99mTc-HSA-D has been developed as a new blood pool scanning agent. Clinical comparison of 99mTc-HSA-D and 99mTc-HSA was made in 16 cases. The activity concentration of 99mTc in blood was measured during 2 hours after the injection in five cases. 99mTc-HSA-D showed higher concentration compared to 99mTc-HSA with the passage of time. Quantitative analysis of contrast between left ventricle and septum was performed on end diastolic frames of gated images 10 minutes after the injection. There was no obvious difference between 99mTc-HSA-D and 99mTc-HSA. The subjective comparison of detectability of lesions between the two agents was performed on three directional gated images. 99mTc-HSA-D was superior to 99mTc-HSA, because the images of the latter deteriorated with the passage of time. On anterior view images 1 hour after the injection, left ventricle/lung and abdominal aorta/background count ratios were greater for 99mTc-HSA-D in many cases. There was no obvious difference in liver/background and kidney/background count ratios between the two agents. Urinary excretion of 99mTc was considerably lesser for 99mTc-HSA-D. The results indicated that 99mTc-HSA-D was superior to 99mTc-HSA for cardiac blood pool imaging.  相似文献

The interaction of bilirubin and99mTc-HIDA and99mTc-IODIDA has been studied in rats. The mechanism of this interaction has been examined at the level of binding with human serum albumin (HSA) by the in vitro method. Percentage of binding with HSA, and affinity constants for99mTc-IODIDA were determined with and without bilirubin. This paper describes biologic distributions, sequential images and macroautoradiograms of 99mTcO4 (pertechnetate), 99mTc-Sn-HSA (human serum albumin) and 99mTc-Sn-TSL (trasylol) in tumor-bearing mice as the first report on tumor affinity of 99mTc-labeled radiopharmaceutical. (1) Maximum tumor concentration (% administered dose/g of tissue weight) of 99mTcO4, 99mTc-HSA and 99mTc-TSL in Ehrlich's tumor-bearing mice resulted in 2.03+/-0.57 at 1 hr, 4.02+/-0.19 at 3 hr and 1.97+/-0.31 at 1 hr respectively. (2) However, tumor to blood concentration ratio of 99mTc-HSA was lowest among them. (3) The corrected tumor accumulation (% 100g dose/g of tissue wt.=% dose/% body weight) of 99mTc-TSL to Ehrlich's tumor in mouse was not different from that of Yoshida's sarcoma in rat, on the contrary to our expectation that the tumor concentration of 99mTc-TSL in them might be different due to differency of the tissue fibrinolytic activity between the respective tumors. (4) Sequential images of the implanted tumor in mouse was best positively delineated with 99mTc-HSA. (5) Macroautoradiograms of Ehrlich's tumor with 99mTcO4, 99mTc-HSA and 99mTc-TSL demonstrated the following findings: all of them were not only accumulated markedly into the tumor cells which were shown as basophilic tissue with H?matoxylin-Eosin staining but also accumulated around the tumor tissue and on the interstitial tissue which were stained as eosinophilic tissue with the above same staining.  相似文献

用亲和色谱研究了两种中药小分子阿魏酸(FA)、丹皮酚(PAE)在人体生理条件缓冲溶液(pH7.4)条件下与人血清白蛋白(HSA)的相互作用.从药物分子在蛋白质分子上有多种类型相互独立的结合位点的假定出发,应用Langmuir吸附模型和竞争置换分析研究了FA,PAE与HSA的竞争性相互作用.结果表明,FA,PAE与HSA之间存在一类位点,且FA与PAE竞争HSA上的indole位点(siteⅡ).根据热力学参数推测出FA,PAE与HSA之间的作用力主要为氢键作用.从FA,PAE竞争HSA上同一位点的角度,对中医用药中常将含有FA与含有PAE的中药配伍使用,以提高疗效的临床用药现象进行了解释.  相似文献

