A novel, general, and stereoselective route to functionalized bridged bicyclo[2.2.2]octanes, bicyclo[3.3.0]octanes, bicyclo[4.2.0]octanes, and tricyclo[4.3.1.0(3,7)]decanes has been described. Various functionalized and substituted bicyclo[2.2.2]octanes endowed with a beta,gamma-enone chromophore were synthesized via cycloaddition of in situ generated cyclohexa-2,4-dienones with electron-deficient 2pi partners and manipulation of the resulting adducts. Triplet sensitized irradiation of bridged bicyclooctenones led to synthesis of bicyclo[3.3.0]octanoids, whereas the direct irradiation furnished bicyclo[4.2.0]octanes in stereoselective fashion as a result of modulation of reactivity in excited states. Further, manipulation of the adducts led to appropriately appended and functionalized bicyclo[2.2.2]octanes that upon radical induced cyclization provided an efficient and stereoselective route to the tricyclo[4.3.1.0(3,7)]decane (isotwistane) framework of pupukeananes.  相似文献

The type 2 intramolecular Diels-Alder (IMDA) reaction is a valuable method for synthesis of both carbocyclic and heterocyclic bridged bicyclo[5.3.1]undecane and bicyclo[4.3.1]decane ring systems. These structures are common to a number of biologically important natural products. Asymmetric variants of the type 2 IMDA reaction incorporating oxazolidinone chiral auxiliaries have been evaluated. This study has resulted in systems that deliver bridged bicyclic [5.3.1] and [4.3.1] ring systems in high diastereomeric (97-99% de) and enantiomeric purity.  相似文献

[reaction: see text] We have developed highly versatile, homochiral oxabicyclo[3.2.1]octadiene building blocks for the synthesis of natural products. We have found that these bridged alkenes undergo exceptionally facile Diels-Alder reactions and react faster than several well studied bicyclo[2.2.1]heptene dienophiles. The reaction proceeds with high levels of stereochemical control and in very good to excellent yields, providing access to bicyclo[5.4.0]undecane and bicyclo[5.3.0]decane systems. This reactivity is attributed to strain and homoconjugation effects.  相似文献

A highly efficient and practical synergistically metal/proton‐catalyzed Conia–ene reaction for the synthesis of bicyclo[3.n.1]alkanones has been developed. This synergistic catalysis was successfully utilized in modifying natural compounds such as methyl dihydrojasmonate, α,β‐thujone, and 5α‐cholestan‐3‐one. Furthermore, the bridged carbonyl group of bicyclo[3.2.1]alkanones could be easily attacked by nucleophiles to give the ring‐opened cycloheptenone products or bicyclo[4.2.1]amide in excellent yields. These reactions provide rapid access to a diverse range of cyclic structures from simple starting materials or naturally occurring compounds.  相似文献

A type II intramolecular oxidopyrylium‐mediated [5+2] cycloaddition reaction allows the efficient and diastereoselective formation of various highly functionalized and synthetically challenging bridged seven‐membered ring systems (such as bicyclo[4.4.1]undecane, bicyclo[4.3.1]decane, bicyclo[5.4.1]dodecane, and bicyclo[6.4.1]]tridecane). This simple, thermal, direct transformation has a broad substrate scope and is high yielding, with high functional‐group tolerance and unique endo selectivity. The highly strained tricyclic cores of ingenol mebutate (picato) and cyclocitrinol are synthesized efficiently and diastereoselectively using this methodology.  相似文献

The rosebengal-sensitized photooxygenation of 2-methoxynorborn-2-ene(1)in acetaldehyde gave cis-1-carboxaldehyde-3-carbomethoxycyclopentane (31%) and the cis and trans-Me derivatives of the cis-fuscd exo-1,2,4-trioxanes arising by addition of a molecule of oxygen and acetaldehyde to 1 at C3 and C2 respectively (13%) Similar photooxygenation of 2-(methoxymethylidene)adamantane in the presence of acetaldehyde, propionaldehyde and pivalaldehyde gave adamantanone (31–42%), and the cis and trans tricylo[3.3.1.13,7]decane-2-spiro-6'-[3-alkyl-5-methoxy-1,2,4-trioxanes] in yields of 32–53% Trioxane formation under similar conditions was experienced for 1,1-di-t-butyl-2-methoxyethene and 2-(methylmercaptomethylidene) adamantane. The results are discussed in terms of an intermediate zwitterionic peroxide which can either close directly to a 1,2-dioxetane or, if aldehyde is present, condense across the CO function giving the 1,2,4-trioxane.  相似文献

Optically active 1,3-bridged cyclobutanes 10 of the bicyclo[3.1.1]heptane ring system and 1,2-bridged cyclobutanes 11 of the bicyclo[3.2.0]heptane ring system were produced by UV irradiation of alpha,beta,gamma,delta-unsaturated esters 9a and 9c-f. The preference of endo-stereochemistry at C-6 bridged head was observed in cross-adducts 10. On the other hand, irradiation of conjugated dienol 9b led via only parallel cycloaddition to 1,2-bridged cyclobutane 11.  相似文献

