For the comprehensive metabolite profiling of human plasma, sample preparation is a crucial step. In this investigation, we have compared 10 different extraction techniques for metabolite profiling by GC–MS. Six one-dimensional (1D) and four two-dimensional (2D) extraction techniques involving solvent precipitation, molecular weight cut off tube (MWCOT) and solid phase extraction (SPE) by using silica, RP C18, cation and anion were investigated. Pooled samples of 50 Healthy Male Plasma (HMP), 50 Healthy Female Plasma (HFP) and 100 Healthy Pakistani Plasma (HPP) were subjected to these extraction methods for comparison purposes. Metabolites obtained were identified through NIST mass spectral (Wiley registry), METLIN and Fiehn RTL libraries. XCMS Software was used for the detection of metabolic features, retention time correction, alignment, annotation and statistical analysis in each method. 116–34 peaks were detected by various methods and approx 33% of the peaks were characterized in each method. Hierarchical clustering of the 10 extraction methods showed a low similarity index (50.1%) which indicated different chemical nature of metabolites, resulting from different methods. Venn diagram highlights the GC–MS peaks (33–77%) common in various methods. Metabolites which were different in male and female groups were detected using a threshold value of p ≤ 0.0001, q ≤ 0.001 and fold change ≥3 by employing Welch's t-test and identified through METLIN. Results indicated that 2D-C18 and 2D-silica offers a comprehensive metabolite profile in term of reproducibility, number of peaks and difference in metabolite pattern of male and female.  相似文献

Boron is an important element in nuclear technology. A comparative study was carried out for the determination of boron in borosilicate glass, boron carbide and graphite samples by wet-chemical and nuclear analytical methods. Wet chemical methods namely titrimetry, Inductively Coupled Plasma Mass Spectrometry and ICP Optical Emission Spectrometry and nuclear analytical methods namely Particle Induced Gamma-Ray Emission and Nuclear Reaction Analysis were used. Plasma spray-physical vapor deposition (PS-PVD) is a novel coating process based on plasma spraying. In contrast to conventional methods, deposition takes place not only from liquid splats but also from nano-sized clusters and from the vapor phase. This offers new opportunities to obtain advanced microstructures and thus to comply with growing demands on modern functional coatings. In this study, different process conditions were investigated with regard to the application of the PS-PVD process for ceramic thermal barrier coatings. Plasma characteristics were calculated under chemical equilibrium conditions by minimizing the Gibbs energy. The plasma-feedstock interaction was modeled taking into account the particular conditions at very low pressure. Since the plasma is highly rarefied, the small feedstock particles are in the free molecular flow regime. Hence, continuum methods commonly used in fluid mechanics and heat transfer approaches with continuous boundary conditions are not appropriate; alternative methods based on the kinetic theory of gases are required. The experimental results confirm the predictions about the degree of vaporization made by such calculations. In particular, they show that the feedstock treatment mainly takes place within the very first trajectory segment between injector and jet expansion.  相似文献

Ex situ determinations of free radicals by 2,2-Diphenyl-1-pricylhydrazyl (DPPH) method and of peroxides by the iodine method applied to plasma treated surfaces were examined. Plasma polymers and plasma-treated polymers, which are already exposed to ambient oxygen, the original free radicals could be detected as peroxides. It was found that the specificity of the both methods was not high enough for quantitative determination. Chemical cleaning of aluminum alloy surfaces, such as alkaline cleaning and deoxidizing treatment, was found to leave large amount of DPPH scavengers, which could be removed by argon or oxygen plasma treatment, implying that the reactivity of DPPH is not exclusive to free radicals. DPPH method should be viewed as an analytical tool to detect scavengers of DPPH (rather than free radicals) existing on the surface region accessible to the DPPH solution. The iodine method should be likewise viewed as an analytical tool for iodine-liberating species existing on the surface region. Plasma treatment (O2 or Ar) of aluminum surface did not remove the species, and in some cases increased the amount. The both methods could be used as tools to assess the overall surface reactivity, particularly of metals, which could influence the adhesion of coatings.  相似文献

Plasma samples from 60 transplant patients on mycophenolate mofetil therapy were analysed using validated capillary electrophoresis (CE) method to determine mycophenolic acid (MPA) and its main metabolite mycophenolic acid glucuronide. Enzyme multiplied immunoassay technique (EMIT) values were available for the same samples. The differences between the results from the two methods was clarified by using Bland–Altman analysis and further statistical analysis. More than 90% of the results showed a positive bias, with EMIT giving higher levels than CE.  相似文献

Elemental analysis of ancient artefacts is of considerable benefit to the field of Archaeology and of general interest to earth scientists. Several techniques are currently available for this purpose, and in this paper the capabilities of PIXE (Particle Induced X-ray Emission), XRF (X-ray Fluorescence) and ICP-MS (Inductively Coupled Plasma - Mass Spectrometry) were evaluated to establish which of these instrumental methods was best suited especially for routine on-line usage.
