Bioactive constituents from Chinese natural medicines. XXXII. aminopeptidase N and aldose reductase inhibitors from Sinocrassula indica: structures of sinocrassosides B(4), B(5), C(1), and D(1)-D(3)

From the methanolic extract of the whole plant of Sinocrassula indica (Crassulaceae), six new flavonol glycosides, sinocrassosides B(4) (1), B(5) (2), C(1) (3), D(1) (4), D(2) (5), and D(3) (6), were isolated together with 30 compounds. The structures of 1-6 were elucidated on the basis of chemical and physicochemical evidence. In addition, several constituents were found to show inhibitory effects on aminopeptidase N and aldose reductase.     

Novel dolabellane-type diterpene alkaloids with lipid metabolism promoting activities from the seeds of Nigella sativa

[structure: see text] Four new dolabellane-type diterpene alkaloids, nigellamines A(1) (1), A(2) (2), B(1) (3), and B(2) (4), were isolated from the seeds of Nigella sativa. Their absolute stereostructures were determined on the basis of chemical and physicochemical evidence. Nigellamines A(1) (1), B(1) (3), and B(2) (4) were found to show potent lipid metabolism promoting activity in primary cultured mouse hepatocytes, and their activities were equivalent to that of a PPAR-alpha agonist, clofibrate.     

New determination methods of halides and cyanide ions by electrospray ionization mass spectrometry based on ternary complex formation

Determination methods of halide ions (X(-) = F(-), Cl(-), Br(-) and I(-)) by electrospray ionization mass spectrometry (ESIMS) were developed, where negative ions of the ternary complexes of group-13 elements, nitrilotriacetic acid (NTA), and halides were measured. In particular, these halides were simultaneously determined by measuring [InX(nta)](-), and the limits of detection (LODs) were 1.1 μmol dm(-3) for F(-), 0.32 μmol dm(-3) for Cl(-), 3.8 nmol dm(-3) for Br(-), and 1.6 nmol dm(-3) for I(-), respectively. This approach was extended to the determination of CN(-), where the ternary complex of Cu(II), CN(-) and 4-(2-pyridylazo)resorcinol (PAR), i.e., [(63)Cu(II)(CN)(par)](-) (m/z 302) was measured. The LOD for CN(-) was 20 nmol dm(-3).     

New triterpenoid saponins from fruit specimens of Panax japonicus collected in Toyama prefecture and Hokkaido (2)

Four new dammarane-type triterpenoid saponins, chikusetsusaponin FT(1) (1), chikusetsusaponin FT(2) (2), chikusetsusaponin FT(3) (3), chikusetsusaponin FT(4) (4), and six known triterpenoid saponins, chikusetsusaponin FK(4) (8), chikusetsusaponin FK(5) (9), chikusetsusaponin FK(2) (10), chikusetsusaponin FK(3) (11), chikusetsusaponin LN(4) (12), and chikusetsusaponin IVa (14), were isolated from the fruits of Panax japonicus C. A. MEYER, collected in Toyama prefecture, Japan, and five new dammarane-type triterpenoid saponins, chikusetsusaponin FT(1) (1), chikusetsusaponin FT(3) (3), chikusetsusaponin FT(4) (4), chikusetsusaponin FH(1) (5), chikusetsusaponin FH(2) (6), and eight known triterpenoid saponins, ginsenoside Re (7), chikusetsusaponin FK(5) (9), chikusetsusaponin FK(2) (10), chikusetsusaponin FK(3) (11), chikusetsusaponin LN(4) (12), 28-desglucosylchikusetsusaponin IVa (13), chikusetsusaponin IVa (14), and chikusetsusaponin V (15), were isolated from the fruits of P. japonicus C. A. MEYER, collected in Hokkaido, Japan. The structures of new chikusetsusaponins were elucidated on the basis of chemical and physicochemical evidences.     

New flavonoids from Oxytropis myriophylla

Eight compounds were isolated from Oxytropis myriophylla. On the basis of spectral analyses, their structures were elucidated to be (6R,9R)-roseoside (1), (6R,9S)-roseoside (2), adenosine (3), myriophylloside B (4), myriophylloside C (5), myriophylloside D (6), myriophylloside E (7), and myriophylloside F (8). Five flavonoids (4-8) were new compounds, and the three known compounds were isolated from this plant for the first time.     

