A facile synthesis of benzo[b][1,4]thiazepine derivatives by palladium acetate catalyzed reaction

The preparation of steroid/nonsteroid fused benzo[b][1,4]thiazepines and 2-arylsubstituted benzo[b][1,4]thiazepines is described from Pd(OAc)₂ catalyzed reaction of steroidal/nonsteroidal β-halovinyl aldehydes and 2-aminothiophenols in DMF under heating condition.     

Synthesis of 1,3-disubstituted benzo[c]thiophene analogs containing benzo[b]thiophene/benzo[b]pyrrole

The synthesis of 1,3-disubstituted benzo[c]thiophene analogs incorporating benzo[b]thiophene/benzo[b]pyrrole units is described.     

Synthesis and characterization of benzo[c]thiophene analogs incorporating benzo[b]thiophene/1-hexylindole/benzo[b]furan

Synthesis of 1,3-disubstituted benzo[c]thiophene analogs incorporating heterocycles such as benzo[b]thiophene/1-hexylindole/benzo[b]furan and thiophene units is described. Optical and electrochemical studies of the benzo[c]thiophene analogs are also reported.     

Microwave-assisted one-pot synthesis of benzo[b][1,4]thiazin-3(4H)-ones via Smiles rearrangement

The synthesis of benzo[b][1,4]thiazin-3(4H)-one derivatives in a simple and efficient method from the one-pot reaction of substituted 2-chlorobenzenthiols, chloroacetyl chloride, and primary amines via Smiles rearrangement under microwave irradiation gave high yields (65-92%) of the products with short reaction time (15-20 min).     

The first synthesis of the benzo[b]indolo[1,2-h][1,7]naphthyridine ring system

The first syntheses of representatives of the benzo[b]indolo[1,2-h][1,7]naphthyridine ring system have been accomplished using the Friedländer reaction.     

Synthesis of polyfluorodibenz[b,f][1,4]oxazepines by the cyclization of 2-[(polyfluorobenzylidene)amino]phenols

Thermolysis of 2-[(polyfluorobenzylidene)amino]phenols in ethanol in the presence of triethylamine affords the corresponding fluorinated dibenz[b,f][1,4]oxazepines in moderate to high yields. The structures of 2-[(2,3,4,5,6-pentafluorobenzylidene)amino]phenol and 1,2,3,4-tetrafluorodibenzo[b,f][1,4]oxazepine have been determined by single-crystal X-ray diffraction.     

Novel synthesis of benzo[b]carbazoles

A new method was adopted for the synthesis of benzo[b]carbazoles by Claisen condensation followed by Fischer indole cyclization. Newly synthesized benzo[b]carbazoles were treated with ethanol amine in the presence of polyphosphoric acid which leads to the formation of pyrazino carbazoles. All the synthesized compounds were characterized by all spectral means.     

Synthesis of dibenzo-[b,f][1,5]diazocine-based hosts and their assembly behaviors with C60

Diaryl[b,f][1,5]diazocine-based macrocycles 5 and 6 with inner cavities have been synthesized via Hay coupling method. Single crystal of 5 shows that it forms a dimeric aggregate via the weak intermolecular interactions between two adjacent rings, which is rarely reported in such macrocycles. Moreover, SEM results reveal that both macrocycles 5 and 6 assemble with C₆₀ to form well-ordered fullerene-based nano-rod structures.     

Synthesis of naphtho[b]cyclobutenes from 1,2-bis(3-propynol)benzenes

We have demonstrated that the reaction of benzene-bridged bis(propargyl alcohol)s with chlorodialkylphosphines exclusively afforded 3,8-bis(dialkylphosphinyl)naphtho[b]cyclobutenes via the [2+2] cycloaddition of 1,2-bis(α-phosphinylallenyl)benzenes. Dephosphinylation of the product has also been achieved.     

Rapid access to multi-substituted pyrimido[4,5-b][1,4]diazepine-2,4,6-trione and pyrimido[4,5-b][1,4]diazepine-2,4-dione as novel and versatile scaffolds for drug discovery

A novel pyrimido[4,5-b][1,4]diazepine-2,4,6-trione was synthesized with an efficient strategy. Especially, the key intermediate 2,4-dimethoxypyrimido[4,5-b][1,4]diazepin-6-one was promoted by one pot tandem reduction-cyclization with Na₂S₂O₄. Subsequently, reduction of lactams 6 with LiAlH₄ afforded a more flexible scaffold of pyrimidodiazepines. The synthetic strategy was versatile since it facilitated the sequential functionalization on the pyrimidodiazepine at three positions. Thus a convenient and effective method for the rapid preparing of multi-substituted pyrimido[4,5-b][1,4]diazepines was developed.     

Photochromism of diarylethene derivatives bearing a benzo[b]silole unit

Photochromic benzo[b]silole derivatives, 1-(1,1-dimethyl-2-phenylbenzo[b]silol-3-yl)-2-(2-phenylbenzo[b]thien-3-yl)perfluorocyclopentene and 1-(1,1-dimethyl-2-phenylbenzo[b]silol-3-yl)-2-(2-phenyl-1-benzofuran-3-yl)perfluorocyclopentene, were synthesized and their photochromic performance was examined in solution.     

