Carbon-13 NMR Studies of 3-Aryl-4-(5-aryl-Δ 2-1,2,4-oxadiazolin-3-yl)sydnones and 3-Aryl-4-(5-aryl-1,2,4-oxadiazol-3-yl)sydnones

The ¹³C NMR spectra of twelve 3-aryl-4-(5-aryl-Δ²-l,2,4-oxadiazolin-3-yl)sydnones and twelve 3-aryl-4-(5-aryl-l,2,4-oxadiazol-3-yl)sydnones have been measured and assigned by means of proton-noise decoupling and DEPT-experiments. The coupling constants were determined by means of gated decoupling, and NOE effects were observed by comparison of proton-decoupled and inverse-gated decoupled spectra. Differences shown by the oxadiazoline and the oxadiazole rings and the substitution effects are discussed.     

The Synthesis and Reactions of N-Hydroxy-3-Arylsydnone-4-Carboxamide Oximes

N-Hydroxy-3-arylsydnone-4-carboxamide oximes (7) were prepared from the corresponding 3-arylsydnone-4-carbohydroximic acid chlorides (6) and hydroxylamine in high yield. The chemical reactivity of compound (2) is somewhat different from 3-arylsydnone-4-carboxamide oximes (2) in that the former compounds reacted with both aromatic and aliphatic aldehydes in the presence of acid catalyst to give 3-aryl-4-(5-aryl-1,2,4-oxadiazol-3-yl)sydnones (5) and 3-aryl-4-(5-alkyl-1,2,4-oxadiazol-3-yl)sydnones (3).     

The 1,3-Dipolar Cycloadditions of 3-Arylsydnone-4-Carbonitrile Oxides with Nitriles

3-Arylsydnone-4-carbonitrile oxides (2), which are generated in situ by thermal dehydrochlorination of the corresponding hydroximic acid chlorides (1), undergo 1,3-dipolar cycloadditions with sydnone-4-carbonitriles (6) to give 3-aryl-4-[5-(3-arylsydnonyl)-1,2, 4-oxadiazol-3-yl] sydnones (9). These nitrile oxides (2) also react with alkyl- and arylnitriles to give 1,3-dipolar cycloaddition products.     

The 1,3-Dipolar Cycloadditions of 3-Arylsydnone-4-Carbonitrile Oxides with Alkenes

3-Arylsydnone-4-carbonitrile oxides may undergo 1,3-dipolar cycloadditions with alkenes to produce the corresponding 3-aryl-4-(5-substituted-isoxazolin-3-yl)sydnones. The direct reaction of 3-arylsydone-4-carbohydroximic acid chlorides with alkenes may also give the same products, and with higher yield.     

Synthesis of 2-[2-(5-phenyl-1,2,4-oxadiazol-3-yl)-ethyl]benzimidazole and its x-ray analysis

Cyclization of O-benzoyl-2-(benzimidazol-1-yl)propioamidoxime under different temperature conditions gave 2-[2-(5-phenyl-1,2,4-oxadiazol-3-yl)ethyl]benzimidazole whose structure has been determined by X-ray analysis. __________ Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 7, pp. 1057–1061, July, 2006.     

Synthese de 5-Aryl-3-[(1-dialcoxyphosphonyl)methyl]-1,2,4-triazoles a Partir d'Imidates N-Chloroacyles

A variety of 5-aryl-3-[(1-dialcoxyphosphonyl)methyl]-1,2,4-triazoles 3 has been synthesized efficiently by treatment of trialkylphosphites with 5-aryl-3-chlorométhyl-1,2,4-triazoles 2 . Compound 2 has been prepared by action of N-chloroacylimidates 1 with hydrazines. The structure of triazoles 2 and 3 have been unequivocally confirmed by means of IR, 1 H, 13 C, 31 P NMR spectroscopy and mass spectra.     

