A new type of photoinduced nitric oxide (NO) donors was designed from nitrobenzene derivatives. Visible-light irradiation of 2,6-dimethylnitrobenzenes bearing extended pi-electron systems at the 4-position revealed efficient NO release using ESR analysis and the Griess assay. Computational study and ultraviolet spectrum analysis suggested that the NO-releasing activity was closely related to the conformation of the nitro group, the absorption intensity, and the length of the conjugated pi-electron system. Employing the photodependent cytotoxicity of compound 14 against HCT116 human colon cancer cells, it was demonstrated that 4-substituted-2,6-dimethylnitrobenzene analogues are useful NO donors for the time- and site-controlled NO treatment.  相似文献

An investigation of potential photosensitive protecting groups for diazeniumdiolates (R2N-N(O)=NO-) has been initiated, and here the effect of meta electron-donating groups on the photochemistry of O2-benzyl-substituted diazeniumdiolates (R2N-N(O)=NOCH2Ar) is reported. Photolysis of the parent benzyl derivative (Ar = Ph) results almost exclusively in undesired photochemistry-the formation of nitrosamine and an oxynitrene intermediate with very little, if any, photorelease of the diazeniumdiolate. We have been able to use meta substitution to tune the photochemistry of these benzylic systems. The desired diazeniumdiolate photorelease has been shown to become more substantial with stronger pi-donating meta substituents. This effect has been verified by direct observation of the photoreleased diazeniumdiolate with 1H NMR spectroscopy and by NO quantification measurements conducted in high- and low-pH solutions. In addition, the observed rates of NO release are consistent with that expected for normal thermal decomposition of the diazeniumdiolate in aqueous solutions and also show the same pH dependence.  相似文献

Sterically hindered phenols react with nitric oxide under basic condititons to give either cyclohexadienone diazeniumdiolates or oximates. Phenols with 2,6-di-tert-butyl and 4-methyl (butylated hydroxy toluene, BHT), 4-ethyl, or 4-methoxy methylene substituents yield the corresponding 2,6-di-tert-butyl-2, 5-cyclohexadienone-4-alkyl-4-diazeniumdiolate salts (4-methyl 1a, 4-ethyl 3a, 4-methoxymethylene 5a). Phenols with 2,6-di-tert-butyl and 4-methylene (2,6-di-tert-butylphenol) substituents yield 4-methoxymethylenediazeniumdiolate (5a) together with 2, 6-di-tert-butyl benzoquinone oximate (6a), while phenols with 2, 6-di-tert-butyl and 4-methylenedimethylamino or hydrogen substituents yield exclusively 2,6-di-tert-butyl benzoquinone oximate (6a). Alkylation of the silver salts of 1a, or treating the O(2)-protonated diazeniumdiolate with diazomethane, both yield mixtures of O(1)- and O(2)-methylated isomers. All the compounds exhibit exothermic thermal decomposition except the quinuclidinium (1e, 3e, 5e) and triethylenediammonium (1f) salts which decompose endothermically. Three of the compounds namely "O(2)-protonated" (Z)-1-[4-(2, 6-di-tert-butyl-4-methyl-cyclohexadienonyl)]diazen-1-ium+ ++-1, 2-diolinic acid (1b), O(2)-methyl (Z)-1-[4-(2, 6-di-tert-butyl-4-methyl-cyclohexadienonyl)]diazen-1-ium+ ++-1,2-diolate (1c), and "O(2)-protonated" (Z)-1-[4-(2, 6-di-tert-butyl-4-methoxymethylenecyclohexadienonyl)]diazen- 1-ium-1, 2-diolinic acid (5b) were characterized by single-crystal X-ray diffraction studies. The diazeniumdiolate framework in all the structures is coplanar with considerable pi-bonding delocalized over the O-N-N-O framework.  相似文献

We report a novel NO donor (RpNO), containing a 2,6-dimethylnitrobenzene moiety for photocontrollable NO release and a rhodamine moiety for targeting to mitochondria. Photorelease of NO from RpNO in aqueous solution was confirmed by means of ESR analysis. Cellular release of NO from RpNO was confirmed with the aid of DAF-FM DA, an NO-specific fluorescence probe. RpNO was colocalized with MitoTracker Green FM, a mitochondrial stain, in HCT116 colon cancer cells and exhibited photodependent cytotoxicity. Our results indicate that RpNO is an effective NO donor for time-controlled, mitochondria-specific NO treatment.  相似文献

