The synthesis of carbazola substituted N-acylated polyethylenimines, namely, poly[N-(9-carbazolyl)acetylethylenimine] 20 and poly[N-(2-(9-carbazolyl))propanoylethylenimine] 21 by a grafting reaction onto PEI and isomerization polymerization of the carbazole substituted 2-oxazolines is reported. A complete acylation of amino groups in PEI by the 9-carbazolylacetyl groups was achieved by the p-nitrophenyl active ester method but PEI was only partially N-acylated by the 2-(9-carbazolyl)propanoyl groups under similar reaction conditions. The carbazole substituted 2-oxazolines, namely, 2-(9-carbazolyl)methyl-2-oxazoline 18 and (R,S)-2-[1-(9-carbazolyl)]ethyl-2-oxazoline 19 , were prepared by a base induced cyclization of ß-chloroamides. The ring-opening isomerization polymerization of 18 and 19 in the molten state with a cationic initiator (dimethyl sulfate, methyl triflate, or ethylene glycol ditosylate) gave 20 and 21. Gel permeation chromatography of 20 and 21 obtained with different monomerto-initiator ratios gave evidence of a chain transfer reaction with the monomer. The polymers were characterized by elemental analyses, IR, and 1H-NMR spectroscopy.  相似文献

Three new platinum(II) complexes of (1R,2R)-N1-(2-butyl)-1,2-cyclohexanediamine with malonate derivatives as leaving groups have been synthesized and spectrally characterized. They were tested in vitro against four human cancer cell lines. [(1R,2R)-N1-(2-butyl)-1,2-cyclohexanediamine-N,N′](2-ethylmalonato-O,O′)platinum(II) turned out to be more active (IC50 = 4.65 μM) than oxaliplatin (IC50 = 6.55 μM) against the MCF-7 cell line and is superior to its parent complex, [(1R,2R)-N1-(2-butyl)-1,2-cyclohexanediamine-N,N′](malonato-O,O′)platinum(II). In addition, agarose gel electrophoresis study revealed that the interaction of the complex with pET22b plasmid DNA had a different behavior from that of cisplatin or oxaliplatin.  相似文献

The title compound 7 and two related novel ergolines have been synthesised from (5R,8R,10R)-8-(methyl-thiomethyl)ergoline-6-carbonitrile ( 4 ). The guanidine function of 7 induces a boat conformation of ergoline-ring D, as demonstrated by a careful NMR spectroscopic analysis of 7 and its N-hydroxy congener 6 . Diphenylphosphinodithioic acid has been used to convert the cyanamide function of 4 into the thiourea function at (5R,8R,10R)-8-(methylthiomethyl)ergoline-6-thiocarboxamide ( 5 ).  相似文献

Epoxidation of (?)-(1R,2R,4R)-2-endo-cyano-7-oxabicyclo[2.2.1]hept-5-en-2-exo-yl acetate ((?)-5) followed by saponification afforded (+)-(1R,4R,5R,6R)-5,6-exo-epoxy-7-oxabicyclo[2.2.1]heptan-2-one ((+)-7). Reduction of (+)-7 with diisobutylaluminium hydride (DIBAH) gave (+)-1,3:2,5-dianhydroviburnitol ( = (+)-(1R,2R,3S,4R,6S)-4,7-dioxatricyclo[3.2.1.03,6]octan-2-ol; (+)-3). Hydride reductions of (±)-7 were less exo-face selective than reductions of bicyclo[2.2.1]heptan-2-one and its derivatives with NaBH4, AlH3, and LiAlH4 probably because of smaller steric hindrance to endo-face hydride attack when C(5) and C(6) of the bicyclo-[2.2.1]heptan-2-one are part of an exo oxirane ring.  相似文献

The preparation and radical-initiated polymerization of a carbazole-substituted N-acylated dehydroalanine, namely, 2-(9-carbazolyl)acetylaminopropenoic acid ( 7 ) and its methyl ester ( 6 ) is reported. The monomers 6 and 7 were prepared by dehydrochlorination of N-acylated β-chloroalanine derivatives. The monomer model compounds for the two polymers, namely, 2-(9-carbazolyl)acetylamino-2-methylpropanoic acid ( 11 ) and its methyl ester ( 10 ), were also prepared. The polymers and their monomer model compounds were characterized by elemental analysis, IR and 1H-NMR spectra. The polymers 12 and 13 of different molecular weights could be obtained by changing the monomer-to-initiator ratios used in polymerization experiments.  相似文献