The new water-soluble photosensitizer 5,10,15,20-tetrakis[3,4-bis(carboxymethyleneoxy)phenyl]chlorin (T3,4BCPC) has been prepared, characterized and labeled with 99mTc radionuclide. The radiotracer was evaluated for tissue distribution in Wistar rats. Accumulation of administrated activities in the liver, kidney, bladder and large intestine at 4 h post-injection indicated that the labeled ligand was largely eliminated through the renal and partly through the hepatobiliary system. In vivo biodistribution studies of the labeled compound were carried out in rodent and murine tumor models in comparison with other tumor-seeking radiopharmaceuticals such as 99mTc(V)-dimercaptosuccinic acid (DMSA), 201thallous chloride (TlCl) and 99mTc-citrate using a gamma camera computer system. In N-nitrosomethylurea (NMU)-induced rat mammary tumors, the labeled ligand showed a five-fold tumor to muscle (T/M) ratio compared to 99mTc(V)-DMSA (3-fold) and 201TlCl (3-fold). In the case of C(3)H/J virus-induced spontaneous mammary tumors, the differences were not marked. However, in the transplanted rat C(6)-glioma, the T/M ratio of the labeled compound was appreciably higher (four-fold) than that noted with 99mTc(V)-DMSA (two-fold), 201TlCl (three-fold) and 99mTc-citrate (more than three-fold). These findings suggest that the radiolabeled T3,4BCPC may have potential for the detection of cancer. In order to ascertain the efficacy of the compound for photodynamic therapy applications, a preclinical PDT study was carried out in fibrosarcoma-bearing mice after injecting 5.0 mg/kg body weight of the T3,4BCPC. A laser dose of 20 mW for 60 s resulted in 80% destruction of tumors. These data suggest that this molecule could be useful for PDT of cancer. The labeled agent could also be useful in monitoring the progression/regression of tumors before, during, and after chemotherapy, radiation therapy or PDT.  相似文献

Stereoselective interaction between a chiral nonsteroidal antiinflammatory drug, namely carprofen (CP), and human serum albumin (HSA) was studied, and the results were compared with those obtained with model dyads. In the presence of albumin the same triplet-triplet transition was detected for both CP stereoisomers; however, time-resolved measurements revealed a remarkable stereodifferentiation in the CP/HSA interaction. For each stereoisomer, the decay dynamics evidenced the presence of two components with different lifetimes that can be correlated with complexation of CP to the two possible albumin binding sites (site I and site II). This assignment was confirmed by using ibuprofen, a site II displacer. Thus, the shorter lived components, for which stereodifferentiation was more important (tauR/tauS ca. 4), were ascribed to the CP triplet state in site I; the lifetime shortening can be attributed to electron-transfer quenching by the only tryptophan (Trp) of the protein. Laser flash photolysis of model dyads containing covalently linked CP and Trp revealed formation of the expected Trp radical cation, providing support for such a mechanism. Moreover, significant stereodifferentiation was observed between the (R)- and (S)-CP-Trp dyads. In the case of CP/HSA complexes, as well as in the model compounds, the stereodifferentiation detected in the decays is in good agreement with that observed in the formation of the only CP photoproduct, resulting from a photodehalogenation process. Moreover, stereodifferentiation was also found to occur for the photobinding of CP to the protein.  相似文献

Modification of the chelate ring structure of technetium-99m (99mTc) dithiosemicarbazone (DTS) chelate was carried out in pursuit of a more stable and lipophilic compound. A new DTS chelating molecule, pentane-2,4-dione bis(N-methylthiosemicarbazone) (PETS), with a 5-6-5 membered chelate ring structure, was synthesized and labeled with 99mTc, PETS generated two 99mTc compounds as major products. Both had much higher stability and lipophilicity than a 5-5-5 membered 99mTc DTS compound, as well as great stability in plasma. Both 99mTc-PETS compounds were rapidly extracted by the brain and heart when injected into mice. Thus, the modified chelate ring structure afforded a preferable characteristics to DTS chelate as for the chelating site for technetium radiopharmaceuticals.  相似文献

Human serum albumin (HSA) is one of the key components in human blood that may influence drug distribution. As such, it is important to know the affinity of any drug for albumin. Previously, Photofrina mixture of monomeric, dimeric and oligomeric porphyrins, has been subjected to HSA binding studies. However, due to its complex nature, binding studies on Photofrin or other hematoporphyrin derivatives with HSA are inconclusive. In this report, the binding properties of some components (dimers and trimers) of Photofrin® and the relationship between murine photosensitizing efficacy and those binding properties were investigated. The interaction of these porphyrins with HSA was investigated by direct ultrafiltration and fluorescent titration techniques with fluorescent probes such as dansyl-L-proline (DP), which is known to interact selectively with site II on HSA. Porphyrins also were tested for antitumor activity in a mouse model following intravenous administration and exposure to laser light. Together, the results suggest that the photosensitizers that were preferentially bound to site II of HSA were most effective at controlling murine tumor regrowth  相似文献