[reaction: see text] The enediamine tautomer of a variety of substituted amidine free bases reacts with nitric oxide (NO) to produce compounds containing a carbon-bound diazeniumdiolate [R1R2R3C-N(O)=NO-] functional group (previously called "nitrosohydroxylamines"). The new reaction has been shown to be quite general, although the nature of the products does vary. Amidines containing more than one replaceable hydrogen produce polydiazeniumdiolates as intermolecular salts, while those in which only one diazeniumdiolation can occur provide zwitterionic salts. These diazeniumdiolated amidines are shown to be useful NO donor compounds which undergo very slow spontaneous dissociation on dissolution in pH 7.4 phosphate buffer to produce mixtures of NO and nitrous oxide containing mostly NO. The most advantageous manifestation of the new discovery is the preparation of the monodiazeniumdiolated amidine zwitterions. Reaction of the medically relevant alpha-adrenergic agonists tetrahydrozoline and idazoxan produced monodiazeniumdiolated amidine zwitterions from which NO release was observed for up to 28 days and showed little sign of ending. The reaction should be applicable to a variety of pharmaceutical agents, including NO synthase inhibitors, antitumor agents, and antibacterials.  相似文献

We have investigated in this work the kinetics of NO-release from two commercially available and commonly used diazeniumdiolates (so-called NONOates), namely diethylammonium (Z)-1-(N,N-diethylamino)-diazen-1-ium-1,2-diolate (DEA-NONOate) and (Z)-1-[N-(2-aminoepropyl)-N-(2-ammoniopropyl)amino]-diazen-1-ium-1,2-diolate (PAPA-NONOate) by electrochemical measurements. This choice was driven by the distinctive half-life of each compound (2–20 min at 37 °C for DEA- and PAPA-NONOate, respectively) that allow covering a relatively wide range of rate of NO-release. The release of NO from NONOate decomposition was followed using a home-made NO-sensitive electrochemical sensor. The electrochemical features of the starting NONOates and of the by-products of the fully decomposed molecules were also examined. Also, the electrochemical experimental results obtained under aerobic atmosphere were numerically fitted to model the production of NO and its degradation through its oxidation by molecular oxygen. All these information gathered together allows in predicting the real amount of NO released under real aerobic conditions.  相似文献

[reaction: see text] Reactions of nitric oxide with enolates derived from aliphatic methyl ketones containing alpha-methylene or alpha-methine groups and with enolates derived from alpha,alpha'-dimethylene or alpha,alpha'-dimethine ketones yield mono- or bis(diazeniumdiolate) products. Diazeniumdiolation occurs in the following order: alpha-methine > alpha-methylene > alpha-methyl. The amount of the base used alters the extent of diazeniumdiolation and the course of the reaction. Mono- and bis(diazeniumdiolate)-substituted methyl ketones are cleaved in the presence of excess base before and after the subsequent diazeniumdiolation of the alpha-methyl group. Similar to the trihalogenated methyl groups in the base-assisted halogenation reactions of methyl ketones, the bis(diazeniumdiolate)-substituted alpha-methylene and alpha-methyl groups act as leaving groups in the presence of excess base. The reaction of nitric oxide with a (approximately 20:80, cis/trans) mixture of 2,6-cyclohexananone yields the cis and trans isomers of 2,6-dimethylcyclohexanone-2,6-bis(diazeniumdiolate) in 12.9% and 57.6% yield. Single-crystal X-ray diffraction data determined for potassium cis-2,6-dimethylcyclohexanone-2,6-bis(diazeniumdiolate), cis-14b, reveal that the N(2)O(2-) substituent is planar with considerable delocalization of a double bond over the anionic four-atom group. Except for one of the diazeniumdiolate products, namely, potassium propanoate 2,2-bis(diazeniumdiolate), 8b, all are stable in neutral and basic aqueous media. Compound 8b slowly decomposes in neutral aqueous solution releasing nitrous oxide and nitric oxide gases but is stable in basic aqueous media. Differential scanning calorimetry data measured for the diazeniumdiolate products indicate that they decompose exothermally with most of them undergoing explosive decomposition at moderately high temperatures (181-274 degrees C).  相似文献

