons. While the origin of these natural products can be traced to simple linear terpene precursors, the details of the enzymatic cyclization mechanism that forms the stereochemically dense tricyclic skeleton has required extensive biochemical, computational, and synthetic investigation. Parallel efforts to prepare the unique and intriguing structures of these compounds by total synthesis have also inspired novel strategies, thus resulting in four synthetic approaches and two completed syntheses. While the biosynthesis and chemical synthesis studies performed to date have provided much insight into the role and properties of these molecules, emerging questions regarding the biosynthesis of newer members of the family and subtle details of rearrangement mechanisms have yet to be explored.  相似文献

A nearly‐30‐year‐old unanswered synthetic puzzle, astellatol, has been solved in an enantiospecific manner. The highly congested pentacyclic skeleton of this rare sesterterpenoid, which possesses a unique bicyclo[4.1.1]octane motif, ten stereocenters, a cyclobutane that contains two quaternary centers, an exo‐methylene group, and a sterically encumbered isopropyl trans‐hydrindane motif, makes astellatol arguably one of the most challenging targets for sesterterpenoid synthesis. An intramolecular Pauson–Khand reaction was exploited to construct the right‐hand side scaffold of this sesterterpenoid. An unprecedented reductive radical 1,6‐addition, mediated by SmI2, forged the cyclobutane motif. Last, a strategic oxidation/reduction step provided not only the decisive solution for the remarkably challenging late‐stage transformations, but also a highly valuable unravelling of the notorious issue of trans‐hydrindane synthesis. Importantly, the synthesis of astellatol showcases a rapid, scalable strategy to access diverse complex isopropyl trans‐hydrindane sesterterpenoids.  相似文献

The α-metallated ylides [Ph3P−C−Z]M+ (with Z=SO2Tol or CN and M=Na or K) were used as versatile nucleophiles for the facile access to ylide-substituted compounds. Halogenations, alkylations, carbonylations and functionalization reactions with main group element halides were easily accomplished by simple trapping reactions with the appropriate electrophiles. X-ray crystallographic studies of all compounds – including the first structures of α-fluorinated P-ylides – showed remarkable differences in the ylide backbone depending on the substituents. In the fluorinated compounds, a change from a fully planar to a pyramidalized ylidic carbon centre was observed despite the strongly anion-stabilizing ability of the yldiide substituent. π-Donation from the ylide substituent also resulted in geometric restrictions depending on the steric and electronic properties of the introduced substituents.  相似文献

The impressive biological profile of secondary metabolites isolated from strains of Sorangium cellulosum prompted us to initiate synthetic studies on kulkenon, also isolated from Sorangium cellulosum. The synthesis features a syn‐selective vinylogous Kobayashi aldol reaction, recently developed by us, and a ring‐closing intramolecular Heck reaction as the pivotal transformations. Comparison of the NMR spectra of the authentic and synthetic material revealed that the proposed configuration had to be revised. A combination of molecular modeling and NOE experiments was used to propose the revised configuration, which was confirmed by a new synthesis.  相似文献
