Herein, we describe an accurate method for protein quantification based on conventional acid hydrolysis and an isotope dilution-ultra performance liquid chromatography–tandem mass spectrometry method. The analyte protein, recombinant human erythropoietin (rhEPO), was effectively hydrolyzed by incubation with 8 mol/L hydrochloric acid at 130 °C for 48 h, in which at least 1 μmol/kg of rhEPO was treated to avoid possible degradation of released amino acids during hydrolysis. Prior to hydrolysis, sample solution was subjected to ultrafiltration to eliminate potential interfering substances. In a reversed-phase column, the analytes (phenylalanine, proline, and valine) were separated within 3 min using gradient elution comprising 20 % (v/v) acetonitrile and 10 mmol/L ammonium acetate, both containing 0.3 % (v/v) trifluoroacetic acid. The optimized hydrolysis and analytical conditions in our study were strictly validated in terms of accuracy and precision, and were suitable for the accurate quantification of rhEPO. Certified rhEPO was analyzed using a conventional biochemical assay kit as an additional working calibrant for the quantification of EPO and improved the accuracy. The optimized protocol is suitable for the accurate quantification of rhEPO and satisfactorily serves as a reference analytical procedure for the certification of rhEPO and similar proteins.

The interaction of the enantiopure (R)- and (S)-1-phenyl-N,N-bis(pyridine-3- ylmethyl)ethanamine ligands, R-L 1 and S-L 1 , with copper(II) chloride followed by addition of hexafluorophosphate resulted in the isolation of the corresponding enantiomeric complexes [Cu(R-L 1 )Cl](PF6) (1), [Cu(S-L 1 )Cl](PF6) (2) and [Cu(S-L 1 )Cl](PF6)??0.5Et2O (3), in which dimerization occurs through two long Cu??????Cl interactions, the ??-chloro bridges being thus strongly asymmetric. The organic ligand is bound to the metal centre via its N3-donor dipyridylmethylamine fragment in a planar fashion, such that each copper centre is in a square planar environment (or distorted square pyramidal with a long axial bond length if the additional interaction is considered). When R,S-L 1 was employed in a parallel synthesis, the similar racemic complex [Cu(R,S-L 1 )Cl](PF6)??0.5MeOH (4) was obtained, in which the L 1 ligands in each dimeric unit have opposite hands. In contrast to the complexes of L 1 , the reaction of Cu(II) chloride with the related ligand, (R)-1-cyclohexyl-N,N-bis(pyridine-3-ylmethyl)ethanamine (R-L 2 ), yielded the mononuclear complex [Cu(R,S-L 2 )Cl2] (5), displaying a distorted square pyramidal coordination geometry. The structure of this product along with its corresponding circular dichroism spectrum revealed that racemisation of the starting R-L 2 ligand has occurred under the relatively mild (basic) conditions employed for the synthesis. A temperature-dependent magnetic studies of the complexes 1, 2 and 5 indicate that a week ferromagnetic interaction is operative in each dicopper core in 1 and 2 with 2J?=?1.2?cm?1. (S)-Indoline-2-carboxylic acid was synthesized by use of a nitro amination approach with l-phenylalanine as chiral pool. The first step of the synthesis was nitration of l-phenylalanine, with urea nitrate (UN)/H2SO4 as nitrating reagent, to give 2,4-dinitro-l-phenylalanine in 75.7 % yield in one-pot synthesis and 69.1 % yield by step-wise nitration. Intramolecular nitro amination of 2,4-dinitro-l-phenylalanine gave (S)-6-nitro-indoline-2-carboxylic acid in 65.7 % yield and more than 99.5 % enantiomeric excess (ee). The title compound, (S)-indoline-2-carboxylic acid, was obtained in 85.9 % yield and high ee by one-pot transformation of (S)-6-nitroindoline-2-carboxylic acid. The total synthesis consisted of three operations and gave the title compound in 42 % yield and more than 99.5 % ee.  相似文献

