Plant extracts are one of the best possible sources of bioactive molecules, and are being used globally as an antioxidants and natural antimicrobial compounds. In current study, Schefflera vinosa leaves extract was prepared through Soxhlet extraction procedure using methanol and chloroform as solvents. The extract was investigated for total antioxidant, phenolic and flavonoid contents, free radical scavenging and antimicrobial activities. The free radical scavenging activities were evaluated through 2,2- diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis-3-ethylbenzotiazolin-6-sulfonic acid (ABTS) and Ferric-reducing/ antioxidant power (FRAP) assay. The antimicrobial activity of extract was determined through poisoned food method. The methanolic extract has exhibited high antioxidant, phenolic, and flavonoid activities compared to chloroform extract. Similarly, free radical scavenging activities (ABTS, DPPH and FRAP) were higher in methanolic extract. Further, Fourier-Transform Infrared Spectroscopy (FTIR) used to determine the functional group and Gas chromatography-mass spectrometry (GC–MS) to elucidate volatile composition of the crude extract. Different functional group like N-H, O-H, C-O, C-N, C-H, C=O, C≡C and C-O-H presence indicate the existence of many metabolites in the extracts. GC–MS study identified 61 compounds and subsequently, these molecules were screened virtually using DockThor. Furthermore, antimicrobial study was confirmed against rice pathogens like Magnaporthe oryzae (M. oryzae) and Xanthomonas oryzae pv. oryzae (Xoo). Molecular docking study further suggested that phytomolecules (3-Isopropoxy-1,1,1,7,7,7-hexamethyl-3,5,5-tris (trimethylsiloxy) tetrasiloxane, and 2-Methoxy-5-methylthiophene) targets Histone Deacetylase (HDAC) of M. oryzae and Peptide Deformylase (PDF) of Xoo, which could inhibit their growth. Hence, this study indicated that Schefflera vinosa extracts could be an important ingredient as an antioxidant as well as antimicrobial agent against rice pathogens.  相似文献

The influence of both acidic and basic hydrolysis on the yield, total phenolic content and antioxidative capacity of methanolic extract of germinated brown rice (GBR) was studied. Total phenolic content (TPC), total flavonoid content (TFC), 2,2'-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS) radical cation scavenging, and ferric reducing antioxidant power (FRAP) tests were used for the measurement of antioxidant ability. There was a significant difference p < 0.05) in the TPC and DPPH radical scavenging assay results when comparing neutral with acidic and basic catalysed hydrolysis. The yield of the crude extract was slightly higher in acidic hydrolysis than in basic hydrolysis p > 0.05). The TPC and TFC were highest in acidic hydrolysis. A significant correlation was observed between ABTS radical cation scavenging and FRAP. The antioxidant activity measured using DPPH radical scavenging assay showed high activity in acidic hydrolysis, while the ABTS radical cationscavenging activity and FRAP showed the highest values in basic hydrolysis. The samples were further evaluated using HPLC to determine the individual phenolic concentrations in different hydrolytic media contributing to the antioxidant effects. This study revealed that acidic and basic hydrolysis can improve the yield, phenolic content, and antioxidant activity of germinated brown rice.  相似文献

In this study, composition of essential oil and antioxidant capacity of Centaurea drabifolia subsp. detonsa were investigated. The antioxidant capacity of the methanolic extract was evaluated by various methods including measuring the total phenolic content, total antioxidant capacity, free radical scavenging activity (DPPH assay), β-carotene/linoleic acid bleaching assay and ferric and cupric ion reducing power assay. The composition of essential oil was identified by using gas chromatography-mass spectrometry. Totally, 41 compounds were described in the essential oil. Germacrene D (44.829%) was determined as the major compound of the essential oil. The total phenolic content, total antioxidant capacity, inhibition rate of oxidation of linoleic acid, IC(50) (in DPPH assay) and EC(50) (in reducing power) value were found to be 40.454?mg?GAE/g, 100.840?mg?AAE/g, 65.639%, 39.584?μg?mL(-1) and 0.603?mg?mL(-1), respectively. The results indicated that the extract of C. drabifolia subsp. detonsa has strong antioxidant properties and this species can be used as a natural antioxidant in food processing and pharmaceutical industries.  相似文献

