The SL3 beta-Lactam Test is a 3 min, receptor-based lateral flow Rapid One Step Assay (ROSA) that detects 5 of 6 beta-lactam drugs approved for dairy cattle in the United States. The method was evaluated through the AOAC Research Institute Performance-Tested Method program following a U.S. Food and Drug Administration protocol. Three combined lots detected penicillin G 4.2 parts per billion (ppb), ampicillin 8.7 ppb, amoxicillin 7.8 ppb, cephapirin 16.0 ppb, and ceftiofur (total metabolites) 51 ppb at least 90% of the time, with 95% confidence as determined by dose response probit analysis. These detection levels are less than safe level/tolerances but not more than 50% less. Lot repeatability was within 20%. Incurred residues were detected comparably or more sensitively to fortified samples due to the cumulative effect of biologically active metabolites. There were no interferences from somatic cells at 1 M/mL, bacterial cells 500 000 colony-forming units/mL, or 30 other non-beta-lactam drugs. These performances met approval conditions of the National Conference on Interstate Milk Shipments. Ruggedness conditions were incorporated into public health procedures for annual laboratory proficiency and certification.  相似文献

The biochemicals utilized in the Charm MRL beta-Lactam test (8 min test) were applied to faster flowing lateral components to create a new 3 min, one-step beta-lactam test called Charm MRL-3 (Charm Sciences Inc., Lawrence, MA). This new test was validated at T&V-ILVO according to Commission Decision 2002/657/EC. The following analytical parameters were checked: test specificity, detection capability, and test robustness (impact of deviation of the test protocol, and impact of the milk composition, batch differences of reagents). Further, the suitability of the Charm MRL-3 to screen heat-treated milk or milk from animal species other than the cow was also tested. Finally, the test was integrated in the monitoring of dairy samples to check the occurrence of false-negative or false-positive results, and the test was also included in a national ring trial and an international proficiency study. The results proved that the Charm MRL-3 is a fast, simple, and reliable cows' milk test that can be used at the farm level in order to prevent tanker milk contamination, or at the entrance of the dairy plant to screen tanker milk for the presence of beta-lactam antibiotics.  相似文献

Delvotest SP NT DA is designed to test milk for the presence of antibacterial substances, such as antibiotics. The test is made of an agar gel containing bacterial spores and a color indicator. The milk sample is added onto the agar gel, and the test is incubated at 64 degrees C. The principle of the test is based on the diffusion of possible inhibitory substances that may be present in the milk sample into agar. This reduces growth and acid production by the test organism, and delays or prevents the agar from changing color from purple to yellow. The Delvotest Accelerator is an automated system in which the plates containing the milk to be analyzed are placed for incubation. The Accelerator automatically detects the end of the incubation and reads the results. A sample containing antibiotic will be noted as "positive." A sample without antibiotics or with antibiotics at concentrations below detection level will be noted as "negative." The present report includes all technical details about the Delvotest SP NT DA, and the results of the validation study. The validation study demonstrates that the Delvotest SP NT DA conforms to the product performance claims and confirms the robustness of the test. The Delvotest SP NT DA is, therefore, granted Performance Tested Method certification.  相似文献

A validation study of VitaKit A for quantitation of total vitamin A in 2% fluid milk was carried out according to the guidelines provided by AOAC INTERNATIONAL. The VitaKit A was compared, in terms of repeatability and accuracy, with the U.S. Food and Drug Administration-Interstate Milk Shippers HPLC reference method for determination of total vitamin A in fluid milk with 2% fat. The data obtained by the VitaKit A method are in excellent agreement with the data obtained by the HPLC reference method. Further, a low LOD (0.33 international unit/mL) was obtained for the VitaKit A method; the presence of interferents, like cholesterol and vitamin D3, in the milk had minor influence on the quantitation of total vitamin A by the VitaKit A method. The VitaKit A test kit was found to be stable for 1 year from the date of manufacture when stored at 2-8 degrees C. The method requires 2 h processing time, compared to 1-2 days for the HPLC reference method. The results of this validation study clearly demonstrate that the VitaKit A method is reliable, rapid, and accurate for the quantitation of total vitamin A in fluid milk containing 2% milk fat. An independent study by Q Laboratories Inc., Cincinnati, OH, under the validation guidelines of AOAC INTERNATIONAL, confirmed these findings.  相似文献

