Maillard products of beta-lactoglobulin (betaLg) and fructose oligosaccharide (FOS) were obtained in different degrees of modification depending on incubation time and pH. By use of a variety of biochemical and spectroscopic tools, it was demonstrated that the modification at limited degrees does not significantly affect the secondary, tertiary, and quaternary structure of betaLg. The consequence of the modification on the thermodynamics of the protein was studied using differential scanning calorimetry, circular dichroism, and by monitoring the fluorescence intensity of protein samples with different concentrations of guanidine-HCl. The modification leads to lowering of the denaturation temperature by 5 degrees C and a reduction of the free energy of stabilization of about 30%. Ellipsometry and drop tensiometry demonstrated that upon adsorption to air-water interfaces in equilibrium modified betaLg exerts a lower surface pressure than native betaLg (16 versus 22 mN/m). Moreover, the surface elastic modulus increased with increasing surface pressure but reached significantly smaller values in the case of FOS-betaLg. Compared to native betaLg, modification of the protein with oligofructose moieties results in higher surface loads and thicker surface layers. The consequences of these altered surface rheological properties are discussed in view of the functional behavior in technological applications.  相似文献

Protein assembly at the air-water interface (AWI) occurs naturally in many biological processes and provides a method for creating biomaterials. However, the factors that control protein self-assembly at the AWI and the dynamic processes that occur during adsorption are still underexplored. Using fluorescence microscopy, we investigated assembly at the AWI of a model protein, human serum albumin minimally labeled with Texas Red fluorophore. Static and dynamic information was obtained under low subphase concentrations. By varying the solution protein concentration, ionic strength, and redox state, we changed the microstructure of protein assembly at the AWI accordingly. The addition of pluronic surfactant caused phase segregation to occur at the AWI, with fluid surfactant domains and more rigid protein domains revealed by fluorescence recovery after photobleaching experiments. Protein domains were observed to coalesce during this competitive adsorption process.  相似文献

The adsorption of α1-acid glycoprotein into bilirubin/cholesterol mixed monolayers with various component molar ratios is investigated using surface pressure-area (π-A) isotherms and (dπ/dA)-A curves. The results showed that the surface area per molecule increased after the adsorption/insertion of glycoprotein molecules into the monolayers. The compressibility of mixed monolayers increased as a result of hydrogen bonding between bilirubin and glycoprotein molecules, while the interactions between bilirubin and cholesterol are weakened. The adsorption of glycoprotein into a monolayer induced changes in molecular surface area depending on the surface pressure and molar fraction of bilirubin. The transmission electron microscopy of mixed monolayers confirmed the insertion of glycoprotein particles of spherical shape with an average diameter of about 80 nm into the monolayer. Vibrational sum-frequency spectroscopy and surface tensiometry have been used to study the adsorption of m- and p-cyanophenol at the air-water interface. Spectra of the cyano (CN) group under different polarization schemes are utilized to determine its hydrogen bonding environment and orientation. For both isomers, it is found that the cyano group is hydrogen bonded at the interface but that the CN orientation is independent of surface density. The average CN tilt angle (theta(0)), however, is found to differ between the isomers, such that the CN group points down toward the aqueous phase for m-cyanophenol (theta(0) = 96-106 degrees ) but points up toward the vapor phase for the p-cyanophenol (theta(0) = 65-80 degrees ). In addition, this average tilt angle is distributed over a narrow range, sigma(0) < 10 degrees for the meta isomer and sigma(0) < 16 degrees for the para isomer.  相似文献

Recent advances in understanding dynamic surface tensions (DSTs) of surfactant solutions are discussed. For pre-CMC solutions of non-ionic surfactants, theoretical models and experimental evidence for a mixed diffusion-kinetic adsorption mechanism are covered. For micellar solutions of non-ionics, up to approximately 100 x CMC, the DST behaviour can also be accounted for using a mixed mechanism model. Finally, the first reported measurements of the dynamic surface excess Gamma(t), using the overflowing cylinder in conjunction with neutron reflection, are described.  相似文献