Selective synthesis, structure and oxidation properties of isomeric 1,4-dithiins fused to two benzo[b]thiophenes

Two novel isomers of 1,4-dithiins fused to two benzo[b]thiophenes on both sides were obtained by selective synthetic methods via the corresponding sulfides. The crystal structures of two dithiins were determined by X-ray crystallographic analyses. Examination of oxidation properties of the dithiins showed interesting results by CV measurement. The ESR spectra of chemically oxidized dithiins indicated formation of stable radical cations.     

Novel synthesis of pyridazino[4,5-b][1,4]oxazin-3,8-diones

A novel and effective synthesis of pyridazino[4,5-b][1,4]oxazin-3,8-diones via Smiles rearrangement is presented. Treatment of N-substituted 2-chloro(or hydroxy)acetamide, 2-tetrahydropyranyl-4-chloro-5-hydroxy(or chloro)pyridazin-3-one and cesium carbonate in refluxing acetonitrile was afforded the corresponding pyridazino[4,5-b][1,4]oxazin-3,8-diones in excellent yield.     

A facile one-pot synthesis of 2-substituted-3-aminoquinolines: preparation of benzo[b]naphthyridine-3-carbonitriles

A facile one-pot synthesis of 3-aminoquinolines from ortho-aminobenzaldehydes was developed. Ethyl 6,7-dimethoxy-3-aminoquinoline-2-carboxylate, a key intermediate for the preparation of a 4-anilino-benzo[b][1,5]-naphthyridine-3-carbonitrile, was efficiently prepared by this method. Synthetic routes to 4-anilino-benzo[b][1,5]-naphthyridine-3-carbonitrile and 4-anilino-benzo[b][1,8]-naphthyridine-3-carbonitrile are described     

Tröger’s bases in the acronycine, benzo[a]acronycine, and benzo[b]acronycine series

Reaction of 11-aminoacronycine, 10-aminobenzo[a]acronycine, and 10-aminobenzo[b]acronycine with paraformaldehyde gave the corresponding Tröger’s bases 11, 14, and 16, respectively. The cytotoxic activity of those three new compounds was determined against L-1210 leukemia and KB-3-1 solid tumor cell lines, in comparison with their parent compounds, acronycine, benzo[a]acronycine, and benzo[b]acronycine.     

Application of 2-(2-chloroaroyl)methyleneimidazolidines in domino and multicomponent reaction: new entries to imidazo[1,2-a]pyridines and benzo[b]imidazo[1,2,3-ij][1,8]naphthyridines

A new strategy for the synthesis of tetrahydroimidazo[1,2-a]pyridines and unusual tetrahydrobenzo[b]imidazo[1,2,3-ij][1,8]naphthyridines has been successfully developed by cascade reactions including Knoevenagel condensation, aza-ene reaction, imine-enamine tautomerization, cyclocondensation/oxidation, and intramolecular S_NAr of precursors 2-(2-chloroaroyl)methyleneimidazolidines as new heterocyclic ketene aminals (HKA), which represent a class of polyfunctional scaffolds with four active reaction sites with aromatic aldehydes and malononitrile or ethyl 2-cyanoacetate under mild conditions. In this domino reaction, nine different active sites are involved, and two C-C bonds, two C-N bonds, and two new rings are constructed with all reactants efficiently utilized in the chemical transformation.     

Bromination of 3-substituted benzo[b]thiophenes: access to Raloxifen intermediate

A practical route to prepare halogeno-derivatives is described, starting from easily available 3-cyano-benzo[b]thiophene. Main efforts have been devoted to the optimization of the experimental procedures (solvent, bromination) to promote selectivity. Synthetic studies investigate the potential access to an advanced intermediate of Raloxifen.     

1,3-Dipolar cycloaddition reactions of benzo[b]thiophene 1,1-dioxide with azomethine ylides

New pyrrolo derivatives of benzo[b]thiophene 1,1-dioxide have been synthesised via 1,3-dipolar cycloaddition reactions. Reaction of benzo[b]thiophene 1,1-dioxide with stabilised azomethine ylides gave products in low yield but high stereoselectivity whereas reaction with non-stabilised azomethine ylides gave high overall yields but low stereoselectivity.     

A new synthesis of benzo[b]thiophenes utilizing an interrupted Pummerer reaction

A convenient synthesis of 3-arylbenzo[b]thiophenes utilizing an interrupted Pummerer reaction of 2-(1-arylvinyl)phenyl ethyl sulfoxides is described. Thus, treatment of these sulfoxides, which were readily prepared from 2-sulfanylphenyl ketones or 2-fluoro-5-methoxybenzaldehyde, with acetic anhydride at 100 °C afforded 3-arylbenzo[b]thiophenes in reasonable yields.     

Densely functionalized 1,2-dihydrobenzo[b][1,6]naphthyridines: one-pot synthesis via sequential Ugi and Heck reactions

A facile one-pot synthesis of functionalized 1,2-dihydrobenzo[b][1,6]naphthyridines is described via sequential Ugi four components using 2-chloroquinoline-3-carboxaldehydes, allyl amine, acetic acid, isocyanides and ligand free Heck reactions in good yields.
