石墨炉原子吸收光谱法测定大鼠血清中硒   总被引:2,自引:0,他引:2  
通过考察不同基体改进剂效果,提出了以硝酸锶和硝酸钯做为混合基体改进剂。建立了用石墨炉原子吸收法测定牛血清和大鼠血清中痕量硒的分析方法。线性范围为0-120ng/mL,硒的检出限为0.095ng/mL,方法用于标准牛血清测定。结果与标准值基本吻合,大鼠血清测定标准加入回收率为102%。  相似文献

Essential differences are demonstrated between bilirubin binding to rat serum proteins and to albumin in human serum. Acidimetric titration of rat serum with and without added bilirubin shows that binding of bilirubin acid in the range of pH from 6.8 to 8.8 takes place with release of less than one hydrogen ion per molecule of bound bilirubin. With human serum, two hydrogen ions are released, indicating binding of bilirubin dianion. The binding equilibrium of N-[4-[(4-aminophenyl)-sulfonyl]phenyl]-acetamide (MADDS) to rat serum albumin is influenced slightly by cobinding of bilirubin whereas MADDS and bilirubin bind competitively to human serum albumin. Finally, the rate of oxidation of bilirubin with hydrogen peroxide and peroxidase is decreased moderately by addition of rat serum albumin and strongly by the human protein, indicating that biliribin in its complex with rat serum albumin is subject to oxidation while the complex with human serum albumin is protected. These differences should be considered when rats are used as a model in experimental studies aiming at prevention of bilirubin encephalopathy in human neonates.  相似文献

A rapid micromethod by flameless atomic absorption spectrometry is presented for the determination of total iron in serum or plasma. A clear distinction is drawn between “total iron in serum” and “serum iron”. The results for serum iron are compared with existing spectrophotometric methods. 以人血清为材料,探讨了帕金森氏病与微量元素的关系。采集了62例患者血清和33例健康人血清,用电感耦合高频等离子体发射光谱仪进行测定,结果发现,患者组血清锌明显低于对照组、铜、锰低于对照组,还发现患者组轻、中、重分型病人血清铅有明显差异,上述结果提示微量元素可能在帕金森氏病的发生、发展和转归中起着重要的作用。  相似文献

Experiments with human serum albumin on the strong cation exchange resin Fractogel EMD SE Hicap (M) were carried out. Even though human serum albumin was used at high purity, two peaks in gradient elution experiments occurred. The obtained data can be explained by considering that human serum albumin binds to Fractogel EMD SE Hicap (M) in two different binding conformations: the protein adsorbs instantaneously in the first conformation and then changes into the second one with a kinetic limitation. The two-peak behavior of human serum albumin was analyzed in detail, especially at various gradient lengths, concentrations and temperatures. Breakthrough curves were performed at four modifier concentrations and three velocities. The characteristic adsorption behavior, found for gradient experiments, was confirmed by the breakthrough curves. The two-peak elution pattern of human serum albumin was also found for other strong cation exchange resins, but not for weak cation exchange resins. It is concluded that the described behavior is peculiar for the interaction of human serum albumin with the strong cation exchange ligand of the resin.  相似文献

为探讨糖尿病肾病 (DN)患者血镁水平 ,以 1 5 9例 2型糖尿病 (T2DM)患者为研究对象 ,用xylidylblue比色法测定了其正常白蛋白尿期、微量白蛋白尿期 (早期DN)、临床DN及晚期DN血镁水平 ,与 2 0例正常对照组相比较 ;同时将 1 1 0例DN患者分为肾功能不全代偿组、失代偿组和肾功能衰竭组 ,将其血镁水平与 1 0 2例慢性肾小球肾炎 (CGN)患者相比较 ,组间比较采用t检验 ;对 79例T2DM正常白蛋白尿期及微量白蛋白尿期患者尿白蛋白排泄率 (UAER)与血镁水平进行了等级相关分析。结果表明 ,T2DM正常白蛋白尿期、早期DN及临床DN血镁水平降低 ,晚期DN血镁水平升高 ,各组与正常对照组相比存在显著性差异 (P <0 0 5或P <0 0 1 ) ;T2DMDN及CGN肾功能不全代偿期、失代偿期及肾功能衰竭期血镁水平渐升高 ,且肾功能处于同一期的DN和CGN相比 ,前者血镁水平均较后者显著降低 (P <0 0 1或P <0 0 5 ) ;T2DM正常白蛋白尿期和微量白蛋白尿期血镁水平与UAER呈负相关 (r=0 5 47,P <0 0 1 )。提示T2DM患者肾功能正常时存在低镁倾向 ,但晚期DN血镁水平升高 ;随着肾功能不全进展 ,DN和CGN患者血镁水平升高 ,但前者血镁水平仍较后者低 ,镁代谢紊乱与DM及其并发症的相互关系有待进一步研究 ;血镁水平测定可否作为DN的早期诊断指标亦  相似文献

