Cellulase hyperproducers of Trichoderma reesei can be constructed using autopolyploidization and haploidization techniques. To increase the efficiency of this method, the active nuclear shuffling system in a swollen conidium was effective. A dried mature green conidium of a model strain, T. reesei QM6a (IFO 31326), was swollen to make room for a larger autopolyploid nucleus. After colchicine treatment, a larger autopolyploid nucleus was produced in such a swollen conidium. Benomyl treatment of swollen conidia generated multiple smaller nuclei from one larger autopolyploid nucleus. Those smaller nuclei were transported through conidia to mycelia after germination. This system could contribute to increasing the efficiency of genetic shuffling.  相似文献

The cost of cellulase is still a problem for bioethanol production. As the cellulase of Trichoderma reesei is applicable for producing ethanol from cellulosic materials, the cellulase productivity of this fungus should be increased. Therefore, we attempted to develop a system to isolate the strain with higher degrading ability of a filter paper and superior proliferation characteristics among the conidia treated with the mitotic arrester, colchicine. When green mature conidia of T. reesei RUT C-30 were swollen, autopolyploidized, and incubated in the double-layer selection medium containing Avicel, colonies appeared on the surface earlier than the original strain. When such colonies and the original colony were incubated on the Avicel plates, strain B5, one of the colonies derived from the colchicine-treated conidia, showed superior proliferation characteristics. Moreover, when strain B5 and the original strain were compared in the filter paper degrading ability and the cellulose hydrolyzing activity, strain B5 was also superior to the original strain. It was suspected that superior proliferation characteristics of strain B5 reflects higher filter paper degrading ability. Thus, we concluded that the Trichoderma strain with higher degrading ability of a filter paper and superior proliferation characteristics can be isolated using Avicel plates and the double-layer selection medium.  相似文献

When the swollen conidia of Trichoderma reesei QM 6a are treated with 0.1% (w/v) colchicine solution, huge autopolyploid nuclei can be formed in those swollen conidia. When a mycelial mat derived from such a conidum is treated with a haploidizing reagent, benomyl, many fan-shaped sectors are produced from the colony, and cellulase hyperproducers are selected from conidia on the colony. When colchicine and benomyl treatments are repeated on cellulase hyperproducers, new hyperproducers can be constructed successively and systematically. Moreover, when conidia derived from autopolyploids are treated with ethylmethanesulfonate solution, another type of cellulase hyperproducers (polyploids) can be obtained.  相似文献

M14-2 is a cellulase hyperproducer derived from Trichderma recesei QM 6a, but with a growth rate lower than that of the original strain. When M14-2 was autopolyploidized followed by haploidization and selection, the strain with both a higher cellulase productivity per mycelia and a higher growth rate could be obtained as M14-2B. This strain seemed to be constructed using gene sources amplified by additional autopolyploidization.  相似文献

We have attempted to develop an active selection system for strains that have a higher potential for Avicel degradation using haploidized conidia from colchicine-treated Trichoderma reesei Rut C-30 as a model strain. Avicel, absorbent cotton, and wood powder were used as substrates for selection. It appeared that the strains that degrade Avicel actively could be effectively selected when the solid medium containing the selection substrate and the liquid medium containing Avicel were used.  相似文献

The first-stage heterokaryons, obtaining from intergeneric protoplast fusion between Aspergillus niger (Y-b) and Trichoderma viride (M5S51), showed slow growth and mixed morphologies on minimal medium. The fusants were classified into heterokaryon and prototrophic haploid, showing the morphology as that of A. niger. The heterokaryon strains formed conidia with the same nutritional requirements as those of the original auxotrophic mutant strains. After several subcultivations on minimal medium containing d-camphor, some heterokaryon strains formed larger two to seven nuclei/conidium as compared to one nucleus/conidium of the auxotrophic mutant and prototrophic strains, indicating that the new hybrids were generated. Interestingly, three fusant strains AT 11-2-3, AT 11-2-10, and AT 11-2-14 produce 19.2, 6.1, and 10.5 g/l citric acid, respectively, in semisolid culture containing cellulose, whereas A. niger Yang no. 2 could not use carboxymethyl cellulose as the sole carbon source for citric acid production. In addition, the average maximum beta-glucosidase and carboxymethylcellulase productions from AT 11-2-3, AT 11-2-10, and AT 11-2-14 were about 16- and 4-folds higher than those of A. niger, respectively.  相似文献