In an aim to prove the efficiency of polyphenols of Rosa canina fruits in promoting human health. A methanolic extract of R. canina fruits was prepared by successive maceration with solvents of increasing polarity. The polyphenol composition was analyzed by HPLC–DAD–ESI–MS. The biological activity of this extract on SH-SY5Y cells and HepG2 cells was then studied. The antioxidant activity was tested by various in vitro tests such as DPPH-radical-scavenging activity, FRAP assay, hydroxyl radical scavenging assay and total antioxidant capacity. The subacute toxicity of R. canina was tested on female rats by repeated intraperitoneal administration of various doses. The phenolic profiles showed 25 antioxidants distributed into three classes of phenolic compounds: glycosylated and agglomerated flavonoids/isoflavonoids, tannins and phenanthrenes. Qualitative phytochemical analyses showed that this extract lacks alkaloids. The methanolic extract of R. canina fruits has a total antioxidant capacity of 82.69 ± 1.18 μg EAA/mg of methanol extract and the IC50 of the methods used is in the following increasing order: FRAP assay (61.88 μg/ml), then hydroxyl radical scavenging assay (67.45 μg/ml) and then DPPH radical-scavenging activity (129.81 μg/ml). The extract of R. canina did not cause any phenotypic signs of toxicity or mortality during and after treatment. The LD50 was >5,000 mg/kg, hence, R. canina was considered nontoxic. An in vivo study proved the protective effect of R. canina against cardiac and hepato-renal toxicities. These results drew the importance of a healthy diet, where diets rich in R. canina fruits can be used as a rich natural source of antioxidants and anticarcinogenic phenolic compounds.  相似文献

The extracts of seven Citrus rootstock seeds have been compared regarding fatty acid profile and antioxidant potential. Sour orange (Citrus aurantium L.) was found to contain the highest oil amount (34%), while the Poncirus trifoliata cultivars contained the highest percentage of unsaturated fatty acids (84-87%). In addition, the antioxidant properties of the extracts from defatted seeds have been evaluated by measuring their radical scavenging activity against 2,2'-diphenyl-1-picrylhydrazyl. The highest antioxidant activities were observed in the case of the acetone extract of sour orange and Citrumelo Swingle (76% and 75%, respectively), at a concentration of 0.17?mg?mL(-1). Moreover, the total phenolic content of the extracts, determined using the Folin-Ciocalteau reagent, was found to be correlated with the radical scavenging activity results. The acetone extracts of sour orange and Citrumelo Swingle exhibited the highest phenolic content [112.3 and 103.4?mg gallic acid equivalent?g(-1) dry sample weight, respectively].  相似文献

Miconia is one of the largest genus of the Melastomataceae, with approximately 1,000 species. Studies aiming to describe the diverse biological activities of the Miconia species have shown promising results, such as analgesic, antimicrobial and trypanocidal properties. M. albicans leaves were dried, powdered and extracted to afford chloroformic and methanolic extracts. Total phenolic contents in the methanolic extract were determined according to modified Folin-Ciocalteu method. The antioxidant activity was measured using AAPH and DPPH radical assays. Chemical analysis was performed with the n-butanol fraction of the methanolic extract and the chloroformic extract, using different chromatographic techniques (CC, HPLC). The structural elucidation of compounds was performed using 500 MHz NMR and HPLC methods. The methanolic extract showed a high level of total phenolic contents; the results with antioxidant assays showed that the methanolic extract, the n-butanolic fraction and the isolated flavonoids from M. albicans had a significant scavenging capacity against AAPH and DPPH. Quercetin, quercetin-3-O-glucoside, rutin, 3-(E)-p-coumaroyl-α-amyrin was isolated from the n-butanolic fraction and α-amyrin, epi-betulinic acid, ursolic acid, epi-ursolic acid from the chloroformic extract. The results presented in this study demonstrate that M. albicans is a promising species in the search for biologically active compounds.  相似文献

Lagoecia cuminoides L. belongs to the family of Umbelliferae (Apiaceae), and known also as common wild cumin. The aerial parts of L. cuminoides were collected at the flowering stage and dried, then the methanolic extract was analyzed for polyphenol compounds identified by HPLC-DAD and antioxidant activity (DPPH(2,2-diphenyl-1-picrylhydrazyl) radical scavenging assay). It was found that the predominant phenolic constituents were chlorogenic acid, hesperidin, rosmarinic acid, hesperetin and vanillin. The antioxidant activity of methanolic extract from L. cuminoides was found 1597 μg/mL in DPPH scavenging assay. There is no strict positive relationship between the polyphenolic content and antioxidant activity of extracts.  相似文献