Ellipsometry was used to determine the adsorbed layer thickness (d) and the surface excess (adsorbed amount, Gamma) of a nonionic diblock copolymer, E(106)B(16), of poly(ethylene oxide) (E) and poly(butylene oxide) (B) at the air-water interface. The results were obtained (i) by the conventional ellipsometric evaluation procedure using the change of both ellipsometric angles Psi and Delta and (ii) by using the change of Delta only and assuming values of the layer thickness. It was demonstrated that the calculated surface excesses from the different methods were in close agreement, independent of the evaluation procedure, with a plateau adsorption of about 2.5 mg/m(2) (400 A(2)/molecule). Furthermore, the amount of E(106)B(16) adsorbed at the air-water interface was found to be almost identical to that adsorbed from aqueous solution onto a hydrophobic solid surface. In addition, the possibility to use combined measurements with H(2)O or D(2)O as substrates to calculate values of d and Gamma was investigated and discussed. We also briefly discuss within which limits the Gibbs equation can be used to determine the surface excess of polydisperse block copolymers.  相似文献

We report the X-ray and neutron reflectometry measurements of the structural changes caused by chemical denaturation of a surface excess of the bovine milk protein, beta-lactoglobulin. The thickness of the diffuse protein surface layer was used as an order parameter as there was no corresponding increase in the surface excess as a function of guanidinium chloride (G.HCl) concentration. A thermodynamic analysis performed gave the interfacial free energy of unfolding in the absence of a denaturant (DeltaG(0)). This energy, lower than the free energy of unfolding bulk solution, shows that the air-water interface has a destabilizing effect on protein structure up to 50 kJ mol(-1).  相似文献

Interaction of the hydroxyl radical with the liquid water surface was studied using classical molecular dynamics computer simulations. From a series of scattering trajectories, the thermal and mass accommodation coefficients of OH on liquid water at 300 K were determined to be 0.95 and 0.83, respectively. The calculated free energy profile for transfer of OH across the air-water interface at 300 K exhibits a minimum in the interfacial region, with the free energy of adsorbtion (DeltaGa) being about 1 kcal/mol more negative than the hydration free energy (DeltaGs). The propensity of the hydroxyl radical for the air-water interface manifests itself in partitioning of OH radicals between the bulk water and the surface. The enhancement of the surface concentration of OH relative to its concentration in the aqueous phase suggests that important OH chemistry may be occurring in the interfacial layer of water droplets, aqueous aerosol particles, and thin water films adsorbed on solid surfaces. This has profound consequences for modeling heterogeneous atmospheric chemical processes.  相似文献

In this paper we provide experimental evidence for a phase transition between a liquid- and gas-like phase occurring in an adsorption layer of a soluble surfactant at the air-water interface. The equilibrium surface tension sigma(e) versus bulk concentration sigma(e) (c) isotherm of surface chemically pure sodium 2-[4-(4-trifluoromethyl-phenylazo) phenoxy]-ethane sulfonate was measured at a temperature of 295 K up to the solubility limit of the amphiphile. The sigma(e) (c) isotherm could be fitted by Frumkin's equation of state. The lateral interaction energy is just above the limit for which Frumkin's model predicts a phase transition. The corresponding surface pressure pi versus surface area A isotherm possesses striking similarities to first-order phase transitions in the Langmuir monolayer. The fact that the difference in the two-dimensional density is only a factor of 2 indicates that the system is very close to the critical point. The surface phases were further characterized by surface second harmonic generation. The major structural difference between the two surface phases is the amphiphile's molecular orientation. A mean orientation of the amphiphile of about 80 degrees was found in the gas analogous phase, whereas a molecular tilt of 38 degrees has been identified in the liquid-like phase.  相似文献

The interactions of proteins with solid surfaces occur in a variety of situations. Motivated by the many nanoengineering applications of protein-carbon nanotube hybrids, we investigate the conformational transitions of hen egg white lysozyme adsorbed on a carbon nanotube. Using a C(α) structure-based model and replica exchange molecular dynamics, we show how the folding/unfolding equilibrium of the adsorbed protein varies with the strength of its coupling to the surface. The stability of the native state depends on the balance between the favorable entropy and unfavorable enthalpy change on adsorption. In the case of a weakly attractive surface when the former dominates, the protein is stabilized. In this regime, the protein can fold and unfold while maintaining the same binding fraction. With increasing surface attraction, the unfavorable enthalpic effect dominates, the native state is destabilized, and the protein has to extensively unbind before changing states from unfolded to folded. At the highest surface coupling, the entropic penalty of folding vanishes, and a folding intermediate is strongly stabilized. In this intermediate state, the α-domain of lysozyme is disrupted, while the β-sheet remains fully structured. We rationalize the relative stability of the two domains on the basis of the residue contact order.  相似文献