Preparation and characterization of novel branched beta-cyclodextrins having beta-D-galactose residues on the non-reducing terminal of the side chains and their specific interactions with peanut (Arachis hypogaea) agglutinin

Six novel branched beta-cyclodextrins (betaCDs) having beta-D-galactose residues on the non-reducing terminal of the sugar side chains, namely 6(1),6(4)-di-O-(beta-D-galactosyl)-betaCD (10), 6-O-(beta-D-galactosyl)-betaCD (11), 6(1),6(4)-di-O-(beta-lactosyl)-betaCD (14), 6-O-(beta-lactosyl)-betaCD (15), 6(1),6(4)-di-O-(4'-O-beta-D-galactosyl-beta-lactosyl)-betaCD (18), and 6-O-(4'-O-beta-D-galactosyl-beta-lactosyl)-betaCD (19), were chemically synthesized using the trichloroacetimidate method. The reaction products were separated by HPLC on an amino column into dibranched and monobranched betaCDs. Their structures were confirmed by mass spectrometry (MS) and two-dimensional (2D) NMR spectroscopic analysis. To study the length of the sugar side chains attached to the CD ring, which leads to differences in the functions of the branched CDs, interactions of these compounds with peanut (Arachis hypogaea) agglutinin (PNA) were investigated using an optical biosensor and an inhibition assay based on hemagglutination. The results showed that all branched betaCDs interacted with PNA, and the binding affinity was 18>14>10 and 19>15>11 when the derivatives were compared on the basis of side chain length.     

Synthesis of N3-substituted uridine and related pyrimidine nucleosides and their antinociceptive effects in mice

Seventy eight N(3)-substituted derivatives of uridine (1), thymidine (2), 2'-deoxyuridine (3), 6-azauridine (4), 2',3'-O-isopropylideneuridine (5), and arabinofuranosyluracil (6) were synthesized and their antinociceptive effects were evaluated. N(3)-(2',4'-Dimethoxyphenacyl)uridine (1l), N(3)-(2',4'-dimethoxyphenacyl)2'-deoxyuridine (3l), and N(3)-(2',5'-dimethoxyphenacyl)arabinofuranosyluracil (6m) possessed 93, 86, and 82% of the antinociceptive effects tested by hot plate, respectively. The antinociceptive effects of three derivatives were 5.8, 5.4, and 5.1-folds of the effect of N(3)-phenacyluridine (1h) (16%), respectively. The structure-activity relationship of N(3)-substituted pyrimidine nucleosides was also discussed.     

New galloyl derivative from winged sumac (Rhus copallinum) fruit

Twelve compounds were isolated from Winged Sumac (Rhus copallinum) fruit and their structures were elucidated on the basis of NMR and mass spectral data. The isolates included a new galloyl derivative, (R)-galloyl malic acid dimethyl ester (1), and eleven known compounds, gallic acid (2), methyl gallate (3), glucogallin (4), methyl m-digallate (5), methyl p-digallate (6), quercetin (7), myricetin (8), rhamnazin (9), kaempferol (10), betulinic acid (11), and oleanolic acid (12). All of the compounds were evaluated for antiproliferative effects against human colon tumorigenic (HCT-116, Caco-2) and nontumorigenic (CCD18-Co) cell lines.     

Antifungal agent and other constituents from Cynanchum otophyllum

Seven constituents were isolated from the ethyl acetate extract of the rhizome of Cynanchum otophyllum Schneid (Asclepiadaceae). Their structures were determined as 1-(4-methoxy-3-(6-methoxy-3-acetylphenylperoxy)phenyl)ethanone (1), 1-(3-hydroxy-7-acetylnaphthalen-2-yl)ethanone (2), 1-(3,4-dihydroxyphenyl)ethanone (3), 1-(2,4-dihydroxyphenyl)ethanone (4), 1-(3-(3,6-dihydroxy-2-methylbenzoyl)-2,4-dihydroxyphenyl)ethanone (beishouwubenzophenone) (5), N,N-dimethylethanamine (6), and 2-oxo-2-phenylacetic acid (7), respectively, by spectral methods. Among them, 1 and 2 were new compounds; 1 had antifungal activity.     

Bioactive saponins and glycosides. XXV. Acylated oleanane-type triterpene saponins from the seeds of tea plant (Camellia sinensis)

Seven new acylated oleanane-type triterpene oligoglycosides, theasaponins A(4) (1), A(5) (2), C(1) (3), E(8) (4), E(9) (5), G(1) (6), and H(1) (7), were isolated from the seeds of Japanese tea plant (Camellia sinensis). The structures of 1-7 were elucidated on the basis of chemical and physicochemical evidence.     