Lavandula stricta belongs to the Lamiaceae family and is considered as an endemic medicinal plant in southern Iran. Essential oil composition, total phenolic content and antioxidant activity from two different populations of L. stricta were studied for the first time. A GC and GC/MS analysis of essential oil isolated from the aerial part of L. stricta identified 31 constituents; the major constituents were α-pinene (58.34–63.52%), linalool (8.85–9.36%), 3-methyl butyl 2-methyl butanoate (7.45–7.70%), sabinene (2.84–3.56%), limonene (2.87–3.21%) and myrcene (2.25%). The total phenolic content of methanolic extracts was determined with the Folin–Ciocalteu reagent and the antioxidant activity of methanolic extract and essential oil were determined with the 2, 2-diphenyl-1-picryl hydrazyl free radical scavenging assay, respectively. Total phenols varied from 61.05 to 64.45 mg GAE/g dry weight, and IC50 values in the radical scavenging assay ranged from 334.11 to 395.23 μg/mL in methanolic extracts and 420–475 μg/mL in essential oil.  相似文献

The total phenolic content, antioxidant and antifungal activities of three Inula crithmoides extracts (n-hexane, methylene chloride and MeOH) were investigated. The methanolic extract showed the highest total phenolic content. In the DPPH assay, the methanolic and hexane extracts exhibited the highest DPPH-radical scavenging activity; in the 5-lipoxygenase assay, the hexane extract showed greater inhibitory effect with an IC50 similar to that of Trolox and ascorbic acid. The antifungal activity of the methanolic extract revealed a higher activity against Phytophtora cryptogea and Alternaria solani.  相似文献

In this study, the antioxidant potency of sequential organic and aqueous leaf extracts of Cassia occidentalis was investigated, employing various established in vitro systems such as nitric oxide scavenging (NOS) activity, β-carotene-linoleic acid model system, hydroxyl radical scavenging (HRS) activity, reducing power, metal chelating activity (MCA) and superoxide radical scavenging (SRS) activity. The aqueous extract of the leaves of C. occidentalis was found to be most effective against free radicals, followed by the methanolic, chloroform, petroleum ether and benzene extracts, respectively. This study was conducted to investigate the chemical and nutritional composition of Artemisia annua leaves in addition to determination of antioxidant potential of their extracts prepared in different solvents. Chemical composition was determined by quantifying fat, protein, carbohydrate, fiber, tocopherol, phytate, and tannin contents. Extraction of A. annua leaves, for antioxidant potential evaluation, was carried out using five solvents of different polarities, i.e., hexane, chloroform, ethyl acetate, methanol and water. Antioxidant potential was evaluated by estimating total phenolic (TPC), flavonoid (TFC) contents, ferric reducing antioxidant power (FRAP), Trolox equivalent antioxidant capacity (TEAC), DPPH radical scavenging activity and lipid peroxidation. Efficiency of different solvents was compared for the yield of antioxidant extracts from leaf samples and a clear variation was observed. The highest TPC, TFC, TEAC, DPPH radical scavenging and lowest lipid peroxidation were observed in MeOH extracts, whereas aqueous extract exhibited high ferric reducing antioxidant power; suggesting MeOH to be the most favorable extractant.  相似文献

The present study was designed to evaluate polarity-dependent extraction efficiency and pharmacological profiling of Polygonum glabrum Willd. Crude extracts of leaves, roots, stems, and seeds, prepared from solvents of varying polarities, were subjected to phytochemical, antioxidant, antibacterial, antifungal, antidiabetic, and cytotoxicity assays. Maximum extraction yield (20.0% w/w) was observed in the case of an acetone:methanol (AC:M) root extract. Distilled water:methanol (W:M) leaves extract showed maximum phenolic contents. Maximum flavonoid content and free radical scavenging potential were found in methanolic (M) seed extract. HPLC-DAD quantification displayed the manifestation of substantial quantities of quercetin, rutin, gallic acid, quercetin, catechin, and kaempferol in various extracts. The highest ascorbic acid equivalent total antioxidant capacity and reducing power potential was found in distilled water roots and W:M leaf extracts, respectively. Chloroform (C) seeds extract produced a maximum zone of inhibition against Salmonella typhimurium. Promising protein kinase inhibition and antifungal activity against Mucor sp. were demonstrated by C leaf extract. AC:M leaves extract exhibited significant cytotoxic capability against brine shrimp larvae and α-amylase inhibition. Present results suggest that the nature of pharmacological responses depends upon the polarity of extraction solvents and parts of the plant used. P. glabrum can be considered as a potential candidate for the isolation of bioactive compounds with profound therapeutic importance.  相似文献