In the frame of a project aimed at finding non-steroidal farnesoid X receptor (FXR) agonists, we identified 4-(2,4-dimethoxyphenyl)-3,6-dimethyl-1-(2-tolyl)-4,8-dihydro-1H-pyrazole[3,4-e][1,4]thiazepin-7-one (1) as a hit endowed with FXR activity. Most of the compounds synthesised during the hit-to-lead optimisation work were characterised by the presence of two chiral centres and were therefore obtained as mixtures of anti(±)- and syn(±)-diastereoisomers. A restricted sub-set of species harboured with a carboxylic acid group on the distal phenyl ring of the biphenyl (a(±)5 (A1) and s(±)5 (S1)) or the phenoxyphenyl (a(±)6 (A2) and s(±)6 (S2)) moiety at C-4 position of the pyrazole[3,4-e][1,4]thiazepin-7-one core, resulted in suitable diastereo- and enantioresolution with a quinine (QN) carbamate-derived chiral stationary phase (CSP). Differently from the compounds usually analysed with QN-based CSPs, the couples A1/S1 and A2/S2 were atypical selectands, in which the two chiral carbon atoms reside at a remote position with respect to the carboxylic function, the main “point of attack” to the CSP. We produced evidence that the scarcely employed normal-phase (NP) eluent systems represent the elective choice for achieving the simultaneous diastereo- and enantioseparation of this class of compounds over the usually preferred reversed-phase (RP) and polar-organic (PO) modes of elution. Indeed, after the optimisation of the eluent composition, NP conditions allowed to obtain profitable enantioselectivity profiles, along with excellent diastereoselectivity levels (α(A1)?=?1.07, R S(A1)?=?1.15; α(S1)?=?1.09, R S(S1)?=?1.47; α(A2)?=?1.08, R S(A2)?=?1.31; and α(S2)?=?1.06, R S(S2)?=?1.18). The optimised NP methods are suitable for simultaneously providing information on the diastereo- and enantiopurity of the investigated compounds.

The kinetics of hydrolysis of two alkoxy-NNO-azoxy compounds with geminal position of N2O2 groups, di(methoxy-NNO-azoxy)methane (I) and di(methyl-NON-azoxy)formal (II), as well as isomeric geminal nitramine, 2,4-dinitro-2,4-diazapentane (III) in 64.16% H2SO4 were studied by a manometric method. The relative rates of the hydrolysis at 80°C of compounds I?CIII and methoxy-NNO-azoxymethane (IV) were found to be equal to 4.2:77:??50000:1. The limiting stage of hydrolysis of compound I is the attack of the nucleophile on the carbon atom of the MeO group of the protonated molecule I by S N2 mechanism. According to the parameters of the Arrhenius equation the hydrolysis of compound II proceeds more probably by the S N1  相似文献

Cyanuration of 2-naphthaldehyde (1) and 5-methyl-2-furaldehyde (2) yielded the racemic 2-hydroxy-2-(β-naphthyl)ethanenitrile (R,S)-3 and 2-hydroxy-2-(5-methyl-2-furyl)ethanenitrile (R,S)-5, respectively. The same reaction can be completed by using acetone cyanohydrin (4) as a transcyanating agent. The optically active (R)-3 and (S)-5 could be respectively obtained by hydrocyanation of 1 and 2 using (R)-hydroxynitrile lyase (R)-PaHNL [EC 4.1.2.10] from almonds (Prunus amygdalus) as a chiral catalyst. Cyanohydrins 3 and 5 in their racemic and optically active forms undergo a number of transformations which involve either the hydroxyl group or the cyanide function. Moreover, derivatization of 3 and 5 with (S)-Naproxen®chloride (S)-14 gave the respective diastereoisomers. The optical activity of (R)-3 and (S)-5 as well as their derivatives were recorded. The postulated structures for the new products were supported with compatible elementary and spectroscopic (IR, 1H NMR, 13C NMR, MS, and single crystal X-Ray crystallography) analyses. The antimicrobial activity of some selected racemic new products and their respective optically active analogues were also undertaken.  相似文献

We have developed a specific method for the visual detection of Staphylococcus aureus based on aptamer recognition coupled to tyramine signal amplification technology. A biotinylated aptamer specific for S. aureus was immobilized on the surface of the wells of a microplate via biotin-avidin binding. Then, the target bacteria (S. aureus), the biotinylated-aptamer-streptavidin-HRP conjugates, biotinylated tyramine, hydrogen peroxide and streptavidin-HRP were successively placed in the wells of the microplate. After adding TMB reagent and stop solution, the intensity of the yellow reaction product can be visually inspected or measured with a plate reader. Under optimized conditions, there is a linear relationship between absorbance at 450 nm and the concentration of S. aureus in the 10 to 107 cfu mL?1 concentration range (with an R2 of 0.9976). The limit of detection is 8 cfu mL?1.

