2-Substituted-4a-hydroxy-9H-cycloalkyl[1,2-e]oxazolo[3,2-a]pyrimidin-9-ones 2a-c were synthesized by an one-step cyclocondensation from the 5-substituted-2-amino-2-oxazolines 1a-c with ethyl 2-oxocyclohexanecarboxylate in ethanol at room temperature, and easily dehydrated to provide 2-substituted-9H-cycloalkyl[1,2-e]oxazolo[3,2-a]pyrimidin-9-ones 3. In refluxing xylene, the reaction conducted with various ethyl 2-oxocycloalkanecarboxylates led to the two isomeric 2-substituted-8/9H-cycloalkyl[1,2-e]oxazolo[3,2-a]pyrimidin-8/9-ones 3 and 2-substituted-5H-cycloalkyl[1,2-d]oxazolo[3,2-a]pyrimidin-5-ones 4. The structure of some compounds was unambiguously established using X-ray crystallography. According to results from the DSC analysis of compound 2a, formation of the thermodynamically stable pyrimidinones 4 could be related to an intramolecular rearrangement of kinetically controlled pyrimidinones 3.  相似文献

A convenient synthesis of new 5,6,7,8-tetrahydro-imidazo[1,2-a]pyrimidin-2-ones and 3,4,6,7,8,9-hexahydro-pyrimido[1,2-a]pyrimidin-2-ones from the Baylis-Hillman adducts of acrylonitrile and their derivatives is described. A common strategy employed to achieve the syntheses of title compounds involved generation of diamines from different Baylis-Hillman derivatives followed by treatment with cyanogen bromide at reflux temperature to trigger a double intramolecular cyclization.  相似文献

The total synthesis of chromeno[4,3,2-cd]isoindolin-2-ones 6a-d and chromeno[4,3,2-de]isoquinolin-3-ones 15a-b from 4-methoxy-9H-xanthen-9-one is reported. The construction of the nitrogenated ring was attempted by both intramolecular electrophilic and anionic cyclizations of the corresponding carbamate precursors. Only anionic cyclization was possible for isoindolinones, but for isoquinolinones the electrophilic and anionic routes both afforded excellent yields.  相似文献

A simple synthesis of benzo[d,e]imidazo[2,1-a]isoquinolines is described. Heating a mixture of acenaphthoquinone, a benzil, and ammonium acetate under solvent-free conditions afforded 9,10-diaryl-7H-benzo[d,e]imidazo[2,1-a]isoquinolin-7-ones in good to excellent yields.  相似文献

One-pot synthesis of benzo[a]phenanthridin-5-ones and benzo[k]phenanthridin-6-ones in fairly good yields was achieved by the photocycloaddition reactions of 3-chloroisoquinolin-1-ones and 3-chloroquinolin-2-ones with styrenes. The reactions were proceeded via photoinduced dechlorinative coupling of 3-chloroisoquinolin-1-ones and 3-chloroquinolin-2-ones with styrenes and subsequent photocyclization, oxidative aromatization.  相似文献

The regioselective synthesis of cis and trans stereoisomers of variously functionalized octahydro[1,2,4]triazolo[4,3-a]quinazolin-5-ones was performed. The 2-thioxopyrimidin-4-ones used in the synthesis reacted with hydrazonoyl chlorides in a regioselective manner to produce the angular regioisomers [1,2,4]triazolo[4,3-a]quinazolin-5-ones rather than the linear isomers [1,2,4]triazolo[4,3-a]quinazolin-5-ones. The synthesis process took place with electronic control. The angular regiochemistry of the products was confirmed by X-ray experiments and two-dimensional NMR studies.  相似文献

The reaction of 4-amino-substituted 6-hydrazinyl-1,3,5-triazin-2(1H)-ones with orthoformate proceeds via the Dimroth-type rearrangement to give 5-amino-substituted 1,2,4-triazolo[1,5-a]-1,3,5-triazine-7-ones. IR, NMR and X-ray studies have shown that the only product of the reaction was the [1,5-a]-isomer as prototropic 3H- and 6H-tautomers.  相似文献

2-Hydroxy-4H-benzo[4,5]thiazolo[3,2-a]pyrimidin-4-one 2a and 7-hydroxy-5H-thiazolo[3,2-a]pyrimidin-5-one 2b, were obtained in high yields under mild conditions from the cyclization reactions of bis-(2,4,6-trichlorophenyl) malonate and 2-aminobenzothiazole or 2-aminothiazole, respectively. A new class of compounds, 2,3-dihydro-4H-benzo[4,5]thiazolo[3,2-a]furo[2,3-d]pyrimidin-4-ones and 6,7-dihydro-5H-furo[2,3-d]thiazolo[3,2-a]pyrimidin-5-ones, were synthesized via the microwave assisted radical addition of compounds 2a and 2b to various alkenes using manganese(III) acetate. A preliminary acetylcholine esterase (AchE) inhibition test of compound 4e showed excellent (92%) inhibitory potential, comparable with the standard drug Donapezil®.  相似文献

By means of base-catalyzed ring enlargement of triazaindenoindenes and pentalenoindenes obtained from anhydride-induced ring transformation of 3,4-dihydro-2H-pyrimido- and 2,3-dihydroimidazo[2,1-a]phthalazinium-olates, respectively, a series of pyrazolo[1,5-e]benzo[g][1,5]diazonin-8-ones and pyrazolo[1,5-d]benzo[f][1,4]diazocin-7(6H)-ones were obtained. Alternative pathways and energetics for the ring inversion of symmetrically substituted medium-size ring systems were determined by combined use of DNMR measurements and B3LYP/6-31G(d,p) calculations using the IEFPCM solvent model. One pyrazolobenzodiazonine carrying hydrogen at the C1 position was found to undergo facile ring inversion by a two-step mechanism, while 1-Me and 1-Ph substituents rendered complete rigidity to this ring system. A three-step mechanism was revealed for the ring inversion of the two studied pyrazolobenzodiazocines with energetics practically invariant to the investigated C1-substituents (H and Me). The attempted RCM of the N,O-diallyl derivative of a selected rigid model effected by Grubbs II catalyst led to deallylation and olefin isomerization avoiding the formation of bridged products with enhanced skeletal strain. A tolerable degree of ring strain associated with negligible skeletal distortion could be introduced into the same benzodiazonine by N,O-dialkylation with 1,3-bis(bromomethyl)benzene.  相似文献