It was found that PVC films grafted with methacrylic acid do not swell in either water or methanol, two solvents of poly(methacrylic acid), even for high grafting ratios. The swelling of these films was examined in mixtures of methylene chloride with methanol and curves of different shapes were obtained depending on the grafting ratio. PVC films grafted with acrylic acid readily swell in both water and methanol but they remain hard in the swollen state. The equilibrium swelling increases with swelling temperature but this process is not reversible; films swollen at high temperature keep a high degree of swelling even when the system is cooled.  相似文献

The multinucleate conidia were produced from the green mature conidia of Trichoderma reesei Rut C-30 strain by colchicine treatment. The strain with higher Filter paper degrading ability was selected among those conidia using a double layer selection medium. The selected strain, JS-2 was able to collapse the filter paper within 15 min but the original strain took 25 min to collapse it completely. Moreover, the amount of reducing sugar in the L-type glass tube of the strain, JS-2 was greater than that of the original strain. The Avicel, CMC-Na, and Salicin hydrolyzing activity of the strain, JS-2, increased 2.1 times, 1.2 times, and 3.6 times higher than that of the original strain.  相似文献

Factors in a cultivation medium affecting fungal growth morphology and funmaric acid production by Rhizopus oryzae ATCC 20344 were investigated. These factors included the initial pH value and trace metals such as zinc, magnesium, iron, and manganese in the cultivation medium. It was found that a significant change in the growth morphology of R. oryzae ATCC 20344 occurs when the initial pH value is varied. A lower initial pH value in the cultivation medium was inhibitory to fungal growth, and fast growth in the cultivation medium at a higher initial pH value promoted, the formation of large pellets or filamentous forms. Trace metals in the cultivation media also had significant effects on pellet formation and fumaric acid fermentation.  相似文献

Nanocomposite hydrogels of poly(2-acrylamido-2-methyl-1-propanesulfonic acid) containing graphene were prepared by radical polymerization. Their swelling properties in response to ionic strength and electrical stimuli were assessed. Graphene was obtained through an easy and convenient method lately developed by our research group, which consists in the exfoliation of graphite by sonicating it in a proper solvent medium. It was found that the graphene content influences the swelling properties of hydrogels; in particular, those containing graphene swell more than the filler-free ones; graphene content influences also the swelling ratio variation between the swollen and deswollen states.  相似文献

A new and effective process has been developed for fabrication of novel cage-like multihollow polymer particles by using sulfonated polystyrene (SP) particles as the templates, with heptane as the phase separation agent, in an ethanol/water medium. The ratio of water/ethanol and the heating temperature play important roles in the formation of these multihollow particles. It was found that the cage-like polymer particles could be obtained when the ratio of ethanol/water is 5:5 (w/w), with a temperature above 50?°C. After a detailed study, the formation mechanism was proposed based on an SP swollen (ethanol and heptane penetrating process) and phase separation process. This new method for fabricating the cage-like multihollow polymer particles has a great meaning not only on confirming the formation mechanism, but also on providing an effective way to prepare the special hollow core/porous shell polymer particles, which could have wide range of potential applications, such as catalysts, sensors, and drug release.  相似文献

Escherichia coli NCIM 2569 was evaluated for its potential for amidase production under submerged fermentation. Among the various amide compounds screened, maximum substrate specificity and enzyme yield (8.1 U/mL) were obtained by using 1% acetamide. Fermentation was carried out at 30°C in shake-flask culture under optimized process conditions. A maximum of 0.52 U/mL of intracellular amidase activity was also obtained from cells incubated for 24 h. Studies were also performed to elucidate the optimal conditions (gel concentration, initial biomass, curing period of beads, and calcium ion concentration in the production medium) for immobilization of whole cells. By using E. coli cells entrapped in alginate, a maximum of 6.2 U/mL of enzyme activity was obtained after 12 h of incubation under optimized conditions. Using the immobilized cells, three repeated batches were carried out successfully, and 85% of the initial enzyme activity was retained in the second and third batches. The study indicated that the immobilized E. coli cells offered certain advantages such as less time for maximum enzyme production, more stability in the enzyme production rate, and repeated use of the biocatalyst.  相似文献

