TheO, O-dibenzoyl derivative of (R, R)-tartaric acid shows a good inclusion ability for diethyl and di-n-propol ethers. The two crystalline inclusion compounds have 1:1 stoichiometry and reveal isomorphous structures. Hydrogen bonded host molecules form chains running along thez axis of the unit cell and guest molecules join to these chains by short O–H...O hydrogen bonds. Hydrogen bonding in the crystals is characterized by a C(7)D first-order network. The ether molecules are in a fully extended conformation. They are accommodated in channel-like voids running along thex axis. Atomic displacement parameters are significantly larger for diethyl ether than for the di-n-propyl ether molecule reflecting less dense packing for this inclusion compound.Supplementary Data: relating to this article are deposited with the British Library as Supplementary Publication No. SUP 82195 (19 pages)  相似文献

The absolute stereochemistry of TT-1 (1=rasfonin), an α-pyrone-containing natural product from a Fungi Imperfecti, Trichurus terrophilus, was determined as 5R,6R,7S,9R, and 6′S on the basis of synthesis of diastereomers of two fragments of 1 in optically active forms, and comparison of their spectral and optical data with those of natural specimens.  相似文献

High yielding and remarkably selective alkylations of a suitably protected derivative of (2S,3aS,7aS)-octahydroindole-2-carboxylic acid are described. The fused bicyclic structure of this proline analogue greatly influences the stereochemical outcome of direct alkylation reactions taking place at the α-carbon and provides access to α-substituted analogues with retention of the configuration. The overall procedure allows the preparation of enantiopure α-substituted derivatives of this Oic isomer, suitably protected for their incorporation into peptides, in a straightforward manner.  相似文献

cis-Chrysanthemic acid has been prepared in a few steps from dimethyldimedone via dibromination at alpha positions of each carbonyl carbons. The trans-dibromide which is almost exclusively formed has been isomerized to its cis-stereoisomer by highly chemoselective tandem H/K-K/H exchanges involving potassium bases at low temperature (<−40 °C). Carbocyclization of the potassium enolate intermediate takes place at around −30 °C and provide the bicyclo[3.1.0]-hexane skeleton. Lithiated bases behave differently and mainly lead to Br/M rather than to H/M exchange. We have been unsuccessful in using state of the art enantioselective metallation reactions to achieve the enantioselective synthesis of (1R)-cis-chrysanthemic acid using the disclosed strategy. A chiral β-amino acid containing three contiguous chiral centers was synthesized efficiently in 11 steps, employing enantio-enriched β-ketoester as a key intermediate, via stereoselective catalytic hydrogenation of the corresponding enamide. Stereoselective 1,4-addition of a methyl group and protonation were key to the preparation of the desired acid 12. Mild and efficient reaction conditions were applied to the enamine formation and protection to avoid epimerization at C-4 of compounds 13 and 14. The final compound was found to display potent affinity for the α2δ-protein that is a recognized drug target for the treatment of a variety of diseases.  相似文献

Racemic trans 3-(9-fluorenylmethyloxycarbonylamino)-1-oxyl-2,2,5,5-tetramethylpyrrolidine-4-carboxylic acid (Fmoc-POAC-OH), prepared by conventional methods, was resolved upon esterification with (aR)-2,2′-dihydroxy-1,1′-binaphthyl. Separation of the obtained diastereomeric monoesters Fmoc-(±)-trans-POAC-O-(aR)-binaphthol by crystallization/chromatography, and removal of the chiral auxiliary by saponification of the aryl ester function furnished both enantiomers (+)-(3R,4R)-Fmoc-POAC-OH and (−)-(3S,4S)-Fmoc-POAC-OH. The absolute configuration of the asymmetric C3, C4 carbons of POAC were assigned from the induced circular dichroism of a flexible biphenyl probe present in the terminally protected dipeptide derivatives Boc-Bip-(+)-POAC-OMe and Boc-Bip-(−)-POAC-OMe (Bip, 2′,1′:1,2;1″,2″:3,4-dibenzcyclohepta-1,3-diene-6-amino-6-carboxylic acid). This assignment was confirmed by X-ray diffraction analysis of the diastereomeric monoester Fmoc-(+)-trans-POAC-O-(aR)-binaphthol, shown to be (aR,3R,4R). Solution synthesis of peptides to the hexamer level, based on the (3R,4R)-POAC enantiomer combined with (1S,2S)-2-aminocyclopentane-1-carboxylic acid, was carried out to examine coupling conditions at both C- and N-termini of the POAC residue, in view of further syntheses and 3D-structural investigations.  相似文献

An orthogonally protected β,γ′-diamino acid 6 possessing conformationally-constrained ring system was synthesized as a novel cyclic amino acid analogue. This synthesis involves as key steps chemoselective enzymatic hydrolysis of cis-piperidine-3,5-dicarboxylic ester derivative followed by efficient kinetic resolution of the partially resolved half-acid to afford the C1-symmetric piperidine-3,5-dicarboxylic acid monoester in high enantiomeric excess (>98% ee). The optically active half-acid was transformed to the cyclic amino acid via Curtius-type rearrangement.  相似文献

