The aim of the study was to investigate the whole-body protein turnover, either before or after continuous, moderate ethanol-induced oxidative stress by red wine consumption over a relatively short period in healthy volunteers. Ten healthy adults received an individual regular diet over 20 days. After 10 days, the subjects consumed 0.4 ml ethanol kg?1 day?1 as red wine together with dinner over a 10-day period. After 8 and 18 days, respectively, a 15N-labelled yeast protein was administered in a dosage of 4.2 mg kg?1 body weight. Urine and faeces were collected over 48 h, respectively. The 15N-enrichment was measured by isotope ratio mass spectrometry, whereas the protein flux rates were calculated by a three-compartment model. The whole-body protein turnover without/with red wine consumption amounted to 3.73±0.6 and 3.49±0.6 g kg?1 day?1 (not significant), respectively. Moderate alcohol consumption does not induce significant short-term changes in the whole-body protein turnover of healthy adults.  相似文献

Fourteen adults received [13C]methacetin and [methyl-13C]methionine together with a standardised dinner. Expired air samples were taken over 6 h. After a wash-out period, the subjects consumed 0.4 ml ethanol/kg/day together with dinner over a 10-day period. Thereafter, 13C-tracer administration was repeated under identical conditions. The 13CO2-enrichments were measured by isotope ratio mass spectrometry. The mean cumulative percentage 13C-dose recovery (CPDR) after administration of [13C]methacetin and [methyl-13C]methionine either without or with red wine consumption amounted to 38.2+/-6.3 vs. 36.3+/-6.7% (p=0.363) and 9.5+/-3.3 vs. 8.8+/-2.5% (p=0.47), respectively. Moderate alcohol consumption does not induce significant short-term changes of the microsomal and the mitochondrial functions of the human liver in healthy subjects.  相似文献

The WPTR and the reutilisation rates (R) were investigated in 20 patients divided in three groups of different degrees of proteinuria (groups I-III: < 1, 1-3, > 3 g/m2/day, respectively). [15N]glycine was administered as a single oral pulse. Urine samples were taken over 2 days. After removing urinary proteins by trichloracetic acid, 15N-enrichment in the supernatant was measured by isotope ratio mass spectrometry. A three-compartment model was used to calculate WPTR and R, which showed a statistically significant difference between groups I and III (2.64 vs. 4.63 g/kg/day, and 70.4% vs. 80.8%, respectively, P < 0.01), whereas the net protein gain remained unchanged (0.13 vs. 0.22 g/kg/day). The higher the protein loss the higher the WPTR and the corresponding R. The severe protein loss provokes increased WPTR and R as well.  相似文献

During the 29th Soviet Antarctic Expedition in Novolazarevskaya from March 1984 to March 1985, the protein and energy metabolisms were studied in six expeditioners from the German Democratic Republic. The investigations were carried out at the beginning of the expedition (May), during the polar night (July) and during the polar day (December). The effect of a special stress situation (sledge trek in April 1984) was investigated in one subject. The stable nitrogen isotope (15)N was used to study the protein metabolism. The assessment of the energy metabolism was based on the oxygen consumption, which was determined by means of a spirograph. In addition, the vital capacity, the breath minute volume, the blood pressure, etc. were measured. The following results were obtained: During the polar night, the utilisation of the dietary proteins and the whole body protein synthesis calculated by means of the (15)N excretion of the total nitrogen in urine were greater (73.6±0.9 % and 3.48±0.17?g protein d(-1)?kg(-1), n=3) than the respective values during the polar day (69.7±1.2, p<0.05, n=3 and 3.05±0.07, p<0.05, n=3) and at the beginning of the expedition (69.6±1.4, p<0.02, n=5 and 2.81±0.09, p<0.01, n=5). The lowest values (58.0 % and 2.43?g protein d(-1)?kg(-1)) were obtained in the subject after the trek. The resting metabolic rate (in kJ?d(-1)?m(-2)) was decreased during the polar night (45.6±5.0, n=4) in comparison with the polar day (61.5±11.3, n=3) and the beginning of the expedition (52.3±9.6, n=4) with p<0.01 in both cases.  相似文献