Bicyclo[3.2.1] type 1,2,4-trioxanes are readily synthesized from precursors that may form intramolecular hemiketals using UHP (H2O2-urea complex) as the source of the peroxy bond and p-TsOH or CSA as the catalyst. The ring closure through an intramolecular Michael addition occurred in a highly stereoselective way, giving only one diasteromer as shown by the NMR spectra.  相似文献

A new and highly versatile approach towards the synthesis of bicyclo[6.3.0]undecanes and bicyclo[5.3.0]decanes was accomplished. The methodology adopted involved [6+3] and [3+2] cycloaddition reactions of pentafulvenes with 3-oxidopyridinium betaines generated either by the action of a base on the pyridinium salt or thermally from pyridinium betaine dimer. These well-functionalized bicyclo[6.3.0]undecanes and bicyclo[5.3.0]decanes offer a wide range of synthetic options, which can be expected to translate into a variety of rapid and efficient synthesis of natural products.  相似文献

The synthesis of 3-allylthio-5H-[1,2,4]triazino[5,6-b]indole has been carried out by the reaction of 3-mercapto-5H-[1,2,4]triazino[5,6-b]indole with allyl bromide in the NaOH–H2O–DMSO system and in a one-pot synthesis from isatin-β-thiosemicarbazide. Halocyclization of allylthio-5H-[1,2,4]-triazino[5,6-b]indole synthesized the 3-halomethyl-3,10-dihydro-2H-[1,3]thiazolo[3',2':2,3][1,2,4]-triazino[5,6-b]indolium halides.  相似文献

The application of the Ramberg-Backlund reaction in the conversion of several 3,n-dithiabicyclo[n.3.1]alkatrienes to the corresponding bicyclo[n.3.1]alkapentaenes is reported. Proton magnetic resonance and ultraviolet spectra were utilized in the overall characterization of bicyclo[8.3.1]tetradeca-2,8,10,12,1(14)-pentaene, bicyclo[9.3.1]pentadeca-2,9,11,1(15)-pentaene, and their precursors. The final step in the synthesis was a Ramberg-Backlund of bis-α,α′-chloro sulfone. The intermediate, 2-chloro-3-thiabicyclo[9.3.1]pentadeca-9,11,13,1(15)-tetraene 3,3-dioxide, was isolated and characterized. The structure and stability of the bridged α,α′-dichloro sulfide intermediates were examined.  相似文献

A series of 4a, 10b-dihydronaphtho-[2,1-e]-1,2,4-trioxanes were examined by h.p.l.c. Compounds existing as a pair of diastereomers are only resolved when hydrogen is a substituent at the C3 position. All of the point groups common to organic chemistry except two are illustrated by known compounds that are rigid [2.2]paracyclophane derivatives. Examples are given of transannular directing effects by acetyl, nitro, and acetoxyl substituents attached to [2.2]paracyclophane. In bromination or chloromethylation, proton loss of a sigma complex is rate-determining, and the oxygens already in the molecule remove the proton being substituted. The synthesis of [2.2.2](1,2,4)cyclophane and [3.2.2](1,2,5)cyclophane, and their unusual chemical properties are described. Transannular hydride shifts out of methyl groups due to proximity effects are reported. Torsional racemizations and epimerizations of [2.2]paracyclophane derivatives are reviewed. The diradical intermediates formed have been intercepted by either H· donors, or by addition to substituted olefins. To account for the stereochemical course of addition and substitution reactions in the side-chains of [2.2]- and [4,2]paracyclophanes, new types of bridged carbonium ions are suggested. Conformational equilibria in the four-carbon side-chain of [4.2]paracyclophane derivatives are discussed.  相似文献

A set of new bis-spirofused 1,2,4-trioxanes 4a-d was obtained from the reaction of cyclohexane-1,4-dione with allylic hydroperoxides 2a-d, bearing an additional hydroxy group in the homoallylic position, by diastereoselective photooxygenation of allylic alcohols 1a-d and subsequent BF(3)-catalyzed peroxyacetalization with the diketone. From the reaction of a monoprotected cyclohexane-1,4-dione 5 with the allylic hydroperoxide 6 derived from the singlet oxygenation of methyl hydroxytiglate, one monospiro compound was obtained, the 1,2,4-trioxane ketone 7, as well as a mixture of the diastereoisomeric syn- and anti bis-1,2,4-trioxanes 8. The structures of bis-1,2,4-trioxanes were examined theoretically by DFT methods and compared with X-ray structural data in order to evaluate the preferential trioxane ring conformational orientation.  相似文献

A diversity of new 7 ‐substituted[1,2,4]triazolo[1,5‐a]pyrimidine and 6‐substituted[1,2,4]triazolo[1,5‐a]pyrimidine‐7‐amine derivatives has been synthesized via reaction of 3‐amino‐[1,2,4]triazole with enaminonitriles and enaminones. The regio orientation and the structure of the products were confirmed by spectral and analytical data and synthesis via an alternative route. The procedure proved to be simple, efficient, and high yielding, and diversities of [1,2,4]triazolo[1,5‐a]pyrimidines were obtained.  相似文献