Binding of glycyrrhizin to human serum and human serum albumin   总被引:3,自引:0,他引:3  
The binding of glycyrrhizin (GLZ) to human serum and human serum albumin (HSA) was examined by an ultrafiltration technique. Specific and nonspecific bindings were observed in both human serum and HSA. The association constants (K) for the specific bindings were very similar: 1.31 x 10(5) M-1 in human serum and 3.87 x 10(5) M-1 in HSA. The number of binding sites (n) and the linear binding coefficient (phi) in HSA were 1.95 and 3.09 x 10(3) M-1, respectively. When the human serum protein concentration was assumed to be 4.2% (equal to the measured serum albumin concentration), n in human serum was 3.09, which is similar to the n value in HSA, and phi in human serum was 0.71 x 10(3) M-1, which is reasonably close to that for HSA. The binding pattern of GLZ with human serum protein on Sephadex G-200 column chromatography showed that GLZ binds to only the albumin fraction. It was concluded that the GLZ-binding sites in human serum exist mainly on albumin and GLZ binds to specific and nonspecific binding sites at lower and higher concentrations than approximately 2 mM, respectively.  相似文献

A novel 99mTc labeled complex, [N-[2-((2-oxo-2-(4-(3-phenylpropyl)piperazin-1-yl)ethyl) (2-mercaptoethyl)amino)acetyl]-2-aminoethanethiolato]Technetium(V) oxide (PPPE-MAMA′-99mTcO) ([99mTc]-2) has been designed and prepared based on the integrated approach. The corresponding rhenium complex (PPPE-MAMA′-ReO)(Re-2) has been prepared and characterized. In vitro competition binding assays show moderate affinity of Re-2 towards σ1 and σ2 receptors with Ki values of 8.67 ± 0.07 and 5.71 ± 1.88 μmol, respectively. Planar images obtained at 0.5 h, 4 h, 20 h after I.v. Injection indicate the accumulation of [99mTc]-2 in MCF-7 human breast tumor bearing mice at 20 h. Furthermore, the accumulation of [99mTc]-2 has been inhibited at 20 h after co-injection of [99mTc]-2 plus haloperidol (1 mg/kg). Biodistribution studies of [99mTc]-2 display an in vivo tumor uptake of 0.14% ± 0.01% ID/g at 24 h post I.v. Injection with a tumor/muscle ratio of 6.02 ± 0.87. The above results suggest that [99mTc]-2, derived from a previously published lead compound, retains certain tumor uptake and affinity for σ receptors. [99mTc]-2 may be used as a basis for further structural modifications to develop tumor imaging agents with high affinity for σ receptors.  相似文献

以人血清白蛋白为连接剂和药物载体, 制备了人血清白蛋白-叶酸偶联物(HSA-FA), 其偶联数n≈3. 以SnCl2·H2O作为还原剂, 采用直接标记法制备 99mTc-HSA-FA标记配合物, 其放射化学纯度大于90%. 体外细胞结合实验结果表明, 99mTc-HSA-FA能被叶酸受体高表达的KB细胞特异性摄取. 荷瘤S180小鼠体内生物分布的研究结果表明, 与 99mTc-HSA相比, 99mTc -HSA-FA在小鼠体内的生物分布发生明显变化, 其在肿瘤中的放射性浓集明显增加(t=12.03, P<0.05), 并表现出较高的摄取[(4.37±1.12)%ID/g at 1 h]和滞留[(3.40±0.69)%ID/g at 4 h]; 经注射过量稳定配体使 99mTc-HSA-FA受到抑制后, 其在肿瘤和肾中的摄取明显降低(t=24.17和17.87, P<0.05), 表明人血清白蛋白-叶酸偶联物对叶酸受体有特异性结合.  相似文献

在模拟人体生理酸度(pH=7.4)条件下,运用荧光光谱、紫外光谱和分子模拟技术,研究了磺胺甲恶唑(SMZ)和左氧氟沙星(LVFX)的协同作用.实验结果表明,SMZ、LVFX主要通过氢键和疏水作用与人血清白蛋白(HSA)发生相互作用形成复合物,导致HSA的内源荧光发生静态猝灭.SMZ、LVFX在HSA上有相同的结合位点,即Site I位.在HSA-SMZ(或HSA-LVFX)体系中分别加入LVFX(或SMZ),其结合常数均明显减小,表明LVFX(或SMZ)的存在削弱了HSA-SMZ(或HSA-LVFX)体系的结合能力,使得LVFX(或SMZ)更多被释放,血液中游离的LVFX(或SMZ)浓度增大,SMZ与LVFX共存能够协同增强药效.  相似文献