Reactions of 3-(4-Aryl-2-thiazolyl)- and 3-(2-Benzothiazolyl)-2-iminocoumarins with N-Nucleophiles

The reaction of 3-(4-aryl-2-thiazolyl)- and 3-(2-benzothiazolyl)-2-iminocoumarins with N-nucleophiles was studied. This reaction gives 2-N-substituted 3-(4-aryl-2-thiazolyl)- and 3-(2-benzothiazolyl)iminocourmarins. N-Nucleophiles such as arylamines, heterocyclic amines, and hydrazine derivatives undergo this reaction.

Synthesis,Crystal Structure and Antitumor Activity of 2-(3-Methyl-5-(methylthio)-4H-1,2,4-triazol-4-yl)isoindoline-1,3-dione

The title compound 2-(3-methyl-5-(methylthio)-4H-1,2,4-triazol-4-yl)isoindoline-1,3-dione(C12H10N4O2S, Mr = 274.30) has been synthesized by a three-step procedure including the cyclization, hydrazinolysis and substitution reactions, and its crystal structure was determined by X-ray single-crystal diffraction. The crystal belongs to the monoclinic system, space group P21/c with a = 12.264(3), b = 14.646(3), c = 14.349(4) , β = 91.69(3)°, μ = 0.255 mm-1, Mr = 274.30, V = 2576.2(10) 3, Z = 8, Dc = 1.414 g/cm3, F(000) = 1136, R = 0.0487 and w R = 0.1329 for 4048 observed reflections with Ⅰ 2σ(Ⅰ). In addition, the preliminary bioassay suggested that the title compound 6 exhibits relatively good antitumor activity against HT-29 and MCF-7.     

The Reaction of 3-Arylsydnone-4-Carbohydroximic Acid Chlorides with Active Methylene Compounds (II)

In the presence of triethylamine, 3-arylsydnone-4-carbohydroximic acid chlorides react not only with active methylene compounds containing keto groups to give 3-aryl-4-(4′,5′-disubstituted-isoxazol-3′-yl)sydnones, but also with compounds containing cyano groups to produce 3-aryl-4-(4′-substituted-5′-aminoisoxazol-3′-yl)sydnones or 3-aryl-4-(4′-cyano-5′-substituted-isoxazol-3′-yl)sydnones.     

Products from condensation of 1-(3(4)-acetylphenyl)-4-carboxy-2-pyrrolidinones with <Emphasis Type="Italic">o</Emphasis>-phenylenediamine and their properties

2-Substituted benzimidazoles were synthesized by the reaction of 1-(3(4)-acetylphenyl)-4-carboxy-2-pyrrolidinones with o-phenylenediamine. 3-(N-取代苯基-2-乙酰胺基)硫基-4-N-取代邻羟苯基亚胺基-5-甲基-1,2,4-三唑类化合物的合成及生物活性研究

以对称二氨基硫脲为原料,与冰醋酸反应生成5-甲基-4-氨基-1,2,4-三唑-3-硫酮(1);在弱酸性条件下,1与取代水杨醛反应生成席夫碱中间体5-甲基-4-(N-取代邻羟苯基)亚胺基-1,2,4-三唑-3-硫酮(2a～2c);最后在碱性条件下分别与N-取代苯基-2-氯乙酰胺发生烷基化反应生成15种未见报道的目标化合物3-(N-取代苯基-2-乙酰胺基)硫基-4-(N-取代邻羟苯基)亚胺基-5-甲基-1,2,4-三唑(3a～3o),其结构经IR,1H NMR,13C NMR确证.初步生物测试表明,质量分数为0.01%时,3a～3o对白色念珠菌的抑菌率均达90%以上,具有很强的抑菌活性;对金黄色葡萄球菌、大肠杆菌的抑菌率达80%以上,具有较强的抑菌活性.     

Synthesis of 3-(3-Alkyl-5-thioxo-1H-4,5-dihydro-1,2,4-triazol-4-yl)aminocarbonylchromones

A series of 3-(3-alkyl-5-thioxo-1H-4,5-dihydro-1,2,4-triazol-4-yl)aminocarbonylchromones has been prepared by oxidation of 3-formylchromone with Jones' reagent followed by reaction with 3-alkyl-4-amino-4,5-dihydro-1,2,4-triazole-5(1H)-thione in the presence of POCl³. The structures of the compounds were confirmed by IR, LC-MS, and ¹H NMR spectra and elemental analyses.     