This paper concerns the numerical behavior of the solution to a problem including a linear mixed kinetic-diffusion model for surfactant adsorption at the air-water interface. The existence and uniqueness of a weak solution is recalled. Then, fully discrete approximations are obtained by using a finite element method and the backward Euler scheme. Error estimates are stated from which, under adequate additional regularity conditions, the linear convergence of the algorithm is deduced. Finally, several numerical simulations are presented in order to demonstrate the behavior of the solution for commercially available surfactants.  相似文献

Phosvitin, an egg yolk protein constituted by 50% of phosphorylated serines, presents good emulsifying properties whereas its interfacial properties are not yet clearly elucidated and remain object of discussion. Phosvitin has a high charge density and naturally forms aggregates through phosphocalcic bridges in egg yolk. This high charge density, doubled by this capacity to aggregate, limits the adsorption of the protein at the air-water interface. In this work, we investigated the aggregation impact by calcium ions on the organization of the phosvitin interfacial film using the atomic force microscopy. Phosvitin interfacial films without calcium ions are compared to phosvitin interfacial films formed in the presence of calcium ions in the subphase. We demonstrated that phosvitin is able to anchor at air-water interfaces in spite of its numerous negative charges. In the compression isotherm a transition was observed just before 28 mN/m signifying a possible modification of the interfacial film structure or organization. Calcium ions induce a reorganization towards a greater compaction of the phosvitin interfacial film even at low surface pressure. In conclusion we suggest that, in diluted regime, phosvitin molecules could adsorb by their two hydrophobic extremities exhibiting loops in the aqueous phase, whereas in concentred regime (high interfacial concentration) it would be adsorbed at the interface by only one extremity (brush model).  相似文献

We present the adsorption kinetics and the surface phase behavior of n-hexadecyl dihydrogen phosphate (n-HDP) at the air-water interface by film balance and Brewster angle microscopy (BAM). A phase diagram, which shows a triple point at about 25.8 degrees C, is constructed by measuring the surface pressure (pi)-time (t) adsorption isotherms. Below 25.8 degrees C, each of the pi-t curves shows a plateau at about zero surface pressure indicating the existence of a first-order phase transition. The BAM observation confirms the order of this phase transition by presenting two-surface phases during this plateau. However, the BAM observation also shows clearly another second-order phase transition from an isotropic phase to a mosaic-textured liquid condensed (LC) phase. The initial phase is a gas (G) phase. Considering the peculiarity of the middle phase, we suggest this phase as an intermediate (I) phase. Above the triple point, the pi-t curves predict the existence of two-step first-order phase transitions. Similar to the results at lower temperatures, the BAM images show two-surface phases during these first-order phase transitions together with a second-order phase transition from an isotropic phase to an LC phase. These transitions are classified as a first-order G-LE (liquid expanded) phase transition, which is followed by another first-order LE-I phase transition. The second-order phase transition is an I-LC phase transition. Contrary to these results, at 36 degrees C both the pi-t measurements and the BAM observation present only two first-order phase transitions, which are G-LE at zero surface pressure and LE-LC transition at higher surface pressure. The shape of the domains during the main transitions shows a peculiar change from a circular at 20 degrees C to an elongated at 24 degrees C and finally to a circular shape at 36 degrees C. Such a change in the domain shapes has been explained considering the dehydration effect at higher temperatures as well as the nature of phases.  相似文献