An activity-guided isolation and purification process was used to identify the DPPH (l,l-diphenyl-2-picrylhydrazyl) radical-scavenging components of Swertia chirayita. A dry, whole plant of S. chirayita was extracted with different solvents and tested for its DPPH radical-scavenging activity. The acetone?:?water (8?:?2) extract showed the highest total phenolic content (TPC) and DPPH radical-scavenging activity, which was column chromatographed to obtain decussatin, swertianin, bellidifolin, isobellidifolin, amarogentin, swertianolin and mangiferin as active components. Good correlation was observed between TPC and DPPH scavenging activity among the extracts. The unique structure of xanthones, including the catecholic moiety and the completely conjugated system, enables them to be promising antioxidants.  相似文献

The antioxidant properties of extracts of Sybaris liquorice roots have been assessed using 2,2'-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity assay. The extracts, obtained by Soxhlet extraction (Et(2)O, AcOEt, MeOH and BuOH) of the yellow (inner part) and brown (cortex) root powders ensuing from decortication of the raw dry roots, followed by separation and powderisation, were analysed for their scavenging activity by evaluating the colourimetric decrease in the absorbance of DPPH. The highest antioxidant activity (98.39?±?0.56%) was observed in the case of the Et(2)O extract of the brown powder, at a concentration of 3.33?mg?mL(-1). Moreover, the total phenolic content of the extracts was determined using the Folin-Ciocalteu reagent and expressed as milligram gallic acid equivalent per gram of dry extract. Our results show that the Et(2)O extract of the liquorice root cortex could be used as an attractive natural source of antioxidant additives for food, cosmetic or pharmaceutical applications.  相似文献

Abstract

Rhaponticum acaule (L.) DC. is a medicinal plant commonly used for the treatment of some illnesses such as gastrointestinal infections. In this work, we report the composition of different parts of this plant on phenolic compounds, their quantification, and antioxidant activity. The obtained results reported that methanolic extracts of the three parts studied revealed high phenolic contents. For flavonoid contents, the highest contents were reported in organic extracts of leaf part. In addition, results obtained from the study of the antioxidant activity showed that methanolic extract of root presented the highest activity, in DPPH? scavenging ability test with an IC50 of 0.31?±?0.04?mg/mL and in FRAP test with an EC50 of 1.06?±?0.02?mg/mL. The RP-HPLC-PDA analysis revealed the presence of five phenolic acids (sinapic, caffeic, chlorogenic, ferulic and syringic acids), one flavanone (naringenin), one flavonol (rutin) and vanillin.  相似文献

The aim of this study was to adopt pyrogallol red bleaching test for LC-DAD capable of characterizing chemical constituents and peroxynitrite (ONOO) scavenging activity in parallel for a large number of plant extracts. The hypothesis is that upon reaction with ONOO, the peak areas of compounds in complex mixtures with potential radical scavenging activity in the LC chromatograms will be significantly reduced or disappeared. We validated this approach with a model mixture of 17 phenolic compounds, which mimics a general alcoholic extract of Salvia species. The LC separation conditions were optimized for baseline separation of phenolic reference compounds and for selective detection of the test substrate pyrogallol red. The results indicated that the depletion kinetic of compounds in the model mixture correlates moderately with the individual scavenging activities. Moreover, catechol group dependent relationship between the structure–scavenging activity relationships confirmed the LC based assay parameters. The final demonstration of the assay was by methanolic extract of Salvia miltiorrhiza Bunge, which showed outstanding ONOO scavenging activity. 13 min total analysis time per sample and excellent resolution allow the method to be applied for chemical fingerprinting coupled with rapid screening for natural antioxidants derived from alcoholic extracts of Salvia species.