A new route for (2S,3S,4S)-form, the physiologically active stereoisomer of dehydroxymethylepoxyquinomicin (DHMEQ), a potent NF-κB inhibitor, was established by chemoenzymatic approach. Elaboration on the asymmetric epoxidation of a p-benzoquinone monoketal with benzylcinchonidinium tert-butylhydroperoxide yielded an epoxyenone, in 79.8% ee and 57% yield in reproducible manner. By way of the transformation of this key intermediate to enantiomerically pure (2S,3S,4S)-DHMEQ, the contaminating undesired enantiomer could be effectively removed by applying Burkholderia cepacia lipase-catalyzed hydrolysis of diacylated precursor. The above integrated combination of chemical asymmetric synthesis and enzyme-catalyzed kinetic resolution enabled us to prepare active DHMEQ in a large-scale.  相似文献

The full details of our enantioselective formal synthesis of the biologically active natural product berkelic acid are described. The insertion of the C-18 methyl group proved challenging, with three different approaches investigated to install the correct stereochemistry. Our initial Horner-Wadsworth-Emmons/oxa-Michael approach to the berkelic acid core proved unsuccessful upon translation to the natural product itself. However, addition of a silyl enol ether to an oxonium ion, followed by a one-pot debenzylation/spiroketalisation/thermodynamic equilibration procedure, afforded the tetracyclic structure of the berkelic acid core as a single diastereoisomer.  相似文献

The reaction of N-(3-chloro-3-methyl-2-butylidene)amines (i.e. α-chloroketimines) with lithium diisopropylamide in tetrahydrofuran gave rise to an unexpected reaction leading to 2-(N-alkylimino)-3,3-dimethyl-1-isopropyl-5-(2-methylpropylidene) pyrrolidines in good yields. The reaction mechanism is interpreted in terms of a Favorskii-type 1,3-dehydrochlorination affording intermediate cyclopropylideneamines (N-analogues of cyclopropanones), which “dimerize” in an unusual way. This mechanism entails a “normal” opening as well as an “abnormal” opening of incipient cyclopropylideneamine adducts. This rearrangement represents an example of the scarcely reported abnormal opening of functionalized cyclopropylamines.  相似文献

The aldol condensation reactions of α,α-diacetyl ketene-(S,S)-acetals, 1a and 1b, with aromatic aldehydes 2 in the presence of NaOH in water have been investigated. At room temperature, the base-mediated condensations proceed smoothly to afford the corresponding mono-condensed products, α-alkenoyl ketene-(S,S)-acetals 3, in high yields. At reflux temperature, the reactions produce high yields of symmetric double condensed α,α-dialkenoyl ketene-(S,S)-acetals 4 when 2 equiv of aldehyde are employed. Moreover, compounds 3 can condense with a different aldehyde to give unsymmetric double condensed α,α-dialkenoyl ketene-(S,S)-acetals 5.  相似文献

A simple methodology to a unique macrocyclic α-amino acid (AAA) derivative involving three step synthetic sequence has been reported. In addition, various ruthenium based catalysts were studied to enhance the selectivity of the desired macrocyclic AAA derivative 6. This Letter, describes the complexation of α-, β- and γ-cyclodextrins (1-3) with TTF derivative 4 in water. In particular, we show using 1H, 13C NMR, UV-vis spectroscopy and isothermal titration calorimetry that β-cyclodextrin 2 forms an effective complex with 4. Complex 2·4 can be conveniently disassembled upon the addition of 1-adamantanol.  相似文献

Microbial transformation of betulonic acid by Nocardia sp. NRRL 5646 in preparative scale resulted in the isolation of one unexpected asymmetric α-hydroxylation product, methyl 2α-acetoxy-3-oxo-lup-20(29)-en-28-oate, and one known compound methyl 3-oxo-lup-20(29)-en-28-oate. The structures of metabolites were elucidated unambiguously by ESI-MS, 2D-NMR spectroscopy. Thermal decompositions of polyisoprene, poly(p-isopropyl α-methylstyrene) (PPIPαMS), and poly(isoprene/p-isopropyl α-methylstyrene) (sample M-32) were carried out at various temperatures in the range 200–340° C in a differential thermo-gravimetric apparatus. The undecomposed polymers as well as their decomposed residues were analyzed by gel-permeation chromatography (GPC), infrared spectroscopy (IR), and nuclear magnetic resonance (NMR). Based on the changes observed in the distribution of molecular weights, depolymerization is the predominant step in the decomposition of PPIPAMS and polymer M-32, whereas random scissions predominate in the case of polyisoprene. The combined data of GPC, IR, and NMR indicate that only interchain reactions leading to the formation of cyclized products are present in the decomposition of polyisoprene while interchain as well as intrachain reactions are operative in the case of polymer M-32.  相似文献