The magnesium and calcium binding on human serum albumin (HSA) was studied using an affinity chromatography approach. The effects of the mobile phase pH, its ionic strength and column temperature on the transfer equilibrium constants were studied. The thermodynamic data corresponding to the electrostatic interactions occurring during the HSA-ion binding were determined. Enthalpy-entropy compensation revealed that the ion binding mechanism at HSA was independent of the ionic strength, the same at four pH values (6.5, 8, 8.5 and 9), but presented a weak change at physiological pH around 7-7.5 due to a HSA phase transition. A theoretical model based on the Gouy-Chapman theory allows to determine the relative charge density of the HSA surface implied in the binding process and the variation of the number of ions bound to one albumin molecule with the pH.  相似文献

通过紫外、荧光和CD光谱, 研究了船体防污漆中主要防污成分——三丁基锡(TBT)化合物及其降解产物——一丁基锡(MBT)和二丁基锡(DBT)与人血清白蛋白(HSA)相互作用的方式与程度, 考察了浓度、酸度、有机溶剂和离子强度等因素对相互作用的影响. 结果表明, 丁基锡与HSA的相互作用是双重的, 既有丁基锡中锡离子与HSA的配位作用, 又有丁基锡中丁基基团的疏水作用, 导致HSA二级结构破坏, α-螺旋结构减少和构象转变. 一般情况下, MBT以配位作用为主, TBT以疏水作用为主, DBT两者兼而有之.  相似文献

A nanocylindrical wall structure was obtained by layer‐by‐layer (LbL) assembly of poly‐L ‐arginine (PLA) and human serum albumin (HSA) and characterized by scanning electron microscopy (SEM), scanning force microscopy (SFM), and cryogenic transmission electron microscopy (cryo‐TEM). SEM and SFM measurements of a lyophilized powder of (PLA/HSA)3 nanotubes yielded images of round, chimney‐like architectures with approximately 100 nm wall thickness. Cryo‐TEM images of the hydrated sample revealed that the tube walls are composed of densely packed HSA molecules. Moreover, when small‐angle X‐ray scattering was used to characterize the individual PLA and HSA components in aqueous solutions, maximum diameters of approximately 28 nm and 8 nm were obtained, respectively. These values indicate the minimum thickness of wall layers consisting of PLA and HSA. It can also be concluded from SEM as well as from cryo‐TEM images that the protein cylinders are considerably swollen in the presence of water. Furthermore, HSA retains esterase activity if assembled in nanotubes, as indicated by measurements of para‐nitrophenyl acetate hydrolysis under semi‐physiological conditions (pH 7.4, 22 °C). The enzyme activity parameters (Michaelis constant, Km, and catalytic constant, kcat) were comparable to those of free HSA.  相似文献

In the present work, the adsorption of human serum albumin (HSA) on commercially pure titanium with a titanium oxide layer formed in a H(2)O(2) solution (TiO(2) cp) and on TiO(2) sputtered on Si (TiO(2) sp) was analyzed. Adsorption isotherms, kinetic studies, and work of adhesion determinations were carried out. HSA exchangeability was also evaluated. Surface characterization was performed by atomic force microscopy (AFM), X-ray photoelectron spectroscopy (XPS), and wettability studies. The two TiO(2) surfaces have very distinct roughnesses, the TiO(2) sp having a mean R(a) value 14 times smaller than the one of TiO(2) cp. XPS analysis revealed consistent peaks representative of TiO(2) on sputtered samples as well as on Ti cp substrate after 48 h of H(2)O(2) immersion. Nitrogen was observed as soon as protein was present, while sulfur, present in disulfide bonds in HSA, was observed for concentrations of protein higher than 0.30 mg/mL. The work of adhesion was determined from contact angle measurements. As expected from the surface free energy values, the work of adhesion of HSA solution is higher for the TiO(2) cp substrate, the more hydrophilic one, and lower for the TiO(2) sp substrate, the more hydrophobic one. The work of adhesion between plasma and the substrates assumed even higher values for the TiO(2) cp surface, indicating a greater interaction between the surface and the complex protein solutions. Adsorption studies by radiolabeling of albumin ((125)I-HSA) suggest that rapid HSA adsorption takes place on both surfaces, reaching a maximum value after approximately 60 min of incubation. For the higher HSA concentrations in solution, a multilayer coverage was observed on both substrates. After the adsorption step from single HSA solutions, the exchangeability of adsorbed HSA molecules by HSA in solution was evaluated. The HSA molecules adsorbed on TiO(2) sp seem to be more easily exchanged by HSA itself than those adsorbed on TiO(2) cp after 24 h. In contrast, after 72 h, nearly all the adsorbed albumin molecules effectively exchange with other albumin molecules.  相似文献