The self-assembly in solution and adsorption at the air-water interface, measured by small-angle neutron scattering, SANS, and neutron reflectivity, NR, of the monorhamnose and dirhamnose rhamnolipids (R1, R2) and their mixtures, are discussed. The production of the deuterium-labeled rhamnolipids (required for the NR studies) from a Pseudomonas aeruginosa culture and their separation into the pure R1 and R2 components is described. At the air-water interface, R1 and R2 exhibit Langmuir-like adsorption isotherms, with saturated area/molecule values of about 60 and 75 ?(2), respectively. In R1/R2 mixtures, there is a strong partitioning of R1 to the surface and R2 competes less favorably because of the steric or packing constraints of the larger R2 dirhamnose headgroup. In dilute solution (<20 mM), R1 and R2 form small globular micelles, L(1), with aggregation numbers of about 50 and 30, respectively. At higher solution concentrations, R1 has a predominantly planar structure, L(α) (unilamellar, ULV, or bilamellar, BLV, vesicles) whereas R2 remains globular, with an aggregation number that increases with increasing surfactant concentration. For R1/R2 mixtures, solutions rich in R2 are predominantly micellar whereas solutions rich in R1 have a more planar structure. At an intermediate composition (60 to 80 mol % R1), there are mixed L(α)/L(1) and L(1)/L(α) regions. However, the higher preferred curvature associated with R2 tends to dominate the mixed R1/R2 microstructure and its associated phase behavior.  相似文献

The coalescence stability of poly(dimethylsiloxane) emulsion droplets in the presence of silica nanoparticles ( approximately 50 nm) of varying contact angles has been investigated. Nanoparticle adsorption isotherms were determined by depletion from solution. The coalescence kinetics (determined under coagulation conditions at high salt concentration) and the physical structure of coalesced droplets were determined from optical microscopy. Fully hydrated silica nanoparticles adsorb with low affinity, reaching a maximum surface coverage that corresponds to a close packed monolayer, based on the effective particle radius and controlled by the salt concentration. Adsorbed layers of hydrophilic nanoparticles introduce a barrier to coalescence of approximately 1 kT, only slightly reduce the coalescence kinetics, and form kinetically unstable networks at high salt concentrations. Chemically hydrophobized silica nanoparticles, over a wide range of contact angles (25 to >90 degrees ), adsorb at the droplet interface with high affinity and to coverages equivalent to close-packed multilayers. Adsorption isotherms are independent of the contact angle, suggesting that hydrophobic attraction overcomes electrostatic repulsion in all cases. The highly structured and rigid adsorbed layers significantly reduce coalescence kinetics: at or above monolayer surface coverage, stable flocculated networks of droplets form and, regardless of their wettability, particles are not detached from the interface during coalescence. At sub-monolayer nanoparticle coverages, limited coalescence is observed and interfacial saturation restricts the droplet size increase. When the nanoparticle interfacial coverage is >0.7 and <1.0, mesophase-like microstructures have been noted, the physical form and stability of which depends on the contact angle. Adsorbed nanoparticle layers at monolayer coverage and composed of a mixture of nanoparticles with different hydrophobisation levels form stable networks of droplets, whereas mixtures of hydrophobized and hydrophilic nanoparticles do not effectively stabilize emulsion droplets.  相似文献

The effect of pH on the adsorption of catalase and lysozyme at the air-water interface has been studied using a combined surface pressure-interfacial shear rheology technique. The results presented show that the rate of development of interfacial phenomena increases as the pH of the subphase approaches the isoelectric point of the protein under investigation. The development of the measured interfacial rheological parameters is due to an increased rate of cross-link formation within the resultant interfacial gel. The formation of the interfacial gels has been modeled using a combination of the Smoluchowski theory for the coagulation of an aerosol or fog and classic rubber elasticity theory. The enhanced rate of cross-link formation at the isoelectric point is a result of an in-surface phase separation brought about by cooperative deionization of the protein molecules near their isoelectric point. Simultaneous measurements of surface pressure and interfacial rheology have enabled us to show that the development of a gel-like interfacial network coincides with observed increases in surface pressure.  相似文献

In situ infrared visible sum frequency generation spectroscopy (SFG) is used to examine the structure of water at the Ag-water interface in NaF and KF electrolyte solutions. Water is observed in environments associated with both the electrode surface and the diffuse double layer. Peaks are observed that are correlated with low-order water, water interacting with electrolyte ions, specifically adsorbed water to the electrode surface, and hydronium. Spectra obtained from a thiol-modified Ag surface enabled discrimination between surface-bound water and that in the double layer. The water organization is dependent on applied potential, with the observed intensities for specifically adsorbed and ion solvating water diminishing near the pzc.  相似文献