The purpose of this study was to investigate the feasibility of a noninvasive approach that combines magnetic resonance imaging (MRI) oximetry and flow measurement to obtain the oxygen consumption in the myocardium and in the whole body. Thirteen healthy male volunteers [mean (+/-S.D.) age: 35+/-7 years] underwent this MR study, which included myocardial oxygen consumption (MVO(2)) measurements in 11 subjects and whole-body oxygen consumption (VO(2)) measurements in 8 subjects. In six subjects, both measurements were obtained. Five subjects had repeated MRI measurements of global MVO(2) in order to verify the reproducibility of this approach. The protocol included in vitro blood sample T(2)-%O(2) calibration, coronary sinus (CS) and main pulmonary artery (MPA) T(2) and phase contrast flow measurement and left ventricular (LV) mass calculation. Based on Fick's law, a global measurement of LV MVO(2) and whole-body VO(2) using MRI was feasible. The MVO(2) values were 11+/-3 ml/min per 100 g LV mass. For repeated measurements, differences in MVO(2) of 1 ml/min per 100 g LV mass appear detectable. The whole-body VO(2) values were 3.8+/-0.8 ml/min/kg body weight. MRI techniques that combine CS and MPA T(2), flow and LV mass measurements to quantify MVO(2) and whole-body VO(2) noninvasively in healthy subjects appear feasible, based on their correspondence to previously published work.  相似文献

A sensitive rapid analytical method was established and validated to determine the bakkenolide A (BA) in rat plasma. This method was further applied to assess the pharmacokinetics of BA in rats receiving a single dose of BA. Liquid chromatography tandem mass spectrometry in multiple reaction monitoring mode was used in the method, and costundide was used as internal standard. A simple protein precipitation based on methanol was employed. The combination of a simple sample cleanup and short chromatographic running time (2.4?min) increased the throughput of the method substantially. The method was validated over the range of 1-1000?ng/mL with a correlation coefficient?>?0.99. The lower limit of quantification was 1?ng/mL for BA in plasma. Intra- and inter-day accuracies for BA were 93-112% and 103-104%, respectively, and the inter-day precision was less than 15%. After a single oral dose of 20?mg/kg of BA, the mean peak plasma concentration (C(max) ) of BA was 234.7?±?161?ng/mL at 0.25?h. The area under the plasma concentration-time curve (AUC(0-24 h) ) was 535.8?±?223.7?h·ng/mL, and the elimination half-life (T(1/2) ) was 5.0?±?0.36?h. In case of intravenous administration of BA at a dosage of 2?mg/kg, the area under the plasma concentration-time curve (AUC(0-24 h) ) was 342?±?98 h?ng/mL, and the elimination half-life (T(1/2) ) was 5.8?±?0.7?h. The aim of the study was, to evaluate the metabolic effect of HAY's diet on protein turnover, fat oxidation, respiratory quotient, body fat and weight loss. Twelve healthy adults received an individually regular diet and thereafter a corresponding isocaloric and isonitrogenous 10-day HAY-diet. Protein turnover and 13C-fat oxidation were investigated after administration of [15N]glycine and an [U-13C]algae lipid mixture. The 15N and 13C enrichment in urine and breath were measured by isotope ratio mass spectrometry. The respiratory quotient was measured by indirect calorimetry. Body fat, total body water and lean body mass were estimated by bio-electric impedance analysis. HAY's diet led to a significantly higher 13C-fat oxidation (15.4 vs. 22.0% P < 0.01), corresponding to a lower respiratory quotient (0.88 vs. 0.81; P < 0.01), whereas the protein turnover remained constant in both diets (3.06 vs. 3.05 g/kg/day). HAY's diet did not reduce total body water, lean body mass, body fat and body weight (72.2 vs. 71.4 kg).  相似文献

Our research group of the Children's Hospital of the University of Rostock (Rostock group) has long-time experience in (15)N-labelling and in using yeast protein and its hydrolysates for tracer kinetic studies to evaluate parameters of the whole-body protein metabolism in premature infants. The particular advantage of applying an economically convenient, highly (15)N-enriched, and completely labelled yeast protein for evaluating protein turnover rates is the fact that the (15)N dose is spread among all proteinogenic amino acids. The absorption has been improved by hydrolysing [(15)N]yeast protein with thermitase into a mixture of amino acids, dipeptides and tripeptides so that faecal analysis becomes unnecessary when determining turnover rates. The review shows that, in contrast to the application of single (15)N-labelled amino acids with resulting overestimation of protein turnover rates, the (15)N-labelled yeast protein thermitase hydrolysate represents the amino acid metabolism more closely without causing amino acid imbalances. The (15)N-labelled yeast protein thermitase hydrolysate leads to the estimation of reliable protein turnover rates, particularly in premature infants.  相似文献