Novel superstructure of nondiscoid mesogens: uneven-parallel association of half-disk molecules, 3,4,5-trialkoxybenzoic anhydrides, to a columnar structure and its one-directionally geared interdigitation

Compact and simple nondiscoid mesogens, trialkoxybenzoic anhydrides 2-11 ((RO)(3)C(6)H(2)-CO-O-CO-C(6)H(2)(OR)(3), R = C(2)H(5), n-C(3)H(7), n-C(4)H(9), n-C(6)H(13), n-C(8)H(17), n-C(10)H(21), n-C(12)H(25), n-C(14)H(29), n-C(16)H(33), and (S)- and (R)-3,7-dimethyloctyl) were designed and synthesized, and their superstructures were investigated by polarized light microscopy, differential scanning calorimetry, calculation (MM2 and AM1), circular dichroism spectroscopy, and X-ray diffraction. As the result, in the cases of 5-9 and 11, the half-disk molecules in the liquid crystal phases were self-assembled by the dipole-dipole interaction between their carbonyl groups to a column in which the molecules were piled up in an alternately antiparallel manner, and the columns were interdigitated each other. The observed interdigitations were highly directional in the perpendicular direction to the column axis. In the case of compound 11 which has (S)- or (R)-3,7-dimethyloctyl groups, a helical organization of the molecules in the column was estimated from the peaks in the circular dichroism spectra. This is the first report of a one-directionally interdigitated columnar phase, and these compounds are the first liquid crystalline acid anhydrides. It was demonstrated that a -CO-O-CO- moiety is useful as a polar junction in liquid crystalline compounds.     

New triterpene constituents, foliasalacins A(1)-A(4), B(1)-B(3), and C, from the leaves of Salacia chinensis

Four dammarane-type, three lupane-type, and an oleanane-type triterpenes named foliasalacins A(1) (1), A(2) (2), A(3) (3), A(4) (4), B(1) (5), B(2) (6), B(3) (7), and C (8) were isolated from the leaves of Salacia chinensis LINN. collected in Thailand. The structures of new triterpene constituents (1-8) were characterized on the basis of chemical and physiochemical evidence.     

The bakkenolides from the root of Petasites formosanus and their cytotoxicity.

Thirty-two new bakkenolides, bakkenolides-Db (1)--Dh(7), -Fa(8), -Fb(9), -I(10)--M(14), -Na(15), -Nb(16), -O(17)--T(22), -Ua (23), -Ub(24), -V(25)--X(27), -Ya(28), -Yb(29), -Za(30), -Zb(31) and -III(32), from the roots of Petasites formosanus together with thirty known compounds were isolated. The structures were characterized by spectral analysis. The locations, C-1 and/or C-9 of bakkenolide skeleton, of the substituents, such as acetoxy, isobutyroyloxy and isovaleroyloxy groups, can be determined by the chemical shifts of their signals and the H-1 and/H-9 in the 1H-NMR spectra. The cytotoxicity was also discussed.     

Sesquiterpenes from the secondary metabolites of Streptomyces sp. (YIM 56130)

Three new sesquiterpenes were isolated from the fermentation broth of Streptomyces sp. and their structures were determined as caryolane-1,7α-diol (1), 1,6,11-eudesmanetriol; (1α,6β)-form (2), 11-eudesmene-1,6-diol; (1α,6β)-form (3), together with nine known compounds as caryolane-1,9α-diol (4), 2-methyl-5-nonanol (5), soyasaponin I (6), cyclo (Ala-Leu) (7), homononatinic acid (8), β-sitosteryl glucoside-3'-O-heptadecoicate (9), 2'-deoxythymidine (10), 2'-deoxyuridine (11), trehalose (12). The structures were elucidated by spectral analysis.     