An improved method for the determination of aluminum in serum by atomic absorption spectrometry with a graphite furnace is presented. The method is suitable for the analysis of serum samples having normal and elevated aluminum content. The serum sample can be analyzed diluted or undiluted because the selected temperatures program minimizes the matrix interferences. High temperatures are made possible by the use of graphite Zr-coated tubes. The detection limit is 0.7 μg Al/liter. The coefficient of variation is 3.2% for serum concentration of 5 μg Al/liter and 1% for serum concentration of 50 μg Al/liter.  相似文献

糖尿病与体内微量元素的不平衡密切相关。妇女在妊娠期中容易发生糖耐量异常和妊娠期糖尿病,因此研究这些孕妇血清中元素含量的变化,对弄清妊娠期糖尿病的机理和进行防治有重要意义。本工作测定了糖耐量异常和妊娠期糖尿病孕妇血清中硒、铜、锶、镁、锌、钙、磷、铁和铝等九种元素持含量,研究了其与正常孕妇的差异,并观察了元素含量与孕期的相关性。结果表明,糖耐量异常和妊娠期糖尿病孕妇血清中硒的含量与正常孕妇相比明显降低。妊娠期糖尿病孕妇血清中铜比正常孕妇显著升高,而锌有降低趋势,其它元素则未明显变化。此外,观察到正常孕妇和血清异常孕妇血清中硒和钙的含量都与孕期呈明显的负相关性。  相似文献

Blood serum from people suffering from cancer and healthy subjects was subjected to a comparative study by X-ray diffraction. The diffraction patterns were referred to that of pure water. The patterns obtained for blood serum of healthy subjects were similar to that of pure water, while those of cancer patients (two kinds of cancer changes) were different. The former similarity is accounted for by the dominant interaction of water molecules in healthy blood serum with non-polar side chains of amino acids, stabilising the serum structure. In samples from cancer patients the structure of water in blood serum is destroyed because of enhanced interactions of water with polar molecules of conformationally changed proteins. This observation suggests X-ray examination of blood serum from cancer patients and healthy subjects, indicating X-ray diffraction as a diagnostic tool for the occurrence of cancer. The results of this work have shown that the presence of protein affected by cancer has a destructive effect on the structure of water in human serum. The results reported confirm the earlier finding relating cancer changes to optical circular birefringence effects.  相似文献

测定68例恶性淋巴瘤患者血清铜、锌、硒及铜/锌比值水平,采用t检验和非条件Logistic回归因素分析方法对测定结果进行分析。结果表明恶性淋巴瘤患者血清铜、铜/锌比值升高,而血清锌及硒水平降低。血清铜和铜/锌比值升高与肿瘤的负荷状态及病期明显有关,提示血清铜和铜/锌比值的测定对监测恶性淋巴瘤患者的病情变化有一定价值,非条件Logistic多因素分析表明,血清锌、硒可能是恶性淋巴瘤发病的保护因素;而  相似文献

A gas chromatographic method with electron capture detection has been developed for the analysis of nitrazepam in serum. N-Desmethyldiazepam is used as internal standard. Nitrazepam isolated from serum is converted by acid hydrolysis into 2-amino 5 nitrobenzophenone, which is chromatographed. Metabolites of nitrazepam (7-amino and 7-acetamido compounds) are not included in the determination. Recovery experiments showed that the method is quantitative. The limit of detection is 5 ng/ml of nitrazepam in serum. The method has been used for measuring serum concentrations of nitrazepam in bioavailability studies on subjects given a single dose of nitrazepam tablets.  相似文献