Abstract For verifying catabolic states in insulin-dependent patients and dogs the method estimating urea production rates with (13)C and with doubly (15)N labeled urea, respectively, has been established. For a fast steady state of urea tracer dilution, a prime of 600 times the continuous infusion rate had to be injected. Urea was isolated from plasma samples by protein precipitation and cation exchange chromatography with a consecutive derivatization of the dried urea fraction (trimethylsilyl derivatives). The masses of the fragment ions m/z 189 ((14)N(14)N), 190 ((14)N(15)N) and 191 ((15)N(15)N) urea are monitored to estimate the [(15)N(2)]urea frequency in the overall body urea pool in mol percent excess (MPE). 1 to 15 ng of derivatized urea were measured efficiently. An excellent correlation between expected standard and measured MPE (r = 0.9977) was achieved from solutions containing 1 to 7% [(15)N(2)]urea. The interassay coefficient of variation amounted to < 10% for a [(15)N(2)]urea portion of ≥ 3%. Normoglycemic diabetic patients who were treated with insulin overnight showed significantly higher urea production compared to healthy controls (9.22 ± 2.07 vs. 5.4 ± 0.32 μmol·kg(-1) · min(-1); p < 0.05). Measurements in chronic diabetic dogs proved an increased rate of amino acid catabolism (+ 20% urea production) in systemic versus portal application of insulin in paired studies. This increased nitrogen load in diabetics may accelerate progression of diabetic nephropathy. - Thus, the established stable isotope technique may serve as a sensitive and useful indicator of amino acid catabolism in clinical and experimental research.  相似文献

The ultraprecise Gravity Probe B experiment measured the frame-dragging effect and geodetic precession on four quartz gyros. We use this result to test WEP II (weak equivalence principle II) which includes rotation in the universal free-fall motion. The free-fall E?tv?s parameter η for a rotating body is ≤10(-11) with a four-order improvement over previous results. The anomalous torque per unit angular momentum parameter λ is constrained to (-0.05±3.67)×10(-15) s(-1), (0.24±0.98)×10(-15) s(-1), and (0±3.6)×10(-13) s(-1), respectively, in the directions of geodetic effect, frame-dragging effect, and angular momentum axis; the dimensionless frequency-dependence parameter κ is constrained to (1.75±4.96)×10(-17), (1.80±1.34)×10(-17), and (0±3)×10(-14), respectively.  相似文献

Abstract A seven compartment model was applied for evaluation of oral L-[1-(13)C]leucine loading tests (38 μmol/kg body wt.) in healthy volunteers. The model comprises transport and absorption in stomach and gut into a central L-leucine-compartment which is connected to a protein compartment and to the compartment of the corresponding 2-oxo acid. CO(2) release from the latter occurs in a fast and a slow compartment into the central CO(2) compartment for exhalation. Using the fmins routine of MATLAB for parameter estimation, a good agreement was obtained between calculated and actually measured kinetics of (13)C-labelled metabolites and a mean in vivo L-leucine oxidation of 0.365 ± 0.071 μmol/kg per min (n = 5) was computed. Plausibility of the model was checked by predicting in vivo leucine oxidation rates from primed continuous infusion tests (priming: L-[1-(13)C]leucine, 5 μmol/kg; NaH(13)CO(2), 1.2 μmol/kg; infusion: L-[1-(13)C]leucine, 5 μmol/kg per h). In 5 tested volunteers, the experimental L-leucine oxidation rate amounted to 0.358 ± 0.105 μmol/kg per min versus predicted 0.324±0.099 μmol/kg per min. Possible causes for some observed intraindividual variations are discussed.  相似文献

Natural and anthropogenic radioactivity of sand and water samples collected from the four big rivers of Punjab province, Pakistan, was measured using a high-purity germanium detector coupled with a high resolution multichannel analyser. The average concentration of the naturally occurring radionuclides (226)Ra, (232)Th and (40)K in all the sand samples from the rivers Jhelum, Chenab, Ravi and Indus was found to be 22±0.6, 36±1 and 287±10 Bq kg (-1), respectively, while the concentration of the anthropogenic radionuclide (137)Cs was found to be below the minimum detectable activity, i.e. ~1.2 Bq?kg (-1). All the sand samples have Ra(eq) concentrations lower than the limit of 370 Bq kg (-1). Indoor (H (in)) and outdoor (H (out)) radiation hazard indices were calculated for the samples to assess the radiation hazards arising due to the use of this sand in construction, and were found to be less than unity in the study area. Calculated values of the absorbed dose rate were less than the typical world average value of 59 nGy h (-1), and the annual effective dose rate was also less than the typical world value of 70 μSv, except in the Indus river, in which it is slightly higher then the world average. Results show that the measured values are comparable with other global radioactivity measurements. None of the studied riverbeds was considered a radiological hazard, and their sand can be safely used in construction.  相似文献