The morphologically undifferentiated cells of nonregenerant callous tissue of Cereus peruvianus cultured in the original medium and in medium supplemented with tyrosine were used as an alkaloid source. Comparison of alkaloid production by C. peruvianus plants and by callous tissues indicated that alkaloid levels were almost twice as high in callous tissues as in shoots of C. peruvianus plants. The ratio of alkaloid concentration between mature plant and morphologically und ifferentiated cells of callous tissue was 1∶1.7. A relationship between culture medium containing tyrosine and alkaloid production was also observed in the callous tissues of C. peruvianus. Since increased alkaloid production may be induced by additional factors such as tyrosine, increasing levels of tyrosine or other conditions of the culture medium may be considered factors for inducing higher alkaloid production by C. peruvianus callous tissues.  相似文献

Abstract— It was found that killing of Neurospora crassa conidia by singlet molecular oxygen generated by a photosensitized reaction in the presence of toluidine blue O, accompanied inhibition of respiration and uptake of the photosensitizing dye by conidia. Both the inactivation of respiration and the uptake of toluidine blue O were stimulated by deuterium oxide and suppressed by azide. Mitochondria of irradiated conidia were swollen and showed matrix of low density under an electron microscope.
These results suggest that singlet molecular oxygen induces the inhibition of respiration and the loss of membrane integrity of Neurospora conidia.  相似文献

Cellulases are a complex group of enzymes that are fundamental for the degradation of amorphous and crystalline cellulose in lignocellulosic material. Unfortunately, cellulases have a low catalytic efficiency on their substrates when compared to similar enzymes such as amylases, which has led to a strong interest in improving their activities. Thermobifida fusca secretes six cellulose degrading enzymes: two exo- and three endocellulases and an endo/exocellulase Cel9A (formerly called E4). Cel9A shows unique properties because of its endo- and exocellulase characteristics, strong activity on crystalline cellulose, and good synergistic properties. Therefore, it is an excellent target for mutagenesis techniques to improve crystalline cellulose degradation. In this article, we describe research conducted to improve Cel9A catalytic efficiency using a rational design and computer modeling. A computer model of Cel9A was created using the program CHARMM plus its PDB structure and a cellohexose molecule attached to the catalytic site as a starting model. Initially molecular graphics and energy minimization were used to extend the cellulose chain to 18 glucose residues spanning the catalytic domain and cellulose-binding domain (CBD). The interaction between this cellulose chain and conserved CBD residues was determined in the model, and mutations likely to improve the binding properties of the CBD were selected. Site-directed mutations were carried out using the pET vector pET26b, Escherichia coli DH5-α, and the QuickChange mutagenesis method. E. coli BL21-DE3 was used for protein production and expression. The purified proteins were assayed for enzymatic activity on filter paper, swollen cellulose, bacterial microcrystalline cellulose, and carboxymethylcellulose (CMC). Mutation of the conserved residue F476 to Y476 gave a 40% improved activity in assays with soluble and amorphous cellulose such as CMC and swollen cellulose.  相似文献