Binding of glycyrrhizin to human serum and human serum albumin   总被引:3,自引:0,他引:3  
The binding of glycyrrhizin (GLZ) to human serum and human serum albumin (HSA) was examined by an ultrafiltration technique. Specific and nonspecific bindings were observed in both human serum and HSA. The association constants (K) for the specific bindings were very similar: 1.31 x 10(5) M-1 in human serum and 3.87 x 10(5) M-1 in HSA. The number of binding sites (n) and the linear binding coefficient (phi) in HSA were 1.95 and 3.09 x 10(3) M-1, respectively. When the human serum protein concentration was assumed to be 4.2% (equal to the measured serum albumin concentration), n in human serum was 3.09, which is similar to the n value in HSA, and phi in human serum was 0.71 x 10(3) M-1, which is reasonably close to that for HSA. The binding pattern of GLZ with human serum protein on Sephadex G-200 column chromatography showed that GLZ binds to only the albumin fraction. It was concluded that the GLZ-binding sites in human serum exist mainly on albumin and GLZ binds to specific and nonspecific binding sites at lower and higher concentrations than approximately 2 mM, respectively.  相似文献

A simple and cheap device for colorimetric determination of serum iron and TIBC (total iron‐binding capacity) was devised. The proposed device employs an LED as a light source and a common light dependent resistor (LDR) as a detector. This device functions on the basis of the light intensity received by LDR, connected to a digit multimeter, yielding resistance readings. The serum, standard, and blank solutions were prepared according to the kits instructions and introduced into the colorimeter with a disposable syringe. The iron content of the serum was calculated from the resistance difference of serum and standard solutions. The precision of the method was assessed with two commercially available serum‐based chemistry controls. The values obtained with the proposed device indicated that the serum iron concentrations correlated well with the values obtained with the commercial automated analyzer. The calibration graph was linear for iron concentrations up to 90 μmol/L (500 μg/dL). The proposed fabricated colorimeter is considerable cheaper, requires smaller sample volumes, and is suitable for serum iron assay.  相似文献

A method for the determination of SQ 27,519 (II), the active phosphinic acid-carboxylic acid of the prodrug SQ 28,555 (I), in human serum is presented. Compounds I and II are simultaneously extracted from acidified serum into ethyl acetate, and II is back-extracted into aqueous sodium bicarbonate. Compound I, in ethyl acetate, can be subsequently hydrolyzed and measured as II. The two acidic groups of II are selectively esterified, first by methylation of the carboxylic acid with methanolic hydrochloric acid and then by formation of the hexafluoroisopropyl ester of the phosphinic acid. The resulting product is measured by splitless-injection capillary gas chromatography with nitrogen-phosphorus detection. Linear standard curves were obtained for II with a detection limit of less than 10 ng/ml of serum. The method was successfully applied to the analysis of serum samples obtained from normal individuals after administration of I. In an ascending-dose study involving several human subjects the serum levels of II ranged from less than 10 to 7000 ng/ml of serum.  相似文献

A procedure for the determination of low levels of methadone (6-dimethylamino-4,4-diphenylheptanone-3) in serum has been developed. Methadone is extracted from serum into n-heptane and re-extracted into an acidic aqueous phase. Methadone is oxidized to benzopheone with barium peroxide in sulphuric acid, during which procedure an n-heptane phase is present into which the oxidation product is continuously extracted. The benzophenone formed is determined by means of electron-capture gas chromatography. The recoveries are 100 +/- 3% and 100 +/- 4.5% at the 120 and 16 ng levels, respectively. The minimum amount that can be determined in 1 ml of serum is 4 ng. Interferences from possible metabolies are probably minor. The main cyclic metabolite is only co-determined to a minor extent if the oxidation time is optimized. Comparison of this oxidation method with a combined gas chromatographic-mass spectrometric determination with selected ion monitoring showed identical serum levels.  相似文献

This paper describes an analytical method for the determination of ceruloplasmin (Cp) in human serum. The method uses immunoaffinity chromatography and size-exclusion chromatography (SEC) to “purify” the serum sample prior to analysis of 63Cu and 65Cu by inductively-coupled plasma mass spectrometry (ICPMS). By removing the six most abundant proteins from serum with immunoaffinity chromatography and by using SEC to separate Cu bound by Cp from any free Cu that might be present in the serum sample, we demonstrated that SEC-ICPMS can accurately and reproducibly measure Cp in the ERM DA470 reference serum. Cp identification is based on retention time match of the unknown in the serum sample with the Cp external standard and the presence of 63Cu and 65Cu at a ratio of 2.2±0.1. This method was used to analyze a reference serum certified for Cp, 47 serum samples from four different diseases and a set of normal controls. The reference serum and a serum sample from a patient with myocardial infarction, as well as a Cp standard, were also analyzed by electrospray mass spectrometry to confirm the presence of Cp in the SEC fraction known to contain 63Cu.  相似文献