A selective and low organic-solvent-consuming method of sample preparation combined with high-performance liquid chromatography with diode-array detection is introduced for analysis of phthalic acid esters in edible oils. Sample treatment involves initial liquid–liquid partitioning with acetonitrile, then QuEChERS cleanup by dispersive solid-phase extraction with primary secondary amine as sorbent. Preconcentration of the analytes is performed by ionic-liquid-based dispersive liquid–liquid microextraction, with the cleaned-up extract as disperser solvent and 1-hexyl-3-methylimidazolium hexafluorophosphate as extraction solvent. Under the optimized conditions, correlation coefficients (r) were 0.998–0.999 and standard errors (S y/x ) were 2.67–3.37?×?103 for calibration curves in the range 50–1000 ng g?1. Detection limits, at a signal-to-noise ratio of 3, ranged from 6 to 9 ng g?1. Intra-day and inter-day repeatability, expressed as relative standard deviation, were in the ranges 1.0–6.9 % and 2.4–9.4 %, respectively. Recovery varied between 84 % and 106 %. The developed method was successfully used for analysis of the analytes in 28 edible oils. The dibutyl phthalate content of four of the 28 samples (14 %) exceeded the specific migration limit established by domestic and international regulations.

Oxidation of the α- and β-4-phenyl-1,2,4-triazolin-3,5-dione adducts of vitamin D3 (2 and1) withMCPBA yields two diastereomeric mixtures of the (5,10)-(7,8)-dioxiranes3 a,3 b,3 c and4 a,4 b respectively. The corresponding benzoates5 a,5 b,6 a and6 b were prepared and the X-ray crystal structure of5 b was determined. This analysis proved5 b to be the (5R, 1 OS)-(7R, 8R)-dioxirane of the β-resp. Gold nanoparticles were in-situ decorated on top of a polyaniline film (GNPs–PANI) via the direct electroreduction of the adsorbed AuCl 4 - ions on a glassy carbon electrode that previously was coated with PANI by electropolymerization. The GNPs–PANI composite and the performance of the resultant sensors were investigated in some detail. The sensor was applied to the oxidation of dopamine (DA) with improved catalytic activity. Its catalytic current showed wide linear response toward dopamine ranging from 3 to 115 μM, with a low detection limit of 0.8 μM (S/N=3). In addition, the sensor exhibits easy-operation, fast response to dopamine, as well as excellent reproducibility and stability.
Gold nanoparticles decorated polyaniline (GNPs-PANI) was prepared via electrosynthesis. The GNPs-PANI composite showed good catalytic activity toward the oxidation of dopamine.  相似文献

This work reported an efficient electrochemical treatment for drinking water disinfection using a pyrolytic graphite electrode modified with ferrocenyl tethered poly(amidoamine) dendrimers–multiwalled carbon nanotubes–chitosan nanocomposite. The influence parameters of electrochemical disinfection of Escherichia coli and Staphylococcus aureus, such as applied potential and sterilization time, were investigated. Further investigation indicated that almost all (99.99 %) of the initial bacteria were killed after applying a low potential of 0.4 V for 10 min. During the electrochemical disinfection process, the oxidized form of ferrocene was formed on electrode, which played a key role in the disinfection towards E. coli and S. aureus. Hence, the proposed method may provide potential application for the disinfection of drinking water.

An electrophoretically mediated microanalysis assay (EMMA) for the determination of the stereoselective reduction of l-methionine sulfoxide diastereomers by methionine sulfoxide reductase enzymes was developed using fluorenylmethyloxycarbonyl (Fmoc)-l-methionine sulfoxide as substrate. The separation of the diastereomers of Fmoc-l-methionine sulfoxide and the product Fmoc-l-methionine was achieved in a successive multiple ionic-polymer layer-coated capillary using a 50 mM Tris buffer, pH 8.0, containing 30 mM sodium dodecyl sulfate as background electrolyte and an applied voltage of 25 kV. 4-Aminobenzoic acid was employed as internal standard. An injection sequence of incubation buffer, enzyme, substrate, enzyme, and incubation buffer was selected. The assay was optimized with regard to mixing time and mixing voltage and subsequently applied for the analysis of stereoselective reduction of Fmoc-l-methionine-(S)-sulfoxide by human methionine sulfoxide reductase A and of the Fmoc-l-methionine-(R)-sulfoxide by human methionine sulfoxide reductase B. The Michaelis–Menten constant, K m, and the maximum velocity, v max, were determined. Essentially identical data were determined by the electrophoretically mediated microanalysis assay and the analysis of the samples by CE upon offline incubation. Furthermore, it was shown for the first time that Fmoc-methionine-(R)-sulfoxide is a substrate of human methionine sulfoxide reductase B.