An efficient synthesis of enantiomerically pure (R)- and (S)-2-(aminomethyl)alanine ((R)- and (S)-Ama) 1a and (R)- and (S)-2-(aminomethyl)leucine ((R)- and (S)-Aml) 1b is described (Schemes 1 and 2). Resolution of the racemic amino acids was achieved using L -phenylalanine cyclohexylamide ( 2 ) as chiral auxiliary. The free amino acids 1a, b were converted to the Nα-Boc,Nγ-Z-protected derivatives 11a, b (Scheme 3) ready for incorporation into peptides. Based on the three crystal structures of the diastereoisomeric peptides 8a, 8b , and 9b , the absolute configurations in both series were determined. β-Turn type-I geometries were observed for structures 8b and 9b , whereas 8a crystallized in an extended backbone conformation.  相似文献

The synthesis of (R)-2,10-dihydroxyaporphine ( 3a ), (R)-2,10-dihydroxy-N-n-propylnoraporphine ( 3b ) from thebaine and (R)-2,11-dihydroxyaporphine (7), and 1,2,11-trihydroxyaporphine ( 9 ), from pukateine is reported. The rearrangement of thebaine and northebaine with methanesulfonic acid to 1a and 1b with subsequent N-propylation gave 1b . Tetrazolyation of 1a, 1b and hydrogenolysis of 2a and 2b on Pd/C in acetic acid with subsequent O-demethylation with hydrobromic acid (48%) led to 3a and 3b . R(-)-2,11-Dihydroxyaporphine ( 7 ) was prepared by lithium/ammonia reduction of pukateine. (R)-1,2,11-Trihydroxyaporphine ( 9 ) was synthesized by reaction of pukateine with boron tribromide in dichloromethane.  相似文献

(+)-(1S, 3S, 6S, 8S)-and (?)-(1R, 3R, 6R, 8R)-4, 9-Twistadiene: Synthesis and Absolute Configuration A synthesis and the determination of the absolute configuration of (+)-(1S, 3S, 6S, 8S)- and (?)-(1R, 3R, 6R, 8R)-4, 9-twistadiene ((+)- and (?)- 4 , respectively) is described. Their chiroptical properties are compared with those of saturated twistane ((+)- and (?)- 5 ) as well as with those of the unsaturated and saturated 2, 7-dioxatwistane analogs (+)- and (?)- 9 , and (+)- and (?)- 10 , respectively, which also are compounds of known absolute configurations.  相似文献

Developmental efforts to improve the yield of the chemical synthesis of (3R,3′R)-zeaxanthin resulted in the isolation, partly by chromatography from reaction mixtures, and full spectroscopic characterization by 1H-NMR, UV/VIS, and CD spectrosocpy of eleven (Z/E)-isomers of zeaxanthin: (7Z)-, (9Z), (13Z)-, (15Z)-, (7Z,7′Z)-, (9Z,9′Z)- (7Z,9Z,7Z)-, (7Z,11Z,7′Z)-, (9Z,13Z,9′Z)-, (7Z,9Z,7′Z,9′Z)-, and (7Z,9Z,11Z,7′Z,9′Z)-zeaxanthin. Five of these isomers were obtained by specific synthesis, namely the (7Z)-, (7Z,7′Z)-, (9Z,9′Z)-, (7Z,9Z,7′Z)-, and (7Z,9Z,7′Z-9Z)-isomers.  相似文献

An efficient method was employed for the preparation of 2-(benzylideneamino)-N-[(R)-2-hydroxy-2-methyl-1-phenylpropyl] acetamide utilizing optically pure (R)-5,5-dimethyl-4-phenyloxazolidin-2-one. The reaction of (1S,2S)-2-amino-1-(4-nitrophenyl)-1,3-propanediol with glutaraldehyde has been studied. It has been established on the basis of AM1 and PM3 calculations and 1H NMR spectra recorded in the presence of the shift reagent Eu(fod)3 that (1S,3S,4S,7R,11R)-3-(4-nitrophenyl)-11-aza-2,6-dioxatricyclo[5,3,1,04,11]undecane is formed as the result of the reaction.  相似文献