自行搭建的拉曼光谱点扫描系统,以柠檬酸钠还原法配制的SC银溶胶为表面增强剂,建立了桂花酒中山梨酸钾的定量预测模型,模型校正集决定系数(R2C)和均方根误差(RMSEC)分别为0.978 9和0.070 3 g·kg-1,验证集决定系数(R2P)和均方根误差(RMSEP)分别为0.934和0.165 7 g·kg-1。桂花酒中山梨酸钾的定量预测模型为主光谱模型,结合K/S算法,探讨了基于DS算法和PDS算法将桂花酒主光谱模型向杨梅酒的修正传递方法。结果显示,用K/S算法选取4个杨梅酒样品,基于DS算法传递桂花酒主光谱模型验证结果RP和RMSEP值分别为0.906 1和0.215 0 g·kg-1。K/S算法选取3个杨梅酒样品(窗口宽度为5),基于PDS算法传递桂花酒主光谱模型验证结果RP和RMSEP值分别为0.905 5和0.225 0 g·kg-1。DS算法和PDS算法均可以用少量样品将桂花酒中山梨酸钾的主光谱预测模型有效传递给杨梅酒,实现了一种被测物预测模型在同类物种间的传递,具有重要实用意义。  相似文献

The analysis of gamma-emitting radionuclides in nature, i.e. (226)Ra, (232)Th, (40)K and (137)Cs, has been carried out in soil samples collected from Peshawar University Campus and surrounding areas using a high purity germanium detector coupled with a computer-based high-resolution multichannel analyser. The activity concentrations in soil ranged from 30.20±0.65 to 61.90±0.95, 50.10±0.54 to 102.80±1.04, 373.60±4.56 to 1082±11.38 and 9.50±0.11 to 46.60±0.42?Bq?kg(-1) for (226)Ra, (232)Th, (40)K and (137)Cs, with a mean value of 45±7.70, 67±12.50, 878±180 and 19±9.20?Bq?kg(-1), respectively. The radium equivalent activity, internal and external hazard indices have mean values of 203.40±29.40?Bq?kg(-1), 0.56 and 0.68, respectively. The mean values of outdoor and indoor absorbed dose rates in air and the annual effective dose equivalents were found to be 106.50 and 128?nGy?h(-1) and 0.19 and 0.54?mSv?y(-1), respectively. In the present study, (40)K was the major radionuclide present in soil samples. The presence of (137)Cs indicates that this area also received some fallout from the nuclear accident of the Chernobyl power plant in 1986. The activity concentrations of radionuclides found in soil samples during the current investigation were nominal. Therefore, they are not associated with any potential source of health hazard to the public.  相似文献

We observed, for the first time, solar neutrinos in the 1.0-1.5 MeV energy range. We determined the rate of pep solar neutrino interactions in Borexino to be 3.1±0.6{stat}±0.3{syst} counts/(day·100 ton). Assuming the pep neutrino flux predicted by the standard solar model, we obtained a constraint on the CNO solar neutrino interaction rate of <7.9 counts/(day·100 ton) (95% C.L.). The absence of the solar neutrino signal is disfavored at 99.97% C.L., while the absence of the pep signal is disfavored at 98% C.L. The necessary sensitivity was achieved by adopting data analysis techniques for the rejection of cosmogenic {11}C, the dominant background in the 1-2 MeV region. Assuming the Mikheyev-Smirnov-Wolfenstein large mixing angle solution to solar neutrino oscillations, these values correspond to solar neutrino fluxes of (1.6±0.3)×10{8} cm{-2}?s^{-1} and <7.7×10{8} cm{-2}?s{-1} (95% C.L.), respectively, in agreement with both the high and low metallicity standard solar models. These results represent the first direct evidence of the pep neutrino signal and the strongest constraint of the CNO solar neutrino flux to date.  相似文献

In Rechna Doab, samples of the most common vegetation, perennial grass Desmostachya bipinnata (dab), were collected along with soil samples from 29 sites. Natural radioactivity of 22?Ra, 232Th/22?Ac and ??K was measured by using high purity germanium-based gamma ray spectrometer. Activity concentration levels of 22?Ra, 232Th/22?Ac and ??K in soil were found to be 46.8 ± 6.2 (36.0-57.6), 61.4 ± 5.9 (48.2-73.2) and 644.8 ± 73.9 (537.7-868.4) Bq kg?1 (dry mass), respectively, and those in vegetation were 2.74 ± 1.70 (1.00-6.39), 2.24 ± 0.59 (1.56-2.61) and 172.72±113.37 (53.14-469.24) Bq kg?1 (dry mass), respectively. The measured values of the activity concentration in vegetation are comparable with some other international data. Calculated soil to vegetation transfer factors of 22?Ra, 22?Ac and ??K were 0.06 ± 0.03 (0.02-0.14), 0.04 ± 0.01 (0.03-0.04) and 0.26 ± 0.16 (0.09-0.69). The mean outdoor absorbed dose rate in air for the area under study was determined as 8.22 nGy h?1 and the mean indoor absorbed dose rate in air was 11.52 nGy h?1. The total annual effective dose to the general public from the vegetation was found to be (0.02-0.16) mSv, which is below the recommended limit value of 1 mSv y?1 for the general public. The dab vegetation under study was found to be radiologically safe for the population and environment.  相似文献