We have synthesized a cobalt(II) coordination polymer and have characterized it by various methods including X-ray single crystal structural analysis. The polymer was used as modifier to fabricate a carbon paste electrode that displays electrochemical activity towards tryptophan (Trp). Trp is oxidized at the surface of the electrode in buffer solution of pH 4.2, yielding a single peak at 814 mV. The experimental conditions such as the concentration, the composition and the pH values of the supporting electrolyte, accumulation time, and the scan rate were optimized. Under the optimized conditions, the current of peak is linearly related to the concentration of Trp in the range from 0.2 to 8.0 μM, and from 8.0 to 80.0 μM. The detection limit (at S/N?=?3) is 0.1 μM at an accumulation time of 60 s. The determination of Trp in amino acid injection solutions was evaluated and the results were satisfactory. The recoveries were in the range of 97.5% to 103.0%.

As shown in Fig. 1, a well-defined and very sharp oxidation peak at 814 mV was obtained in the curve the curve b. Whereas, a relatively weak anodic peak at 835 mV can be observed under the same conditions with an bare electrode. By comparing curve b and c, it can be seen that, the peak current was about 0.720 μA with the bare CPE. However, the peak current was increased to 10.39 μA with the modified CPE. This significant improvement of peak current may be related to the effective catalytic fashion of the cobalt (II) coordination polymer film modified on the surface of the CPE in the electrochemical oxidation of Trp.  相似文献

The interaction of Cu(II), Fe(III) and Co(III) with 6,6,13-trimethyl-13-amino-1,4,8,11-tetraazacyclotetradecane (L 3 ) incorporating a pendent amine group has led to isolation of the new octahedral complexes [Cu(HL 3 )(ClO4)2]Cl·H2O (1), [Fe(L 3 )Cl](S2O6)·H2O (2), [Co(L 3 )Cl](ClO4)1.5Cl0.5·0.25H2O (3), [Co(HL 3 )Cl2](ClO4)2·H2O (4) and [Co(L 3 )Cl]2(S2O4)(ClO4)2 (5). In (1) the copper ion occupies the macrocyclic cavity of protonated (–NH3 +) L 3 which is present in its trans-III configuration; weakly bound ClO4 ? ligands occupy the axial positions. The X-ray structure of (2) showed that Fe(III) occupies the N4-macrocyclic cavity of L 3 in a trans-III configuration, with the pendent amine group binding in an axial position. The remaining axial position is occupied by a Cl? ligand. Chromatography of the product obtained from the reaction of Na3[Co(CO3)3] with L 3 yielded three fractions. Fraction 1 yielded crystals (3) composed of three crystallographically independent species incorporating cations of type [Co(L 3 )Cl]2+ with very similar structures; in each case the macrocyclic ring nitrogens of L 3 are bound to the Co(III) in an asymmetric cis-fashion. Fraction 2 yielded the trans-III octahedral cationic complex (4) incorporating L 3 in its protonated form. The Co(III) complex (5) from fraction 3 shows a different coordination arrangement to the products from fractions 1 or 2. The macrocyclic ring coordinates in its trans-III form, but the axial sites in this case are occupied by the pendent-NH2 group and a Cl? ligand.  相似文献