(3Z,9Z,6S,7R)-6,7-epoxy-3,9-octadecadiene (1) and (3Z,9Z,6R,7S)-6,7-epoxy-3,9-octadecadiene (2) have been stereoselectively synthesized in eight steps from 2-pentyn-1-ol with an overall yield of 8%. The key steps involved the Sharpless asymmetric dihydroxylation of (2E)-oct-2-en-5-yn-1-ol (6). The new synthetic method is suitable for multigram-scale preparation of 1 and 2 and might be used for producing sufficient quantities of the sex pheromone components for management of the pest of tea plantations.  相似文献

The first synthesis of (R)- and (S)-4-hydroxyisophorone by catalytic transfer hydrogenation of ketoisophorone is reported. Ruthenium catalysts containing commercially available chiral amino alcohols afforded 4-hydroxyisophorone in up to 97% selectivity and 97% ee. (R)- or (S)-4-Hydroxyisophorones with >99% ee were isolated by crystallization. The catalyst precursors [RuCl2((S,R)-ADPE)(η6-p-cymene)] ((S,R)-ADPE=(1S,2R)-amino-1,2-diphenylethanol-N) and (RRu)-[RuCl((S,R)-ADPE−1)(η6-p-cymene)] (ADPE−1=amino-1,2-diphenylethanolato-N,O) were isolated for the first time and the X-ray crystal structure of the latter determined.  相似文献

The Diels-Alder adduct of furan and 1-cyanovinyl (1′R)-camphanate was converted into methyl [(tert-butyl)-dimethylsilyl 5-deoxy-2, 3-O-isopropylidene-β-L -ribo-hexofuranosid] uronate ((+)- 4 ). Reduction with diisobutyl-aluminium hydride gave the corresponding aldehyde which was condensed with the ylide derived from triphenyl-(propyl)phosphonium bromide to give (1R, 2S, 3S, 4S)-1-[(tert-butyl)dimethylsilyloxy]tetrahedro-2, 3-(isopropyl-idenedioxy)-4-[(Z)-pent-2′ -enyl]furan ((+)- 7 ). Removal of the silyl protective group gave a mixture of the corresponding furanose that underwent Wittig reaction with the ylide derived from [8-(methoxycarbonyl)-octyl]triphenylphosphonium bromide to yield methyl (11R, 12S, 13S, 9Z, 15Z)-13-hydroxy-11, 12-(isopropylidene-dioxy)octadeca-9, 15-dienoate ((?)- 9 ). Acidic hydrolysis, then saponification afforded (11R, 12S, 13S, 9Z, 15Z)-11, 12, 13-trihydroxyoctadeca-9, 15-dienoic acid ( 1 ).  相似文献

This paper describes the synthesis and preliminary metabolic studies on N,N-dimethyl-N′-2-imidazolyl-N′-benzyl-1,2-ethanediamine (compound 12 ), an imidazole analog of the carcinogenic antihistamine methapyrilene. The 2-aminoimidazole starting material is carried through a five-step reaction sequence which involves introduction of the benzyl and dimethylaminoethyl side chains via sequential acylation of the 2-amino group and reduction of each intermediate amide. Metabolic studies on compound 12 and a d2-analog were performed with rabbit liver microsomes. Chemical ionization mass spectral analysis indicates the presence of metabolites formed by N-demethylation and imidazole C-oxidation. In addition, a seven membered ring metabolite has been identified which apparently is formed by intramolecular cyclization of an intermediate methylene iminium ion.  相似文献

A series of novel dinuclear platinum(II) complexes with a chiral tetradentate ligand, (1R,1′R,2R,2′R)-N1,N1′-(1,2-phenylenebis(methylene))dicyclohexane-1,2-diamine (HL), and mono-carboxylic acid derivatives as ligands have been designed, synthesized, and characterized. In vitro cytotoxicity evaluation of synthesized complexes against human HepG-2, A549, HCT-116, and MCF-7 cancer cell lines has been conducted by MTT assays. All compounds showed antitumor activity to HepG-2 and HCT-116 cell lines. Compound L2 exhibited better cytotoxicity than that of carboplatin against HepG-2 and A549 cell lines and also showed comparable activity against HCT-116 cell line.  相似文献