Reveal Salmonella 2.0 is an improved version of the original Reveal Salmonella lateral flow immunoassay and is applicable to the detection of Salmonella enterica serogroups A-E in a variety of food and environmental samples. A Performance Tested Method validation study was conducted to compare performance of the Reveal 2.0 method with that of the U.S. Department of Agriculture-Food Safety and Inspection Service or U.S. Food and Drug Administration/Bacteriological Analytical Manual reference culture methods for detection of Salmonella spp. in chicken carcass rinse, raw ground turkey, raw ground beef, hot dogs, raw shrimp, a ready-to-eat meal product, dry pet food, ice cream, spinach, cantaloupe, peanut butter, stainless steel surface, and sprout irrigation water. In a total of 17 trials performed internally and four trials performed in an independent laboratory, there were no statistically significant differences in performance of the Reveal 2.0 and reference culture procedures as determined by Chi-square analysis, with the exception of one trial with stainless steel surface and one trial with sprout irrigation water where there were significantly more positive results by the Reveal 2.0 method. Considering all data generated in testing food samples using enrichment procedures specifically designed for the Reveal method, overall sensitivity of the Reveal method relative to the reference culture methods was 99%. In testing environmental samples, sensitivity of the Reveal method relative to the reference culture method was 164%. For select foods, use of the Reveal test in conjunction with reference method enrichment resulted in overall sensitivity of 92%. There were no unconfirmed positive results on uninoculated control samples in any trials for specificity of 100%. In inclusivity testing, 102 different Salmonella serovars belonging to serogroups A-E were tested and 99 were consistently positive in the Reveal test. In exclusivity testing of 33 strains of non-salmonellae representing 14 genera, 32 were negative when tested with Reveal following nonselective enrichment, and the remaining strain was found to be substantially inhibited by the enrichment media used with the Reveal method. Results of ruggedness testing showed that the Reveal test produces accurate results even with substantial deviation in sample volume or device development time.  相似文献

A validation study designed to meet the requirements of the AOAC Research Institute and the U.S. Food and Drug Administration, Center for Veterinary Medicine (FDA/CVM) was conducted for a receptor and antibody-based, immunochromatographic method (BetaStar Plus) for detection of beta-lactam antibiotic residues in raw, commingled bovine milk. The assay was found to detect amoxicillin, ampicillin, ceftiofur, cephapirin, cloxacillin, and penicillin G at levels below the FDA tolerance/safe levels, but above the maximum sensitivity thresholds established by the National Conference on Interstate Milk Shipments (NCIMS). Results of the part I (internal) and part II (independent laboratory) dose-response studies employing spiked samples were in close agreement. The test was able to detect all six drugs at the approximate 90/95% sensitivity levels when presented as incurred residues in milk collected from cows that had been treated with the specific drug. Selectivity of the assay was 100%, as no false-positive results were obtained in testing of 1031 control milk samples. Results of ruggedness experiments established the operating parameter tolerances for the BetaStar Plus assay. Results of cross-reactivity testing established that the assay detects certain other beta-lactam drugs (dicloxacillin and ticarcillin), but it does not cross-react with any of 30 drugs belonging to other classes. Abnormally high bacterial or somatic cell counts in raw milk produced no interference with the ability of the test to detect beta-lactams at tolerance/safe levels.  相似文献

Fourier transform infrared (FTIR) spectroscopy with attenuated total reflectance (ATR) sampling was evaluated for use in screening for adulteration in raw materials used in the formulation and manufacture of dietary supplements. ATR requires minimal-to-no sample preparation and the method runs in less than ten minutes, providing a robust, rapid screening test for a variety of possible adulterants in the raw materials of dietary supplements. Spectral comparison methods targeting structural similarities of known adulterants were developed. In this study, FTIR-ATR was used to detect the presence of known adulterants intentionally spiked into dietary ingredients, including erectile dysfunction drugs, steroids, weight loss drugs and Melamine.  相似文献

Performance Tested Method multiple laboratory validations for the detection of peanut protein in 4 different food matrixes were conducted under the auspices of the AOAC Research Institute. In this blind study, 3 commercially available ELISA test kits were validated: Neogen Veratox for Peanut, R-Biopharm RIDASCREEN FAST Peanut, and Tepnel BioKits for Peanut Assay. The food matrixes used were breakfast cereal, cookies, ice cream, and milk chocolate spiked at 0 and 5 ppm peanut. Analyses of the samples were conducted by laboratories representing industry and international and U.S governmental agencies. All 3 commercial test kits successfully identified spiked and peanut-free samples. The validation study required 60 analyses on test samples at the target level 5 microg peanut/g food and 60 analyses at a peanut-free level, which was designed to ensure that the lower 95% confidence limit for the sensitivity and specificity would not be <90%. The probability that a test sample contains an allergen given a prevalence rate of 5% and a positive test result using a single test kit analysis with 95% sensitivity and 95% specificity, which was demonstrated for these test kits, would be 50%. When 2 test kits are run simultaneously on all samples, the probability becomes 95%. It is therefore recommended that all field samples be analyzed with at least 2 of the validated kits.  相似文献