The photochemistry of O2-naphthylmethyl- and O2-naphthylallyl-substituted diazeniumdiolates has been investigated. Electron-donating methoxy group substitution is shown to have a significant effect on the observed photochemistry, with the appropriate substitution pattern resulting in efficient diazeniumdiolate photorelease. Observed nitric oxide release rates from these photoprecursors are consistent with those expected for normal thermal dissociation of the diazeniumdiolate in aqueous solutions and show the same pH dependence.  相似文献

We have investigated the kinetics of NO escape from Geobacillus stearothermophilus nitric oxide synthase (gsNOS). Previous work indicated that NO release was gated at position 223 in mammalian enzymes; our kinetics experiments include mutants at that position along with measurements on the wild type enzyme. Employing stopped-flow UV-vis methods, reactions were triggered by mixing a reduced enzyme/N-hydroxy-l-arginine complex with an aerated buffer solution. NO release kinetics were obtained for wt NOS and three mutants (H134S, I223V, H134S/I223V). We have confirmed that wt gsNOS has the lowest NO release rate of known NOS enzymes, whether bacterial or mammalian. We also have found that steric clashes at positions 223 and 134 hinder NO escape, as judged by enhanced rates in the single mutants. The empirical rate of NO release from the gsNOS double mutant (H134/I223V) is nearly as rapid as that of the fastest mammalian enzymes, demonstrating that both positions 223 and 134 function as gates for escape of the product diatomic molecule.  相似文献

All major properties of the aqueous hyponitrite radicals (ONNO- and ONNOH), the adducts of nitric oxide (NO) and nitroxyl (3NO- and 1HNO), are revised. In this work, the radicals are produced by oxidation of various hyponitrite species in the 2-14 pH range with the OH, N3, or SO4- radicals. The estimated rate constants with OH are 4 x 10(7), 4.2 x 10(9), and 8.8 x 10(9) M(-1) s(-1) for oxidations of HONNOH, HONNO-, and ONNO2-, respectively. The rate constants for N3 + ONNO2- and SO4- + HONNO- are 1.1 x 10(9) and 6.4 x 10(8) M(-1) s(-1), respectively. The ONNO- radical exhibits a strong characteristic absorption spectrum with maxima at 280 and 420 nm (epsilon280 = 7.6 x 10(3) and epsilon420 = 1.2 x 10(3) M(-1) cm(-1)). This spectrum differs drastically from those reported, suggesting the radical misassignment in prior work. The ONNOH radical is weakly acidic; its pKa of 5.5 is obtained from the spectral changes with pH. Both ONNO- and ONNOH are shown to be over 3 orders of magnitude more stable with respect to elimination of NO than it has been suggested previously. The aqueous thermodynamic properties of ONNO- and ONNOH radicals are derived by means of the gas-phase ab initio calculations, justified estimates for ONNOH hydration, and its pKa. The radicals are found to be both strongly oxidizing, E degrees (ONNO-/ONNO2-) = 0.96 V and E degrees (ONNOH, H+/HONNOH) = 1.75 V, and moderately reducing, E degrees (2NO/ONNO-) = -0.38 V and E degrees (2NO, H+/ONNOH) = -0.06 V, all vs NHE. Collectively, these properties make the hyponitrite radical an important intermediate in the aqueous redox chemistry leading to or originating from nitric oxide.  相似文献

Although many organic/inorganic compounds that release nitric oxide (NO) upon photoirradiation (phototriggered caged-NOs) have been reported, their photoabsorption wavelengths mostly lie in the UV region, because X-NO bonds (X=heteroatom and metal) generally have rather strong π-bond character. Thus, it is intrinsically difficult to generate organic compounds that release NO under visible light irradiation. Herein, the structures and properties of N-pyramidal nitrosamine derivatives of 7-azabicyclo[2.2.1]heptanes that release NO under visible light irradiation are described. Bathochromic shifts of the absorptions of these nitrosamines, attributed to HOMO (n)-LUMO (π*) transitions associated with the nonplanar structure of the N-NO moiety, enable the molecules to absorb visible light, which results in N-NO bond cleavage. Thus, these compounds are innate organic caged-NOs that are uncaged by visible light.  相似文献