A poly(vinyl alcohol) hydrogel was prepared by coupling poly(vinyl alcohol) with epichlorohydrin as the cross-linking agent. The structure of the hydrogel was characterized by FTIR and GPC techniques. Various amounts of water were added into the dry gel to swell it, and the quantity of water in various states in the partially swollen hydrogel was determined by DSC technique. The analytical results indicate that the water introduced into the dry gel first combines with the hydrophilic groups of the network chains through hydrogen bond forming non-freezable water. The weight ratio of the non-freezable water to dry gel in the hydrogels is about 0.20. After the non-freezable water is saturated, the additional water penetrates the network space and exists simultaneously both in the freezable and free water states until reaching equilibrium swelling. A method for the determination of selected amino acids in culture medium using HPLC with fluorescence detection is described. Twenty hours after intra‐cytoplasmic sperm injection, one randomly selected zygote was transferred to the culture medium. After incubation (72 h after fertilization), the culture medium in which the embryo was incubated and blank medium was immediately stored at −80°C. Filtered medium samples were derivatized with ortho ‐phthalaldehyde (naphthalene‐2,3‐dicarboxaldehyde), forming highly fluorescent amino acids derivatives. Reverse‐phase columns (LichroCART, Purospher STAR RP18e or Ascentis Express C18) were used for the separation. The derivatives were analyzed by gradient elution with a mobile phase containing ethanol and sodium dihydrogen phosphate. The analytical performance of this method is satisfactory for all amino acids; the intra‐assay coefficients of variation were <10% and quantitative recoveries were between 95.5 and 104.4%. Changes in the levels of selected amino acids before and after human embryo cultivation were observed. After embryo incubation, the levels of all amino acids in the medium were increased, apart from aspartate and asparagine. After the cultivation of some embryos, amino acids which were not part of the medium were detected. Low amino acids turnover was observed in some embryos.  相似文献

Polyacrylate gels such as: N-isopropylacrylamide (NIPA), ionic copolymer NIPA-co-sodium acrylate (NIPA-co-SA) and ionic copolymer NIPA-co-[2-(acryloyloxy)ethyl]trimethylammonium chloride (NIPA-co-XCl) were swollen by methanol, ethanol and N,N-dimethylformamide. These solvents were oxidized voltammetrically in the gels at platinum microelectrodes. The blocking coefficients, defined as the ratio of the currents observed in the presence and absence of the polymeric chains, were different for the solvents in the charged and uncharged polymers and did not differ for the electroactive probe, ferrocene, present in the solvents at millimolar level. This may mean that the micro ionic layer that is formed at microelectrodes at the plateau of the voltammetric solvent waves leads to a kind of collapse/phase transition of the polymer in the depletion layer. For methanol, the gel structure was found to prevent occasionally the formation of large gas bubbles at the potentials positive versus the first wave and then three one-electron waves could be obtained. Experiments done with a capillary cell and an optical microscope indicated that before formation of large bubbles at the Pt microelectrode a very dynamic, dense stream of microbubbles leaves the electrode surface.Dedicated to Zbigniew Galus on the occasion of his 70th birthday  相似文献

Inclusion complex between the essential oil of Mentha x villosa Hudson and β-cyclodextrin, with a 1:9 mass/mass oil–β-cyclodextrin ratio was prepared by co-precipitation and kneading methods in a hydroethanolic medium. The GC/MS analysis showed a total volatile content of 99.5% in the Mentha x villosa oil. The characterization of the complex involved the analysis of the original essential oil, the surface and the total extracted oils. Among 28 detected compounds in the original essential oil, 13 are monoterpenes and 10 sesquiterpenes, furthermore, piperitenone-oxide is the major component (35.4%). 12 compounds were totally and 11 partially complexed, 3 have been adsorbed only on the surface of the β-CD and 2 have not been detected neither in the surface oil nor in the complexed oil. A 13.6% encapsulation efficiency was observed, while the total oil and volatiles retention was 15 and 77%, respectively. Non-parametric statistic analysis of the data showed that the profile of the volatiles were not significantly different comparing the original oil and the complexed oil (p>0.04). The results of thermogravimetry-mass spectrometry and XRD analysis have proven the inclusion complex formation between the essential oil and cyclodextrin.  相似文献

TH‐11, a bacterial strain with strong depolymerase activity that breaks down aliphatic esters such as poly(3‐hydroxybutyrate) (PHB) and poly(ethylene succinate) (PES) was isolated from a soil sample collected from the sediment of Tou‐Chain River, Taiwan, R.O.C. It was phenotypically and genetically characterized to be a Streptomyces strain. The degradation of PHB and PES were tested both using emulsified polymers in solid agar and thin polymer films in liquid culture media. The degradations were measured by clear‐zone formation on solid agar plates, or direct weight measurements and electromicroscope inspection of the incubated polymer films in the liquid culture. The depolymerase activities can be detected in the cell‐free preparation of the culture medium, and can be enhanced by gelatin.  相似文献