Glycerol derivatives and sterols from Sargassum parvivesiculosum

Five glycerol derivatives (1-5) and three sterols (6-8) were isolated from the EtOH extraction of the brown alga of Sargassum parvivesiculosum. On the basis of spectroscopic methods, their structures were elucidated as 1,3-di-O-[2',2'-di-(p-phenylene) isopropylidene] glycerol (1), (2S)-1-O-heptatriacontanoyl glycerol (2), (2S)-1,2-di-O-palmitoyl-3-O-(6-sulpho-alpha-D-quinovopyranosyl) glycerol (3), (2S)-1-O-palmitoyl glycerol (4), (2S)-1,3-di-(O-palmitoyl)-2-O-octadecanoyl glycerol (5), 24-ethylcholest-5,23Z-dien-3beta,28zeta-diol (6), 24-vinylcholest-5-en-24zeta-hydroperoxy (7), 24-ethylcholest-4,24(28)-dien-3beta-ol (8), respectively. Among them, 1 and 2 were new.     

4-[(1,3-二氧代丁基)氨基]苯甲酸稀土配合物的合成、表征及发光性质

在乙醇体系中,由主配体4-[(1,3-二氧代丁基)氨基]苯甲酸(H2L,C11H11NO4)、稀土硝酸盐及辅助配体邻菲啰啉(phen)反应合成了两个系列8个配合物[Ln2(L)3(H2O)4]n(Ln=Sm(1),Eu(2),Tb(3),Dy(4));[Ln2(NO3)2(L)2(phen)2]n(Ln=Sm(5),Eu(6),Tb(7),Dy(8))。用元素分析、红外光谱、摩尔电导、热重分析进行表征,确定了产物的化学组成,推断了相应的结构。测定了室温时固体产物的激发和发射光谱,结果表明:由主辅配体共同配位的三元配合物的发光强度好于无辅助配体参与的二元配合物。测定了三元配合物的荧光寿命,其中铕和铽配合物显示较长的荧光寿命。     

Solvent extraction of several metal ions with mercaptotropone

Mercaptotropone was synthesized from tropone, and its acid dissociation constant (K_a) and distribution coefficient (K_D) between benze and aqueous solution were spectrophotometrically determined as 5.75 (pK_a) and 2.46 (log K_D); Extraction behaviour of Mn(II), Co(II), Ni(II), Cu(II), Zn(II), Pb(II), Cr(III), Fe(III), Y(III), and Zr(IV) with this reagent into benzene was examined. Cu(II) and Fe(III) were completely extracted from acidic solution, Mn(II), Co(II), Ni(II), Zn(II), Pb(II), and Zr(IV) were also extracted from intermediate pH region, a part of Cr(III) was extracted, but Y(III) was not extracted.     

Capillary gas chromatographic analysis of pyrrolidinedithiocarbamate metal chelates

Pyrrolidinedithiocarbamate (PDTC) chelates of Zn(II), Cu(II), Ni(II), Co(III), Fe(III), Mn(II), Cr(III), and VO(II) were analysed by capillary GC on a DB-1701 column (30 m x 0.25 mm id) with flame ionisation detection (FID). Linear calibrations were attained within "1-30 microg/mL" for Ni(II), Fe(III), Mn(II), Cr(III), Cu(II), and VO(II), and within "2-50 microg/mL" for Co(III) and Zn(II). The limits of detection were in the "150-500 ng/mL" range, corresponding to 15-50 pg amounts reaching the FID system. The optimised method was applied to the determination of Cu(II) and Ni(II) in coins, and that of Zn(II), Cu(II), Ni(II), Fe(III), Mn(II), Cr(III), and VO(II) in pharmaceutical preparations with relative standard deviations within 1.1-5.2%. The results obtained are in good agreement with sewage water samples and the declared values for the pharmaceutical formulations, or with the results of AAS of metal contents in coins, pharmaceutical preparations, and sewage water samples.     

Phenolic constituents from the rhizomes of Gastrodia elata

Gastrodia elata Bl. (Orchidaceae) is an important traditional medicinal plant as well as a famous foodstuff in China. In the present article, the HPLC chromatograms of different preparation processes were reported, and structures of nine phenolic compounds, isolated from Gastrodia elata, were assigned. The isolated compounds were identified as 1,3-bis(4-hydroxybenzyl)citrate (1), gastrodin (2), 4-hydroxybenzyl alcohol (3), 1-(4-beta-D-glucopyranosyloxybenzyl)citrate (4), 4-hydroxybenzaldehyde (5), parishin B (6), 4-hydroxybenzyl methyl ether (7), 4-hydroxybenzyl ethyl ether (8), and 4-(4'-hydroxybenzyl)phenol (9). Compounds 1 and 4, named as parishin D and E, were new. Their structures were elucidated on the basis of spectral analyses, including 2D NMR spectroscopy.