The water-soluble manganese(III) meso-tetrakis (N-ethylpyridinium-2-yl) porphyrin (Mn(III)TEPyP) and manganese(III) meso-(tetrakis(4-sulfonato-phenyl)) porphyrinate (Mn(III)TPPS) are able to chemically distinguish between HNO and NO donors, reacting with the former in a fast, efficient, and selective manner with concomitant formation of the {MnNO}(7) complex (k(on(HNO)) approximately equal to 10(5) M(-1) s(-1)), while they are inert or react very slowly with NO donors. DFT calculations and kinetic data suggest that HNO trapping is operative at least in the case of Mn(III)TPPS, while catalytic decomposition of the HNO donors (sodium trioxodinitrate and toluene sulfohydroxamic acid) seems to be the main pathway for Mn(III)TEPyP. In the presence of oxygen, the product Mn(II)TEPyP(NO) oxidizes back to Mn(III)TEPyP, making it possible to process large ratios of nitroxyl donor with small amounts of porphyrin.  相似文献

Diazeniumdiolate ions of structure R(2)N[N(O)NO](-) (1) are of pharmacological interest because they spontaneously generate the natural bioregulatory species, nitric oxide (NO), when dissolved in aqueous media. Here we report the kinetic details for four representative reactivity patterns: (a) straightforward dissociation of the otherwise unfunctionalized diethylamine derivative 2 (anion 1, where R = Et) to diethylamine and NO; (b) results for the zwitterionic piperazin-1-yl analogue 4, for which the protonation state of the neighboring basic amine site is an important determinant of dissociation rate; (c) data for 5, a diazeniumdiolate derived from the polyamine spermine, whose complex rate equation can include terms for a variety of medium effects; and (d) the outcome for triamine 6 (R = CH(2)CH(2)NH(3)(+)), the most stable structure 1 ion identified to date. All of these dissociations are acid-catalyzed, with equilibrium protonation of the substrate preceding release of NO. Specific rate constants and pK(a) values for 2-6 have been determined from pH/rate profiles. Additionally, a hypsochromic shift (from approximately 250 to approximately 230 nm) was observed on acidifying these ions, allowing determination of a separate pK(a) for each substrate. For 6, the pK(a) value obtained kinetically was 2-3 pK(a) units higher than the value obtained from the spectral shift. Comparison of the ultraviolet spectra for 6 at various pH values with those for O- and N-alkylated diazeniumdiolates suggests that protonation at the R(2)N nitrogen initiates dissociation to NO at physiological pH, with a second protonation (at oxygen) accounting for both the spectral change and the enhanced dissociation rate at pH <4. Our results help to explain the previously noted variability in dissociation rate of 5, whose half-life we found to increase by an order of magnitude when its concentration was raised from near-zero to 1 mM, and provide mechanistic insight into the factors that govern dissociation rates among diazeniumdiolates of importance as pharmacologic progenitors of NO.  相似文献

Nitric oxide (NO) is an important cell-signaling molecule whose role in a variety of cellular processes such as differentiation and apoptosis depends strongly on its concentration and flux levels. This work describes and characterizes a novel nitric oxide releasing nanocomposite, capable of photostimulated NO flux that can by dynamically modulated in within a range of biological levels. This material mimics the common compartmentalization strategies used by living cells to achieve its novel features. The material is constructed by encapsulating a photosensitive nitric oxide donor within lipid vesicles with an average diameter of 150 nm. The vesicles are then doped into the interstitial liquid phase of a solid porous silica matrix, which has previously demonstrated biological compatibility and capabilities as a growth surface for mammalian cells. Stimulation by a light source produces a step increase in NO concentration within seconds. The NO flux at the surface of the material is measured to be 14 pmol-cm(-2) sec(-1) using a NO selective self-referencing amperometric microsensor. The NO concentration profile decreases with distance perpendicular to the surface as expected for diffusion from a surface through an aqueous environment. A pattern of one minute light pulses produced uniform pulses of increased NO concentration of one minute duration. A linear relationship exists between NO surface concentration and photon flux, and this relationship can be used to tune the material response.  相似文献