This work involves a comprehensive chemical composition analysis of leaf and cone samples of Lithuanian hop varieties. This study aimed to determine the chemometric properties of the leaves and cones of five Lithuanian hop varieties. Determined properties were the following: (a) xanthohumol content, (b) phenolic compounds, (c) flavonoids, (d) radical scavenging activity, and (e) the qualitative composition of volatile compounds. The total content of phenolic compounds in aqueous 75% methanolic extracts varied between 31.4–78.2 mg of rutin equivalents (RE)/g, and the concentration of flavonoids was between 11.0–23.3 mg RE/g. Radical scavenging activity varied between 34.4–87.2 mg RE/g. A QUENCHER analysis procedure showed 91.7–168.5 mg RE/g of the total phenolic compound content, 12.7–21.4 mg RE/g of flavonoids, and 48.4–121.0 mg RE/g of radical scavenging activity. ‘Fredos taurieji’ and ‘Fredos derlingieji’ varieties have shown maximum values of phenolic compounds and radical scavenging activity both in leaf and cone suspensions. These varieties accumulated a higher amount of xanthohumol in leaves. The concentration of xanthohumol in the samples varied between 0.0014–0.2136% of dry mass, with the highest concentration in the cones of ‘Kauno gražieji’. We identified 19 volatile compounds in leaves, and in cones, we identified 32. In both of them, α-humulene and β caryophyllene dominated. ‘Raudoniai’ leaves were exceptional in their aroma due to dominating compound nagina ketone (Kovats index 1306). The QUENCHER procedure has shown a great potential for the unextractable residue of hop raw material. Further investigation and valorization of different hop biomass components, not only cones, are essential.  相似文献

The main task of this work was to create a rapid, simple and less material and time consuming method involving capillary electrophoretic separation in order to separate analytes and evaluate antioxidant activity within a single analytical run. Several interfaces were developed and used to couple CE to the reaction detector. The method developed enables simultaneous electrophoretic separation and evaluation of antioxidant activity of each separated compound in the mixture. The analysis was performed using DPPH (2,2-diphenyl-1-picrylhydrazyl) as synthetic radical reagent. The on-line capillary electrophoresis-reaction (antioxidant activity) detection method can be used for a rapid evaluation of individual antioxidants in complex mixtures, particularly extracts of natural products. Possibility to evaluate radical scavenging activity of extracts of natural products components is demonstrated on an example of aqueous propolis extract. Four phenolic acids where separated and their radical scavenging activity was on-line evaluated.  相似文献

The antioxidant activities of the water and ethanol extracts of F. koreana flowers were evaluated using DPPH, superoxide anion (?O(-2)), and nitric oxide (?NO) radical scavenging activity assays. The SC(50) values of the water extract of F. koreana on DPPH, ?O(-2) and ?NO were 48.39, 24.36 and 100.21 μg mL(-1), respectively. The SC(50) values of the ethanol extract of F. koreana on the aforesaid free radicals were 57.50, 49.00 and 146.08 μg mL(-1), respectively. Further, the total phenolic contents of both extracts were determined and expressed as milligram gallic acid equivalent (GAE) per gram of extract. The water extract exhibited a higher phenolic content (113.78 mg GAE?g(-1)), while the ethanol extract showed 94.53?mg GAE?g(-1). Our findings demonstrate that the water and ethanol extracts of F. koreana flowers might be potential natural sources of antioxidative additives for use in the food and other allied industries.  相似文献

This study investigates the in vitro antioxidant and antimicrobial activities of eight extracts obtained from the dried barks of Commiphora berryi and Commiphora caudata (Burseraceae). The radical scavenging activity was assessed by 1,1-diphenyl-2-picryl hydrazyl (DPPH) and nitric oxide assays. The methanolic extracts of C. berryi and C. caudata showed significant DPPH radical scavenging activity, with IC?? values of 26.92 and 21.16?μg?mL?1, respectively, and low radical scavenging activity against the nitric oxide assay. The antimicrobial activity of the plants was tested against the Gram-positive and Gram-negative bacteria. The ethyl acetate, chloroform and petroleum ether extracts of C. berryi showed good antibacterial activity against Pseudomonas aeruginosa, with a minimum inhibitory concentration (MIC) of 0.26?mg?mL?1, whereas the ethyl acetate and methanol extracts of C. caudata showed moderate antimicrobial activity with an MIC of more than 2.0?mg?mL?1 against P. aeruginosa compared to the petroleum ether and chloroform extracts, which showed an MIC of 1.1?mg?mL?1. The methanolic extracts of C. berryi and C. caudata also showed moderate cytotoxic activity against a human mammary carcinoma cell line (MCF-7), with values IC?? of 82.6 and 88.4?μg?mL?1, respectively.  相似文献