Synthesis of 1-(2,4,6-trichlorophenyl)-2-(1,2,4-triazol-4-yl)-diazene-1-oxide

By reaction of 4-amino-1,2,4-triazole with 1-nitroso-2,4,6-trichlorobenzene in the presence of excess dibromoisocyanurate the title compound was obtained.N. D. Synthesis of （S）-2-（3-Arylacrylamido）-3-{4-[2-（5-methyl-2-phenyloxazol-4-yl）ethoxy]phenyl}propanoic Acids

The synthesis of （S）-2-（3-arylacrylamido）-3-{4-[2-（5-methyl-2-phenyloxazol-4-yl）etho- xy]phenyl}propanoic acids is described. Their structures were confirmed by ^1H-NMR.     

Synthesis and Antibacterial Activities of N-[(1-Aryl-3-phenyl-pyrazol-4-yl)methylene]-2-(halo-o-hydroxyphenyl)hydrazide Derivatives

A series of novel N-[(1-aryl-3-phenyl-pyrazol-4-yl)methylene]-2-(halo-o-hydroxyphenyl)hydrazide derivatives was synthesized and the antibacterial activity of each of them was evaluated. The supposed reaction mechanism of acquiring compounds 3a—3d is that catalytic activity is enhanced by the electron-donating groups of the first phenyl ring while decreased by electron-withdrawing groups of that ring. The result of preliminary bioassay shows that the lowest minimal inhibitory concentration(MIC) of the title compounds against Escherichia coli is 2 μg/mL. MIC values against Monilia albican and Staphlococcus aureus are as low as 4 μg/mL. They will be a series of potential antibacterial compounds against fungi and gram-negative bacteria.     

The Reaction of 3-Arylsydnone-4-Carbohydroximic Acid Chlorides with Active Methylene Compounds

3-Arylsydnone-4-carbohydroximic acid chlorides react with compounds containing activated methylene groups in the presence of bases to give 3-aryl-4-(4′,5′-disubstituted-iso-xazol-3′-yl)sydnones which have not been reported in the literature. All the products were obtained in good yields, and their structures were confirmed by spectral analysis and elemental analysis.     

Synthesis and structures of 4,6-disubstituted 2-(5-methyl-1,2,4-oxadiazol-3-yl)-1,3,5-triazines

New 1,2,4-oxadiazolyl-1,3,5-triazines were synthesized from amidoximes derived from sym-triazine mononitriles. The structure of one of the resulting compounds was studied in detail by X-ray diffraction. __________ Published in Russian in Izvestiya Akademii Nauk. DIASTEREOSELECTIVE SYNTHESIS AND STEREOCHEMISTRY OF (Z)-1-[3-ARYL-2-(PHENYLSULFANYL)-2-OXIRANYL]-1-ETHANONES

Diastereoselective synthesis of a series of (Z)-1-[3-aryl-2-(phenylsulfanyl)-2-oxiranyl]-1-ethanones was effected from the reaction of (Z)-4-aryl-3-(phenylsulfanyl)-3-buten-2-ones with alkaline hydrogen peroxide in tetrahydrofuran. The stereochemistry of the oxiranes has been deduced from two-dimensional NOESY spectrum.     

Synthesis of 5-Hydroxymethyl-8-methyl-3-(3-aryl-[1,2,4]oxadiazol-5-yl)-2H-pyrano[2,3-c]pyridin-2-ones and Their Esters

New 5-hydroxymethyl-8-methyl-3-(3-aryl-[1,2,4]oxadiazol-5-yl)-2H-pyrano-[2,3-c]pyridin-2-ones and their esters were synthesized. The structure of obtained compounds was determined through a complete ¹H NMR analysis.