Two new fluorescein-tethered nitrosyls derived from designed tetradentate ligands with carboxamido-N donors have been synthesized and characterized by spectroscopic techniques. These two diamagnetic {Ru-NO}(6) nitrosyls, namely, [(Me(2)bpb)Ru(NO)(FlEt)] (1-FlEt, Me(2)bpb = 1,2-bis(pyridine-2-carboxamido)5-dimethylbenzene, FlEt = fluorescein ethyl ester) and [((OMe)(2)IQ1)Ru(NO)(FlEt)] (2-FlEt, (OMe)(2)IQ1 = 1,2-bis(isoquinoline-1-carboxamido)-4,5-dimethoxybenzene), display NO stretching frequencies (ν(NO)) at 1846 and 1832 cm(-1) in addition to their FlEt carbonyl stretching frequencies (ν(CO)) at 1715 and 1712 cm(-1), respectively. Coordination of the dye ligand enhances the absorptivity and NO photolability of these two nitrosyls in the visible region (450-600 nm) of light. Exposure to visible light promotes rapid loss of NO from both {Ru-NO}(6) nitrosyls to generate Ru(III) photoproducts in dry aprotic solvents, such as MeCN and DMF. The FlEt(-) moiety remains bound to the paramagnetic Ru(III) center in such cases, and hence, the photoproducts exhibit very weak fluorescence from the dye unit. In the presence of water, the Ru(III) photoproducts undergo further aquation and loss of the FlEt(-) moiety via protonation. These steps lead to turn-ON fluorescence (from the free FlEt unit) and provide a visual signal of the NO photorelease from 1-FlEt and 2-FlEt in aqueous media.  相似文献

The release of nitric oxide (NO) from polymers has proven to be highly effective at inhibiting platelet adhesion and thus enhancing the blood compatibility of medical implants. Micropatterning techniques were used to design surfaces that release NO while preserving the underlying substrate for other applications (e.g., sensors). Micropatterned NO-releasing substrates based on aminosilane-containing methyltrimethoxysilane sol-gels were prepared and characterized in terms of stability, NO surface flux, and resistance to in vitro platelet adhesion. We have found that surface-localized NO release from substrates modified with sol-gel micropatterns exhibit enhanced blood compatibility relative to controls.  相似文献

The preparation and characterization of nitric oxide (NO)-releasing dendrimer conjugates are reported. Generation 3 and 5 polypropylenimine dendrimers (DAB-Am-16 and DAB-Am-64) were modified at the exterior to impart different amine functionalities. The ability to store NO on a dendritic scaffold using N-diazeniumdiolate NO donors was examined via the reaction of primary amine, secondary amine, and amide functionalities with high pressures of NO (5 atm). The secondary amine dendrimer conjugates exhibited a high storage capacity for NO (up to 5.6 micromol NO/mg), greatly increasing the "payload" of released NO over existing macromolecular NO donors. The mechanism of diazeniumdiolate decomposition was proton initiated, generating NO spontaneously under physiological conditions (pH 7.4, 37 degrees C). The NO release durations (>16 h) observed for the secondary amine dendrimers were significantly longer compared to small molecule alkyl secondary amine diazeniumdiolates, thus illustrating a dendritic effect on NO release kinetics. The multivalent exterior of dendrimers allows for the future combination of NO donors and other functionalities on a single molecular scaffold, enabling diverse utility as NO storage/delivery systems.  相似文献

The release of NO by [Fe(NO)(Et2NpyS4)], where (Et2NpyS4)2? = 2,6-bis(2-mercaptophenylthiomethyl)-4-substituted pyridine(2-), has been studied in the absence and presence of a trapping agent. The results show that [Fe(NO)(Et2NpyS4)] releases NO spontaneously in solution with a slow rate, k-NO = 1.7 × 10?4 s?1 at 23 °C, in a reversible reaction. NO release becomes faster when the reaction intermediate [Fe(Et2 NpyS4)] was trapped by CO, thereby preventing the back reaction. The release of NO was studied as a function of CO concentration and temperature. The reported activation parameters, especially the positive activation entropy values for the release of NO, favor the operation of a dissociative interchange (Id) mechanism. Thus, [Fe(NO)(Et2NpyS4)] can serve as a NO deliverer.  相似文献