In this study, lead in raw cow's milk has been determined by validated electrothermal atomic absorption spectrometry (ETAAS) with Zeeman-effect background correction. Maximum pyrolysis and optimum atomization temperatures of lead were determined in the presence of modifiers. Pd + Mg(NO3)2 has been found a powerful modifier mixture for the determination of lead. The analytical parameters of the method such as limit of detection, limit of quantification and the effect of interfering ions have been investigated. The detection limit (3sigma) achieved by the method was calculated to be 0.62 ng/mL for Pb. Repeatability of the method evaluated as the relative standard deviation of 16-17 replicates using 5 ng/mL, under optimum experimental conditions were about 1.5% for synthetic sample solution and about 3% for real sample (N = 5). The described method has been validated by analyzing certified reference material (BCR-CRM 150) and by comparing the results with those obtained by inductively coupled plasma mass spectrometry (ICP-MS). The validated method was applied to raw cow's milk samples produced in 7 different regions of Turkey in 2003-2004. Raw cow's milk contained a mean (range) of 31.4 (2.5 - 313) microg kg(-1) lead with a relative error below 2%.  相似文献

Increasingly, more food companies are relying on molecular methods, such as PCR, for pathogen detection due to their improved simplicity, sensitivity, and rapid time to results. This report describes the validation of a new Real-Time PCR method to detect Listeria monocytogenes in nine different food matrixes. The complete system consists of the MicroSEQ L. monocytogenes Detection Kit, sample preparation, the Applied Biosystems 7500 Fast Real-Time PCR instrument, and RapidFinder Express software. Two sample preparation methods were validated: the PrepSEQ Nucleic Acid extraction kit and the PrepSEQ Rapid Spin sample preparation kit. The test method was compared to the ISO 11290-1 reference method using an unpaired-study design to detect L. monocytogenes in roast beef, cured bacon, lox (smoked salmon), lettuce, whole cow's milk, dry infant formula, ice cream, salad dressing, and mayonnaise. The MicroSEQ L. monocytogenes Detection Kit and the ISO 11290-1 reference method showed equivalent detection based on Chi-square analysis for all food matrixes when the samples were prepared using either of the two sample preparation methods. An independent validation confirmed these findings on smoked salmon and whole cow's milk. The MicroSEQ kit detected all 50 L. monocytogenes strains tested, and none of the 30 nontargeted bacteria strains.  相似文献

An interlaboratory study of 21 public health, state agriculture, and industry laboratories in the United States tested raw commingled bovine milk containing aflatoxin M1 using the Charm Rapid One Step Assay (ROSA) Safe Level Aflatoxin M1 Quantitative lateral flow method. Blind coded sample pairs were fortified with 0, 300, 350, 400, 450, 500, and 550 parts per trillion (ppt) aflatoxin M1. A ROSA reader quantitatively interpreted test strips with ppt readings. Readings < or = 400 ppt were interpreted as negative, and readings >400 ppt were interpreted as positive. Initial positive samples were subsequently assayed 2 additional times. If both retest results were >400 ppt, the sample was called positive/ actionable relative to U.S. and Codex levels, 500 ppt. The concentration of 400 ppt was chosen for the positive/negative interpretation to provide 90% sensitivity with 95% confidence at the 500 ppt legislative level. The combined false negative rate was <5% (4 of 83) for samples at 500 and 550 ppt. The false violatives at 0, 300, 350, 400, and 450 ppt (n = 42 at each level) were 0, 0, 21, 14, and 93%, respectively. The 90% positive concentration with 95% confidence was 503 ppt by probit analysis. The average intralaboratory repeatability was 11% and average interlaboratory reproducibility was 13% for the fortified sample pairs. High-performance liquid chromatography analysis of the study samples by 5 laboratories showed 38% false negatives with the 500 and 550 ppt samples, and a 0% false-violative rate with samples less than the 500 ppt action level.  相似文献