Reaction of a macrocyclic copper(II) complex [Cu(L)](ClO4)2 · 3H2O (I) (L = 1,3,10,12,16,19-hexaazatetracyclotetracosane) with a hexapod carboxylate ligand H6TTHA (H6TTHA = 1,3,5-triazine-2,4,6-triamine hexaacetic acid) and a tripod carboxylate ligand H3TATB (H3TATB = 4,4′,4″-S-triazine-2,4,6-triyl-tribenzoic acid) yielded two mononuclear copper(II) complexes [Cu(L)][H4TTHA] · 4H2O (II) and [Cu(L)][HTATB] · 4H2O (III). The complexes I–III have been structurally characterized. The crystal structures of complexes II and III show the copper(II) ion has a distorted pentacoordinate square-pyramidal geometry with two secondary and two tertiary amines from the macrocyclic complex [Cu(L)]2+ and one oxygen atom from the carboxylate ligand group at the axial position. The UV-Vis spectra are utilized to discuss the hydrolysis of the complex II.  相似文献

Two series of poly(imide-ester)s (PIEs) and poly(ether-imide-ester)s (PEIEs), having benzoxazole or benzothiazole pendent groups, were conveniently prepared by the diphenylchlorophosphate-activated direct polyesterification of two bis(imide-carboxylic acid)s (1), such as 2-[3,5-bis(N-trimellitimidoyl)phenyl]benzoxazole (1 O ) and 2-[3,5-bis(Ntrimellitimidoyl) phenyl]benzothiazole (1 S ) and two bis(imide-ether-carboxylic acid)s (2), such as 2-[3,5-bis(4-trimellitimidophenoxy)-phenyl]benzoxazole (2 O ), and 2-[3,5-bis(4-trimellitimidophenoxy)-phenyl]benzothiazole (2 S ) with various aromatic dihydroxy compounds in the presence of pyridine and lithium chloride. The structures, solubilities and thermal properties of obtained polymers were investigated in detail. All of the resulting polymers were characterized by FTIR and 1H-NMR spectroscopy and elemental analysis. All of the resulting polymers exhibited excellent solubility in common organic solvents, such as pyridine, tetrahydrofuran and m-cresol, as well as in polar organic solvents, such as N-methyl-2-pyrrolidone, N,N-dimethylacetamide, N,N-dimethylformamide and dimethyl sulfoxide. The modified polymers were obtained in quantitative yields with inherent viscosities between 0.47 and 0.67 dl·g?1. Experimental results indicated that all the polymers had glass transition temperature between 198 °C and 262 °C, the decomposition temperature at 10% weight loss between 398 °C and 531 °C under nitrogen.  相似文献

A new chromatographic method for the enantioseparation and the determination of (?)-trans-paroxetine and (+)-trans-paroxetine has been developed with the aid of amylose ovomucoid-based chiral stationary phase. The method is faster and five times more sensitive than procedures recommended previously: limit of detection and limit of quantification are 5 and 16 ng/mL, respectively [modified (Ferretti et al. in J Chromatogr B 710:157–164, 1998): 20 and 60 ng/mL]. It was carefully validated and applied for the determination of (?)-trans-paroxetine and (+)-trans-paroxetine in Parogen (Mc Dermott Laboratories Ltd.) and Xetanor (Actavis) coated tablets.

Stereoselective (exo-specific) synthesis, dynamic 1H NMR and computational analysis of exo-N??-{3-azatricyclo[3.2.1.0.2,4]oct-3-yl)mesithyloxy)methylene}-1-benzensulfunamide (3) were investigated. Aziridine nitrogen inversion gives rise to two sets of configurations where the N-substituent is Syn (S) or Anti (A) to C7 of the norbornyl ring. At lower temperature, the proton signals of aziridine exo-E-3 decoalesces to show two syn conformers and one anti conformer (exo-E-3 1 S ? $ \leftrightharpoons $ ?exo-E-3 2 S ? $ \leftrightharpoons $ ?exo-E-3 3 A ) with ratio of 60:20:20, respectively. Experimentally, the Gibbs free energy of activations [??G ? (kcal/mol) ?±?0.08] were calculated 11.96, 12.45 for 3 isomerizations. The standard Gibbs free energy (??G o kcal/mol) 0.174, 0, 0.174, and 0.298 at 213?K and energy minimum 6.64, 4.77 and 1.78 were calculated for 3 1S? $ \leftrightharpoons $ ?3 2S, 3 2S? $ \leftrightharpoons $ ?3 3 A , 3 1 S? $ \leftrightharpoons $ ?3 3 A isomerizations, respectively. The enthalpy (??H ?, kcal/mol) and entropy (??S ?, cal?mol?1?K?1) of activation for the nitrogen inversion of aziridine of 3 were calculated 11.2 and ?0.80, respectively.  相似文献