2-Alkoxyiminoimidazolidines 2-3 react with acetylene dicarboxylates and ethyl phenylpropiolate to give 8-alkoxy-imidazo[1,2-a]pyrimidin-5(3H)-ones C, which subsequently undergo a sterically induced multihetero-retro-ene fragmentation to give imidazo[1,2-a]pyrimidin-5(1H)-ones 4-7 together with formaldehyde or benzaldehyde. On the other hand, a similar reaction of 2-3 with ethyl propiolate gives corresponding 8-alkoxy-imidazo[1,2-a]pyrimidin-5(3H)-ones 8-10. The unsubstituted imidazo[1,2-a]pyrimidin-5(1H)-one 11 can be prepared by retro-ene reaction of 9 upon prolonged heating in refluxing ethanol. A direct synthetic approach to 1-formyl-7-phenyl-imidazo[1,2-a]pyrimidine-5(1H)-one 14 is reported using DMF/sulfonyl chloride as a new Vilsmeier-type N-formylating reagent.  相似文献

1-Hydroxy-9,9a-dihydro-1H-imidazo[1,2-a]indol-2(3H)-ones, as a new type of azaheterocyclic hydroxamic acids, have been synthesized regioselectively from 1-carbamoylmethyl- or 1-(methoxycarbonyl)methyl-2,3,3-trimethyl-3H-indolium salts by reaction with hydroxylamine in the presence of a strong base. The alkylation and reduction with sodium borohydride of these novel heterocycles have been investigated. When treated with protic acids 1-hydroxy- or 1-alkoxy-9,9a-dihydro-1H-imidazo[1,2-a]indol-2(3H)-ones underwent ring opening of the imidazolidine to afford 1-[2-(hydroxyamino)-2-oxoethyl]-2,3,3-trimethyl-3H-indolium salts. The structural assignments are based on extensive 1H, 13C and 15N NMR spectroscopic studies and single crystal X-ray analyses.  相似文献

Reactions of 3-substituted-2-chloroquinoxalines, terminal alkynes, and 1,3-dicarbonyl compounds in the presence of palladium catalyst afforded a number of 6,8,9-trisubstituted-10H-pyrido[1,2-a]quinoxalin-10-ones in 60–86% reaction yields. This one-pot protocol provided an efficient, practical, and direct method for the synthesis of pyrido[1,2-a]quinoxalines from readily available starting materials in the absence of a copper salt.  相似文献

A series of 3,7-dihydroimidazo[1,2a]pyrazine-3-ones were prepared from 2-amino-3,5-dibromopyrazine. The concise synthesis of coelenterazine (5j), in three steps, 48% overall yield and >99% purity exemplifies the strategy. Further, the synthetic approach facilitated the regiospecific incorporation of carboxyalkyl linkers on the 3,7-dihydroimidazo[1,2a]pyrazine-3-one nucleus that are required for bioconjugation. Peroxymonocarbonate, an electrophilic oxidant, was used to initiate ‘pseudo-flash’ chemiluminescence from this class of molecules.  相似文献

A simple and an efficient eco-friendly methodology has been developed for the synthesis of benzo[4,5]imidazo[1,2-a]-pyrimido[4,5-d]pyrimidin-4(1H)-ones in excellent yields under catalyst-free-conditions using inexpensive and non-toxic polyethylene glycol (PEG-400) medium. This new protocol offers an environmental acceptability, shorter reaction time, room temperature, low cost, high yields, and recyclability of the PEG-400 medium are the important features. Although 1-ethynyl-2-allenylbenzenes readily undergo Myers-Saito or Schmittel cyclization under mild conditions, cyclocarbonylation of these moieties to 1H-cyclopenta[a]inden-2-ones proceeds smoothly using suitable molybdenum carbonyl reagents, with Mo(CO)3(CH3CN)3 being the most efficient. A straightforward, two-step synthesis of fluoro substituted chromeno[2,3-c]pyrazol- and [1]benzothieno[2′,3′:5,6]pyrano[2,3-c]pyrazol-4(1H)-ones, respectively, is presented. Hence, treatment of 1-substituted or 1,3-disubstituted 2-pyrazolin-5-ones with fluoro substituted 2-fluorobenzoyl chlorides or 3-chloro-6-fluoro-1-benzothiophene-2-carbonyl chloride using calcium hydroxide in refluxing 1,4-dioxane gave the corresponding 4-aroylpyrazol-5-ols, which were cyclized into the fused ring systems. 5-Fluorochromeno[2,3-c]pyrazol-4(1H)-one was obtained upon treatment of the 1-(4-methoxybenzyl) protected congener with trifluoroacetic acid. Treatment of 5-fluorochromeno[2,3-c]pyrazol-4(1H)-ones with methylhydrazine afforded novel tetracyclic ring systems such as 2-methyl-7-phenyl-2,7-dihydropyrazolo[4′,3′:5,6]pyrano[4,3,2-cd]indazole. Detailed NMR spectroscopic investigations (1H, 13C, 15N, 19F) with the obtained compounds were undertaken.  相似文献