The BAX System PCR assay for Salmonella detection in foods was previously validated as AOAC Research Institute (RI) Performance Tested Method (PTM) 100201. New studies were conducted on beef and produce using the same media and protocol currently approved for the BAX System PCR assay for E. coli O157:H7 multiplex (MP). Additionally, soy protein isolate was tested for matrix extension using the U.S. Food and Drug Administration-Bacteriological Analytical Manual (FDA-BAM) enrichment protocols. The studies compared the BAX System method to the U.S. Department of Agriculture culture method for detecting Salmonella in beef and the FDA-BAM culture method for detecting Salmonella in produce and soy protein isolate. Method comparison studies on low-level inoculates showed that the BAX System assay for Salmonella performed as well as or better than the reference method for detecting Salmonella in beef and produce in 8-24 h enrichment when the BAX System E. coli O157:H7 MP media was used, and soy protein isolate in 20 h enrichment with lactose broth followed by 3 h regrowth in brain heart infusion broth. An inclusivity panel of 104 Salmonella strains with diverse serotypes was tested by the BAX System using the proprietary BAX System media and returned all positive results. Ruggedness factors involved in the enrichment phase were also evaluated by testing outside the specified parameters, and none of the factors examined affected the performance of the assay.  相似文献

氟白普洛芬(FB)为一具有光敏感特性的非类固醇抗发炎药物(NSAID s)。当将氟白普洛芬的甲醇溶液以低压汞灯照射,可观察到氟白普洛芬有3种光解产物的产生。建立了一种灵敏度高、准确的、稳定的高效液相色谱分析法(HPLC),并藉此观察氟白普洛芬的光解反应。日内、日间实验的相对标准偏差(RSD)值皆可低于1.1%和1.7%,回收率在98.2%~102.0%之间。结果显示,HPLC分析方法具有良好的选择性和专一性。氟白普洛芬的光解反应在不同醇性溶剂中并无显著的差异。  相似文献

This project is undertaken to fully optimize the U.S. Environmental Protection Agency Method 531.1 post-column chemistries and to incorporate recent advances in liquid chromatographic separation, post-column derivatization, and detection techniques. Sample preservation and storage stability studies establish citric acid as a suitable replacement for the caustic monochloroacetic acid in the current method and confirm its antimicrobial effectiveness. Performance of an alternate set of commercially available post-column reagents is also investigated. This research has resulted in the publication of Method 531.2, a high-performance liquid chromatographic direct injection method for the analysis of N-methylcarbamoyloximes and N-methylcarbamates using post-column derivatization and fluorescence detection.  相似文献

Two common extraction solvent systems, namely acidified aqueous methanol and acidified aqueous acetone, were used to extract blackberry phenolics, and the antioxidant properties of the recovered extracts were compared. The crude extracts were fractionated into low- and high-molecular-weight phenolics by Sephadex LH-20 column chromatography. The hydrophilic-oxygen radical absorbance capacity (H-ORACFL), ferric reducing antioxidant power (FRAP), and the cellular antioxidant activity (CAA) assays were employed as indices to assess antioxidant capacity of the extracts and their respective fractions. The methanolic solvent system displayed a greater efficiency at extracting anthocyanin and flavonol constituents from the blackberries, while the acetonic solvent system was better at extracting flavan-3-ols and tannins. Anthocyanins were the dominant phenolic class found in the blackberries with 138.7 ± 9.8 mg C3G eq./100 g f.w. when using methanol as the extractant and 114.6 ± 3.4 mg C3G eq./100 g f.w. when using acetone. In terms of overall antioxidant capacity of blackberry phenolics, the acetonic solvent system was superior. Though present only as a small percentage of the total phenolics in each crude extract, the flavan-3-ols (42.37 ± 2.44 and 51.44 ± 3.15 mg/100 g f.w. in MLF and ALF, respectively) and ellagitannins (5.15 ± 0.78 and 9.31 ± 0.63 mg/100 g f.w. in MHF and AHF, respectively) appear to account for the differences in the observed antioxidant activity between the two solvent systems.  相似文献

This paper describes the validation of an isocratic HPLC method for the assay of voriconazole in tablets. The method employs a Merck LiChrospher? 100 RP-8 (125 × 4.6 mm I.D., 5 μm particle size) column, with a mobile phase of methanol : triethylamine solutions 0.6 %, pH 6.0 (50:50, v/v) and UV detection at 255 nm. A linear response (r > 0.9999) was observed in the range of 20.0–100.0 μg mL−1. The method showed good recoveries (average 100.4%) and the relative standard deviation intra and inter-day were ≤ 1.0 %. Validation parameters as specificity and robustness were also determined. The method can be used for both quality control assay of voriconazole in tablets and for stability studies as the method separates voriconazole from its degradation products and tablet excipients.  相似文献
